참깨의 리그난 화합물의 항염증 효과 (Effects of Lignan Compound of Sesame on LPS-induced Nitric Oxide Generation in Murine Macrophage RAW 264.7 Cells)

  • 참깨(Sesamum indicum L.)는 1년생 초본식물로서 볶은 참깨나 참기름은 우리나라 식단에서 아주 중요한 조미용 양념 및 식용유지로서 이용되고 있다. 약효로는 동맥경화, 고혈압 및 노화 예방 등으로 알려져 왔으나, 참깨에 함유되어 있는 어떤 성분이, 이들 약효에 관여하고 있는지의 과학적인 규명이 미흡한 실정이다. 본 연구에서는 천연물 유래의 새로운 노화방지제를 개발하기 위해 참깨의 리그난 화합물(sesamin, sesamol, sesaminol, sesaminol diglucosides (SDG), sesaminol triglucosides (STG))의 항염증 활성을 검색하고, 그 중에서 항염증 효과가 가장 높게 나타난 sesamin을 전처치 방법과 동시처치 방법으로 NO 생성억제효과, 세포독성, inducible nitric oxide synthase (iNOS) 및 cyclooxygenases-2 (COX-2) 발현에 미치는 영향을 탐색하고 작용기작을 규명하고자 하였다. 리그난 화합물의 NO 생성억제효과를 탐색한 결과, sesamol, sesaminol, sesamin에서 NO 생성에 미치는 억제율이 높았으나, sesamol, sesaminol은 세포독성이 나타났다. 하지만 sesamin ($IC_{50}: 64{\mu}M$)은 세포독성은 없으면서 농도의존적으로 NO 생성을 억제시켰으며, iNOS 발현에서도 억제 효과를 보여주었다. 결론적으로 sesamin은 피부노화에 원인 중 하나이며 체내 염증반응의 중요 매개인자인 NO의 생성을 억제하는 항염증 물질로서 이와 같은 연구 결과는 향후 임상에서 항염증과 관련된 새로운 항노화 전략으로 개발하기 위해 기초자료로 응응될 수 있음을 시사한다.

분심기음(分心氣飮)이 고혈압 백서와 인간유래 혈관내피세포주(ECV 304)에 미치는 영향에 대한 연구 (An Experimental Study of Effect on ECV 304 Cells, Platelet Rich Plasma and Rats treated with L-NAME by Boonsimgieum extract)

  • Object : This study was designed to research whether the protection and inhibitory effects of cardiovascular diseases in L-NAME induced rat or ECV 304 cell lines through the Cell morphological pattern, Tunel assay, LDH activity, heart rate, blood pressure and immunohistochemistric analysis by Boonsimgieum water extract Methods : Nitric oxide(NO) play an important role in normal and pathophysiological cells including as a messenger molecule, neurotransmitter, microbiocidal agent, or dilator of blood vessels and artheriosclerosis, hypertension, myocardial infarction, respectively. Endothelial cell products can modulate the magnitude of a response to a vasoconstrictor, as evinced by the greater constriction after endothelium removal or NO synthesis blockade. To investigate that Boonsimgieum in the potential contribution of the levels of nitric oxide generated by endothelial nitric oxide synthase (eNOS) and the mechanisms of protection against NG-nitro-L-arginine methyl ester (L-NAME), human ECV 304 cells, which normally do not express eNOS, were expressed by L-NAME. L-NAME stimulated rat or cells were found to be resistant to injury and delayed death following the Boonsimgieum. Inhibition of nitric oxide synthesis abolished the protective effect against L-NAME, thrombin and collagen exposure. Interestingly, such effects have been observed during stimulation with agents such as phenylephrine and KCl on L-NAME mediate rats, were damaged by the NOS inhibitor L-NAME. Result : As the result of this study, In group, the anti-apoptosis and necrosis in the cardiovascular system have a potential capacity for prevented, protected and treating the diseases of cardiovascular system, against the necrosis of rat and ECV 304 cells with Caspase 3 and calpain expression by L-NAME is promoted. Conclusion : these results demonstrate neuroprotective and memory enhancing effects of ZIBU, suggesting its beneficial actions for the treatment of AD.

Administration of red ginseng ameliorates memory decline in aged mice

  • Background: It has been known that ginseng can be applied as a potential nutraceutical for memory impairment; however, experiments with animals of old age are few. Methods: To determine the memory enhancing effect of red ginseng, C57BL/6 mice (21 mo old) were given experimental diet pellets containing 0.12% red ginseng extract (approximately 200 mg/kg/d) for 3 mo. Young and old mice (4 mo and 21 mo old, respectively) were used as the control group. The effect of red ginseng, which ameliorated memory impairment in aged mice, was quantified using Y-maze test, novel objective test, and Morris water maze. Red ginseng ameliorated age-related declines in learning and memory in older mice. In addition, red ginseng's effect on the induction of inducible nitric oxide synthase and proinflammatory cytokines was investigated in the hippocampus of aged mice. Results: Red ginseng treatment suppressed the production of age-processed inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis factor-${\alpha}$, and interleukin-$1{\beta}$ expressions. Moreover, it was observed that red ginseng had an antioxidative effect on aged mice. The suppressed glutathione level in aged mice was restored with red ginseng treatment. The antioxidative-related enzymes Nrf2 and HO-1 were increased with red ginseng treatment. Conclusion: The results revealed that when red ginseng is administered over long periods, age-related decline of learning and memory is ameliorated through anti-inflammatory activity.

태생후 당귀약침 자극이 태생전 소음스트레스로 인한 태아쥐의 NOS 신경세포 발현에 미치는 영향 (Influence of Postnatal Angelicae Gigantis Radix Herb-acupuncture on Prenatal Noise Stress-induced NOS Expression in the Offspring Rats)

  • 목적 : 당귀는 전통적으로 학습과 기억능력 증진 및 각종 질병치료에 우수한 효과가 있는 것으로 알려져있다. 본 연구에서는 이러한 인삼약침이 소음스트레스가 유발된 태아쥐의 해마 및 시상하부에서 nitric oxide synthase에 미치는 영향을 관찰하였다. 방법 : 당귀약침이 소음스트레스에 미치는 영향을 연구하기 위하여 태아기에 소음스트레스를 유발하고 생후 4주후 7일간 당귀약침을 투여한 후 NADPH-d 조직화학법을 시행하였다. 결과 : 1. 소음스트레스는 태아쥐의 해마 및 시상하부에서 NOS 발현이 유의하게 증가하였다. 2. 당귀약침은 해마 CAI에서만 소음스트레스에 의해서 증가된 NOS 발현을 유의하게 억제하였다. 3. 당귀약침은 시상하부 PVN, DMH 및 LHA에서 소음스트레스에 의해서 증가된 NOS 발현을 유의하게 억제하였다. 결론 : 본 연구를 통하여 당귀약침이 소음스트레스가 유발된 태아쥐의 해마 및 시상하부에서 증가된 NOS 발현을 유의하게 억제시켜 태아의 스트레스 자극에 당귀약침치료가 유의한 효과가 있음을 확인하였다.

Alterations in Cerebrovascular Reactivity by Trigeminovascular System Injury in Rats

  • Trigeminovascular system plays an important role for the cerebral memodynamics. The aim of this study was to investigate the alterations in cerebrovascular reactivity by trigeminovascular system injury in rats. Trigeminovascular system of male Sprague-Dawley rats was injured by either denervation of nasocilliary nerve or neonatal capsaicin treatment. Trigeminovascular system was stimulated by controlled hemorrhagic hypotension or somatosensory (whisker) stimulation. Changes in regional cerebral blood flow (rCBF) and pial arterial diameter were continuously measured by laser-Doppler flowmetry and videomicroscopy, respectively. Nitric oxide synthase (NOS) activity in cerebral cortex was determined by measuring the conversion of $L-^3H-arginine\;to\;L-^3H-citrulline$. Cyclic GMP levels in cerebral cortex and pial artery were determined using the cyclic GMP $^{125}I$ scintillation proximity assay system. rCBF autoregulation was impaired or almost abolished by trigeminovascular system injury. rCBF response to whisker stimulation was significantly attenuated by trigeminovascular system injury. NOS activity as well as cyclic GMP level in cerebral cortex and pial artery were significantly reduced in the group of trigeminovascular system injury. These results suggest that trigeminovascular system injury causes prominent alterations in cerebrovascular reactivity, and that NO, which is generated by neuronal NOS in the trigeminovascular system, is implicated in the regulation of rCBF.

Dendrobium moniliforme Stem Extract Inhibits Lipoteichoic Acid-Induced Inflammatory Responses by Upregulation of Heme Oxygenase-1

  • The stems of Dendrobium moniliforme have been used in traditional herbal medicine for the treatment of fever and lack of body fluid in Korea. In this study, we investigated anti-inflammatory effects of the aqueous extract of D. moniliforme stems (DM) in response to lipoteichoic acid (LTA), a major constituent of the cell wall of Gram-positive bacteria. DM inhibited LTA-induced expression of a pro-inflammatory mediator inducible nitric oxide synthase (iNOS) in the murine macrophages. And DM induced expression of heme oxygenase-1 (HO-1) at the transcriptional level. Conversely, the knockdown of HO-1 expression by siRNA markedly reversed the inhibitory effects of DM on LTA-induced iNOS expression. We also demonstrated that nuclear translocation of Nrf2 was increased following treatment with DM. In addition, DM-mediated Nrf2 activation and HO-1 expression were suppressed by PI3K/Akt and p38 inhibitors; treatment with DM also resulted in phosphorylation of Akt and p38. These results suggest that DM inhibits the expression of iNOS in LTA-stimulated macrophages, and that these effects are mediated by the upregulation of HO-1 expression via PI3K/Akt/p38-Nrf2 signaling.

Effects of Acute Exercise on Nitric Oxide Generation from Mouse Macrophages

  • Physical activity is a primary cancer control strategy that has received little attention to date. However, an Increasing number of epidemiological studies have proposed that physical exercise may be beneficial by enhancing anticancer immune system responses. We investigated the effects of acute exercise on changes in nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression. The amounts of NO generated by abdominal macrophages in mice were measured after exercise. Thirty-two mice, which were challenged with thioglycollate broth to activate peritoneal macrophages, were randomly assigned to control, exercise and recovery groups. The mice exercised on a motor-driven treadmill for 3 consecutive days, either moderately (18m/min, 30 min/day, 5% grade) or severely (18-35m/min, 60 min/day, 5% grade). The mice were killed immediately after exercise or after 6 hrs of recovery. Nitric oxide was quantified by the Griess assay. The exercised mice showed higher levels of NO generation than those of the control mice, but the intensity of exercise had no significant effect on NO generation. Mice allowed six hours of recovery after exercise showed higher levels of NO generation than that of animals sacrificed immediately after exercise, but there were no significant differences in NO generation with variations in the intensity of exercise. Increased levels of iNOS were found in the exercised groups, and this was greatest in the groups allowed six hours of recovery compared to those groups sacrificed immediately after exercise. The results of this study suggest that acute exercise may enhance an immune response by inducing macrophage-derived NO generation; these results support the epidemiological findings which support the benefits of exercise in the prevention and control of cancer. Further study is needed to determine the physiological significance of these findings, which could be applied to the use of therapeutic exercises to assist in the prevention and control of cancer.

Nitric Oxide의 합성 억제제인 N$_G$-Nitro-L-Arginine의 항이뇨작용 기전에 관한 연구 (Studies on Mechanism of Antidiuretic Action of N$_G$ Nitro-L-Arginine, Nitric Oxide Synthase Inhibitor, in Dog)

  • This studies were performed for investigation of mechanism on central antidiuretic action of L$_{G}$-Nitro-L-arginine (L-NOARG), nitic oxide systhase inhibitor, in dog. Antidiuretic action of L-NOARG infused into the carotid artery was not affected by renal denervation but inhibited by pretreatment with arginine, NO Precusor. Furthermore, L-NOARG inhibited the diuretic action of dopamine induced by hemodynamic development. Above results suggest that antidiuretic actions of L-NOARG mediated by endogenous substances not associated with renal nerve. Therefore, it is demonstrated that those endogenous substances might be associated with NO which mediate the diuretic action of dopamine.e.

천연 암반 심해수의 항산화 활성 및 면역반응에 대한 연구 (Study of Deep Ground Sea-Like Water on Antioxidant Activity and the Immune Response in RAW264.7 Macrophages.)

  • 해양성 심층수(KDSW)는 해양심층수(DSW)와 유사한 mineral 조성을 가지고 있어 식품 및 의약분야에서 그 유용성에 대한 관심이 증대되고 있다. 본 연구에서는 KDSW 및 탈염한 Danasoo의 항산화력, 면역활성, 함암 및 당뇨에 대한 효과를 연구하였다. KDSW와 Danasoo의 항산화 활성은 DPPH radical scavenging 활성, SOD-like 활성 및 PCL 법에 의하여 항산화 활성을 조사하였다. KDSW와 Danasoo의 항산화 활성은 첨가된 농도에 의존적으로 증가하였으며, 특히 PCL법에 의한 KDSW와 Danasoo의 항산화능은 85.32과 14.02(nmol of ascorbic acid equivalent/ml)로 높은 항산화 활성을 나타내었다. Macrophage RAW 264.7 cell에서 LPS에 의하여 유도된 NO 활성은 Danasoo (20%)를 첨가에 의하여 약 30%정도의 NO 합성이 저해되었으며, 이 농도에서 세포 독성이 없는 것으로 MTT assay에 의하여 확인하였다. NO는 nitric oxide synthase에 의하여 합성되며, tumor성장 및 angiogenesis에서 중요한 역할을 하는 것으로 밝혀져 있으며, Danasoon (20%)는 위암세포와 폐암세포의 iNOS 발현을 현저히 감소시켰다. STZ에 의하여 유도된 당뇨 쥐의 혈당량은 Danasoo (20%) 식이에 의하여 대조구와 유사한 당뇨효과를 나타내었다. 이러한 결과들은 KDSW가 우수한 biological 활성을 가지고 있으며, 또한 천연 기능성 제품생산을 위한 소재로 사용될 수 있는 높은 가능성올 시사하고 있다.

구아바잎 추출물이 난소절제 흰쥐에 미치는 항산화 및 혈관보호 효과 (Effect of oral guava leaf extract administration on antioxidant and vasculoprotective activity in ovariectomized rats)

  • 본 연구에서는 난소절제 흰쥐에 구아바잎 추출물을 8주 동안 경구투여한 결과 난소절제 대조군인 OVX군과 비교시 체중 증가량 및 혈중 유리지방산이 유의적으로 감소된 것을 확인하였다. 또한 간 내 중성지방 농도가 $OVX{\cdot}GL$군과 $OVX{\cdot}GH$군에서 모두 유의적으로 감소하였으며 혈중 항산화 효소인 GPx 농도가 유의적으로 증가하였다. 간 내 항산화 효소 및 eNOS의 mRNA 발현 정도를 측정한 결과 OVX군에 비해 구아바잎 추출물 급여군인 $OVX{\cdot}GL$군과 $OVX{\cdot}GH$군에서 모두 Nrf2 및 CAT의 mRNA 발현 정도가 유의적으로 증가하였으며 eNOS또한 $OVX{\cdot}GH$군에서 유의적으로 증가함을 확인할 수 있었다. 따라서 본 연구의 결과를 종합해 볼 때 구아바잎 추출물 경구투여는 항산화 효소의 활성을 증가시키고 혈관내피세포의 기능을 향상시킴으로써 폐경 후 나타날 수 있는 혈관질환과 산화스트레스로 인한 대사적 장애 개선에 도움이 될 것으로 사료된다.
