Effect of the Rosae laevigatae Fructus extract on the nitric oxide synthase activity and antioxidan action in Rat's corpus cavernosum penis (금앵자(金櫻子) 추출물(抽出物)이 음경해선체(陰莖海線體)의Nitric oxide synthase 활성(活性) 및 항산화효과(抗酸化效果)에 미치는 영향(影響))

  • Rosae laevigatae Fructus extract (RLF) was tested for the effects on the urethral nitric oxide synthase (NOS) activity and Antioxidation in streptozotocin (STZ) induced diabetic rats. RLF was treated firstly into samples, and then STZ induced diabetic rats were set with them. In vitro, the urethral NOS activity was not noted but the type O activity and type conversion ratio of xanthine oxidase and the level of urethral lipid peroxide were decreased in the level of Dose of extract prepared from RLF. In vivo, after the extract was administered to the animal model for fifteen days, the urethral NOS activity increased in STZ induced diabetic rats to the level of normal rats. The content of urethral nitrite and glutathione followed by RLF pre-medicating administration, increased as highly as normal group in compare with the group treated with STZ. The type O activity and type conversion ratio of xanthine oxidase and the level of urethral lipid peroxide followed by RLF pre-medicating administration, decreased as lowly as normal group in compare with the group treated with STZ. In conclusion, the extract of RLF will be able to restore erectile dysfunction of STZ induced diabetic rats.

Anti-inflammatory effect of Distylium racemosum leaf biorenovate extract in LPS-stimulated RAW 264.7 macrophages cells (LPS로 유도된 RAW 264.7 세포에 대한 조록나무 잎 Biorenovation 추출물의 항염증 활성)

  • Biorenovation is a microbial enzyme-based structural modification of component compounds in natural products and synthetic compounds including plant extracts with the potential benefits of improved biological activities compared with its reaction substrates. In this study, we investigated the anti-inflammatory activity of Distylium racemosum leaf extract and D. racemosum leaf biorenovation extract (DLB). As a result, DLB inhibited nitric oxide, prostaglandin E2, and inflammatory cytokines including tumor necrosis factor-α, interleukin-6, interleukin-1β at non-toxic concentrations. In addition, DLB significantly inhibited inducible nitric oxide synthase and cyclooxygenase-2 on LPS-treated RAW 264.7 macrophages. Based on these results, we suggest that the DLB could be used as a potent anti-inflammatory agents. It also suggests that the application of biological evolution has potential usefulness to increase the practical value of natural products.

Diesel Exhaust Particles and Airway Inflammation: Effect of Nitric Oxide Synthase Inhibitors

  • This study was carried out to investigate if nitric oxide synthase (NOS) inhibitors modulate airway inflammation induced by diesel exhaust particles (DEP). N$\^$G/-nitro-L-arginine methyl ester (L-NAME), a potent constitutive NOS (cNOS) inhibitor, and aminoguanidine (AG), a selective inducible NOS (iNOS) inhibitor, were administered to mice in their drinking water for 7 weeks. Airway inflammation was elicited by the repeated intratracheal administration of DEP. The results showed that macrophages, inflammatory eosinophils and neutrophils in bronchoalveolar lavage (BAL) fluids by intratracheal DEP instillation were significantly suppressed in the mice treated with two NOS inhibitors toghther with DEP. The suppression of these cells was more effective in AG treated groups than in L -NAME treated groups. NOS inhibitor treatment also reduced interleukin -5 (IL-5 in the BAL fluids and lung homogenates. Additionally, it was found that eosinophil peroxidase (EPO) activity in the BAL fluids was also decreased by NOS inhibitor treatment. These results suggest that nitric oxide (NO) is produced in airway inflammation by repeated DEP instillation, and that iNOS inhibition as well as cNOS inhibition can play a modulating role in this airway inflammation by DEP.

The Expressional Changes of Nitric Oxide Synthase in the Rat Brain Following Food Restriction

  • This study investigated the changes in the neuronal nitric oxide synthase (nNOS) and nicotinamide adenine dinucleotide phosphate-diaphorase (NADPH-d) activities during food restriction in the rat brain such as cerebral cortex, cerebellum, caudate pautamen and hypothalamus. The rats were placed on a restricted feeding schedule consisting of half the ad libitum quantity for 3 days and 1, 2, 4, 6 and 9 weeks, and a free feeding schedule for 4 weeks. The loss of body weight peaked after 1 week of food restriction and persisted during the entire 9-week period of food restriction. The dramatic weight change in the first week ($12\%$) and the reduction in weight changes thereafter suggest that major adaptation changes occur early and body maintenance occurs subsequently. In the hypothalamus, the optical densities of the NADPH-d and nNOS immunoreactivities were found to be significantly higher in the 1-week and lower in the 9-week food restricted group than in the ad libitum fed control rats. In contrast, in the cerebral cortex, the optical densities of the NADPH-d- and nNOS-positive neurons were not changed significantly during the period of food restriction. This study provides the morphological evidence showing that food restriction has a significant effect on the nitric oxide synthesizing system of the hypothalamus.
