Effect of Ginsenoside Rd on Nitric Oxide System Induced by Lipopolysaccharide Plus $TNF-{\alpha}$ in C6 Rat Glioma Cells

  • Effects of ginsenosides on nitric oxide (NO) production induced by lipopolysaccharide plus TNF-$\alpha$ (LNT) were examined in C6 rat glioma cells. Among several ginsenosides, ginsenoside Rd showed a complete inhibition against LNT-induced NO production. Ginsenoside Rd attenuated LNT-induced increased phosphorylation of ERK. Among several immediate early gene products, only Jun Band Fra-1 protein levels were increased by LNT, and ginsenoside Rd attenuated Jun Band Fra-1 protein levels induced by LNT. Furthermore, LNT increased AP-1 DNA binding activities, which were partially inhibited by ginsenoside Rd. Our results suggest that ginsenoside Rd exerts an inhibitory action against NO production via blocking phosphorylation of ERK, in turn, suppressing immediate early gene products such as Jun Band Fra-1 in C6 glioma cells.

Anti-inflammatory activity of methanol extract isolated from stem bark of Albizia julibrissin

  • Albizia julibrissin (AJ) has been used widely as a traditional medicine. In macrophages nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions including inflammation and carcinogenesis. In this study we have examined the NO inhibition effect of 85% methanol extracts of AJ in mouse macrophage. Lipopolysaccharide (LPS) has been reported to induce production of NO. Extracts of AJ (1, 10, $100{\mu}g/ml$) suppressed nitric oxide production in LPS-stimulated ($100{\mu}g/ml$) mouse (C57BL/6) macrophages and analyzed by ELISA. In addition, it also attenuated the expression of inflammatory products like Interleukin-6 (IL-6), cyclooxygenase-2 (COX-2), and inducible NOS (iNOS) as assessed by immunoblotting with specific antibodies. These results suggest that 85% methanol extracts of AJ would be useful in inflammatory diseases.

Structural Effects of Sulfated-Glycoproteins from Stichopus japonicus on the Nitric Oxide Secretion Ability of RAW 264.7 Cells

  • The effect of various levels of proteins, sulfates, and molecular weight ($M_w$) of a sulfated-glycoprotein ($NF_3$) from a sea cucumber, Stichopus japonicus, on nitric oxide (NO) releasing capacity from RAW 264.7 cells was investigated. The $NF_3$ derivatives had various amounts of proteins (4.8~11.2%) and sulfates (6.8~25.2%) as well as different $M_w$ ($640.3{\times}10^3{\sim}109.2{\times}10^3g/mol$). $NF_3$ was able to stimulate RAW 264.7 cells to release NO with lower protein contents, indicating that the protein moiety was not an important factor to stimulate macrophages. On the other hand, the NO inducing capacity was significantly reduced with decreased levels of sulfates and $M_w$, implying that sulfates and $M_w$ played a pivotal role in activating RAW 264.7 cells. It was not clear why sulfates and a certain range of $M_w$ were essential for stimulating macrophages. It appeared that certain levels of sulfates and $M_w$ of sulfated-glycoproteins were required to bind to the surface receptors on RAW 264.7 cells.

Water Extract of Flowers of Magnolia Denudata Inhibits LPS-induced Nitric Oxide and Pro-inflammatory Cytokines Production in Murine Peritoneal Macrophage by Inhibiting $NF-{\kappa}B$ Activation (LPS로 활성화된 복강 대식세포에서 신이 추출물의 염증성 사이토카인 및 NO 억제 효과)

  • Flowers of Magnolia denudata has been reported to possess a variety of pharmacological activities. In this study, we investigated the anti-inflammatory effects and mechanism of the water extract of Flowers of Magnolia denudata(MD) in lipopolysacchride (LPS)-mediated inflammatory mediators in murine peritoneal macrophages. MD itself does not have any toxic effects in murine peritoneal macrophages. MD inhibits LPS-induced nitric oxide (NO), tumor necrosis factor $(TNF)-{\alpha}$, IL-6 and IL-12 production in murine peritoneal macrophages. Furthermore, we have found that MD inhibited LPS-induced $NF-{\kappa}B$ but not c-Jun N-terminal kinase (JNK), p38 and extracellular signal-ragulated kinase (ERK) activation. These results suggested that MD inhibit LPS-induced production of $TNF-{\alpha}$, IL-6 and IL-12 via suppression of the $NF-{\kappa}B$ activation.

  • The purpose of this Study was to investigate effect of TakliSodokSan(TSS) on the anti-tumor, immunocytes and nitric oxide(NO) production from mice peritoneal macrophages. This Study estimated the proliferation of L1210 cell lines, A431 cell lines, Hep-G2 cell lines, K562 cell lines, 3T3 cell lines, mouse thymocytes and mouse splenocytes and NO production from pcritoneal macrophages in vitro, and estimated the proliferation of L1210 cells, thymocytes and splenocytcs, NO production from peritoneal macrophages and body weight in L1210 cells-transplanted mice in vivo. The results were obtained as follows; 1. TSS inhibited significantly the proliferation of L1210, A431, Hep-G2, K562 cell lines in vitro. 2. TSS accelerated the proliferation of mice thymocytes and splenocytes in vitro. 3. TSS was not increased the nitric oxide production from mice peritoneal macrophages in vitro. 4. TSS inhibited significantly the proliferation of L1210 cells in Ll210 cells∼transplanted mice. 5. TSS accelerated the proliferation of mice thymocytes and splenocytes In L1210 cells-transplanted mice. 6. TSS was increased significantly the nitric oxide production from mice peritoneal macrophages in L1210 cells-transplanted mice. 7. TSS was increased the body weight as comparing with control group in Ll210 cells-transplanted mice.

Effects of Far Infrared on Nitric Oxide level at a rat model of Autoimmune Arthritis Induced (원적외선이 자가면역성 관절염 흰쥐모델의 산화질소정량에 미치는 영향)

  • OBJECTIVE: This study was Far infrared(FIR) irradiating to a rat collagen-induced autoimmune arthritis and observed nitric oxide level of spine. METHODS: Type II collagen(CII) in adjuvant was injected into rats on day 0. The results showed that rats injected with CII plus adjuvant had signs of autoimmune arthritis on day 14. Thereafter, Far infrared(FIR) irradiation was begun on day 15. The rats were irradiated FIR twice per day for 20 min each time. RESULTS: Seven days later(Day 22), the level of NO at FIR irradiated group(FRG; Arth+FIR) was decreased more than FIR-non irradiated group(FNRG; Arth). Twenty-one days later(Day 36), the level of NO at FRG was more closed to the control group's NO level than the FNRG. CONCLUSION: These results indicated that Far infrared irradiating appears to reduce the nitric oxide level at a rat model of autoimmune arthritis induced.

Gold Sodium Thiomalate Inhibits iNOS Gene Expression in RAW 264.7 Macrophage: Differential Regulation by Gold Sodium Thiomalate and Sodium Salicylate (Gold Sodium Thiomalate에 의한 유도성 Nitric Oxide Synthase Gene의 발현억제: Gold 제제와 Sodium Salicylate의 차이점)

  • Gold sodium thiomalate (GST, gold compound) is a widely used anti-arthritic, anti-rheumatic and anti-inflammatory drug that is considered a good alternative to sodium salicylate (NaSA) for individuals who cannot tolerate salicylates. Nitric oxide (NO) synthesized by inducible nitric oxide synthase (iNOS) has been implicated as a mediator of inflammation. Recent evidence suggests that anti-inflammatory effect of NaSA lies in the inhibition of iNOS, but nothing has been reported about the direct effect of iNOS expression by GST. The present study was designed to elucidate sequentially the action mechanisms of GST and NaSA on lipopolysaccharide (LPS) plus interferon-gamma (IFN-$\gamma$) induced iNOS expression in RAW 264.7 macrophages. Both GST and NaSA inhibited NO production and iNOS protein expression in a dose dependent manner. GST inhibited iNOS mRNA expression induced by LPS plus IFN-$\gamma$, whereas NaSA did not. These findings suggest that GST may exert anti-arthritic, anti-rheumatic and anti-inflammatory effect by inhibiting iNOS expression induced by LPS plus IFN-$\gamma$ at transcriptional level, whereas NaSA exert its effect by inhibiting iNOS expression at the translational or posttranslational level.

Modulation of Suppressive Activity of Lipopolysaccharide-Induced Nitric Oxide Production by Glycosidation of Flavonoids

  • Flavonoids have been demonstrated to exhibit a wide range of biological activities including anti-inflammatory and neuroprotective actions. Although a significant amount of flavonoids has been identified to be present as glycosides in medicinal plants, determinations of the biological activities of flavonoids were mainly carried out with aglycones of flavonoids. Therefore, the exact role of the glycosidation of flavonoid aglycones needs to be established. In an attempt to understand the possible role of glycosidation on the modulation of the biological activities of flavonoids, diverse glycosides of kaempferol, quercetin, and aromadendrin were examined in terms of their anti-inflammatory activity determined with the suppression of lipopolysaccharide (LPS)-induced nitric oxide (NO) production in BV2 microglial cells. The results indicated that glycosidation of aglycones attenuated the suppressive activity of aglycones on LPS-induced NO production. Although attenuated, some of glycosides, depending on the position and degree of glycosidation, maintained the inhibitory capability of LPS-induced NO production. These findings suggest that glycosidation of flavonoid aglycones should be considered as an important modulator of the biological activities of flavonoids.

Effect of Dipsaci Radix Water Extract on LPS-induced Inflammatory Response in RAW264.7 Mouse Macrophages (속단(續斷)의 RAW264.7 세포에서 LPS에 의해 유도되는 염증반응에 대한 효과)

  • Objectives : In this study, the effect of Dipsaci Radix(DR, Dipsacus asperoides C.Y. Cheng et T. M. Ai) water extract on LPS-induced inflammatory response in RAW264.7 cells were investigated. Methods : Dried roots of DR was extracted with water for 3 h(DR-W extract). RAW264.7 cells, a mouse macrophage line, were incubated with different concentrations of DR-W extract for 30 min and then stimulated with LPS at indicated times. Cell toxicity was determined by MTT assay. The concentrations of nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) were measured by Griess assay and enzyme immunoassay (EIA), respectively. The expression of inducible nitric oxide synthease (iNOS) and cyclooxyganase (COX)-2 mRNA and protein was determined by RT-PCR and Western blot, respectively. Results : DR-W extract was significantly inhibited LPS-induced productions of NO and PGE2 in RAW264.7 cells. DR-W extract was not suppressed the expressions of iNOS mRNA and protein in LPS-stimulated RAW264.7 cells. Conclusions : This study suggests that DR-W extract can attenuate inflammatory response via inhibition of the NO and PGE2 production in activated macrophages.

Alleviating Effects of Nitric Oxide on Cadmium Toxicity in White Poplar (Populus alba)

  • Cadmium (Cd) is non-essential heavy metal that negatively affects plant metabolism. Nitric oxide (NO) is an increasingly important molecule for plant metabolism that makes signaling. In this study, it was aimed to investigate the alleviating effect of sodium nitroprusside (SNP) application as NO donor in white poplar (Populus alba) under Cd stress conditions. SNP and without SNP treatments increased the Cd accumulation in root tissue. While photosynthetic pigments (Chl a, Chl b, Chl a+b, and carotenoid) content decreased by only Cd application, SNP+Cd application decreased the rate of photosynthetic pigments reduction. When the results of Cd and Cd+SNP applications were evaluated for mineral (Fe, Zn, Mn and Cu) uptake, it was found that the positive effect of SNP was heterogeneously affected. Depending on SNP application, it was found that malondialdehyde (MDA) amount decreased in leaf in 100 µM Cd applications while hydrogen peroxide (H2O2) amount decreased in 100 and 500 µM Cd applications. When antioxidant enzyme activities were examined, it was found that catalase (CAT) and ascorbate peroxidase (APX) enzyme activities increased with 100 µM SNP applications under all Cd applications. As a result, it was found that SNP application under Cd stress generally supports physiological processes positively in white poplar, suggesting that NO molecule plays important alleviating roles in plant metabolism.
