Asarum sieboldii has been used for treatment of fever, pain, common cold, and chronic sinusitis in Korea. In this study, we performed quantification analysis of seven major constituents including aristolochic acid I, aristolochic acid II, ${\alpha}$-asarone, ${\beta}$-asarone, elemicin, methyl eugenol, and safrole in the 70% ethanol extract of Asarum sieboldii and its solvent fractions, n-hexane, ethylacetate, n-butanol, and water ones using a ultra-performance liquid chromatography-electrospray ionization-mass spectrometer(UPLC-ESI-MS) and gas chromatography-mass spectrometer(GC-MS). Regression equations of seven components were acquired with $r^2$ values >0.99. The values of limit of detection(LOD) and quantification(LOQ) were 0.1-3.9 ng/mL and 0.3-11.7 mg/mL, respectively. The amount of the seven compounds in Asarum sieboldii were not detected -143.66 mg/g. The established LC-MS/MS and GC-MS methods will be helpful to improve quality control of Asarum sieboldii.
A noble method for pre-concentration and speciation of ultra trace As (III) and As (V) in urine and whole blood samples based on dispersive liquid-liquid microextraction (DLLME) has been developed. In this method, As (III) was complexed with ammonium pyrrolidine dithiocarbamate at pH = 4 and Then, As (III) was extracted into the ionic liquid (IL). Finally, As (III) was back-extracted from the IL with hydrochloric acid (HCl) and its concentration was determined by flow injection coupled with hydride generation atomic absorption spectrometry (FI-HGAAS). Total amount of arsenic was determined by reducing As (V) to As (III) with potassium iodide (KI) and ascorbic acid in HCl solution and then, As (V) was calculated by the subtracting the total arsenic and As (III) content. Under the optimum conditions, for 5-15 mL of blood and urine samples, the detection limit ($3{\sigma}$) and linear range were achieved 5 ng $L^{-1}$ and 0.02-10 ${\mu}g\;L^{-1}$, respectively. The method was applied successfully to the speciation and determination of As (III) and As (V) in biological samples of multiple sclerosis patients with suitable precision results (RSD < 5%). Validation of the methodology was performed by the standard reference material (CRM).
The dynamics of nitrogen compounds and RNA-based functional genes were characterized in the nitrification-denitrification coupling process. For the removal of residual ammonium, intermittent aeration was introduced in the denitrification reactor. N2O production was not observed in both reactors. In both reactors, the nitrifying genes (achaeal-amoA, bacterial-amoA and hor) and denitrifying genes (narG, nirK, norB and nosZ) had a copy number of 3.92 × 102-7.25 × 105 and 2.85 × 102-3.06 × 104 per ng of DNA, respectively. These results suggest that denitrification and nitrification reactions occur in both the nitrification and denitrification reactors, respectively. Therefore, the coupling process is a promising one for the conversion of ammonium to nitrogen without generating N2O.
In this study, an expanded graphite-titanium oxide composite is developed from expanded graphite (EG) and titanium n-butoxide (TNB). EG is synthesized by chemical intercalation of natural graphite (NG) and rapid expansion at high temperature. TNB is used as the titanium source. The performances of the prepared EG-$TiO_2$ composite are characterized by BET surface area measurements, scan electron microscope (SEM), X-ray diffraction patterns (XRD) and energy dispersive X-ray analysis (EDX). The catalytic activities of the EG-$TiO_2$ composite are investigated by analysis of the degradation of methylene blue (MB) in aqueous solution under irradiation of UV light. Compared with the pristine $TiO_2$ and activity carbon-$TiO_2$ (AC-$TiO_2$) composite, the EG-$TiO_2$ composite shows very high efficiency against MB solution, and the EG could improve the photocatalytic effect of $TiO_2$ in the MB degradation reaction under the irradiation of UV light.
Expanded graphite (EG) is synthesized by chemical intercalation of natural graphite (NG) and rapid expansion at high temperature, with titanium n-butoxide (TNB) used as titanium source by a sol-gel method to prepare EG-TiO$_2$ composite. The performances of the prepared EG-TiO$_2$ composite are characterized by BET surface area measurement, scanning electron microscopy (SEM), X-ray diffraction patterns (XRD) and energy dispersive X-ray analysis (EDX). To compare the photocatalytic activities of the EG-TiO$_2$ composite, three kinds of dye solutions, methylene blue (MB), methylene orange (MO) and rhodamine B (RhB), and two kinds of light source, UV light and visible light (VL), are used. Comparing the results, it can be clearly seen that the degradation of all of the dye solutions under irradiation by UV light is much better than that under irradiation by visible light, and the decomposition of MB solution was better than that of both of MO and RhB solution.
The present study was prospectively designed to assess the clinical effect of leukocyte-depleted blood cardioplegic solution (BCS) on myocardium during cardiac surgery with cardiopulmonary bypass (CPB). 30 adult patients scheduled for elective cardiac surgery were divided into control group (n=15), which infused routine BCS, and leukocyte-depleted (LD) group (n=15), which infused leukocyte-depleted BCS. Total and differential leukocyte counts in BCS, malondialdehyde (MDA) and troponin-T (TnT) concentrations in coronary sinus blood, and cardiac index (CI) were measured at preoperative and postoperative period. The BCS in LD group had less total leukocyte counts with neutropenia than that in control group (P<0.01). MDA (3.70$\pm$0.35 vs 5.90$\pm$0.57 $\mu$mol/L, p<0.05) and TnT (0.42$\pm$0.03 vs 0.60$\pm$0.09 ng/mL, p<0.05) were significantly low in LD group compared with control group, while LD group had higher CI (3.28$\pm$0.16 L/min/$m^2$, p<0.05) than control group (2.69$\pm$0.18 L/min/$m^2$). These results suggest that leukocyte-depleted blood cardioplegic solution has a better myocardial protective effect with less generations of oxygen free radicals and ischemia/reperfusion injury.
The effects of some nutrients and culture conditions on the growth and the production of fumonisin B$_{1}$, (FB$_{1}$) from Fusarium moniliforme NRRL 13569 were investigated in liquid culture. Xylose and soytone yielded the highest mycelial growth as the C- and N-source, respectively. The highest level of FB$_{1}$, was obtained when yeast extract was used as the N-source but no FB$_{1}$, from NaNO$_{3}$. While Fe$^{+++}$ showed inhibition effect on FB$_{1}$, production, Zn$^{++}$ enhanced the FB$_{1}$, production as well as the mycelial growth. FB$_{1}$, was maximally produced when the initial pH value and the specific surface area of the medium was adjusted to 5 and 1.4 cm$^{2}$/ml, respectively. FB$_{1}$ formation reached the maximum value (210, 000 ng/ml) in 30 days and then decreased in Czapek medium substitued with 1% xylose and 0.3% yeast extract, and supplemented with 0.2% NH$_{4}$H$_{2}$PO$_{4}$ where the initial pH value and the specific surface area of the medium are optimally controlled.
Background: : Chemotherapy-induced thrombocytopenia (CIT) is an important cause of morbitity in patients with cancer. Aim: To investigate the effect of the baseline plasma D-dimer level, an important marker for thrombotic activity, on chemotherapy-induced thrombocytopenia in patients with stage III colon cancer. Materials and Methods: A total of 43 (28 men) eligible patients were divided into two groups according to whether they exhibited chemotherapy-induced thrombocytopenia: Group 1 (n=21) and Group 2 (n=22). Comparison was made using demographic, histopathologic, and laboratory variables. Additionally, baseline plasma D-dimer levels underwent receiver operation characteristics curve analysis, and areas under the curve were calculated. Sensitivity, specificity, and positive and negative likelihood rates were then determined. Results: The incidence of chemotherapy-induced thrombocytopenia had a significant correlation with baseline platelet count (r=0.568, P=0.031) and baseline plasma D-dimer levels (r=0.617, P=0.036). When the cut-off point for the latter was set as 498 ng/mL, the area under the curve was 0.89 (95%CI: 0.74-0.93), the sensitivity was 91.4%, the specificity was 89.7%, the positive likelihood rate was 3.64 and the negative likelihood rate was 0.24 for chemotherapy-induced thrombocytopenia diagnosis. Conclusions: The baseline level of plasma D-dimer could help to differentiate high-risk patients for chemotherapy-induced thrombocytopenia.
Various single solvents were tested to find the effective solvent for the extraction of algae oil from wet-form Chlorella minutissima. In the case of single solvents, their extractabilities for algae oil were increased with their polarity because the water in wet algae cell is to form a solvent shell around the lipids. Based on these results, the wet-form algae samples were treated with a polar alcohol solvent and then a nonpolar solvent was added in algae residue. In the algae oil extraction by ethanol/n-hexane, total lipid contents were 40-50% and composition of triglyceride in extracted oil was 46.50%. Considering solvent toxicity of conventional solvent mixture such as chloroform and methanol for algae oil extraction, the ethanol/n-hexane system was identified as the effective one for the oil extraction from wet-form Chlorella minutissima.
We report a novel HPLC method for analyzing the capsaicinoids and nonivamide. The following conditions were used: a total column and column temperature of $3^{\circ}C$, a detection wavelength of 280 nm, an eluent containing acetonitrile : 0.5% formic acid(31:69, v/v). The experimental data was compared with standard nonivamide and capsaicin. The results showed that nonivamide was eluted in 96.7 minutes with a recovery ratio of 96.5% after adding known nonivamide. In addition, the detection limit for nonivamide was 18.6 ng. The capsaicinoids from Korean pepper were extracted (after blooming 60th) from 3 breeds cultivated in Korea, Buchon, Chungyang and Hanbando. and crisis it analyzed sympathizes the nonivamide in the result nonivamide from the Buchon and the Chungyang which are not detected from the Hanbando $0.16{\sim}0.24{\mu}g/g$ it detects with very small amount it contains it confirmed the each capsaicinoid with the HPLC and the LC-MS. These results are expected contribute to the quality control of various product processes, which includes product delivery inspection as well as examining the capsaicinoids present in the product for possible medicine manufacture and various foodstuff, and the nonivamide content, which may be a feed for various manufacturing processes involving chemical synthesis.
