Study on the Effect of Density Ratio of Gas and Liquid in Sloshing Experiment (기체-액체 밀도차에 대한 슬로싱 충격압력의 실험적 고찰)

  • This paper presents the results of sloshing experiments having different fluids in model tanks with various density ratios. The experimental model consisting water and air at ambient, which has been commonly used, is not consistent in density ratio with that of an actual LNG cargo tank. Therefore, an advanced experimental scheme is developed to consider the same density ratio of LNG and NG by using a mixed gas of sulfur hexafluoride ($SF_6$) and nitrogen ($N_2$). For experimental observation, a two-dimensional model tank of 1/40 scale and a three-dimensional model tank of 1/50 scale have been manufactured and tested at various conditions. Two different fillings with various excitation frequencies under regular motions have been considered for the two-dimensional model tank, and three different filling levels under irregular motions have been imposed for the three-dimensional model tank. The density ratio between gas and liquid varies from the ratio of the ambient air and water to that of the actual LNG cargo container, and the different composition of gas is used for this variation. Based on the present experimental results, it is found that the decrease of sloshing pressure is predicted when the density ratio increases.

Simultaneous Determination of Cannabidiol, Cannabinol, and ${\Delta}^9-Tetrahydrocannabinol$ in Human Hair by Gas Chromatography-Mass Spectrometry

  • An analytical method was developed for evaluating the cannabidiol (CBO), cannabinol (CBN), ${\Delta}^9-tetrahydrocannabinol$ $({\Delta}^9-THC)$ level in human hair using gas chromatography-mass spectrometry (GC-MS). Hair samples (50mg) were washed with isopropyl alcohol and cut into small fragments (< 1mm). After adding a deuterated internal standard, the hair samples were incubated in 1.0M NaOH for 10 min at $95^{\circ}C$. The analytes from the resulting hydrolyzed samples were extracted using a mixture of n-hexane-ethyl acetate (75:25, v/v). The extracts were then evaporated, derivatized, and injected into the GC-MS. The recovery ranges of CBD, CBN, and ${\Delta}^9-THC$ at three concentration levels were $37.9-94.5\%$ with good correlation coefficients $(r^2>0.9989)$. The intra-day precision and accuracy ranged from $-9.4\%\;to\;17.7\%$, and the inter-day precision and accuracy ranged from $-15.5\%\;to\;14.5\%$, respectively. The limits of detection (LOD) for CBD, CBN, and ${\Delta}^9-THC$ were 0.005, 0.002, and 0.006 ng/mg, respectively. The applicability of this method of analyzing the hair samples from cannabis abusers was demonstrated.

The study on the analysis of α-naphthylamine in urine (요중 알파나프틸아민 분석에 관한 연구)

  • This study was performed to analyze the purity of technical grade ${\alpha}$-naphthylamine, to establish optimal analytical condition of ${\alpha}$-naphthylamine in urine and to determine the urine sample of workers exposed to ${\alpha}$-naphthylamine. The purity of technical grade ${\alpha}$-naphthylamine were $96.5{\pm}2.38%$, $94.1{\pm}0.97%$, $97.0{\pm}0.02%$ by gas chromatography-mass selective detector. To analyze ${\alpha}$-naphthylamine in urine, high performance liquid chromatography-electrochemical detector and gas chromatography-electron capture detector operating conditions have been optimized by preliminary expriment. In high performance liquid chromatography-electrochemical detector, the mobile phase was consisted of acetonitrile(35%) and water(65%), and the flow rate was maintained at 1.0ml per minute. Optimal detective condition was 9.0V(10nA/V) of electrochemical detector. The recovery of sep-pak treatment method was highly estimated as pretreatment of ${\alpha}$-naphthylamine in urine. The free amine was isolated by gas chromatography-electron capture detector after basic hydrosis, sep-pak treatment, toluene elution and HFBA(heptafluoro-butyric anhydride) derivatization of urine. The recovery of ${\alpha}$-naphthylamine in urine was $98.73{\pm}3.29%$ by gas chromatography-electron capture detector. The sensitivity was more higher than that of high performance liquid chromatography-electrochemical detector. Urinary ${\alpha}$-naphthylamine was detected in only one worker among nine workers. The level of ${\alpha}$-naphthylamine in urine was 6.42 ng/ml.

Impact of High Fat Diet-induced Obesity on the Plasma Levels of Monoamine Neurotransmitters in C57BL/6 Mice

  • Obesity is one of the most serious health problems in developed countries. It negatively affects diverse aspects of human wellbeing. Of these, a relationship between obesity and depression is widely recognized but biomarkers for assessment of obesity-associated mood changes in animal obesity models are rarely known. Here we explored the link between obesity and the plasma levels of monoamine neurotransmitters involved in mood control using a sensitive UPLC/MSMS technique in high fat diet (HFD)-induced obesity model in male C57BL/6 mice to explore the potential utility of plasma tests for obesity-associated mood change. HFD (60% of total calories, 8 weeks) induced significantly higher weight gains in body (+37.8%) and fat tissue (+306%) in male C57BL/6 mice. Bioanalysis of serotonin, dopamine and norepinephrine in plasma at 8 weeks of HFD revealed that serotonin decreased significantly in the obese mice when compared to normal diet-fed mice ($2.7{\pm}0.6$ vs $4.3{\pm}2.0ng/ml$, N=8). Notably, a negative correlation was found between the levels of serotonin and body weight gains. Furthermore, principal component analysis (PCA) with the individual levels of neurotransmitters revealed that plasma levels of dopamine and serotonin could apparently differentiate the obese mice from lean ones. Our study demonstrated that blood plasma levels of neurotransmitters can be employed to evaluate the mood changes associated with obesity and more importantly, provided an important clue for understanding of the relationship between obesity and mood disorders.

Determination of urinary cortisol : creatinine ratios by sequential thin layer chromatography and ELISA in dogs (개에서 thin layer chromatography 박층크로마토그라피 및 ELISA를 이용한 요중의 cortisol : creatinine비 측정)

  • This study was conducted to evaluate the ELISA kit for measuring the level of cortisol in the urine. The CV of within-run variation and day to day variation were 0.4~2.8 and 1.8~5.7, respectively. The minimum limitation of measurement was 1ng/ml. The cross reaction was high ($CR_{50}(%)=11.4{\sim}43.2$) in prednisolone, 11-deoxycortisol, 21-deoxycortisol and predinosone. There was low and no cross reaction in other steroid. To develop the ELISA kit we measured the cortisol level in diluted urine with PBS (procedure I), extracted urine with methylene chloride (procedure II) and extracted methylene chloride-extracted urine from thin-layer chromatography (procedure III). The CV value of procedure I, II, III was 9.4~28.3%, 7.2~8.9% and 2.5~5.7%, respectively. There was significant difference between procedure I with II, and pro-cedure I with III(p < 0.01), but no difference between procedure II with III significantly(p < 0.01). The mean UCCR of urine collected through am 8 to 10 was $9.5{\pm}7.6$(0.14~28.0) in 12-month-old dog(n = 47). In this study we can measure the cortisol level in extracted urine with methylene chloride and sequential thin-layer chromatography accurately using ELISA kit.

An Efficient VLSI Architecture for the Discrete Wavelet Transform (이산 웨이브렛 변환을 위한 효율적인 VLSI 구조)

  • This paper proposes efficient VLSI architecture for computation of the 1-D discrete wavelet transform (DWT). The proposed VLSI architecture computes the wavelet lowpass and highpass output sequences using the product term anhm, $n,m{\ge}0$, where an and hm denote the imput sequence and the wavelet lowpass filter coefficient, respectively. Whereas the conventional architectures compute the lowpass and highpass output sequences using the product terms anhm and angm, respectively, where gm denotes the wavelet highpass filter coefficient. The proposed architecture is applied to computation of the Daubechies 4-tap wavelet transform using the relationships between the Daubechies wavelet filter coefficients. Performance comparison of various architectures for computation of the 1-D DWT are presented. Note that the proposed architecture does not require extra processing units whereas the conventional architectures need them. Also it is modeled in very high speed integrated circuit hardware description language (VHDL) and simulated to show its functional validity.

Effect of Mitogens, Supplement of β-mercaptoethanol and Fetal Bovine Serum Supplementation in Whole Blood Culture Medium for Bovine Chromosome Analysis

  • We investigated the effects of the mitogen supplements of 3 types, pokeweed mitogen (PWM), phytohemagglutinin (PHA) and concanavalin A (ConA), to a whole blood culture system on the number of metaphase spreads obtained in perinatal bovine chromosome analysis. In addition, the supplementation of ${\beta}$-mercaptoethanol (${\beta}$-ME) and FBS was examined in such system. Significant differences (p<0.05) were seen in the number of metaphase spreads with PHA stimulation compared to both PWM and ConA stimulation. When examined the effects of ${\beta}$-ME supplementation, the number of metaphase spreads was significantly (p<0.05) increased at $30{\mu}M$ ${\beta}$-ME compared to control. When evaluated FBS supplementation during PWM stimulation, no significant effect of the supplementation was found. Finally, the effects of the cortisol concentration (10-20, 20-30 and >30 ng/ml) of the blood samples were examined. There was no significant effect of cortisol concentration (p>0.05) among these 3 cortisol concentration groups. The mean percentages of normal metaphase plates (2n=60) from each calf 1) with ${\beta}$-ME, 2) without ${\beta}$-ME and 3) with FBS stimulated with PWM were not significantly different (p>0.05). In conclusion, these findings may be useful in cytogenetic screening programs for not only perinatal calves but also for mature cattle.

Tetramine Analysis using Liquid Chromatography-Tandem Mass Spectrometry and Ion Chromatography (LC-MS/MS와 Ion Chromatography를 이용한 테트라민 분석)

  • As a first step toward identifying a new method for testing sea snail tissue for toxins, and thus prevent food poisoning due to the ingestion of contaminated snails, we measured the tetramine [$(CH_3)_4N^+$] contents of sea snails from the Korean coast using both liquid chromatography-tandem mass spectrometry (LC-MS/MS) and ion chromatography. For tetramine tested, good linearity ($r^2$ = 0.9996) was observed between the amounts in the injected samples and the peak areas of standard toxins, which ranged from 0.1 to 100 ng. The recovery (%) of tetramine from spiked tissue and mid-gut gland samples ranged from 84.0 to 95.3%. The quantitative results for tetramine using this method were in good agreement with the theoretical values. LC-MS/MS has both high sensitivity and selectivity, which makes it possible to measure trace quantities of tetramine in samples.

Localized Surface Plasmon Resonance (LSPR) Biosensors on Metal Nanoparticles with the Design of Bioreceptors

  • Label-free biomolecular assay based localized surface plasmon resonance (LSPR) of noble metal nanoparticles enables simple and rapid detection with the use of simple equipment. Nanosized metal nanoparticles exhibit a strong absorption band when the incident light frequency is resonant with the collective oscillation of the electrons, which is known as the LSPR. Here we demonstrate localized surface plasmon resonance (LSPR) substrates such as plasmonic Au nanodisks fabricated by a nanoimprinting process and gold nanorod-immobilized surfaces and their applications to highly sensitive and/or label-free biosensing. To increase detection sensitivity various bioreceptors weree designed. A single chain variable fragment (scFv) was used as a receptor to bind C-reactive protein (CRP). The results of this effort showed that CRP in human serum could be quantitatively detected lower than 1 ng/ml. Aptamers, which were immobilized on gold nanorods, were used to detect mycotoxins. The specific binding of ochratoxin A (OTA) to the aptamer was monitored by the longitudinal wavelength shift of LSPR peak in the UV-Vis spectra resulting from the changes of local refractive index near the GNR surface induced by accumulation of OTA and G-quadruplex structure formation of the aptamer. According to our results, OTA could be quantitatively detected lower than 1 nM level. Additionally, aptamer-functionalized GNR substrate was quite robust and can be regenerated many times by rinsing at 70 OC to remove bound target. During seven times of washing steps, the developed OTA sensing system could be reusable. Moreover, the proposed biosensor exhibited selectivity over other mycotoxins with an excellent recovery for detection in grinded corn samples, suggesting that the proposed LSPR based aptasensor plays an important role in label-free detection of mycotoxins.

Anti-inflammatory Activity of Solvent Fractions from Ginseng Berry Extract in LPS-Induced RAW264.7 Cells (인삼열매추출물의 용매분획물이 LPS로 유도된 RAW264.7 세포에 대한 항염활성)

  • Anti-inflammatory activity of the extracts of ginseng berry (GBE) was investigated through the evaluation of its inhibitory effect on the production of inflammatory meditator, nitric oxide(NO), tumor necrocis factor-alpha (TNF-${\alpha}$), interleukin-6 (IL-6) in LPS-induced RAW264.7 macrophage cells. GBE was fractionated using n-hexane, chloroform, ethylacetate, buthanol and $H_2O$, sequentially. RAW264.7 cells were induced $100ng/m{\ell}$ of lipopolysaccharide (LPS) and treated with 0, 1.6, 8, 40 and $200{\mu}g/m{\ell}$ of GBE fractions. LPS-induced NO production on all of GBE fractions was inhibited with increasing added concentration of GBE fractions. Chloroform fraction of GBE was the most effective in inhibiting LPS-induced TNF-${\alpha}$ production. Hexane, chloroform and $H_2O$ fractions of GBE exhibit strong inhibition LPS-induced IL-6 production. Especially, $H_2O$ fractions of GBE was the most effective in inhibiting LPD-induced IL-6 production without significant cytotoxicity in RAW264.7 cells, and reduced the activation of mitogen-activated protein kinases (MAPK) and IkB phosphorylation. These results indicate that $H_2O$ fractions of GBE exhibits strong anti-inflammatory effects by inhibition of NF-kB by inhibition of p-38 on MAPK and IkB phosphorylation.
