글루타메이트로 유발한 세포독성에 대한 삼채추출물의 뇌세포 보호 효과 (Effects of Allium hookeri Extracts on Glutamate-induced Neurotoxicity in HT22 Cells)

  • Glutamate-induced oxidative stress results in neuro-degenerative disorders in many central nervous system (CNS) such as Alzheimer's disease, ischemia, Huntington's disease, and Parkinson's disease. Our study was performed to investigate neuroprotective effects of Allium hookeri extracts (leaf, root, and whole) on glutamate-induced HT22 cells. In this study, ethanol extract of A. hookeri showed the outstanding neuroprotective effect in HT22 cells. In addition, we found that ethanol extract of A. hookeri root increased heme oxygenase (HO)-1 in HT22 cells. Moreover, ethanol extract of A. hookeri root also upregulated nuclear accumulation of nuclear factor E2-related factor 2 (Nrf2) in HT22 cells. These results demonstrate that ethanol extract of A. hookeri root contributes neuroprotective effects against glutamate-induced oxidative stress in HT22 cells, via Nrf2-mediated HO-1 expression. Our study suggests that ethanol extract of A. hookeri root could be the potential agent for the treatment of many neuro-degenerative diseases.

백서의 국부 뇌경색에 대한 당귀의 신경보호 효과 (The Neuroprotective Effects of Angelicae gigantis Radix on Focal Cerebral Ischemia in the Rat)

  • Current therapy for acute ischemic stroke is highly focused on neuroprotective agents, and many herbal medicines have been challenged for experimental models. The aim of this study is to investigate whether Angelicae gigantis Radix can protect nerve cells against ischemic neural damage of middle cerebral artery occlusion (MCAO) in rats' brains. Rats were treated with Angelicae gigantis Radix immediately after 2 hours of MCAO for 7 days. On the 7th day, the brains of the rats were sliced through the hippocampus and dyedby c-Fos immunohistochemistry stain and cresyl violet stain for microscopic examination. The number of viable neurons and c-Fos immunoreactive cells in CA1 regions was counted. MCAO caused significant decrease in density of neurons and c-Fos immunoreactive cells compared to those of sham-operated rats. Administration of Angelicae gigantis Radix significantly elevated MCAO-induced decrease in density of neurons and c-Fos immunoreactive cells. These results suggest that the neuroprotective effect of Angelicae gigantis Radix against focal cerebral ischemia is related to c-Fos gene expression. Thus, these findings indicate that Angelicae gigantis Radix can be used for treatment and prevention of cerebral ischemia.

  • Objectives: This study was designed to assess neuroprotective effects of herb medicine against Alzhheimer's disease related brain damage in organotypic hippocampal slice culture. Methods: We induced dementia related brain damage in organotypic hippocampal slices by $\beta$-amyloid. Those slices were treated with herb medicines - Hwangryeonhaedoktang, Sopungsoongiwon. Using by PI staining, the extents of cell death were assessed. After that, we selected the best effective one among those herb medicines and the major components of that medicine were studied to reveal neuroprotective effects and related proteins by using PI stating. Results: In PI staining, Sopungsoongiwon is the best effective herb medicine between Hwangryeonhaedoktang and Sopungsoongiwon. Notopterygii Rhizoma, Corni Fructus, Areca Catechu, Aurantii Fructus Immaturus, Plantaginis Semen is the best effective one among the components of Sopungsoongiwon. Conclusions: We suggested that purgative effect would be the best effetive medicine on dementia related brain damage between clearing heat and toxic materials.

Neuroprotective Effects of N-Acetyldopamine Dimers from Cicadidae Periostracum

  • The chemical investigation of the 90% EtOH extract from Cicadidae Periostracum led to the isolation and identification of seven known N-acetyldopamine dimers (1-7). These compounds were identified by comparing mass spectrometry data and NMR spectroscopic data with those previously reported. In this study, complete interpretation of 1D and 2D NMR data of 1 and 2 were reported for the first time. In addition, compounds 3 and 4 were isolated from this material for the first time. All isolates were obtained as racemic mixtures, as confirmed by chiral HPLC. Furthermore, we evaluated the neuroprotective activities of compounds 1-7 and found that compounds 1, 5, and 6 significantly attenuated rotenone-induced death of SH-SY5Y neuroblastoma cells at a concentration of 100 μM. Parallel to this result, compounds 3 and 6 displayed antioxidant effects in the cytoplasm, as determined by CM-H2DCFDA fluorescence intensity, while compounds 1 and 5 showed antioxidant effects in the mitochondria, as assessed by MitoSox fluorescence intensity. Overall, these results suggest that some of these compounds protect neuroblastoma cells by ameliorating the release of reactive oxygen species. Further studies are warranted to elucidate the underlying mechanisms by which these compounds exhibit antioxidant and neuroprotective actions.

호장근의 일시적 국소뇌허혈 흰쥐 모델에 대한 신경보호효과 (Neuroprotective Effect of the Roots of Polygonum Cuspidatum on Transient Focal Cerebral Ischemia in Rats)

  • Objectives : The purpose of the present study was to development of neuroprotective antioxidant agents. For the purpose, we investigated the neuroprotective effect of anti oxidant herb, the root of Polygonum cuspidatum on transient focal cerebral ischemia in rats. Methods : The roots of Polygonum cuspidatum were extracted by 85% MeOH (PCE). Radical scavenging effects were investigated using DPPH assay and TBARs (Thiobarbituric acid reaction substance) assay in brian homogenates. Neuroprotective effect was investigated using transient focal cerebral ischemia rat model (2 h of ischemia, 22 h of reperfusion) by behavioral test and measurement of brain damage using 2, 3, 5-triphenyltetrazolium chloride staining. Results : PCE showed potent and dose dependent radical scavenging effects in DPPH and TBARs assay. Oral administration of PCE reduced brain infarct volume by 29.7% and improved the sensory motor functional deficit by 29% compared with vehicle treated group. Conclusions : PCE showed radical scavenging effects and neuroprotective effect on stroke rat model. Therefore, Polygonum cuspidatum could be a candidate for the development of neuroprotective-antioxidant agents.

증숙 과정을 통한 소청룡탕의 뇌신경세포 보호 활성 변화에 관한 연구 (Neuroprotective Activity of So Cheong Ryong Tang with Steaming Process)

  • So Cheong Ryong Tang (SCRT) has been used as a traditional herbal medicine for the treatment of the bronchial asthma. In this study, the effect of steaming process on changes in 11 major compounds (homogentisic acid, ephedrine HCl, paeoniflorin, cinnamic acid, cinnam aldehyde, glycyrrhizin, 6-gingerol, schizandrin, methyleugenol, gomisin A and gomisin N) content and neuroprotective activity of SCRT were evaluated. Major compound content was slightly different with steaming produce. The contents of paeoniflorin, cinnamic acid, cinnam aldehyde, 6-gingerol and methyleugenol were decreased and homogentisic acid, ephedrine HCl, glycyrrhizin, schizandrin and gomisin A were increased by steaming process. The neuroprotective activity of steaming SCRT was determined in mouse hippocampal HT22 cells by MTT assay. As a result, neuroprotective activity of a steaming SCRT was higher than that of a non steaming SCRT. This study demonstrated that neuroprotective activity of SCRT can be improved through steaming process.

증숙에 따른 소시호탕의 활성 변화 (Enhancement of Bioactivity of Sosihotang with Steaming Process)

  • Sosihotang is a herbal medicine traditionally used to treat chronic liver disease. This study was performed to investigate the effect of steaming Sosihotang at a $121^{\circ}C$ on its major compounds and neuroprotective activity. We evaluated quantitative changes of 6 major compounds in Soshihotang, saikosaponin A, homogentisic acid, baicalin, ginsenoside $Rg_3$, glycyrrhizin and 6-gingerol using HPLC-DAD. After steaming produce, saikosaponin A, homogentisic acid, baicalin, glycyrrhizin and 6-gingerol in Sosihotang were decreased and ginsenoside $Rg_3$ was increased. The neuroprotective activity of steamed Sosihotang was determined by MTT assay, comparing to Sosihotang. The steamed sosihotang showed a higher neuroprotective activity than Sosihotang. In conclusion, steam produce could improve neuroprotective activity of Soshihotang.

뇌해마의 장기양 조직배양을 이용한 한약물의 뇌신경세포손상 보호효능 연구 (Neuroprotective Effects of Medicinal Herbs in Organotypic Hippocampal Slice Cultures)

  • Objectives : For the screening of neuroprotective effects of medicinal herbs, the complex system of animal models suffer some disadvantages in controlling critical parameters such as blood pressure and body temperature. Additionally, application of drugs to the appropriate brain area sometimes is difficult, due to poor permeability though the blood brain barrier, and so potential protective effects might be masked. Methods : Organotypic hippocampal slice culture (OHSC) method has the advantages of being relatively easy to prepare and of maintaining the general structure, including tissue integrity and the connections between cells. Drugs can easily be applied and neuronal damage can easily be quantified by using tissues and culture media. This study demonstrates neuroprotective effects of Puerariae radix (葛根, PR), Salviae miltiorrhizae radix (丹蔘, SR), Rhei rhizoma (大黃, RR), and Bupleuri radix (柴胡, BR). These were screenedand compared to MK-801, antagonist of NMDA receptors, by using OHSC of 1 week-old Sprague-Dawley rats. Oxygen/glucose deprivation (OGD) were conducted in an anaerobic chamber $(85%\;N_2,\;10%\;CO_2\;and\;5%\;H_2)$ in a deoxygenated glucose-free medium for 60 minutes. Water extracts of each herbs were treated to culture media with $5\;{\mu}g/ml$ for 48 hours. Results : Neuronal cell death in the cultures was monitored by densitometric measurements of the cellular uptake of propidium iodide (PI). PI fluorescence images were obtained at 48 hours after the OGD and medicinal herb treatment. Also TUNEL-positive cells in the CAI and DG regions and LDH concentrations in culture media were measured at 48 hours after the OGD. According to measured data, MK-801, PR, SR and BR demonstrated significant neuroprotective effect against excessive neuronal cell death and apoptosis induced by the OGD insult. Especially, PR revealed similar neuroprotective effect to MK-801 and RR demonstrated weak neuroprotective effect. Conclusions : These results suggest that OHSC can be a suitable method for screening of neuroprotective effects of medicinal herbs. 가감보양환오탕(加減補陽還五湯)의 N2a 뇌신경세포에 대한 보호효과 (Neuroprotective effects of modified Bo-Yang-Hwan-Oh-Tang in N2a neuroblastoma cells)

  • Objectives : To evaluate the neuroprotective effects of modified Bo-Yang-Hwan-O-Tang (BHT), we investigated the neuronal death protection effects to oxidative damages in N2a neuroblastoma cells. Methods : To study the cytotoxic effect of BHT on N2a neuronal cells, the cell viability was determined by MTT assay. To investigate the neuronal death protection of BHT, N2a neuronal cells were induced oxidative damages by H2O2, and assayed the cell viability and DNA fragmentation. We also investigated DPPH free radical scavenging effects of BHT by tube test. Results : In MTT assay, $500{\mu}g/ml$ of BHT was not showed cytotoxic effect on N2a neuronal cells. BHT protected N2a neuronal cells from H2O2-induced cell death in a dose-dependent manner. BHT also protected N2a neuronal cells from H2O2-induced DNA fragmentation. BHT scavenged DPPH free radicals in a dose-dependent manner. Conclusion : These data suggest that BHT may have strong anti-oxidant effects through the free radical scavenging and neuroprotective effects in neuronal cells.

양격산화탕(凉膈散火湯)이 고지혈증 흰쥐의 중대뇌동맥 폐쇄에 의한 뇌경색에 미치는 영향 (Neuroprotective Effect Yanggyuksanhwa-tang on Cerebral Infarction Induced by MCAO in Hyperlipidemic Rats)

  • Objectives : Yanggyuksanhwa-tang is a prescription used for cerebral infarction clinically it is known that this formula reduces body fat, serum cholesterol and triglyceride in hyperglycemia and obesity patients. According to previous research data, controlling these types of lipid is considered to decrease the risk of cerebral infarction. Based on this fact, we investigated the relationship between hyperlipidemia and cerebral infarction, and the effect of Yanggyuksanhwa-tang on hyperlipidemic cerebral infarction. Methods : We induced cerebral infarction by middle cerebral artery occlusion (MCAO) in high-fat diet rats, and the rats were administered Yanggyuksanhwa-tang. Results : Infarct area and serum lipid were measured, and the level of elements such as c-Fos, Bax and caspase-3 in penumbra of infarct were expressed by immunohistochemical staining. Conclusions : Yanggyuksanhwa-tang showed neuroprotective effect through preventing neuronal cell apoptosis as well as reducing serum lipid level in hyperlipidemic condition.
