NMR Structure of Syndecan-4L reveals structural requirement for PKC signalling

  • Syndecans, transmembrane heparan sulfate proteoglycans, are coreceptors with integrin in cell adhesion process. It forms a ternary signaling complex with protein kinase C and phosphatidylinositol 4,5 bisphosphate (PIP2) for integrin signaling. NMR data indicates that cytoplasmic domain of syndecan-4 (4L) undergoes a conformational transition in the presence of PIP2, forming oligomeric conformation. The structure based on NMR data demonstrated that syndecan-4L itself forms a compact intertwined symmetric dimer with an unusual clamp shape for residues Leu$^{186}$ -Ala$^{195}$ . The molecular surface of the syndecan-4L dimer is highly positively charged. In addition, no inter-subunit NOEs in membrane proximal amino acid resides (Cl region) has been observed, demonstrating that the Cl region is mostly unstructured in syndecan-4L dimmer. However, the complex structure in the presence of PIP2 induced a high order multimeric conformation in solution. In addition, phosphorylation of cytoplasmic domain induces conformational change of syndecan-4, resulting inhibition of PKC signaling. The NMR structural data strongly suggest that PIP2 promotes oligomerization of syndecan-4 cytoplasmic domain for PKC activation and further induces structural reorganization of syndecan for mediating signaling network in cell adhesion procedure.

Shear-induced structure and dynamics of hydrophobically modified hydroxy ethyl cellulose (hmHEC) in the presence of SDS

  • The interaction between hydrophobically modified hydroxyethyl cellulose (hmHEC), containing approximately 1 wt% side-alkyl chains of $C_{16}$, and an anionic sodium dodecyl sulphate (SDS) surfactant was investigated. For a semi-dilute solution of 0.5 wt% hmHEC, the previously observed behaviour of a maximum in solution viscosity at intermediate SDS concentrations, followed by a drop at higher SDS concentrations, until above the cmc of surfactant when the solution resembles that of the unsubstituted polymer, was confirmed. Additionally, a two-phase region containing a hydrogel phase and a water-like supernatant was found at low SDS concentrations up to 0.2 wt%, a concentration which is akin to the critical association concentration, cac, of SDS in the presence of hmHEC. Above this concentration, SDS molecules bind strongly to form mixed micellar aggregates with the polymer alkyl side-chains, thus strengthening the network junctions, resulting in the observed increase in viscosity and elastic modulus of the solution. The shear behaviour of this polymer-surfactant complex during steady and step stress experiments was examined In great detail. Between SDS concentrations of 0.2 and 0.25 wt%, the shear viscosity of the hmHEC-polymer complex network undergoes shear-induced thickening, followed by a two-stage shear-induced fracture or break-up of the network. The thickening is thought to be due to structural rearrangement, causing the network of flexible polymers to expand, enabling some polymer hydrophobic groups to be converted from intra- to inter-chain associations. At higher applied stress, a partial local break-up of the network occurs, while at even higher stress, above the critical or network yield stress, a complete fracture of the network into small microgel-like units, Is believed to occur. This second network rupture is progressive with time of shear and no steady state in viscosity was observed even after 300 s. The structure which was reformed after the cessation of shear is found to be significantly different from the original state.

  • The purpose of this study are to examine the educational use of the Facebook and to examine the effects of the learners perceived usefulness, attitude and self-efficacy on learning flow and learning presence at university classes using social network service. The subjects of this study are 155 university students attending a class at a 4-year university in Seoul, and certain questions verified in the existing studies were modified, complemented, and used as a tool for measurement. The details of the purpose are as follows. First, it turned out that there were significant differences in learning flow and learning presence in accordance with the levels of the usefulness that learners perceived. It can be said that the higher the perceived level of usefulness, the higher the learning flow and learning presence. Second, it turned out that there were significant differences in learning flow and learning presence in accordance with the levels of learners' perceived attitudes. We can interpret such a result as suggesting that the higher a learner's perceived attitude, the higher the learning flow and learning presence.

  • In this paper, an algorithm is presented to recogniz the driving direction of a vehicle and obstacles in front of it based on highway road image. The algorithm employs a neural network with 27 sub sets obtained from the road image as its input. The outputs include the direction of the vehicle movement and presence or absence of obstacles. The road image, obtained by a video camera, was digitized and processed by a personal computer equipped with an image processing board.

  • In the power system state estimation, the J(x)-index test and normalized residuals ${\gamma}$S1NT have been the presence of bad measurements and identify their location. But, these methods require the complete re-estimation of system states whenever bad data is identified. This paper presents back-propagation neural network medel using autoregressive filter for identification of bad measurements. The performances of neural network method are compared with those of conventional mehtods and simulation results show the geed performance in the bad data identification based on the neural network under sample power system.

  • Collaboration in wireless sensor networks must be fault-tolerant due to the harsh environmental conditions in which such networks can be deployed. This paper focuses on finding signal processing algorithms for collaborative target detection based on the generalized approach to signal processing in the presence of noise that are efficient in terms of communication cost, precision, accuracy, and number of faulty sensors tolerable in the wireless sensor network. Two algorithms, namely, value fusion and decision fusion constructed according to the generalized approach to signal processing in the presence of noise, are identified first. When comparing their performance and communication overhead, decision fusion is found to become superior to value fusion as the ratio of faulty sensors to fault free sensors increases. The use of the generalized approach to signal processing in the presence of noise under designing value and decision fusion algorithms in wireless sensor networks allows us to obtain the same performance, but at low values of signal energy, as under the employment of universally adopted signal processing algorithms widely used in practice.

  • If RCS-e subscribers and SMS subscribers want to exchange instant messages between each other, RCS-e app should know the presence information of the SMS subscribers. However, a RCS-e specification does not have the way to identify the information. This paper propose a software framework that makes RCS-e app can identify the information without having to modify RCS-e app. It describes the call flows between network equipments to manage the information. Also, it defines RCS-e service tags and service tuple for SMS subscribers. Performance evaluation proves that the proposed framework does not add loads to the existing network.

  • The reliability of wireless body area networks is an important research issue since it may jeopardize the vital human life, unless managed properly. In this article, a new modeling and analysis of node misbehaviors in wireless body area networks is presented, in the presence of multi-type failures. First, the nodes are classified into types in accordance with routing capability. Then, the node behavior in the presence of failures such as energy exhaustion and/or malicious attacks has been modeled using a novel Semi-Markov process. The proposed model is very useful in analyzing reliability of WBANs in the presence of multi-type failures.

  • Presence information provides various informations about users such as on-line status, current location, network connection method and connection address, and there are two kinds of presence information, SIP(Session Initiation Protocol) based presence information and XMPP(Extensible Massaging and Presence Protocol) based presence information. In this paper, a multiple server architecture that can handle these two kinds of presence information has been proposed. In this architecture, severs are added dynamically according to number of users to provide system scalability, and load of each server can be effectively controlled. In this system, a new XMPP stanza architecture and presence information data format are designed for load control. Furthermore message exchanging procedures between servers and users for dynamic server control has been also suggested. The performance of the proposed system has been analysed by simulation.
