Ameliorative Potential of Rengyolone Against CCI-induced Neuropathic Pain in Rats

  • The sciatic nerve is the largest nerve among the peripheral nerves, and the damage to the sciatic nerve is caused by mechanical and physical pressure. This is an important disease that consumes a lot of time and money in the treatment process. Among them, research on relieving nerve pain caused by damage to the peripheral sciatic nerve has been made efforts to prevent and treat this disease through various methods such as drugs, natural products, electrical stimulation, exercise therapy, and massage. Existing treatments are not very effective in neurological pain, and countermeasures are needed. Forsythia Fructus, used in this study, has been used as a therapeutic agent for infectious diseases and a pain reliever for cancer from the past, and in past studies, it has been known to properly control the inflammatory response. In this study, rengyolone, a physiologically active substance of Forsythiae Fructus, was administered to rats that caused chronic left nerve pain to verify the pain relief effect. As a result of the experiment, it was found that mechanical pain and cold stimulation pain were significantly reduced in the rengyolone-treated group compared to the non-administered group. In addition, it was found that nerve growth factor (NGF) mRNA expression was significantly reduced and Cyclin-dependent kinase 2 (Cdc2) expression was increased in the rengyolone administration group. This increase in NGF expression is thought to be related to rengyolone's anti-inflammatory regulatory mechanism. It is expected that the reduced NGF was directly involved in pain relief.

사회적으로 고립된 쥐의 해마에서 NGF와 Synapsin I, ChAT의 단백질 수준에 미치는 사전운동효과 (Effects of Exercise Preconditioning on the Expression of NGF, Synapsin I, and ChAT in the Hippocampus of Socially Isolated Rats)

  • 본 연구는 사회적 고립 스트레스로 인한 해마에서의 nerve growth factor (NGF), Synapsin I 및 choline acetyltranferase (ChAT) 감소에 있어서 사전운동경험(exercise preconditioning: EPC)이 미치는 영향을 규명하고자 실시되었다. 목적을 위해 Sprague-Dawley (SD) 쥐(수컷, 22주령, $500.1{\pm}48.41$ g)를 이용해 크게 통제집단(Con)과 운동(Ex)집단으로 구분하여 운동(트레드밀, 5일/주, 최대 18-20 m/min; 50분까지 점진적 증가, 경사 없음, 8주)을 적용하였으며, 이후 각각 사회적 고립(Isolation, 8주)을 적용하여 분석하였다(Group/Con: GC, Group/Ex: GE, Isolation/Con: IC, Isolation/Ex: IE, 각 집단별 n=8). 실험결과, IC집단에서 GC집단에 비해 해마에서 NGF, Synapsin I 및 ChAT가 유의하게 감소한 것으로 나타났다. 반면 IE집단에서 IC집단에 비해 NGF, Synapsin I 및 ChAT의 감소가 유의하게 개선된 것으로 나타났다. 이상의 결과 사회적 고립에 의한 해마에서의 NGF, Synapsin I 및 ChAT 단백질 감소는 EPC에 의해 개선되며, 이를 통해 해마의 기능 저하를 일부 완충 시킬 수 있을 것으로 판단된다.

신경세포에서 신경성장인자(nerve growth factor)의 조절에 미치는 천문동(Asparagus cochinchinensis) 열수추출물의 영향 (Effects of an Aqueous Extract of Asparagus cochinchinensis on the Regulation of Nerve Growth Factor in Neuronal Cells)

  • 천문동(Asparagus cochinchinensis)은 북아시아 지역에서 열병, 감기, 신장질환, 유방암, 염증질환, 뇌질환 등의 치료에 오랫동안 사용되어온 약용식물(medicinal plant)이다. 비록, 천문동의 항염증(ani-inflammatory) 효능에 대한 일부 연구들이 수행되었지만, 신경세포에서 항염증작용과 신경성장인자(nerve growth factor, NGF)의 연관성에 대한 연구는 수행된바 없다. 따라서, 본 연구에서는 신경세포에서 신경성장인자의 분비와 작용기전에 대한 천문동 열수추출물(aqueous extract from A. cochinchinensis, AEAC)의 영향을 연구하였다. AEAC로 처리된 B35세포의 배양액에 NGF단백질의 농도는 대조물질(vehicle) 처리군에 비하여 유의적으로 증가하였으며, 특별한 독성은 관찰되지 않았다. 또한, NGF mRNA의 발현도 단백질의 농도변화와 유사한 양상을 나타내었다. 더불어, B35세포로부터 분비된 NGF의 생리활성을 확인하기 위해, AEAC-조정배지(conditioned medium)를 미분화된 PC12세포에 처리한 후 이들 세포의 신경염성 성장(neuritic outgrowth)을 관찰하였다. PC12세포의 수상돌기 길이(dendritic length)는 vehicle처리군에 비하여 AEAC-조정배지처리군에서 유의적으로 증가하였다. 또한, High affinity NGF 수용체의 하위신호전달에 포함된 p-TrkA와 p-ERK의 발현은 AEAC-조정배지처리군에서 높았지만, low affinity NGF 수용체의 하위신호전달에서는 낮은 수준으로 관찰되었다. 이러한 결과는 AEAC가 신경세포에서 NGF발현과 분비의 조절에 기여하기 때문에 신경퇴행성질환(neurodegenerative disease) 치료제로서 우수한 후보물질임을 제시하고 있다.

Morphological Analyses on Retinal Glial Responses to Glaucomatous Injury Evoked by Venous Cauterization

  • Retinal glial responses to hypertensive glaucomatous injury were spatiotemporally surveyed. Retinas as a whole or vertical sections were processed for anti-glial fibrillary acidic protein (GFAP), anti-Iba1, anti-nerve growth factor (NGF), and anti-tumor necrosis factor (TNF)-${\alpha}$ immunohistochemistry for confocal microscopic analyses. The optic nerve head of paired controls was processed for electron microscopy. GFAP positive astrocytes appeared in the nerve fiber layer in the glaucomatous and control retinas, changing from fine protoplasmic to stout fibrous parallel to glaucomatous duration. Iba1 positive microglia appeared in both retinas, and enormous reaction appeared at the latest glaucomatous. M$\ddot{u}$ller reaction detected by GFAP reactivity expanded from the end feet to whole profile following to duration in the glaucomatous. NGF reactivity expended from the end feet to the proximal radial processes of the M$\ddot{u}$ller cells in both retinas according to glaucomatous duration. TNF-${\alpha}$ immunoreactivity in the nerve fiber layer was stronger in both the glaucomatous and controls than in the normal, and exceptionally at the latest glaucomatous was even lower than the normal. The astrocytes in the optic nerve head are interconnected with each other via gap junction. These results demonstrate that astrocyte reaction propagates to the contralateral via physical links, and TNF-${\alpha}$ is correlated with NGF production for neuroprotection in response to hypertensive glaucomatous injury.

해인탕 추출물이 흰쥐 좌골신경 손상 모델에서 기능회복과 뇌의 c-Fos 발현에 미치는 영향 (Effects of Haein-tang(Hairen-tang) Extract on Functional Recovery in Sciatic Nerve and c-Fos Expression in the Brain after Crushed Sciatic Nerve Injury in Rats)

  • Objectives : Peripheral nerve injuries are commonly encountered clinical problems and often result in severe functional deficits. The purpose of this study was to evaluate the effects of Haein-tang(Hairen-tang) extract on functional recovery and pain release in the sciatic nerve after crushed sciatic nerve injury in rats. Methods : 1. Sciatic functional index(SFI) were performed on functional recovery. 2. c-Fos immunohistochemistry were performed on c-Fos expressions in the paraventricular nucleus(PVN) and ventrolateral periaqueductal gray(vIPAG). 3. Neurofilament immunohistochemistry were performed on neurofilament regeneration. 4. Western blot were performed on brain-derived neurotrophic factor(BDNF) and nerve growth factor(NGF) expression. Results : 1. Haein-tang(Hairen-tang) extract significantly enhanced the SFI value in the sciatic nerve injury and 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. 2. Haein-tang(Hairen-tang) extract significantly suppressed the sciatic nerve injury-induced increment of c-Fos expressions in the PVN and vIPAG in the sciatic nerve injury and 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. 3. Haein-tang(Hairen-tang) extract significantly increased neurofilament expression in the sciatic nerve injury and 50 mg/kg, 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. 4. Haein-tang(Hairen-tang) extract significantly controled the sciatic nerve injury-induced increment of BDNF and NGF expressions in the sciatic nerve injury and 100 mg/kg, 200 mg/kg Haein-tang(Hairen-tang)-treated group. Conclusions : These results suggest that Haein-tang(Hairen-tang) treatment after sciatic nerve injury is effective for the functional recovery by enhancing of axonal regeneration and suppressing of pain.

다양한 구강안면통증환자의 혈장 및 타액에서의 신경성장인자와 감각성 신경펩티드 농도에 관한 연구 (Nerve Growth Factor and Sensory Neuropeptide Levels in Plasma and Saliva of Various Orofacial Pain Patients)

  • 신경섬유 말단에서의 유해자극 인지과정에는 신경성장인자 (nerve growth factor [NGF])와 감각성 신경펩티드가 관여하고, 또한 이들은 중추신경계에도 광범위하게 분포되어 있다. 본 연구는 인체 혈액과 타액에서 NGF와 감각성 신경펩티드(substance P [SP], calcitonin gene-related peptide [CGRP])의 농도를 조사하여 다양한 구강안면통증 증상들과의 관계를 알아보고자 시행되었다. 67명의 구강안면통증 환자 (관절 통증, 치아 혹은 치주 통증, 점막 통증)와 36명의 건강한 성인에서 혈장과 안정시 전타액을 채취하여 효소면역분석법 (enzyme immunoassay)을 이용하여 NGF, SP, CGRP의 농도를 측정하였다. 만성통증척도 (Graded Chronic Pain Scale) 설문지를 이용하여 각 피실험자들의 통증강도를 조사하였으며 안정시 전타액의 타액분비율 또한 측정하였다. 관절 통증 환자군은 치아 통증 환자군, 점막 통증 환자군, 대조군 각각에 비하여 유의하게 높은 혈장 내 NGF 농도를 나타내었다. 치아 통증 환자군의 혈장 내 NGF 농도는 대조군에 비하여 유의하게 높았다. 치아 통증 환자군의 타액 내 SP 농도와 점막 통증 환자군의 타액 내 CGRP 농도 또한 대조군에 비하여 유의하게 높았다. 관절 통증 환자군의 혈장 및 타액 내 SP 농도는 통증 강도와 유의한 상관관계를 보였다. 치아 통증 환자군과 점막 통증 환자군에서, 혈장 내 SP 농도, 타액 내 SP 농도, 타액 내 CGRP 농도는 연령에 따라 증가하였다. 혈장과 타액에서의 신경성장인자와 신경펩티드 검사는 다양한 구강안면통증의 특성과 예후를 평가하는데 도움을 줄 수 있으리라 생각된다.

Tensile stress regulation of NGF and NT3 in human dermal fibroblast

  • Fibroblast is constantly subjected to mechanical loads in connective tissues where mechanical signals are converted to intercellular biochemical events. The aim of this study is to understand the effects of tensile stress on the neurotrophin (NT) and transforming growth factor (TGF) expression of fibroblast in vitro. Nerve growth factor (NGF) stimulates fibroblast migration, and TGF is related to tissue repair. In this study, at the uniaxial stretch of 10% strain and frequency of 0.5 Hz, different resting times of 0, 20, and 60 min are placed in between 10 min stimulations periods. Results show increase in NGF mRNA levels and a substantial decrease in NT3 mRNA after 1 hr of stimulation, indicating that the tensile stress may regulate NGF and NT3, key factors for the neurocosmetic applications. The mRNA level for TGF-${\alpha}$ and TGF-${\beta}2$ had increased up to two-folds after 1 hr of stimulation, showing that the tensile stress may control TGF, an important part of wound healing.

말초신경재생을 위한 hNGF-$\beta$ recombinant Adenovirus의 제작 및 수종세포주에서 신경성장인자의 발현 (CONSTRUCTION OF HNGF-$\beta$ RECOMBINANT ADENOVIRUS & SCREENING OF ITS EXPRESSION AFTER TRANSFECTION INTO VARIOUS CELL LINES)

  • Nerve growth factor(NGF) has a critical role in peripheral nerve regeneration. The aim of this study is to construct a well-functioning hNGF-$\beta$ recombinat adenovirus for the ultimate development of improved method to promote peripheral nerve regeneration with adenovirus mediated hNGF-$\beta$ gene transfection into Schwann cells. First PCR associated cloning of GFP-tagged hNGF-$\beta$ which was ligated into E1/E3 deleted adenoviral vector was performed and tranfected into E. coli to construct hNGF-$\beta$ recombinant adenovirus. After production of recombinat adenovirus in a large scale, its transfection efficiency, expression, and function were evaluated using cell lines or primarily cultured cells of HEK293 cells, Schwann cells, fibroblast(NIH3T3) and myocyte(CRH cells). GFP expression was observed in 90% of infected cells compared to uninfected cells. Total mRNA isolated from hNGF-$\beta$ recombinat adenoviru infected cells showed strong RT-PCR band, however, LacZ recombinant adenovirus infected or uninfected cells did not. NGF quantification by ELISA showed a maximal release of 18.865 +/- 0.31ng/mL at 4th day. PC-12 cells exposed to media with hNGF-$\beta$ recombinant adenovirus infected Schwann cell demonstrated higher levels of differentiation compared with controls. We generated hNGF-$\beta$ recombinant adenovirus and induced over expression of NGF successfully in nonneuronal and neuronal cells. Following these result, it is expected to develop an improved treatment strategy peripheral nerve regeneration using the hNGF-$\beta$ gene transfected cells.
