병원내 공기중 미생물의 농도에 관한 조사연구 (An Investigation on Concentration of Airborne Microbes in a Hospital)

  • A survey was conducted to measure concentration of airborne microbe in a hospital using RSC air sampler during October~November 1991.The result was as follows: 1) In an agar strip GK-A media for total counts of microbial particles. The highest count were 1384 CFU/m$^3$ in the main lobby, followed by 912 CFU/m$^3$, in the obstetric room, 688 CFU/m$^3$ in 1CU. By gram staining, the distribution for organisms in the air were shown 74.1% in gram possitive cocci followed by 16.8%, in gram possitive bacilli 6.7% in gram negative bacilli and 4.7% in yeast, but low organism was detected in recovery room with 194 CFU/m$^3$. 2) In agar strip S media for Staphylococci the count at the main lobby was detected in the recovery room with 92 CFU/m$^3$, Tests of coagulase were negative Staphylococci with 78%, and positive Staphylococci with 22%. The Staphylococci were highly resistance to penicillin, ampicillin and sensitive to amikacin, cefazolin, gentamycin and chloramphenicol. 3) In agar strip C media for coliform bacteria the colony counts at the main lobby was 139 CFU/m$^3$ and treatment room was 190 CFU/m$^3$, most frequently isolated microorganisms were non fermentative bacilli. 4) In agar strip HS media for yeast and molds. Most frequently colony counts 17~76 CFU/m$^3$, 0.5% lactophenol cotton blue stains were shown unidentified 77.2%, 8.1%, in Penicillium 8.1% in Aspergillus, and 3.8% in mucor.

  • The high surface area polymer, AquaMats$^{circledR}$ was used for the leachate purification process originated from the Samsan Dong Landfill in Ulsan Metropolitan City. And then, three species of dominant microbes were isolated and identified from AquaMats$^{circledR}$. Gram staining revealed these microbes to be Gram-negative rod strains: They were identified as Agrobacterium radiobacter, Pseudomonas cepacia, Flavobacterium indologenes. All they showed no growth on media in which the leachate was added alone, but a rapid proliferation rate on media with yeast extract as nutrient requirements.

  • The incidence of aseptic meningitis infection is ensuing and threatening the health of children. Enteroviruses are the major agents of aseptic meningitis and identification of virus has been a clue to diagnosis and epidemiology. The outbreak of aseptic meningitis occurred in Pusan, 1998. Patients were concentrated from April through November. Children were more susceptible than adults. Among 306 cases of specimens from stool, throat swab tested, only 7.2% were positive on virus isolation, 12 cases from stool and 10 from throat, respectively. All isolated 7 serotypes of viruses represented cytopathic effect on cultured cells. Three types of echovirus 6.25, 30 and coxsackievirus B2, B3, B4, B6 were identified by neutralizing antibody test. Isolated coxsackievirus and echovirus were observed by an electron microscope with negative staining.

수종(數種) Rhizobium의 편모에 대한 전자현미경적(電子顯微鏡的) 관찰(觀察) (Electron Microscopical Observation on the Flagella of Several Species of the Genus Rhizobium)

  • Five strains of the Genus Rhizobium were isolated from the nodules of five leguminous plants respectively. They were identified according to Bergey's Manual together with the results of Vincent. The flagella of each strains were observed by electron microscope using negative staining with PTA and metal shadowing with chromium. Five host plants and identified Rhizobium strains were as. follows. Pisum sativum.....R. leguminosarum Phaseolus vulgaris.....R. phaseoli Trifolium repens.....R. trifolii Glycine max.....R. japonicum Lupinus grandiflorus.....R. lupini Electron micrographs showed that R. leguminosarum and R. phaseoli had 4 peritrichous flagella, where as R. trifolii had 5 peritrichous flagella. On the other hand, R. japonicum and R. lupini had 1 subpolar flagellum.

  • Cryo-transmission electron microscopy (cryo-TEM) allows us to perform structural analysis of a analyses of large protein complexes, which are difficult to analyze using X-ray crystallography or nuclear magnetic resonance. The most common examples of proteins used are ribosomes and proteasomes. In this paper, we briefly describe the advantage of cryo-TEM and the process of two-dimensional classification by considering a human proteasome as an example.

해양미생물로부터 면역증강물질 생산의 최적화

  • A halophilic bacterium was isolated from domestic marine. The bacterium was gram negative and motile. Transmission electron micrograph after cultivating for 6-, 20-, 72-, and 144 hrs showed that it was bacilli and contained intracellular granules, which were pleomorphic, larger in density by the time and considered to be PHB since they were positive on the sudan black B staining. The presence of sodium chloride was critical, because the isolated marine bacterium could not multiply and even produce any immunostimulant in the deficiency of sodium chloride. The strain produced an immunostimulant, which was investigated for the biological characteristics. The optimal conditions for the production of the immunostimulant were 1 % dextrose and 1 % yeast extract in artificial sea water for carbon and nitrogen sources, respectively. The initial pH and growth temperature for the production were 8.0 and $30^{\circ}C$ under the presence of oxigen, respectively.

Characterization of haemocytes in the surf clam Mactra veneriformis

  • Haemocyte characterization in the surf clam, Mectra veneriformis was carried out based on morphological, cytochemical, and phagocytic characteristics. The haemocytes were classified into two cell types, granulocytes and agranulocytes on the basis of presence of cytoplasmic granules. Granulocytes were then classified again into 2 types, large eosinophilic granulocytes and small eosinophilic granulocytes after staining with May-Grünwald Giemsa. In electron microscopy, both types of granulocytes contained electron-dense and electron-lucent cytoplasmic granules. Agranulocytes (hyalinocytes) were also divided into two cell types, large agranulocytes and small agranulocytes based on their sizes. Both cell types did not have granules in cytoplasm. Granulocyte and agranulocyte were negative for the enzyme activities of alkaline phosphatase, peroxidase, and $\beta$-glucuronidase but positive for phenoloxidase and acid phosphatase activities. Both types of haemocytes have phagocytic activity, with the exception of small agranulocyte, and granulocytes seemed more active in this respect than agranulocytes. This present study is the first study to characterize haemocytes of M. veneriformis.

The Effects of Electron Beam Exposure Time on Transmission Electron Microscopy Imaging of Negatively Stained Biological Samples

  • Negative staining electron microscopy facilitates the visualization of small bio-materials such as proteins; thus, many electron microscopists have used this conventional method to visualize the morphologies and structures of biological materials. To achieve sufficient contrast of the materials, a number of imaging parameters must be considered. Here, we examined the effects of one of the fundamental imaging parameters, electron beam exposure time, on electron densities generated using transmission electron microscopy. A single site of a negatively stained biological sample was illuminated with the electron beam for different times (1, 2, or 4 seconds) and sets of micrographs were collected. Computational image processing demonstrated that longer exposure times provide better electron densities at the molecular level. This report describes technical procedures for testing parameters that allow enhanced evaluations of the densities of electron microscopy images.

Bacterial Sepsis Associated with a Captive State Caused by Edwardsiella tarda in a Eurasian Brown Bear (Ursus arctos arctos)

  • Edwardsiella (E) tarda belongs to the Enterobacteriaceae family and is a motile, gram-negative, rod-shaped, facultative anaerobe regarded as an opportunistic and food-borne pathogen in animals and humans. A 21-year-old male Eurasian brown bear (Ursus arctos arctos) died suddenly without any preliminary signs. Necropsy performed according to standard protocol revealed swollen abdomen with hemorrhagic congestions of the gastroenteric organs, ascites, and hemorrhagic exudates around the mouth. The liver showed discoloration, along with a severely swollen and multiple hemorrhages of the spleen, an elongated gallbladder, and a congested cortex and medullar lesion of kidneys. The stomach contained semi-liquid exudates and undigested chicken exuding a decayed odor. The stomach membranes were dark-gray in color with several cysts in the fundus lesions. Rod-shaped bacteria were found in the major organs by Giemsa staining, identified as E. tarda using a biochemical rapid diagnostic identification kit.

공기중에서 분리된 포도구균의 항생제 감수성 (Antibiotics Sensitivity to Isolated Staphylococcus sp. from the Air)

  • This investigation was performed to isolate and identify Staphylococcus sp. from air at the 25 sites in one hospital in Daegu. Drug sensitivity tests bacteriologically were also studied. The results are summarized as follows: 1. In gram staining of isolated colonies, the number of staphylococci strains was 959(70.2%) among 1367 collected in the morning, and 653(66.2%) among 987 collected in the afternoon. 2. The number of isolates was slightly higher in samples collected from the outpatient treatment rooms and wards in the main building than in samples from the other sites. 3. In biochemical tests of 1612 strains of isolates gram positive cocci, the number of coagulase positive strains was 584(36.2%) and coagulase negative 1028(63.8%). In experiments on growth on mannitol agar containing 7.5% NaCl and mannitol fermentation tests, the number of mannitol fermentation strains was 423(30.8%) and nonfermenter was 951(59.2%). 4. In antibiotic sensitivity tests of 746 identified strains of staphylococci, coagulase positive strains showed resistance in. higher portions to penicillin(89.7%), chloramphenicol(64.6%), gentamycin (52.1%) and tetracycline(45.2%), whereas in lower portions to kanamycin(39.2%) and apmicillin(33.1%). 5. Among coagulase negative and mannitol fermenting strains, higher portions showed resistance to chloramphenicol(76.5%), penicillin(69.8%) and ampicillin(63.6%), whereas lower portions showed resistance to kanamycin(48.8%), tetracycline(39.5%) and gentamycin(34.0%). 6. The number of strains showing multiple resistance to above 6 antibiotics were 15(52(19.8%) among coagulase positive and mannitol non-fermenter and 36(22.2%) among coagulase negative and mannitol fermenter.
