Reconstructed Adeno-Associated Virus with the Extracellular Domain of Murine PD-1 Induces Antitumor Immunity

  • Background: The negative signaling provided by interactions of the co-inhibitory molecule, programmed death-1 (PD-1), and its ligands, B7-H1 (PD-L1) and B7-DC (PD-L2), is a critical mechanism contributing to tumor evasion; blockade of this pathway has been proven to enhance cytotoxic activity and mediate antitumor therapy. Here we evaluated the anti-tumor efficacy of AAV-mediated delivery of the extracellular domain of murine PD-1 (sPD-1) to a tumor site. Material and Methods: An rAAV vector was constructed in which the expression of sPD-1, a known negative regulator of TCR signals, is driven by human cytomegalovirus immediate early promoter (CMV-P), using a triple plasmid transfection system. Tumor-bearing mice were then treated with the AAV/sPD1 construct and expression of sPD-1 in tumor tissues was determined by semi quantitative RT-PCR, and tumor weights and cytotoxic activity of splenocytes were measured. Results: Analysis of tumor homogenates revealed sPD-1 mRNA to be significantly overexpressed in rAAV/sPD-1 treated mice as compared with control levels. Its use for local gene therapy at the inoculation site of H22 hepatoma cells could inhibit tumor growth, also enhancing lysis of tumor cells by lymphocytes stimulated specifically with an antigen. In addition, PD-1 was also found expressed on the surfaces of activated CD8+ T cells. Conclusion: This study confirmed that expression of the soluble extracellular domain of PD-1 molecule could reduce tumor microenvironment inhibitory effects on T cells and enhance cytotoxicity. This suggests that it might be a potential target for development of therapies to augment T-cell responses in patients with malignancies.

Enhancement of Anticancer Activities of Ephedra sinica Stapf Extracts by Nano-encapsulation (마황 추출물의 나노 입자화를 통한 항암 활성 증진)

  • This study showed the increase of antitumor activities of water soluble E. sinica extract by nano-encapsulation process with lecithin. Five groups of lecithin only group (LO), lecithin nano-encapsulated E. sinica group (LE), E. sinica only group (EO), one negative control group (NCO) and positive control group (PCO) were set for several anticancer experiment and fed into Sarcoma-180 injected mice. The cytotoxicity of LE on the human normal kidney cell (HEK293) showed 14.8% lower than 19.2% of EO and 18.4% of LO. Growth of human liver carcinoma cell and human stomach carcinoma cell as representative of digestive system in vitro was inhibited up to about 85.1% and 87.3%, in adding 1.0 mg/$m{\ell}$ of LE, which values 15% higher than that from conventional EO. The survival rates of each mice group were 40%, 63%, 48%, 33% and 100%, respectively after 40 days of injecting Sarcoma-180. The increment of their body weights of the extract feeding groups was suppressed down to 10~15%, compared to the negative control. The nano-particles also reduced the hypertrophy of the internal organs such as spleen and liver down to 15~20%, compared to those as the other groups. Among them, LE effectively reduced the size of tumor form to 20%. From these results, in vitro and in vivo antitumor activities of E. sinica could be enhanced by using nano-encapsulation process with lecithin because of better permeation into the cancer cells by confocal observations.

Multi-carbohydrase application into energy and amino acid deficient broiler diets: A strategy to enhance performance of broiler chickens

  • The effect of Multi-Carbohydrase (MC) supplementation on growth performance, visceral organ weights, blood metabolites, jejunum morphology, nutrient digestibility, and carcass parameters of broiler chickens fed nutrient-deficient corn soybean-meal based diets containing high levels of non-starch polysaccharides from wheat and wheat by-products was investigated. A total of 378 one-day-old Ross 308 broiler chickens were randomly assigned to one of seven dietary treatments to give six replicates per treatment (nine birds per pen). Dietary treatments were as follows: (1) positive control (PC; commercial standard diet); (2) negative control 1 (NC-1; PC-120 kcal/kg metabolizable energy); (3) NC-2 (PC-3% standardized ileal digestibility [SID] amino acids). The remaining four dietary treatments were formulated with the addition of MC (MC; Superzyme-CSTM) into two negative controls along with two supplementation levels of MC (i.e., 0.025% and 0.05%, respectively). Improved body weight, average daily gain, and feed conversion ratio (p < 0.05) were observed in broiler chickens fed a reduced energy diet supplemented with MC compared to birds fed NC-1 diet from days 1-35. Additionally, birds fed a reduced energy diet with 0.05% MC showed comparable (p > 0.05) growth performance with birds fed PC for 35-day post-hatch. Furthermore, the addition of MC into reduced amino acid diets improved (p < 0.05) growth performance. Broiler chickens fed MC supplemented nutrient-deficient diets showed a greater (p < 0.05) villus height to crypt depth ratio than birds fed diets without MC on days 21 and 35. Similarly, improved (p < 0.05) nutrient digestibility was observed in birds fed reduced energy diets supplemented with MC compared to birds fed NC-1 on days 21 and 35. Our results suggest that MC supplementation into reduced energy or reduced amino acid diets containing wheat and wheat by-products has the potential to improve growth performance and nutrient digestibility while maintaining healthier gut morphology in broiler chickens from 1 to 35 days of age.

Effects of Dietary Supplementation of Coffee Meal on Growth Performance, Blood Biochemical Profiles and Antioxidant Defense System in Broiler Chickens (사료 내 항산화원으로서 커피박 첨가가 닭의 사양성적, 혈액생화학성상 및 항산화 작용에 미치는 영향)

  • The effects of dietary supplementation of dried coffee meal (CM) on growth performance, blood biochemical profiles, the weights of immune-related organs, and the antioxidant defense system in broiler chicks were examined. A total of 162, 3-day-old male broiler chickens were assigned to three dietary groups: control group (CON), control diet added with 0.5% CM (CM0.5), and control diet added with 1.0% CM (CM1.0). In vitro antioxidant activity test, coffee extracts showed concentration-dependent increase in radical scavenging activity. Dietary addition of 0.5 and 1.0% of CM did not have negative effects on growth performance and feed conversion during the experimental periods, whereas dietary CM significantly (P<0.05) increased the relative weight of thymus without changes in the other organ weights. In addition, birds fed the diet supplemented with CM (0.5 and 1.0%) significantly increased blood albumin without affecting other components including glucose, triglyceride and cholesterol compared with those fed control diet. In antioxidant defense system, the specific activities of superoxide dismutase, glutathione peroxidase and glutathione S-transferase and the level of glutathione in the small intestine and liver were not affected by dietary supplementation of CM. However, hepatic lipid peroxidation in birds fed the diet supplemented with 0.5% CM was significantly (P<0.05) decreased compared with that in control birds. In conclusion, dietary supplementation of CM(0.5~1.0%) has potential for use as a natural antioxidant source without negative effect on growth performance in broiler chickens.

Effect of Buan Mulberry on Metabolic Improvement in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨쥐의 신진대사 증진에 대한 부안 오디의 효과)

  • This study was designed to evaluate the anti-diabetic effects of Buan mulberries by using an insulin-dependent diabetes mellitus animal model. Several studies have shown that mulberries have metabolism-improving, antioxidant, and lipid-lowering properties in rats with streptozotocin (STZ)-induced diabetes. In this study, Sprague-Dawley male rats were randomly assigned to 1 normal control group and 5 STZ-induced diabetes groups: rats that had STZ-induced diabetes and did not receive any agents (diabetic group; negative control), rats that had STZ-induced diabetes and received insulin (insulin group; positive control), rats that had STZ-induced diabetes and received 0.5% mulberry extract (0.5% mulberry group), rats that had STZ-induced diabetes and received 1.0% mulberry extract (1.0% mulberry group), and rats that had STZ-induced diabetes and received 2.0% mulberry extract (2.0% mulberry group). Mulberry extracts were administered to the diabetic animals for 4 weeks. The rats that received mulberry extracts showed lower body weights and insulin levels, as well as higher kidney weights, blood glucose levels, urine quantities, and water intake in comparison with the normal controls. Further, the insulin concentrations in the mulberry-fed animals were higher than those in the diabetic group, and the kidney weights, blood glucose levels, urine quantities, and water intake in the mulberry-fed animals were lower than the corresponding values in the diabetic controls. These results suggest that mulberry may be an effective functional food to prevent diabetes-related complications.

Prediction Formulas of Pulmonary Function Parameters Derived from the Forced Expiratory Spirogram for Healthy Nonsmoking and Smoking Adults and Effect of Smoking on Pulmonary Function Parameters (비흡연 및 흡연 성년 한국인에서의 노력성호기곡선을 이용한 폐활량측정법 검사지표들의 추정상치 및 이에 대한 흡연의 효과)

  • Background : The past studies on prediction formulas of pulmonary function parameters in healthy nonsmoking Korean adults have been performed in relatively small number of subjects and the reported results were restricted on a few parameters. Also there was no systematic investigation into the effect of smoking on pulmonary function parameters in smokers who have no respiratory symptoms. Therefore we attempted to establish prediction formulas of pulmonary function parameters and examined the effect of smoking on pulmonary function parameters. Methods We analyzed the result of parameters derived from the forced expiratory spirogram in 1,067 nonsmoking subjects from June in 1990 to December in 1991. They consisted of 306 males and 761 females and had neither respitatory symptoms nor history of respiratory disease. We derived prediction formulas by multiple linear regression method from their age, heights, and weights in each sex. To examine the effect of smoking on pulmonary function parameters, we classified 383 smoking men into three groups according to the past amount of smoking as follows : i.e. group of smokers who have smoked below 10 pack-years, 10-20 pack-years and above 20 pack-years. Regarding each group of past smoking as an independent dummy variable, we analyzed pulmonary function parameters including nonsmoking men as a baseline by multiple linear regression. We evaluated the smoking effect on pulmonary function parameters according to estimated p-value. Result : 1) Prediction formulas for pulmonary function parameters in each sex were derived. 2) The past smoking less than 10 pack-years does not give any effect on pulmonary function parameters. The past smoking of 10~20 pack-years showed significant negative correlation with $FEV_1$/FVC and FEF 25~75%, and the smoking above 20 pack years showed negative correlation with $FEV_1$ and $FEV_1$/FVC. Conclusion : We have got prediction formulas of pulmonary function parameters which is driven from forced expiratory spirogram in nonsmoking Korean adults by multiple linear regression from age, heights and weights of subjects. The past smoking more than 10 pack-years showed negative correlation with some pulmonary function parameters of airflow obstruction.

Effects of Egg Yolk Antibody Powder (IgY) Supplementation on Growth Performance, Blood Component Profile, Intestinal Microflora, and Immunoglobulin G in Meat Ducks (난황항체제제(IgY) 첨가가 육용오리의 생산성, 혈액성상, 장내 미생물 및 면역글로불린 함량에 미치는 영향)

  • This study was conducted to investigate the effect of dietary egg yolk antibody (IgY) powder supplementation on the growth performance, blood component profile, intestinal microflora, and immunoglobulin G in ducks. A total of 300 1-day-old ducks (Cherry Valley) were randomly divided into 5 groups, with 3 replicates of 20 birds. The treatment groups were the negative (NC), positive (PC), egg yolk antibody powder 0.1% (T1), egg yolk antibody powder 0.5% (T2), and egg yolk antibody powder 1.0% (T3) groups. In the growth performance of ducks during the entire experimental period, the IgY groups and positive control group (PC) showed significantly higher (P<0.05) body weights and body weight gains compared to the negative control group (NC). However, no significant differences were observed in the feed intake and feed conversion ratio. The blood component profiles showed that the IgY 0.1 and 1.0% groups decreased in total cholesterol content compared to the NC group. The aspartate aminotransferase (AST) and alanine aminotranferease (ALT) contents were lower in the IgY 1.0% group, but there were no significant differences. Regrading the leukocyte content after feeding IgY, the heterophil: lymphocyte ratio decreased in the IgY groups, especially in the IgY 0.1% group, which had a lower content than the other groups. However, these results showed no significant differences. The Lactobacillus count in the intestines significantly increased (P<0.05) in the IgY 0.1 and 0.5% groups, the level of IgY increased, and the Escherichia coli count decreased. However, no significant difference was observed in the total plate count. The immunoglobulin G content was lower in the IgY groups than in the NC group, and compared with the IgY groups, the IgY 0.5% had a lower content, which was not a significant difference.

  • Field experiments were conducted to evaluate the competitive characteristics of weeds with direct-seeded rice. In both dry- and water-seeded rice a weeds) started to grow over the plant height of rice from 45 days after seeding, and there was a great difference in plant height between rice and weeds from 75 days after seeding. Dry weights of weeds drastically increased from 30 days after seeding in both dry- and water-seeded rice, and the increase of dry weight was faster in dry-seeded rice than in water-seeded rice with time. Although weeds competed with rice at earlier growth stages, they did not reduce rice yields and its components until competition period of 75 days after seeding. Required period of weed free at the beginning of the season was 30 to 45 days from seeding in order to avoid any yield loss. The numbers of panicle per plant, spikelets per panicle, grain weight and yield of rice in dry-seeded rice had highly negative correlations with the growth ratios of Echinochloa crus-galli, Ludwigia prostrata, Cyperus difformis, Bidens frondosa, and Cyperus serotinus. In water-seeded rice, the number of plants, panicle per plant, spikelets per panicle and yield had highly negative correlations with growth rate of Echinochloa crus-galli, but the percentage of filled spikelet had negative correlation with Cyperus difformis. Research indicated that in direct-seeded rice weed competition with rice started from 30 days after seeding and significant yield loss by weed competition occurred from 75 days after seeding.

  • This study was conducted to determination of the endocrine distruptor function of 'Parabens' by dosing ethyl paraben, propyl paraben, isopropyl paraben, butyl paraben and isobutyl paraben to the immature SD rats. 18 groups were given vehicle control group, negative control group (Dibutyl phthalate), postive control group ($1'7-{\alpha}$ Ethynylestrdiol) and each paraben groups involved 3 dose level. Rats were injected with 62.5, 250 and 1,000mg/kg from postnatal day 19 till 21 once a day in subcutaneous and a total 3, times. There was no treatment related death. but, subcutaneous nodule, edema, alopecia and scrub formation on injection site was observed. These signs was become worse in high dose level. these signs was cause from physical stimulation by test substance which parabens were mix with com oil as vehicle. In the analysis of organ weights, absolute and relative weights of brain, spleen, liver, thymus, heart, kidneys, adrenals, ovaries and vagina were no difference with control group. but, wet and blotted weight of uterus was increased in every high dose parabens treat group. Especially, all dose level of isobutyl paraben was showed increment of uterus weight. uterus dilatation of parabens treated group was observed in gross anatomic pathology and these result was agree with wet and blotted weight of uterus. In the result of this study, estrogenic effect as endocrine distruptor was observed in ethyl paraben, propyl paraben, isopropyl paraben, butyl paraben and isobutyl paraben. and it was considered isobutyl paraben has highest estrogrnic effect under the condition of this study.

Leaf Shape Index, Growth, and Phytochemicals in Two Leaf Lettuce Cultivars Grown under Monochromatic Light-emitting Diodes (단색 발광다이오드에서 자란 축면상추 두 품종의 엽형, 생장 및 기능성 물질)

  • As an artificial light source, light-emitting diode (LED) with a short wavelength range can be used in closed-type plant production systems. Among various wavelength ranges in visible light, individual light spectra induce distinguishing influences on plant growth and development. In this study, we determined the effects of monochromatic LEDs on leaf shape index, growth and the accumulation of phytochemicals in a red leaf lettuce (Lactuca sativa L. 'Sunmang') and a green leaf lettuce (Lactuca sativa L. 'Grand rapid TBR'). Lettuce seedlings grown under normal growing conditions ($20^{\circ}C$, fluorescent lamp + high pressure sodium lamp, $130{\pm}5{\mu}mmol{\cdot}m^{-2}{\cdot}s^{-1}$, 12 hours photoperiod) for 18 days were transferred into incubators at $20^{\circ}C$ equipped with various monochromatic LEDs (blue LED, 456 nm; green LED, 518 nm; red LED, 654 nm; white LED, 456 nm + 558 nm) under the same light intensity and photoperiod ($130{\pm}7{\mu}mmol{\cdot}m^{-2}{\cdot}s^{-1}$, 12 hours photoperiod). Leaf length, leaf width, leaf area, fresh and dry weights of shoots and roots, shoot/root ratio, SPAD value, total phenolic concentration, antioxidant capacity, and the expression of a key gene involved in the biosynthesis of phenolic compounds, phenylalanine ammonia-lyase (PAL), were measured at 9 and 23 days after transplanting. The leaf shape indexes of both lettuce cultivars subjected to blue or white LEDs were similar with those of control during whole growth stage. However, red and green LEDs induced significantly higher leaf shape index than the other treatments. The green LED had a negative impact on the lettuce growth. Most of growth characteristics such as fresh and dry weights of shoots and leaf area were the highest in both cultivars subjected to red LED treatment. In case of red leaf lettuce plants, shoot fresh weight under red LED was 3.8 times higher than that under green LED at 23 days after transplanting. In contrast, the accumulation of chlorophyll, phenolics including antioxidants in lettuce plants showed an opposite trend compared with growth. SPAD value, total phenolic concentration, and antioxidant capacity of lettuce grown under blue LED were significantly higher than those under other LED treatments. In addition, PAL gene was remarkably activated by blue LED at 9 days after transplanting. Thus, this study suggested that the light quality using LEDs is a crucial factor for morphology, growth, and phytochemicals of two lettuce cultivars.
