FT-NIR spectroscopy를 이용한 현미의 총 식이섬유함량분석 예측모델 개발 (Development of Prediction Model for Total Dietary Fiber Content in Brown Rice by Fourier Transform-Near Infrared Spectroscopy)

  • 분석이 번거로웠던 현미의 총 식이섬유(TDF) 함량을 신속하면서도 친 환경적인 비파괴 분석방법인 FT-NIRS를 이용하여 예측 모델을 개발하였다. 현미는 국내산으로 전남 지방에서 재배된 47개 품종(516개 시료)에 대해서 AOAC 방법에 준한 효소법에 의해 각 측정 시료별 TDF 함량을 분석하였다. 습식 분석된 TDF 함량의 분석오차범위는 0.17-0.72% 이었다. FT.NIRS로 측정된 스렉트럼의 검량식은 빛의 산란 효과를 최소화하기 위해 수학적 처리를 하였고, 몇 개의 특정 파장이 아닌 전 파장 영역(1,000-2,500nm)에 대해서 PLS법으로 작성하였다 회귀분석과 검량식은 NIRCal chemometric software에 의해 작성되었다. 얻어진 검량식의 정확도는 상관계수(r), SEE 및 SEP로 확인하였다. 현미 중 총 식이섬유 함량에 대한 회귀분석을 행한 결과, 상관계수는 0.9780, SEE는 0.0636, SEP는 0.0642로 측정 정확도가 우수함으로 현장 적용을 위한 실용화도 가능할 것으로 판단된다.

근적외선분광법을 이용한 플라스틱류의 연소 잔류물 분석 (Thermal residues analysis of plastics by FT-near infrared spectroscopy)

  • 화재 현장에서 완전 연소하지 않은 잔류물의 성분을 규명하는 것은 화재 원인을 추적하는 중요한 단서가 된다. 화재 현장에서 많이 발견되는 플라스틱 연소 잔류물의 종류를 규명하기 위하여 폴리에틸렌 (Polyethylene, PE) 수지와 Acrylonitrile butadiene styrene (ABS) 수지의 연소 후 잔류물에 관한 연구를 진행 하였다. 콘 칼로리미터 (Cone calorimeter)를 사용하여 플라스틱 시료를 200, 300, 350, 400, $500^{\circ}C$로 각각 3분 동안 연소시킨 후 변화된 무게와 연소생성물을 얻었다. 각 온도에서 얻어진 연소생성물을 동결 건조 후 분쇄한 분말 시료를 FT-near infrared (NIR) spectrometer로 스펙트럼을 측정 하였다. 폴리에틸렌 수지 시료를 연소시켰을 때 $350^{\circ}C$까지는 무게의 변화가 없었으나 $400^{\circ}C$ 이상에서는 급격한 무게의 변화를 측정 할 수 있었다. 각 온도에서 얻어진 폴리에틸렌 수지 및 ABS 수지 시료의 FT-NIR 측정 스펙트럼을 주성분 분석(Principle component Analysis, PCA)를 통해서 연소 잔유물이 폴리에틸렌과 ABS 지임을 확인할 수 있었다. 따라서 근적외선 분광법으로 화재현장에서 발견되는 불연소 플라스틱의 종류를 신속하게 확인할 수 있었다.

  • Fast and dynamic biochemical, enzymatic and morphological changes occur during the so-called generative development and during the vegetative processes in seeds. The most characteristic biochemical and compositional changes of this period are the formation and decline of storage components or their precursors, the change of their degree in polymerization and an extensive change in water content. The aim of the present study was to detect the maturation processes in seed nondestructively and to verify the applicability of near infrared spectroscopic methods in the measurement of physiological, chemical and biochemical changes in wheat seed. The amount and variation of different water “species” has been changed intensively during maturation. Characteristic changes of three water absorption bands (1920, 1420 and 1150 nm) during maturation were analysed. It was concluded that the free/bound transition of water molecules could be followed sensitively in different region of NIR spectra. Kinetic changes of carbohydrate reserves were characteristic during maturation. An intensive formation and decline of carbohydrate reserves were observed during early stage of maturation (0 -13 days, high energy demand). An accelerated formation of storage carbohydrates (starch) was detected in the second phase of maturation. Five characteristic absorption bands were analysed which were sensitive indicators the changes of carbohydrates occurred during maturation. Precursors of protein synthesis and the synthesis of reserve proteins and their kinetic changes during maturation were followed from NIR spectra qualitative and qualitatively. Dynamic formation of amino acids and the changes of N forms were detected by spectroscopic, chromatographic and by capillary electrophoresis methods. Calibration equations were developed and validated in order to measure the optimal maturation time protein and moisture content of developing wheat seeds. The spectroscopic methods are offering chance and measurement potential in order to detect fine details of physiological processes. The spectra have many hidden details, which can help to understand the biochemical background of processes.

Near-infrared Spectroscopy of Young Stellar Objects around the Supernova Remnant G54.1+0.3

  • We present near-infrared (NIR) spectra of 6 young stellar objects (YSOs) around the supernova remnant G54.1+0.3 obtained with TripleSpec, a slit-based NIR cross-dispersion echelle spectrograph on th 5-m Palomar Hale telescope covering the entire NIR atmospheric window of 1-2.4 micron. These YSOs, whose formation was possibly triggered by the progenitor of G54.1+0.3, show significant mid-infrared (MIR) excess and have been proposed to be late O- and early B-type YSOs based on their spectral energy distribution. Our TripleSpec observations reveal the existence of strong H and He I lines, consistent with the previous interpretation of their spectral types, while the absence of Br-gamma emission line indicates that the YSOs do not have a nearby circumstellar disk. We discuss the relation between these YSOs and G54.1+0.3 based on the TripleSpec data and previous photometric data as well.

근적외 분광법을 이용한 양념 야채의 건조 불균일성 관찰 (Observation for drying non-uniformity of allium vegetables using NIR spectroscopy)

  • 본 연구에서는 근적외 분광법을 이용하여 양념용 채소로 많이 사용되는 파, 마늘, 양파의 건조 불균일성을 관찰하였다. 대류식 온풍건조기에서 건조 위치에 따른 양념채소류의 건조 상태 차이를 조사한 결과, 건조 트레이의 높이나 위치에 따라 바람의 온도와 습도에 차이가 발생하여 건조가 균일하게 되지 않았다. 건조 정도가 다른 양념채소류의 2차 미분 근적외 스펙트럼을 비교한 결과 1,390 nm에서 1,400 nm 부근의 수소결합이 약한 물이 먼저 증발되었고 1,420 nm 부근의 수소결합이 강한 물은 건조 후에도 잔존함을 관찰할 수 있었다. 초분광 NIR 이미징 기법으로 측정한 스펙트럼을 주성분분석함으로써 건조채소의 건조도를 가시적으로 판별할 수 있었다.

  • 화석연료에 대한 의존성을 줄이기 위한 대안으로서 재생에너지 사용이 요구되고 있다. 청정에너지원의 하나인 바이오매스는 그 물성치의 실시간 파악이 중요하기 때문에 다양한 종류의 바이오매스에 대해 널리 연구되어 왔으며, 방법론적인 측면에서는 비침투성이며 많은 정보를 가진 특징으로 인하여 근적외선 분광법이 성공적으로 적용되었다. 본 논문에서는 여러 바이오매스 종류에 대한 물성치의 빠른 예측을 위해 근적외선 데이터에 기반한 비선형 방법론의 적용성을 평가하였다. 다양한 방법론에 기반한 예측 모델들을 근적외선 데이터의 전처리방법과 조합하여 예측 성능을 평가하였다. 바이오매스 물성 예측 모델의 성능에서는 선형 모델보다는 비선형 모델에서 예측오차가 최소화되었으며 전처리 방법과 결합되었을 때 최적의 예측결과를 얻을 수 있었다.

근적외 분석법에 의한 참깨의 원산지 판별 (Identification of geographical origin of sesame seeds by near infrared spectroscopy)

  • 근적외 분석법을 응용하여 참깨의 원산지 판별 가능성을 조사한 결과, 한국산, 중국산 및 일본산 참깨를 판별할 수 있었으며, 필터형 근적외 분석장치로도 연구용 장치와 비슷한 정확도로 참깨의 원산지를 판별할 수 있었다. 중국산 참깨가 혼합된 경우 혼입율은 10% 오차 범위에서 측정할 수 있었다. 근적외 분광분석법에 의해 참깨의 원산지가 판별되는 근거는 참깨의 기름성분 보다는 참깨박 성분에 그 요인이 있는 것으로 판단되었다.

Near-infrared Spectroscopy of Metal-enriched Supernova Ejecta in Cassiopeia A

  • The supernova remnant Cassiopeia A (Cas A) provides a unique opportunity to observe the fine details of the explosion of core-collapse supernova (SN). Previous optical and near-infrared (NIR) observations of Cas A have shown that the spatial distribution of the metal-enriched SN ejecta is very complicated, indicating that the SN explosion should have been asymmetric and turbulent, especially near the core. Recently, we obtained a long-exposure (~10 hr) image of Cas A by using the UKIRT 3.6-m telescope with a narrow-band filter centered at [Fe II] 1.644 um emission. This 'deep [Fe II] image' provides an unprecedented panoramic view of Cas A, revealing the distribution of dense SN ejecta over the entire remnant. We have carried out NIR multi-object spectroscopic observations of the dense ejecta knots in the northeastern (NE) and eastern (E) outer regions of the remnant using the MMIRS attached on the MMT 6.5-m telescope. A total of 67 ejecta knots are detected. By analyzing their spectra, we have found that the knots in the NE area show strong [S II]/[S III] and [Fe II] lines but little or no [P II] line, while those in the E outer region show strong [Fe II] lines but weak [S II]/[S III] lines. In this talk, we present the preliminary results of our NIR spectroscopic observations and discuss the implications.

Prediction of the content of white clover and perennial ryegrass in fresh or dry mixtures made up from pure botanical samples, by near infrared spectroscopy

  • Pasture composition, an important attribute determining sward condition and value, is normally assessed by hand separation, drying and measuring weight contribution of each species in the mixture. This is a tedious, time and labour consuming procedure. NIRS has demonstrated the potential for predicting botanical composition of swards, but most of the work has been carried out on dry samples. The aim of this work was to evaluate the feasibility of developing NIR models for predicting the white clover and ryegrass content in fresh or dry mixtures artificially prepared from pure samples of both species. Mixtures from pure stands of white clover(Trifolium repens) and perennial ryegrass (Lolium perenne) were prepared with different proportions (0 to 100%) of each species (fresh weight). A total of 55 samples were made (11 mixtures,5 cuts). Spectra (400 to 2500 nm) were taken from fresh chopped (rectangular cuvettes, transport sample module) samples, in a NIR Systems 6500 scanning monochromator controlled by the software NIRS 3 (Infrasoft International), which was also utilized for calibration development. Different math treatments (derivative order, subtraction gap and smooth segment) and a scatter correction treatment of the spectra (SNV and Detrend) were tested. Equations were developed by modified partial least squares. Prediction accuracy evaluated by cross-validation, showed that percentage of clover or ryegrass, as contribution in dry weight, can be successfully percentage of clover or ryegrass, as contribution in dry weight, can be successfully predicted either on fresh or dried samples, with equations developed by different math treatments. Best equations for fresh samples were developed including a first, second, or third derivative, whereas for dry samples best equations included a second or third derivative. Standard errors of ross validation were about 6% for fresh and 3.6% for dry samples, Coefficient of determination of cross validation (1-VR) were over 0.95 times the value of SECV for fresh samples and over 8 times the value of SECV for dry samples. Scatter correction (SNV and Detrend) in general improved prediction accuracy. It is concluded more precise on dried and ground samples, it can be used with an acceptable error level and less time and labour, on fresh samples.

  • The amount of water for the cultivation of citrus is different based on the growing period. The water content in the leaves of citrus can be a index for watering during cultivation. The purpose of this study is to determine non-destructively the water content of Citrus leaves by using near infrared spectroscopy (NIRS). Citrus leaves were prepared from satsuma mandarin leaves (Citrus unshiu Marc. var. okitsu) ranging from 62.20 to 69.98% of water content by loss on drying, NIR reflectance spectra of Citrus leaves were acquired by using a fiber optic probe.
