Functional Expression of the Internal Rotenone-Insensitive NADH-Quinone Oxidoreductase (NDI1) Gene of Saccharomyces cerevisiae in Human HeLa Cells

  • Many studies propose that dysfunction of mitochondrial proton-translocating NADH-ubiquinone oxidoreductase (complex I) is associated with neurodegenerative disorders, such as Parkinson's disease and Huntington's disease. Mammalian mitochondrial proton-translocating NADH-quinone oxidoreductase (complex I) consists of at least 46 different subunits. In contrast, the NDI1 gene of Saccharomyces cerevisiae is a single subunit rotenone-insensitive NADH-quinone oxidoreductase that is located on the matrix side of the inner mitochondrial membrane. With a recombinant adeno-associated virus vector carrying the NDI1 gene (rAAV-NDI1) as the gene delivery method, we were able to attain high transduction efficiencies even in the human epithelial cervical cancer cells that are difficult to transfect by lipofection or calcium phosphate precipitation methods. Using a rAAV-NDI1, we demonstrated that the Ndi1 enzyme is successfully expressed in HeLa cells. The expressed Ndi1 enzyme was recognized to be localized in mitochondria by confocal immunofluorescence microscopic analyses and immunoblotting. Using digitonin-permeabilized cells, it was shown that the NADH oxidase activity of the NDI1-transduced HeLa cells were not affected by rotenone which is inhibitor of complex I, but was inhibited by flavone and antimycin A. The NDI1-transduced cells were able to grow in media containing rotenone. In contrast, control cells that did not receive the NDI1 gene failed to survive. In particular, in the NDI1-transduced cells, the yeast enzyme becomes integrated into the human respiratory chain. It is concluded that the NDI1 gene provides a potentially useful tool for gene therapy of mitochondrial diseases caused by complex I deficiency.

  • Mitochondria diseases have been reported to involve structural and functional defects of complex I-V. Especially, many of these diseases are known to be related to dysfunction of mitochondrial proton-translocating NADH-ubiquinone oxidoreductase (complex I). The dysfunction of mitochondria complex I is associated with neurodegenerative disorders, such as Parkinson's disease, Huntington's disease, and Leber's hereditary optic neuropathy (LHON). Mammalian mitochondrial proton-translocating NADH-quinone oxidoreductase (complex I) is largest and consists of at least 46 different subunits. In contrast, the NDI1 gene of Saccharomyces cerevisiae is a single subunit rotenone-insensitive NADH-quinone oxidoreductase that is located on the matrix side of the inner mitochondrial membrane. The Saccharomyces cerevisiae NDI1 gene using a recombinant adeno-associated virus vector (rAAV-NDI1) was successfully expressed in AML12 mouse liver hepatocytes and the NDI1-transduced cells were able to grow in media containing rotenone. In contrast, control cells that did not receive the NDI1 gene failed to survive. The expressed Ndi1 enzyme was recognized to be localized in mitochondria by confocal immunofluorescence microscopic analyses and immunoblotting. Furthermore, these results indicate that Ndi1 can be functionally expressed in the AML12 mouse liver hepatocytes. It is conceivable that the NDI1 gene is powerful tool for gene therapy of mitochondrial diseases caused by complex I deficiency. In the future, we will attempt to functionally express the NDI1 gene in mouse embryonic stem (mES) cell.

  • Using the fatigue limit (△σunsm) and residual stress (σr) of the UNSM smooth specimen, the harmless maximum crack depth (αhml) according to the crack aspect ratio (As) was evaluated. αhml evaluated the reliability in the relationship between the minimum crack depth (αNDI1, αNDI2) detectable by nondestructive inspection(NDI) and the crack depth (α25, α50) that reduces the fatigue limit by 25% and 50%. All αhml was determined by the crack depth. The αhml of 80N UNSM with high σr and high △σunsm was found to be large. σr in the depth direction had a much effect on αhml. Since αhml50(As=0.6-0.1) and αhml25, α25 and α50(some range) can secure the safety and reliability. Since αhml50 (As=1. 0-0. 6), it cannot be made harmless by UNSM. So safety and reliability cannot be secured. αNDI1 and αNDI2 are larger than α25, α25 cannot be detected by NDI. αNDI1 and αNDI2 are smaller than α50, α25 can detected by NDI.

Anti-Arthritic and Analgesic Effect of NDI10218, a Standardized Extract of Terminalia chebula, on Arthritis and Pain Model

  • The fruit of Terminalia chebula Retzius has been used as a panacea in India and Southeast Asia but its biological activities have not been fully elucidated. Here we report anti-arthritic and analgesic effect of NDI10218, a standardized ethanol extract of Terminalia chebula, on collagen-induced arthritis and acetic acid-induced writhing model, respectively. Arthritis was induced in DBA/1J mice by immunizing bovine type II collagen and mice were treated with NDI10218 daily for 5 weeks after the onset of the disease. NDI10218 reduced the arthritis index and blocked the synovial hyperplasia in a dose-dependent manner. The serum levels of pro-inflammatory cytokines TNF-${\alpha}$, IL-6, and IL-$1{\beta}$ were significantly reduced in mice treated with NDI10218. Production of the inflammatory IL-17, but not immunosuppressive IL-10, was also inhibited in splenocytes isolated from NDI10218-treated arthritis mice. Administration of NDI10218 markedly decreased the number of T cell subpopulations in the regional lymph nodes of the arthritis mice. Finally, NDI10218 reduced the number of abdominal contractions in acetic acid-induced writhing model, suggesting an analgesic effect of this extract. Taken together, these results suggest that NDI10218 can be a new therapeutic candidate for the treatment of rheumatoid arthritis.

  • Using the fatigue limit (∆σunsm) and residual stress (σr) of the UNSM smooth specimen, the harmless maximum crack depth (ahlm) according to the crack aspect ratio (As) was evaluated. In addition, the relationship between the minimum crack depth (aNDI1, aNDI2) detectable by non-destructive inspection(NDI), the crack depth (a25, a50) that reduces the fatigue limit by 25% and 50%, and ahlm were evaluated. The harmless crack condition was determined by the deepest crack point (point A). Since ahlm is larger than a25 and a50, a25 and a50 can secure the safety and reliability of steel via UNSM. Because aNDI1 and aNDI2 are larger than a25 and a50, cracks in a25 and a50 cannot be detected by non-destructive testing. Therefore it is necessary to apply more precise NDI.

경항부 통증과 심리적 상태와의 상관성에 관한 연구 (The relationship between neck pain and psychological state in female office workers.)

  • Objectives : The purpose of this study was to investigate the relationship between pain and psychological state in female office workers with neck pain. Methods : Experimental group of 31 healthy subjects complained of neck and arm discomfort related to computer use which lasted more than 3 months in the past year and was present in the past 7 days as well as on the day of test. Outcomes were assessed by meridian-electromyography(MEMG), Neck Disability Index(NDI), Visual Analog Scale(VAS), Beck Depression Inventory(BDI), Stress Reaction Inventory(SRI), Holmes & Rahe Social Readjustment Rating Scale(SRRS). Results : The NDI score and BDI score had a correlationship(r=0.48), and the NDI score and SRI score also had a correlationship(r=0.48), significantly. The NDI score and contraction power of upper trapezius by MEMG had a correlationship significantly, but with the other muscles the NDI score didn't have correlationships. The NDI score and the SRI score were significantly higher in depression group(BDI score ${\circ}{\surd}$14). The contraction power of trapezius by MEMG was significantly lower in depression group. Conclusions : The results suggest that mental stress can be a major risk factor of neck pain in female office workers.

교통사고 후 발생한 편타 손상 환자에 대한 신바로약침과 중성어혈약침의 치료효과 비교연구 (A Retrospective Study on the Effects of ShinBaro Pharmacopuncture and Jungsongouhyul Pharmacopuncture on Whiplash Injury by Traffic Accident)

  • Objectives The purpose of this study is to compare the effects of ShinBaro pharmacopuncture and Jungsongouhyul pharmacopuncture on whiplash injury by traffic accident. Methods This study was carried out on 30 patients who received treatment in Jaseng Hospital of Korean Medicine. 30 patients were divided ShinBaro pharmacopuncture group and Jungsongouhyul pharmacopuncture group. Visual analog scale (VAS) and neck disability index (NDI) were compared after treatment. Results 1) Both the ShinBaro group and Jungsongouhyul group showed significant improvement in the visual analog scale (VAS) and neck disability index (NDI) after 3 weeks of treatment. 2) The VAS and NDI of the ShinBaro group decreased gradually with treatment. The VAS scores showed significant improvement up to week 2, but although week 3 showed further improvement compared to week 2, the difference did not reach statistical significance. The NDI scores showed significant improvement consistantly throughout the treatment period. 3) The VAS and NDI of the Jungsongouhyul group decreased with treatment also. The VAS scores significantly improved up to week 2, but though week 3 showed further improvement in comparison to week 2, the difference did not reach statistical significance. The NDI scores showed significant improvement consistantly throughout the treatment period. 4) Although the ShinBaro group showed a swifter decline than the Jungsongouhyul group in both VAS and NDI scores, the difference was not statistically significant. Conclusions We found out that ShinBaro pharmacopuncture group and Jungsongouhyul pharmacopuncture group is effective and useful in whiplash injury by traffic accident. And, further studies will be needed.

대학생의 스마트폰 이용 시간과 경항통과의 관련성 연구 (The Correlation Analysis between Hours of Smartphone Use And Neck Pain in the Gachon University Students)

  • Objectives : The purpose of this study was to analysis the relationship between hours of smartphone use and neck pain in university students. Methods : A survey of 2,353 university students was conducted in Gyeonggi province and Incheon city using a self report questionnaire from April 1 to 11 2013. The questionnaire of questions regarding the hours of using smartphone, mainly used function in participant's smartphone and neck discomfort degree. Neck disability index(NDI) was used to evaluate degree of pain. The collected data were analyzed with the t-test and Kendall's tau test using the SPSS 21.0 program and R 3.1.0, respectively. Results : 1. Almost all of the survey participants(99.07 %) were smartphone users. 66.97 % of them answered that they use their smartphone daily more than 2 hours and 48.18 % of them answered that they use their smartphone 10 to 30 minutes everytime they use it. 2. The overall distribution of NDI scores was 'no disability(0~4 score, 62.92 %)', 'mild disability(5~14 score, 32.85 %)', 'moderate disability(15~24 score, 1.19 %)'. As a result of t-test, we found that the average NDI score for female students was significantly higher than the average NDI score for male students(p<0.05). 3. The Kendall's tau test revealed that total time spent daily using smartphones has a strong positive correlation with 'pain intensity', 'lifting', 'reading' 'headache', 'concentration', and 'driving'(p<0.05) in NDI. Also, time duration of one time smartphone usage has a strong positive correlation with 'pain intensity', 'lifting', 'reading', 'headache', 'concentration', 'work', and 'recreation'(p<0.05). 4. The Kendall's tau test revealed that total NDI scores have a significantly strong positive correlation with both of total time spent daily using smartphones(p<0.05) and time duration for one time smartphone usage(p<0.05). This results imply that long-time use of smartphone has a strong relationship with neck pain. Conclusions : The results of the study would be a good starting point for future studies to reduce the risks of chronic neck pain caused by smartphone usage.

주성분 분석을 활용한 자연가뭄지수 산정 및 평가 (Estimation and assessment of natural drought index using principal component analysis)

  • 본 연구에서는 인위적 수공시설물을 고려하지 않는 자연상태의 가뭄해석을 위해 자연가뭄지수(NDI)를 산정하고 활용성을 평가하였다. 자연가뭄지수는 주성분 분석을 이용하여 산정하였으며, 입력자료는 3개월 누적강수량, 자연유량, 토양수분량 자료이다. 강수량은 ASOS 59개 지점 자료이고, 자연유량 및 토양수분량은 지표수문해석모형의 결과를 이용하였다. 가뭄지수의 평가기간은 1977~2012년이며, 활용성 평가를 위해 시계열 분석, 지역별 분석 및 가뭄특성인자 분석을 수행하였다. 시계열 분석결과, 자연가뭄지수는 가뭄의 시작과 끝에 대한 평균절대오차는 0.85로 가장 정확하게 나타났다. 과거 가뭄사례와 가뭄특성인자를 분석한 결과, 자연가뭄지수의 지속기간과 발생간격의 평균절대오차는 각각 1.3, 1.0으로 가뭄의 지속기간 및 발생간격을 적절히 반영하는 것을 확인하였다. 지역별 분석결과, 자연가뭄지수는 단일변량 가뭄지수의 지역별 가뭄상황을 적절히 반영하여 활용성이 높은 것으로 나타났다. 또한, 가뭄의 시작, 끝, 지속기간, 발생간격에 대한 자연가뭄지수, 표준강수지수, 표준유출지수, 표준토양수분지수의 순위를 분석한 결과 자연가뭄지수가 가장 높은 순위로 산정되었다. 자연가뭄지수는 기존 단일변량 가뭄지수의 상이한 가뭄상황을 종합적으로 반영하며, 가뭄의 특성을 정량적으로 제시한다는 점에서 활용성이 높다고 판단된다.

임상실습 학생을 대상으로 한 위전도검사와 Nepean Dyspepsia Index(NDI) 설문검사와의 연관성 연구 (Correlation Study between Electrogastrography and Nepean Dyspepsia Index (NDI) in Clinical Practice Students)

  • Objective: The aim of this study was to find a correlation between the Nepean Dyspepsia Index (NDI) and electrogastrography (EGG) in clinical practice students. Methods: In total, 28 students were enrolled. NDI and EGG were measured in all students. NDI was classified as abnormal when the score was 11 or more. In EGG, standard 1 was classified as abnormal if the preprandial or postprandial percentage of normal gastric slow waves of Ch3 was less than 70% or if power ratio of Ch3 was less than 1. Standard 2 was classified as abnormal if standard 1 was met or if the percentage of normal gastric slow waves of Ch3 decreased after meals. Results: NDI and EGG had no statistically meaningful correlation when standard 1 was used, but NDI and EGG had statistically meaningful correlation when standard 2 was used. Conclusions: These findings suggest that NDI and EGG are correlated and that EGG can be used to evaluate the degree of dyspepsia.
