APPLICATION OF RANDOMLY AMPLIFIED POLYMORPHIC DNA(RAPD) ANALYSIS METHOD FOR CLASSIFICATION AND BREEDING OF THE KOREAN GINSENG

  • Korean ginseng has been widely used as medicine from ancient times in Asia. Current breeding efforts in Korea include the individual plant selection and the subsequent pure - line isolation, and considerable number of lines with desirable traits have thus been isolated. However, there were rare data on genetic maker and its analysis for selection of superior varieties. For taxonomic characterization and development of genetic markers for ginseng breeding, molecular biological methods including the RFLP and RAPD methods were applied. Cytoplasmic DNA of ginseng was analyzed for RFLP analysis. However. there is no different pattern among the chloroplast DNA or mitochondrial DNA of variants. In the case of RAPD analysis, the band patterns using 4 of 10 RAPD primers show the distinctive polymorphism among 9 ginseng variants, and lines, and Similarity Index(SI) on polymorphism was calculated for the extent and nature of these variabilities in ginseng. The sequences of 4 selected primers were TGCCGAGCTG, AATCGGGCTG. GAAACGGGTG, and GTGACGTAGG. By SI based on the polymorphic band patterns, Chungkyung - Chong and Hwangskoog - Chong, and JakyungChong 81783 and Jinjakyung of Russia showed the most close SI. The data of KG10l coincided with the fact that it was released from Hwangskoog - Chong. and Jakyung - Chong 81783 and Jinjakyung of Russia showed the most close SI. The data of KG101 coincided with the fact that it was released from Hwangskoog - Chong by breeding process. The data of Jakyung strains indicated the significant variation among the strains. From these results, RAPD analysis method could be succesively applied to the classification and genetic analysis for breeding of Korean ginseng.

Efficacy of two commonly used potentized homeopathic drugs, Calcarea carbonica and Lycopodium clavatum, used for treating polycystic ovarian syndrome (PCOS) patients: II. Modulating effects on certain associated hormonal levels

  • In view of greater attention given to the incidence of Polycystic Ovarian Syndrome (PCOS) in women of reproductive age, particularly in urban and semi-urban population in India, research works in both the regimens of orthodox and complementary and alternative medicines have been rejuvenated in recent years. We report here relative efficacy of two potentized homeopathic remedies, Calcarea carbonica (Calc) and Lycopodium clavatum (Lyco) used traditionally for the removal of ovarian cysts. These drugs are most frequently used based on guiding symptoms of individual patients. Effects of either of these remedies on its ability of removing cysts, along with amelioration of certain other hormones and hormone-related parameters of PCOS, such as follicle stimulating hormone, luteinizing hormone, Estradiol, Testosterone (Free/Total), Dehydroepiandosterone, Prolactine, Progesterone (17- Hydroxyprogesterone), TSH including T3, T4, and Insulin were studied. The Insulin-related parameters like changes in fasting or postprandial glucose levels were also studied. The mentioned hormones play some- direct or indirect roles in causing irregular menstrual cycle and PCOS. The data collected at three fixation time points, namely, at 6, 12, and 18 months were considered. Results showed that out of 40 patients initially having PCOS, cysts were totally removed in 21 patients along with amelioration of other relevant symptoms. Both Calc and Lyco had amelioration of similar nature. Results of this study therefore validate safe and effective use of both Calc and Lyco in homeopathy, to patients with basic guiding symptoms for either drug, and can be recommended for patients with PCOS as they do not have any reported side-effects.

Ultrastructural Study on the Development of Notochordal Cells in Nucleus Pulposus of Human Fetuses (인태아(人胎兒) 수핵(髓核) 발육(發育)에 관(關)한 전자현미경적(電子顯微鏡的) 연구(硏究))

  • The development of notochordal cells of nucleus pulposus was studied with electron microscope in human fetuses ranging from 30 mm to 260 mm crown-rump length. At 30 mm fetus, primitive notochordal cells were large with central nucleus, few organelles, and their cytoplasm usually contained dense glycogen and fine filaments. Notochordal cells at all ages contained bundles of fine filaments of indeterminate nature. One unusual feature of fetal notochordal cells was the consistent presense of rough endoplasmic reticulum surrounding poorly developed mitochondria. At 50 mm fetus, notochordal cells formed dense masses with interdigitating cell membranes connected by a variety of cell to cell junctions. With increasing age, the cell connections became slender threaded cytoplamic extending from cell and enclosed large extracellular space. Chondrocyte-like cells appeared to be separated by large volumes of extracellular matrix. Viable notochordal and condrocyte-like cells existed in specimen from all age. The extracellular spaces were filled with fibrillar and granular material by 90 mm fetus. Necrotic cells were distinguished by loss of their membrane integrity, vacuolization of their organelles, and the presence of dense osmiophilic masses. In adult tissue, notochordal cells became rounded or irregular in shape and developed a pericellular matrix consisting of collagen fibrile, and dense particle. The structure of notochordal cells and their persistance in the nucleus pulposus after fetal life suggested that they may have a significant role in the formation and maintenance of the nucleus pulposus. The presence of Golgi complex and well-developed endoplasmic reticulum in chondrocyte-like cells suggested that they are capable of producing and maintaining the extracellular matrix.

Dose coefficients of mesh-type ICRP reference computational phantoms for external exposures of neutrons, protons, and helium ions

  • Recently, the International Commission on Radiological Protection (ICRP) has developed the Mesh-type Reference Computational Phantoms (MRCPs) for adult male and female to overcome the limitations of the current Voxel-type Reference Computational Phantoms (VRCPs) described in ICRP Publication 110 due to the limited voxel resolutions and the nature of voxel geometry. In our previous study, the MRCPs were used to calculate the dose coefficients (DCs) for idealized external exposures of photons and electrons. The present study is an extension of the previous study to include three additional particles (i.e., neutrons, protons, and helium ions) into the DC library by conducting Monte Carlo radiation transport simulations with the Geant4 code. The calculated MRCP DCs were compared with the reference DCs of ICRP Publication 116 which are based on the VRCPs, to appreciate the impact of the new reference phantoms on the DC values. We found that the MRCP DCs of organ/tissue doses and effective doses were generally similar to the ICRP-116 DCs for neutrons, whereas there were significant DC differences up to several orders of magnitude for protons and helium ions due mainly to the improved representation of the detailed anatomical structures in the MRCPs over the VRCPs.

  • Hrp1, of Schizosaccharomyces pombe, is a new member of the SW12/SNF2 protein family that contains a chromodomain and a DNA binding domain as well as ATPase/7 helicase domains. This configuration suggests that Hrp1 could be a homolog of mouse CHD1, which is thought to function in altering the chromatin structure to facilitate gene expression. To understand the enzymatic nature of Hrp1 we purified the 6-Histidine-tagged Hrp1 protein (6$\times$His-Hrp1) to homogeneity from a S. pombe Hrp1-overexpressing strain and hen examined its biochemical properties. We demonstrate that the purified 6$\times$His-Hrp1 protein exhibited a DNA-binding activity with a moderate preference to the (A+T)-rich tract in double-stranded NA via a minor groove interaction. However, we failed to detect any intrinsic DNA helicase activity from the purified Hrp1 like other SW12/SNF2 proteins. These observations suggest that the DNA binding activities of Hrp1 may be involved in the remodeling of the chromatin structure with DNA-dependent ATPase. We propose that Hrp1 may function in heterochromatins as other proteins with a chromo- or ATPase/helicase domain and play an important role in the determination of chromatin architecture.

A Research on Li Dong-yuan's Application Rule of Wind Herbs(風藥) (이동원(李東垣)의 풍약(風藥) 활용법(活用法)에 대한 고찰)

  • Objectives : The paper studies the epistemology of Wind Herbs, its origin, its drug category and medicinal property, attempted to understand Lidongyuan's framework of mechanism of internal damage and treatment principle, and why he used it from his viewpoint. Methods : His suggestion was based on this treatment of internal damage suing wind herbs. Therefore, it cannot be viewed as a simple herbological concept. It rather displays the characteristics of medication based on clinical pathology. Therefore, wind herb should be comprehensively understood from the understanding of the mechanism of internal damage. That is why the paper studied around Piweilun, where Lidongyuan's concept on internal damage is established with finality. Additionally, the paper also referred to Wanghaogu's Tangyenbencao, a text that comprehensibly assembles the authors of Zhangyuans, Yixuqiyan's and Yishuixuepi's knowledge o herbology. Results : The origin of wind herb is 'herbs that uses the unique nature of wind in treatment of disease.' Medication unfolds yang qi, and this signifies the Shengyang function. This means that it starts from the lower energizer yin aspect, which is the beginning point of yang qi, and unfolds to the whole body through upbearing and effusion. Instead of producing yang qi directly similar to pungent, sweet and warm formula, however, it contributes to the achievement of the final purpose of the way of Shengyang through forming a ascending mechanism in the whole body via yin aspect's yang qi upbearing and effusion. Conclusions : Wind herb is within the scope of clinical herbology selected by Lidongyuan, for the purpose of achieving comprehensive clinical purposes, in order to treat internal damage. In this way, wind herb is distinguished from the application of other medicines that are limited in usage depending on Qiwei.

The Effect of a Community-Based Self-Management Program for Patients at Thyroid Cancer-Diagnosis Stage : a Pilot Study (재가 갑상선암 환자를 위한 지역암센터 자가관리프로그램 중재 효과에 대한 예비연구)

  • Purpose: This study was conducted to examine the effectiveness of a self-management program on patients with thyroid cancer, particularly during the time of waiting for surgery after cancer diagnosis. Psychological distress, biological responses of immune cell counts, and quality of life were the variables of this study. Methods: One group pre-post test design was used with the nature of a pilot study. Ten newly diagnosed thyroid cancer patients were recruited through physicians' referrals. After drop out of 4 participants, final data were collected from six participants. Small group technique, a one and half hour-session per week for one month (total 4 sessions, 6 hours) was used. Relaxation techniques, meditation training, and strategies to reduce distress were provided by researchers. Standardized questionnaires and an established bio-assay were used for collection of data. Results: Participants showed significant lowering of psychological distress (p<.05) and improvement in global quality of life (p<.05). Biological responses of immune cell counts did not show statistical significance. Conclusion: The self-management program may reduce psychological distress and improve quality of life of patients with thyroid cancer between the time of diagnosis and surgery. The self-management program would be a valuable approach for patients with an unexpected cancer diagnosis to prepare for their disease experience in a community setting.

Comparison of Anti-inflammatory effects between Artemisia capillaris and Artemisia iwayomogi by extraction solvents (인진호(茵蔯蒿)와 한인진(韓茵蔯)의 추출용매별 항염증 효능 비교)

  • Objectives : Artemisia capillaris Thunberg (AC) and Artemisia iwayomogi Kitamura (AI) have been used without distinguishment since ancient times due to similar appearance. In this study, we compared the inhibitory effects of AC and AI on the expression of inflammatory cytokines induced by lipopolysaccharide (LPS) in murine macrophages. Methods : AC and AI were extracted by reflux with distilled water (DW) and 70% ethanol (EtOH). We investigated the inhibitory effects of AC and AI on the expression of nitric oxide (NO), inducible NO synthase (iNOS) and tumor necrosis $factor-{\alpha}$($TNF-{\alpha}$) induced by LPS in macrophages. Results : Firstly, yield of the samples was higher in order of Artemisia iwayomogi DW Extract (AID), Artemisia iwayomogi 70% EtOH Extract (AIE), Artemisia capillaris DW Extract (ACD) and Artemisia capillaris 70% EtOH Extract (ACE). All of the samples were not toxic in macrophages. The inhibitory effect of the samples on LPS-induced NO expression was stronger in the order of AIE, ACE, AID and ACD. The inhibitory effect of the samples on LPS-induced inducible iNOS expression was stronger in the order of AIE, ACE and AID. Effect of ACD was same with that of AID. In addition, inhibitory effect of the samples on LPS induced $TNF-{\alpha}$expression wes stronger in the order of AIE, ACE, AID and ACD. Conclusion: These results showed that AI would be more effective than AC and 70% EtOH would be more effective than DW as an extraction solvent in inflammatory diseases.

Urological Evaluation of Tethered Cord Syndrome

  • To describe how to perform urological evaluation in children with tethered cord syndrome (TCS). Although a common manifestation of TCS is the development of neurogenic bladder in developing children, neurosurgeons often face difficulty in detecting urological problems in patients with TCS. From a urological perspective, diagnosis of TCS in developing children is further complicated due to the differentiation between neurogenic bladder dysfunctions and transient bladder dysfunctions owing to developmental problems. Due to the paucity of evidence regarding evaluation prior to and after untethering, I have shown the purpose and tools for evaluation in my own practice. This may be tailored to the types of neurogenic bladder, developmental status, and risks for deterioration. While the urodynamic study (UDS) is the gold standard test for understanding bladder function, it is not a panacea in revealing the nature of bladder dysfunction. In addition, clinicians should consider the influence of developmental processes on bladder function. Before untethering, UDS should reveal synergic urethral movement, which indicates an intact sacral reflex and lack of TCS. Postoperatively, the measurement of post-void residual urine volume is a key factor for the evaluation of spontaneous voiders. In case of elevation, fecal impaction, which is common in spinal dysraphism, should be addressed. In patients with clean intermittent catheterization, the frequency-volume chart should be monitored to assess the storage function of the bladder. Toilet training is an important sign of maturation, and its achievement should be monitored. Signs of bladder deterioration should be acknowledged, and follow-up schedule should be tailored to prevent upper urinary tract damage and also to determine an adequate timing for intervention. Neurosurgeons should be aware of urological problems related to TCS as well as urologists. Cooperation and regular discussion between the two disciplines could enhance the quality of patient care. Accumulation of experience will improve follow-up strategies.

Prescription Patterns and Factors Related to the Number of Medications in Chronic Obstructive Pulmonary Disease in Non-elderly Adults (비고령 성인환자의 만성폐쇄성폐질환 약물사용 현황과 영향인자)

  • Background: This study is to investigate the prescription patterns and factors related to the number of medications treating chronic obstructive pulmonary disease (COPD) in patients under 65 years old according to GOLD guidelines. Methods: We retrospectively analyzed the medical records of patients aged 40-64 years with a diagnosis of COPD from January to March 2016. Patients were classified by combined assessment of COPD (grades A, B, C, D) using spirometry, exacerbation history, mMRC, and/or CAT results. We analyzed prescribed medications, treatment options and factors related to the numbers of COPD medications. Results: The total number of prescriptions were 251. About 35.5% of patients were classified as GOLD A, 34.2% as GOLD B, 17.1% as GOLD C and 13.2% as GOLD D. Inhaled bronchodilator was prescribed for 86.9% of patients and the most frequent COPD medication was long-acting muscarinic antagonist (LAMA) followed by inhaled corticosteroids/long acting beta agonist (ICS/LABA). The majority of low risk patients (GOLD A/B) were prescribed a monotherapy with LAMA or LABA. For high risk patients (GOLD C/D), combination treatment with ICS+LAMA+LABA was mostly prescribed. The 21.2% of patients in GOLD D received systemic corticosteroid. The average number of medications per prescription was 3.7, and this number increased with increasing COPD grade, COPD duration and lung function reduction ($FEV_1$, $FEV_1/FVC$). Conclusion: Generally high adherence to GOLD guideline recommendations was reported. Given the progressive nature of the disease, results suggest that closer attention to respiratory symptoms for early detection, diagnosis, and appropriate treatment of COPD is warranted.
