Objective: A study was made investigate factors affecting body surface temperature changes after weaning in sows, whether these can be used to aid detection of natural estrus and how they relate to subsequent reproductive performance. Methods: A total of 132 sows were selected during summer from a breeding farm, with mean parity of 3.6±2.3 and 28.5±0.9 days lactation length. Four daily measurements (6:00, 8:00, 16:00, and 18:00) of vulva (VST), udder (UST), ear base and central back skin temperatures for individual sows were taken by an infrared thermometer, continuing up to 8 days post weaning. Results: The VST obtained from sows showing estrus at 4 days post-weaning (4DPW), 5DPW, and 6DPW showed a peak at the fourth day post-weaning, but then started to decrease. The VST of sows not detected in standing heat (NDPW) remained at a lower level during the experiment, but UST was increased soon after weaning. The VST-UST temperature differences during daytime of sows that were showing behavioural standing heat on 4DPW, 5DPW, 6DPW, and 7DPW were 0.46℃±0.123℃, 0.71℃±0.124℃, 0.66℃ ±0.171℃, and 0.58℃±0.223℃, respectively. The NDPW sows had the highest UST observed, but also the lowest VST so that a more negative value of temperature difference (-0.31℃) was seen during first few days post-weaning. A total of 119 sows were observed to show standing heat and were bred. The later the estrus, the smaller the litter size (p = 0.005). Conclusion: Sows which did not show behavior indicative of stable standing heat after weaning had a VST which remained at a lower level, but the UST increased soon after weaning. Therefore, for sow heat detection under field conditions, the changes of VST and UST and difference between the two should be considered together to increase the accuracy of detection.
Toxicity tests in our laboratory are conducted usually with mass-reared organisms. They are under the same environmentel conditions throughout seasons and are supplied at specific age. A total of 38 species of aquatic organisms are being reared. We have attempted to establish pruified strains or to select clones of various parthenogenic organisms. The merits or demerits of our culturing test organisms are discussed. The differences in the susceptibility among clones or strains of test organism are also discussed. For a single species test, algae, daphnia, fish are often used. However, we usually use early stages, but occasionally, adults fish are used for reproduction tests. As an another important aspect, the toxicity through food chains has been studied. In this study, we select a pair of species belonging different trophic levels. The differences between single species tests and multispecies tests will be discussed. Even a single species test intends to assess the effects of chemicals on ecosystem levels, however, this idea is not applicable to ecosystems. Single species tests with standard organisms and multispecies tests are contradictory in concept. One type of multispecles tests is indoor microcosms being composed of severel species artificially assembled, and another is composed of natural components (both indoor and outdoor). We have used three types of outdoor mesocosms using ponds and three types of artificial streams. The mesocosms is useful to not only to analyze the floral or faun61 changes but also to study the fate or behaviour of chemicals in naturd environments. Lastly, usefulness of the field observation or experiments or semi-field experiments will be discussed. This will enhance the exploitation of early warning systems utilizing indicator organisms or animal behaviour.aviour.
This study was to examine whether the vitrified, one-step diluted and direct transferred Hanwoo IVM/IVF/IVC blastocysts can be successfully survived in vivo and they were succeeded into the live birth. For vitrification, blastocysts were serially exposed in glycerol (G) or/and ethylene glycol (EG) mixtures [10% (v/v) G for 5 min, 10% G plus 20% EG (v/v) for 5 min, and 25% G plus 25% EG (v/v) for 30 sect] which is diluted in 10% FBS added D-PBS. Thawing of straw was carried out in air for 10 sec and then in water bath of $25^{\circ}C$ for 20 sec. One-step dilution within the straw was done in water bath of $25^{\circ}C$ for 1 min. Vitrified and one-step diluted embryos were directly transferred into 36 (natural or hormone induced synchronized) recipient cows in 6 areas of Kyungsang Buk-Do. Pregnancies were confirmed at first when recipient cows did not return to the subsequent estrus cycle, and later by manual palpation per rectum on day 45, 90 and then living calves were derived into parturition. Overall pregnancy was 33.3%(12/36), However, higher pregnancy was obtained when the recipients exhibited estrus one day earlier than the age of transferred embryos (53.3 vs 25.0-27.3%), irrespective of synchronization methods. Also, parous recipients became pregnant higher than nulliparous heifers, And, there were not different in pregnancy rates by the aspect of corpus luteum (CL) quality of recipients (good, 29.4; fair, 37.5; poor, 33.3%). One hundred eight of frozen-thawed Hanwoo blastocysts were directly transferred into 36 recipient cows. In 12 of pregnant cows, 3 cows were aborted and 9 cows were calved [single, 66.7% (6/9): twin, 33.3% (3/9)]. Total embryo implantation rate was 11.1% (12/108). However, 9 Hanwoo calves were lived. Therefore, these results demonstrate that direct transfer technique of vitrified and one-step diluted bovine blastocysts can be applied easily and effectively with the higher pregnancy rate on field trial without the equipment and embryological skills.

Production of transgenic animals for studying specific gene has been limited due to a low efficiency, lack of skilled researchers and the need for expensive equipment. Currently, the boar spermatozoa as a vector to deliver exogenous DNA into the oocyte were used to improve the efficiency of transfection rate. In this study, we revealed that the optimal conditions for DNA uptake in spermatozoa by liposome were to 90 min of incubation, $17^{\circ}C$, $10^5$ spermatozoa, 4 ng/ml of exogenous DNA and 0.5% (v/v) liposome, without damage to fertility. In addition, the developmental rate to the blastocyst stage of embryo in control group was significantly higher than those embryos with exogenous DNA and liposome, whereas there were no significant differences in embryo development between the liposome and type of DNA. The transfection rates of embryo using treated spermatozoa with both liposome and circular DNA were higher than those using linear DNA. These findings raise the possibility thattreated spermatozoa with liposome/DNA complexes could be used in in vitro fertilization, and the exogenous DNA transferred into the oocytes. Taken together, we demonstrated that liposome a vector for the uptake of exogenous DNA in boar spermatozoa could improve the efficiency of sperm-mediated gene transfer in creating transgenic pig and the other domestic transgenic animals.
The present study was undertaken to identify changes of plasminogen activators (PAs) in porcine oviductal epithelial cells (POECs) during the estrous cycle classified with post-ovulatory stages (Post-Ov), early to mid-luteal stages (Early-mid L) and pre-ovulatory (Pre-Ov) stages. The urokinase-type plasminogen activator (uPA) was only observed on day 5 and day 7 of culture in the POECs on all the estrous cycles and gradually increased according to increasing culture times, but not Early-mid L. In POECs-conditioned medium, uPA, tissue-type (tPA) and tPA-PA inhibitor (tPA-PAI) activity were observed at all culture times during estrous cycles. The uPA activity of POECs-conditioned medium on Post-Ov stage were significantly (p<0.05) decreased during prolonged cultures. On the other hand, the tPA activity of POECs-conditioned medium at Post-Ov stage was significantly (p<0.05) higher on day 5 than compared to the other days. Although was higher on day 1 at Post-Ov stage, the tPA-PAI activity of POECs-conditioned medium was significantly (p<0.05) higher on day 7 at all stage than that of day 5 of the culture. Taken together, these results suggest that uPA, tPA and tPA-PAI are produced by POECs, and the variations of the PAs activity are regulated in the different stages of the estrous cycle.

Plasminogen activators (PAs) are serine protease that cleave plasminogen to form the active protease plasmin. PA/plasmin system playa role in mammalian fertilization and motility and acrosome reaction of sperm. The present study was undertaken to identify PAs in porcine gametes and investigate a possible role of plasminogen in in vitro fertilization in the pig. When boar spermatozoa were preincubated in a fertilization medium (mTBM) for 0, 2, 4 or 6 h, the activity of tPA-PAI ($110{\sim}117\;kDa$), tPA ($62{\sim}70\;kDa$), and uPA ($34{\sim}38\;kDa$) was observed in the sperm incubation medium and sperm sample. PA activities in the sperm incubation medium significantly (p<0.05) increased according to increasing incubation times, while PA activities in sperm significantly (p<0.05) decreased at the same times. In addition, the rate of acrosome reaction in spermatozoa increased by increasing culture times. When oocytes were separated from porcine cumulus-oocytes complexes at 0, 22 or 44 h of maturation culture, no PA activities were observed in cumulus free-oocyte just after aspiration from follicles. However, the activity of tPA-PAI ($108{\sim}113\;kDa$) and tPA ($75{\sim}83\;kDa$) was observed at 22 h of in vitro culture and significantly (p<0.05) increased as the duration of the culture increased. On the other hand, when porcine oocytes were activated by sperm penetration or calcium ionophore, plasminogen significantly (p<0.05) increased ZP dissolution time (sec) in activated oocytes by sperm penetration. These results suggest that supplementation of plasminogen to fertilization medium may playa positive role in the improvement of in vitro fertilization ability in the pig.

Xenotransplantation is proposed as a solution to the problem of organ shortage. However, transplantation of xenogeneic organs induces an antigen-antibody reaction in ${\alpha}$-1,3-gal structure that are not present in humans and primates, and thus complement is also activated and organs die within minutes or hours. In this study, we used FasL gene, which is involved in the immune response of NK cell, and US11, which suppresses MHC Class I cell membrane surface expression, to inhibit cell mediated rejection in the interspecific immunity rejection, and also hDAF(CD55) was introduced to confirm the response to C3 complement. These genes were tranfeced into Korean native pig fetal fibroblasts using pCAGGS vector. And cytotoxicity of NK cell and human complement was confirmed in each cell line. The US11 inhibited the cytotoxicity of NK cell and, in addition, the simultaneous expression of US11 and Fas ligand showed excellent suppress to T-lymphocyte cytotoxicity, hDAF showed weak resistance to cytotoxicity of natural killer cell but not in CD8+ CTLs. Cytotoxicity study with human complement showed that hDAF was effective for reducing complement reaction. In this studies have demonstrated that each gene is effective in reducing immune rejection.
One of the main goals of small, medium and large farms is to improve the reproductive performance of rabbit does. Stocks of lower productivity can be improved by crossing with intensive breeds. A better nutritional status of both foetuses and suckling kits has a positive effect on their later productivity. Overfeeding young females before first mating can lead to conditions of fattiness. Using restricted feeding or higher fibre content in the feed and changing it for a higher level ad libitum feeding about one week prior to first mating leads to longer lifespan and higher productive level. Intensive reproductive rhythm creates a negative energy balance in does : they are unable to consume enough feed (energy) for the nutritional requirements of foetus and lactation, and therefore lose most of their fat reserves. Furthermore, primiparous does also expend energy because they are still growing. Under intensive conditions, the 42-d reproductive rhythm (re-mating 11 days after parturition) is recommended. Under extensive conditions, the 18 or 25-d mating interval with 35 to 42-d weaning could be suitable. On small farms, natural mating is favoured; on large farms AI is commonly employed. The main advantage of AI is the all-in, all-out system. Hormonal (PMSG) treatment is used with AI to increase receptivity on d 11. Frequent and high level PMSG use can lead to higher anti-PMSG antibody rates. Lower level (max. 20 IU) and less frequent PMSG injection or non-hormonal alternative methods (short dam-litter separation, changing nursing method or lighting programs) are recommended for this reason.
