습지와 유수지를 이용한 하구담수호 유입하천의 수질개선 (Water Quality Improvement of Inflow Stream in Estuary using Wetland and Pond)

  • Wetland systems are widely accepted natural water purification systems around the world in nonpoint sources pollution control. In this study, the field experiment to reduce nonpoint source pollution loadings from agricultural drainage and polluted stream waters using wetland and pond system was performed. The removal rate of $BOD_5$, TSS, T-N and T-P during growing season was 7.2%, 64.3%, 57.0%, and 60.3%, respectively. And removal rate of $BOD_5$, TSS, T-N and T-P during winter was -49.5%, -56.1 %, 30.5%, and 47.1%, respectively. In this study, pond-wetland system is more effective than wetland-pond system to remove nutrient.

p-Coumaroylamino Acids from Yeast-Elicited Ephedra distachya Cultures

  • Three p-coumaroylamino acids (p-CAAs) were isolated from the yeast-elicited Ephedra distachya cultures by consecutive purification using XAD_2, silicagel and RP-HPLC. Retention times on HPLC as well as their UV, IR, NMR and MS spectral data indicated that the yeast-induced p-CAAs wre p-coumaroyl--D-valine, p-coumaroyl-D-serine and p-coumarouyl-D-threonine, respectively. The structures of p-CAAs were confirmed by the comparison of their physico-chemical properties 3with those of synthetic ones. They were isolated and identified for the first time from natural products and supposed to be accumulated as phytoalexins of Ephedra.

Rapid Purification of Recombinant Human Lipocortin-I Secreted from Saccharomyces cerevisiae

  • Human lipocortin-I was expressed as a secretory product by Saccharomyces cerevisiae harboring an expression system consisting of GAL10 promoter, inulinase signal sequence and lipocortin-I terminator. Fed-batch fermentation was carried out to overproduce recombinant human lipocortin-I. The culture medium was desalted and concentrated by ultrafiltration, and then subjected to hydroxyapatite column chromatography. The lipocortin-I was purified to >98% purity by single-step hydroxyapatite column chromato-graphy. However, it was found that the purified lipocortin-I was a proteolytically-cleaved form which was cleaved immediately after the basic amino acid Lys26.

경상남도 함안군, 법수늪의 식물플랑크톤 군집구조의 양상 (Patterns in Phytoplankton Community Structure in the Beopsu Marsh, Haman-gun, Gyeongsangnam-do, Korea)

  • The structure of phytoplankton community was studied for one year round at 6 stations in the Beopsu Marsh, Haman-gun, Gyeongsangnam-do, Korea. One hundred seventy-eight taxa (69 taxa belonging to Chlorophyta, 55 taxa to Bacillariophyta, 33 taxa to Cyanophyta, 15 taxa to Euglenophyta, 2 taxa to Rhodophyta, 2 taxa to Dinophyta and 1 taxon to Xanthophyta) were observed. Phytoplankton bloom occurred at the staions 2, 3 and 4 where waters were polluted by sewage and treated water of purification plants from the vicinity. The species diversity index of phytoplankton was the lowest (0.66) in November at the station 3, which the station showed the most eutrophic water due to the diverse pollutants from surroundings of the marsh. For the whole seasons, Fragilaria sp. and Euglena sp. appeared in pollutant water at the station 3, while Cyclotella sp. and Pinnularia sp. inhabited in relatively clean water at the station 5. To reserve the natural treasure, the wetland should be more specially managed by a local governing institution.

  • The experiment established optimal conditions for over-carbonization. With the use of the electron microscopy and X-ray phase analysis the regularities of carbon deposit formation in process of methane and propane pyrolysis on the zeolites, Kazakhstan natural clays, chrome and bauxite sludge containing metal oxides of iron subgroup, have been studied. In process of over-carbonization the trivalent iron was reduced to metal form. In addition, the carbon tubes of divers morphology had been impregnated with ultra-dispersed metal particles. The kinetic parameters of carbon formation in process of methane decomposition on the zeolite - CoO mixture surface were investigated by method of thermo-gravimetric analysis. The morphology and structure of formed carbon fibrils, with the metal particles fixed at their ends, have been investigated, the formation of branched carbon fibrils pattern, so called octopus, being found. Also, the walnut shells and grape kernel carbonization, their immobilization by the cells of selective absorption of heavy metal and sulfur dioxide ions have been studied. The example of metal-carbon composites used as adsorbents for wastewater purification, C$_3$- C$_4$ hydrocarbon cracking catalysts and refractory materials with improved properties have been considered.

Purification and Glycosylation Pattern of Human L-Ferritin in Pichia pastoris

  • Ferritin is an iron storage protein found in most living organisms. For expression and industrial use, human light chain ferritin (L-ferritin) was cloned from human liver cDNA library and expressed in Pichia pastoris strain GS115. The recombinant L-ferritin in Pichia pastoris was glycosylated. In a fed-batch culture, the cell mass reached about 57 g/l of dry cell weight, and the L-ferritin in the cell was increased to about 95 mg/l after 150 h. In an atomic absorption spectrometry analysis, the intracellular content of iron in the L-ferritin transformant was measured as $1,694{\pm}85\;\mu\textrm{g}g/g$, which is 5.4-fold more than that of the control strain. This L-ferritin transformant could serve as iron-fortified nutrients in animal feed stock.

Compounds from the Aerial Parts of Aceriphyllum rossii and Their Cytotoxic Activity

  • The purification of the MeOH extracts from the aerial parts of Aceriphyllum rossii Engler (Saxifragaceae) using column chromatography furnished fourteen compounds (1 - 14). The chemical structures of the isolated compounds were determined using mainly nuclear magnetic resonance spectra and mass spectrometry. All the isolated compounds were tested for their cytotoxic activity against LH-60, MCF-7 and HeLa cancer cells in vitro using a MTT assay method. Compounds possessing an olean-triterpenoid skeleton entirely exhibit dose dependent cytotoxic activity. These findings partially confirmed the anticancer effect of this medicinal plant, suggesting a further study on the anticancer potential of the triterpenoid structure in compounds from this plant.

무주군 진도리의 생태마을 조성계획에 관한 연구 (A Study on Eco-village Planning in Chindo-ri, Muju-kun)

  • This paper proposes the planning and design of the Eco-village centering around Chindo-ri, Muju-kun. The Eco-village project has three basic objects the Eco-village residents' lives adapting to ecological principles such as regeneration and circulation, life agricultural techniques based on organic cycle, and residents' harmonious lives with the surrounding nature. For constructing of the Eco-village, first, the subject area was analyzed in terms of the natural and cultural environment and then, on this analytic ground the concrete concept and the specific plan were set up. The following details also should be considered: the scale of the residential household, the population, the space arrangement of the village, the material cycling structure, the waste treatment, the purification of the sewage, the energy supply, the landscape architecture, the production activity, the conservation of the ecosystem and so on.

Isolation, purification and characterization of phytohemagglutinating proteins from Korean natural products

  • Seeds or beans of 55 plants belonging to 31 families were screened by using several different types of red blood cells to find new lectins. In this paper, white kidney been (phaseolus vulgaris C.) was chosen to study biochemical properties of hemagglutinating proteins(lectins). An anion exchanger, DEAE Sephadex A-50, and polyacrylamide disc gel electrophoresis were main techniques used. From three main fractions eluted by stepwise NaCl gradient in 25mM Tris-HCI buffer on DEAE Sephadex column, principal lectin was identified.

Lactobacillus helveticus의 자연항생작용에 관한 연구 (Natural Antibiotic Activity of Lactobacillus helveticus)

  • The objective of this study was to extract and purity the antibacterial agent from the fermented milk with Lactobacillus helveticus CH-1. The extraction and purification of antibacterial agent from the Lb. helveticus fermented milk were carried out by methanol extraction, acetone extraction, Sephadex G-200 gel filteration and thin layer chromatography and the results were as followings. The antibacterial activity of methanol-acetone extraction showed antibacterial activity against test organisms, B. subtilis, E. coli, Pseu. fluorescens, Sal. typhimurium, Shi. flexneri, and Sta. aureus. Sephadex G-200 gel chromatography showed only antibacterial activity from 33 to 37th fractions of 60 fractions. The agent purified from TLC plate confirmed the antibacterial activity by the means of bioautography.
