  • Puerariae Radix is one of the medicinal plants used in oriental medicine for hangover, It has been claimed for several pharmacological effects including anti-alcohol abuse, antidipsotropic activity and anti-alcohol intoxication. In connection with Puerariae Radix effects, an activity-guided purification of active substance on alcohol dehydrogenase (hnH) was carried-out. The most active compound was isolated as puerarin (C$_{21}$H$_{20}$ O$_{9}$ ), molecular weight 416. Puerarin inhibited ADH noncompetitively against ethanol or NAD$^{+}$./.

  • Amyloid oligomers are believed to play important causal roles in many types of amyloid-related degenerative diseases. Many different laboratories have reported amyloid oligomers that differ in size, morphology, toxicity, and method of preparation or purification, raising the question of the structural relationships among these oligomer preparations. The structural plasticity that has been reported to occur in amyloid formed from the same protein sequence indicates that it is quite possible that different oligomer preparations may represent distinct structural variants. In view of the difficulty in determining the precise structure of amyloids, conformation- and epitope-specific antibodies may provide a facile means of classifying amyloid oligomer structures. Conformation-dependent antibodies that recognize generic epitopes that are specifically associated with distinct aggregation states of many different amyloid-forming sequences indicate that there are at least two fundamentally distinct types of amyloid oligomers: fibrillar and prefibrillar oligomers. Classification of amyloid oligomers according to their underlying structures may be a more useful and rational approach than relying on differences in size and morphology.

  • Cystocin, a derivative of Puromycin, is a new material derived from Streptomyces sp GCA0001, new strain of Actinomycetes spiecies. This compound has outstanding biological activities in anti-bacteria, anti-tumor and anti-virus than former Puromycin compounds. And it is chosen by natural selection processing through extraction, isolation and purification from , so it may replace old Puromycins in most applications.

  • It is well known that inflammation is associated with neurodegenerative disorders, including Alzheimer' disease, Parkinson's disease and ischemia. Nitric oxide (NO), a pro-inflammatory mediator, is produced by inducible NO synthase (iNOS) in microglia as well as macrophages and appears to account for neurodegeneration. In this study, we aimed to isolate NO inhibitors from Pyrus pyrifolia by activity guided purification. As a result, we identified daucosterol and ${\beta}$-sitosterol, which have not been isolated from this plant before. This article also describes NO inhibitory activities of the methanol extract of Pyrus pyrifolia fruit and the isolated compounds from this, which are lupeol, betulin, betulinic acid, ${\beta}$-sitosterol and daucosterol, in LPS-activated RAW 264.7 and BV2 cell lines. Western blot analysis was performed to clarify the underlying mechanism of NO inhibition in the two cell lines.

Isolation of Angiotensin Converting enzyme inhibitors from Ripe Cucurbita moschata Duch

  • Angiotensin converting enzyme (ACE) inhibitor acts on the inhibition of ACE and causes a decrease in blood pressure. There have been several reports on screening of ACE inhibitors from natural food products and protein hydrolysates of various food sources. Ripe Cucurbita moschata Duch has been used as an oriental medicine in Korea. To isolate ACE inhibitors, crude water extracts of the edible portion of ripe Cucurbita moschata Duch were obtained after heating in water at 95$^{\circ}C$ for 2 h. Crude extracts were then filtered using PM-10 and YM-1 membranes. The membrane-filtered solution was loaded onto Sephadex G-15 column equlibrated with a phosphate buffer. Among the four major fractions of gel permeation chromatography, the second fraction had the highest inhibitory activity of 65%. Further purification of the fraction using reversed-phase HPLC with a $C_{18}$ column produced ACE inhibitors, which were identified as a mixture having molecular mass of 222 and 273 by Tandem mass spectrometry.

  • 박층류 자연정화공법은 수로의 폭을 넓히고 수심을 낮추어 박층류를 조성하고, 이를 이용하여 하천을 정화하는 공법이다. 박층류가 조성되면 낮은 수심에서 상대적으로 느린 유속 조건이 형성되며 이 때 박층부에 침전된 오염물질은 자갈층 표면에 형성된 생물막에 의해 산화 분해되어 제거된다. 박층류에 의한 오염물질 제거는 생물막의 형성, 생물 산화를 위한 용존산소량 등이 중요한 인자로 작용하며, 박층부를 조성하기 위해서는 수심 10 cm, 유속 30~50 cm/s 의 조건이 적절하다. 본 연구에서는 박층류에 의한 자연정화 성능을 평가하기 위하여 용인시 오산천 일부 구간 고수부지에 사석자갈을 이용한 박층류 자연정화공법이 적용된 차집수로를 설치하고, 오산천으로 유입되는 농수로의 물을 공급하여 오염물질의 농도 변화를 측정하였다. 측정된 항목은 T-N, T-P, COD, DO 등이며, 측정 결과 박층류 자연정화 수로를 유출부에서의 오염물질의 농도가 유입부에서보다 저감되는 것을 확인 할 수 있었다.

  • The Malaysian biogas upgrading technologies and policies were examined. In Malaysia, the regulation of palm oil mill effluent (POME) has been enforced to reduce the biochemical oxygen demand to 20 ppm and the biogas capture in the palm oil mills have been recently enforced for renewable energy. A huge amount of organic waste is produced from POME, and 80 million tons from palm oil trees, every year. Due to the renewable energy trends, the Malaysian government is modifying the use of biogases as fuels in favor of their conversion into compressed natural gas (CNG) and other chemicals; various green policies are being promoted because of many advantages of the organic substances. The Korean policies for biogas are a good model for exporting environmental plants after upgrading the digestion and purification technologies. Therefore, this article introduces the current status of POME and biogas production in Malaysia, it could encourage creating a new market for biomethane.

A Novel Approach to Investigating Protein/Protein Interactions and Their Functions by TAP-Tagged Yeast Strains and its Application to Examine Yeast Transcription Machinery

  • Tandem affinity purification (TAP) method combined with LC-MS/MS is the most accurate and reliable way to study the interaction of proteins or proteomics in a genome-wide scale. For the first time, we used a TAP-tag as a mutagenic tool to disrupt protein interactions at the specific site. Although lots of commonly used mutational tools exist to study functions of a gene, such as deletional mutations and site-directed mutagenesis, each method has its own demerit. To test the usefulness of a TAP-tag as a mutagenic tool, we applied a TAP-tag to RNA polymerase II, which is the key enzyme of gene expression and is controlled by hundreds of transcription factors even to transcribe a gene. Our experiment is based on the hypothesis that there will be interrupted interactions between Pol II and transcription factors owing to the TAP-tag attached at the C-terminus of each subunit of Pol II, and the abnormality caused by interrupted protein interactions can be observed by measuring a cell-cycle of each yeast strain. From ten different TAP-tagged strains, Rpb7- and Rpb12-TAP-tagged strains show severe defects in growth rate and morphology. Without a heterodimer of Rpb4/Rpb7, only the ten subunits Pol II can conduct transcription normally, and there is no previously known function of Rpb7. The observed defect of the Rpb7-TAP-tagged strain shows that Rpb7 forms a complex with other proteins or compounds and the interruption of the interaction can interfere with the normal cell cycle and morphology of the cell and nucleus. This is a novel attempt to use a TAP-tag as a proteomic tool to study protein interactions.

Purification and Characterization of Substance P-related Peptide from the Body of the African Lungfish, Protopterus dolloi

  • The peptide with structural similarity to mammalian substance P (M-SP) has been isolated from extract of the body of the African lungfish, Protopterus dolloi, using the rectum of the newt as the bioassay system. The primary structure of the SP-related peptide was identified as Lys-Pro-Arg-Pro-Asp-Gln-Phe-Tyr-Gly-Leu-Met-NH2 (L-SP) and contained four substitutions ($Lys^{1}\rightarrow $ Arg, $Arg^{3}\rightarrow$ Lys, $Asp^{5}\rightarrow$ Gln, and $Tyr^{8}\rightarrow$ Phe) compared with M-SP; this structure is identical to that of the peptide isolated from the gut of the Australian lungfish. Circular dichroism spectra showed that L-SP had an unordered structure in the buffer solution and phospholipid bilayers. This peptide was found to have an excitatory effect on rectal muscle tissues of newt, quail, and fish. L-SP also had a more potent vasodilatory effect on the guinea-pig aorta than that of M-SP. The identification of the peptide provides evidence that SP family, hitherto confined to mammals, have a widespread occurrence in lungfish.
