  • Currently, cancer is the primary cause of death and 50% of cancer patients are incurable by surgery, radiotherapy and chemotherapy. Therefore, immunotherpy is interested as the fourth remedy. Biological response modifier (BRM), such as organometallic compounds, glycoproteins, polysaccharides and other natural products. Is the one which can enhance the immune response against cancer cell. To develop new BRM from natural sources, we investigated 63 species Korean traditional medicines by observing the mitogenic activity to splenocytes, generation of activated killer cells and activation of macrophages. Finally, we selected 9 species including Angelicae gigantis Radix, Mori Cortex Radicis, Arisaematis Tuber, Salviae Radix, Cnidii Rhizoma, Ligusti Fructus, Pasoraliae Semen, Loranthi Ramulus, Ginseng Radix. Bioassay-guided fractionation and purification is undergoing.

Purification and Characterization of TSA from Lumbricus terrestris (지렁이(Lumbricus terrestris)로부터 Thiol-Specific Antioxidant protein(TSA)의 분리 및 정제에 관한 연구)

  • A thiol-specific antioxidant(TSA) protein was purified from Earthworm, Lumbricus terrestris by DEAE-Cellulose, Phenl sepharose, Sephacryl S-200 gel filtration and HPLC S-300 Column Chromatography. This protein showed a thiol-specific antioxidant activity against inactivation of glutamine synthetase by a metal-catalysed oxidation system capable of generation reaction oxygen species. The molecular mass of the protein was determinated to be 51-kDa by SDS-polyacrylamide gel electrophores. Taken together, the purified TSA protein could be a new member of TSA family.

  • $\beta$-Galactosidase(EC 3.2.1.23) from pathogenic Neisseria lactamica 2118 was purified by p-aminopheny1-$\beta$-D-thiogalactopyranoside agarose affinity chromatography. Then some properties of the purified $\beta$-galactosidase were investigated.$\beta$-Galactosidase form Neisseria lactamica 2118 was constitutive enzyme, not induced by lactose and IPTG. Optimal activity was observed at $35^{\circ}C$ and pH 7.5 and the enzyme was stable at the range of pH 6.0-9.0 and at temperature below $50^{\circ}C$. The enzyme activity was inhibited by cations such as $Hg^(2+)$ and $Co^(2+)$.

The Antioxidative Effect and Ingredients of Oil Extracted from Schizandra chinensis Seed (오미자씨에서 추출된 유지의 성분 및 항산화 효과)

  • The objectives of this study were to investigate the antioxidant activity of Schizandra chinensis seed oil and its active ingredients. Schizandra chinensis seed oil content extracted with hexane was 36.06%. Schizandra chinensis seed oil extracted with hexane was purified during 20 min at $85^{\circ}C$ with phosphoric acid 0.15% for degumming and 20 min at $80^{\circ}C$ with 3 M NaOH 1% for deaciding. The purified oil consisted of unsaturated fatty acid (88.7%), fatty acid (9.97%), and so on. The major unsaturated fatty acids of purified oil were linoleic acid (71.1%) followed by oleic acid (15.7%), while the main saturated fatty acid was palmitic acid (6.56%). The purified oil was found that contents of phenolic compounds, vitamin A, and E were 1.45 g/100 g, 1494.86 RE/100 g, and 0.58 mg ${\alpha}$-TE/100 g, respectively. Schizandra chinensis seed oil exhibited strong antioxidant activity (91.7%) as compared to grape seed oil and canola seed oil with 87.4% and 85.1% in the DPPH assays. Present results suggest that Schizandra chinensis seed oil could be potentially used as bioactive source for health and preventing numerous diseases.

Purification of Cellulase from Trichoderma viride and properties of Its Component Enzymes

  • Major cellulase components, such as three endoglucanases (endoglucanases I, II, and III) and one exoglucanase (exoglucanase II), were isolated from a commercial cellulase (Meicelase TP 60) derived from the fungus Trichoderma viride by a series of chromatography procedures. These procedures were the gel filtration on Bio-Gel, the anion exchange on DEAE-Bio-Gel A, the cation exchange on SP-Sephadex C50, and the affinity chromatography on Avicel cellulose. The average molecular weights determined by SDS-polyacrylamide gel electrophoretic analysis were 51,000, 59,000, 41,000 and 62,000 Da for endoglucanases I, II and III and exoglucanase II, respectively. The extinction coefficients, ${\varepsilon}^{1%}$ 280 nm, of these enzymes were 11.7, 3.3, 7.2 and 11.3, respectively. Among them, the endoglucanase II showed the very low value of the coefficient compared with the others. On the other hand, it was found that endoglucanase II and III were of more random hydrolytic mode on carboxymethylcellulose as compared with those of endoglucanase I and exoglucanase II. Especially, endoglucanase I showed less random action than that of exoglucanase II. In the hydrolysis of insoluble cellulose by the enzyme components, cellobiose was the major product, but glucose was the major product by endoglucanase III.

Rat Liver $\beta$-Glucuronidase; Its Purification and Inhibition Studies

  • ${\beta}$-Glucuronidase (EC 3.2.1.31) which hydrolizes D-glucuronate from ${\beta}$-D-glucuronide was purified from rat liver, using ammonium sulfate fractionation, DEAE-cellulose chromatography, Concanavalin-A Sepharose 4B chromatography and gel filtration on Sephadex G-200. This enzyme has the molecular weight of 280,000 daltons by gel filtration and 75,000 daltons by SDS-polyacrylamide gel electrophoresis. As its funtion is reverse of detoxification in the liver, the inhibition of the enzyme was tested with extracts of several food products and medicinal herbs, some are known as anti-cancer agents. Among them, Panax ginseng and Cortnellus shiiake inhibited the enzyme competitively and the $K_1$ values were $9.22 {\times}\;10^{-2}$ and 0.102 mg/ml, respectively. These inhibitors strongly bound to DEAE-cellulose. The negatively charged amino acids, L-aspartate and L-glutamate, inhibited the enzyme, and $K_1$ value of L-aspartate was 0.80 mM. The interaction between ${\beta}$-glucuronidase and p-nitrophenyl-${\beta}$-D-glucuronide was found to involve ionic forces by the effect of ionic strength on the kinetic constant, Vmax/Km. It was inferred from these findings that cationic group at the active center of the enzyme is probably involved in attacking the substrate.

  • A methanol soluble extract from the dried hooks and stems of Uncaria rhynchophylla showed a strong inhibitory activity against monoamine oxidase in mouse brain. Using a bioassay-guided purification of this extract, a known ${\beta}-carboline$ type alkaloid, harman (1), was obtained as an active constituent. In addition, five known indole alkaloids, isocorynoxeine (2), isorhynchophylline (3), corynoxeine (4), cadambine (5), and $3{\alpha}-dihydrocadambine$ (6), were isolated and found to be weakly active or inactive.

A study on determining the optimum mixing ratio of carbon source for anaerobic treatment (혐기성처리를 위한 탄소원의 최적 혼합비율 결정에 관한 연구)

  • The purpose of this study were to determine the optimum mixing ratio of sewage sludge and papermill sludge as carbon source required to SRB in treating abandoned mine drainage with natural purification wetland. If mixing sewage sludge/papermill sludge 2.0 SO42- reduced 46.2%, and then 30% in mixing ratio 0.5.Because sewage sludge was faster biodegradability than papermill sludge, effluent SCOD was 40mg/L in mixing ratio 0.5, and after that was all but regular. pH and ORP were almost neutral and -160mV, but after that was all but regular and it indicated that SRB activity was suitable. Fe removal rate was 60% in mixing ratio 2.0, and 54% in mixing ratio 0.5. In point of carbon source supply, It indicated that mixing ration 0.5 was considered as the most appropriate, because degradability of swewage sludge under short time was higher than that of papermill sludge.

  • Anthocyanins are water-soluble glycosides and acylglycosides of anthocyanidins, having different color variations due to its substitution patterns. Anthocyanins, present in various fruits, vegetables and crops as natural colorant, have been well characterized for its bioactive properties, anti-oxidant, anti-cancer, anti-proliferative and anti-inflammatory properties. During extraction and purification, the factors, such as pH, temperature, oxygen, light, enzymes, nucleophilic agents, sugar derivatives and co-pigments, have affected on anthocyanin stability. For this reason, the extraction method should be thoroughly checked for the qualitative/quantitative analysis of anthocyanin in particular plant material. To identify the optimum extraction method of cyanidin-3-glucoside, major anthocyanin of dark purple-colored grains, Oryza sativa cv. Heugjinjubyeo, Phaselous vulgaris, Phynchosia gngularis, Sesamum indium, Rhynchosia nulubilis and Lablab purpureus, reversed-phase HPLC analysis using solvent system of acetonitrile, methanol and water were accomplished.

Radical Scavenging Activity of Gallic Acid from Woodfordia Fruticosa Flowers

  • Woodfordia fruticosa Kurz (Lythraceae) is used in the treatment of various ailments in traditional medicines. DPPH activity guided fractionation and purification process was used to identify the free radical-scavenging components from the flowers of this plant. The methanolic extract of the plant was first fractionated into four extracts; namely, n-hexane, chloroform, ethyl acetate and water fractions. Among them, the ethyl acetate fraction was found to be the most effective and was further subjected to activity guided-fractionation and isolation procedures. After successive column chromatography on silica gel and Sephadex LH-20, gallic acid, which is responsible for the radical scavenging activity, was isolated and its structure was elucidated by spectral methods ($^1H$ NMR, $^{13}C$ NMR) and by comparison with literature.
