The purpose of this study was to evaluate tyrosinase inhibitory activity of plant extracts, especially Korean indigenous plants, for the cosmetic use of skin whitening. When 50 plant extracts were tested, the methanol extracts of Agrimonia pilosa, Aster scaber; Dianthus sinensis, Fatsia japonica, Hemistepta lyrata, Lespedeza cuneata, Osmunda japonicum, Pyrvla japvnica, Rodgersia phodophylla and Veratrum grandiforum possessed the considerable tyrosinase inhibitory activity at 3-300 $\mu\textrm{g}$/mL. Especially, L. cuneata, aerial part of O. japonicum and V. gandiforum exhibited the strong inhibition (>50% inhibition at 300 $\mu\textrm{g}$/mL). In particular, the methanol extract of V. grandiforum and its ethylacetate fraction showed potent inhibition ($IC_{50}$/=30 and 13$\mu\textrm{g}$/mL, respectively), while the reference compound, kojic acid, showed $IC_{50}$/ value of 26$\mu\textrm{g}$/mL. These plant extracts may be used as tyrosinase inhibitors in cosmetics.
The aim of study was to elucidate the antibiosis of the Sophora Radix extracts by ethanol solvent on bacterias. The antibiosis on bacterias was examined in moisture water included the Sophora Radix extract. The results are as follows; Minimum inhibitory concentration (MIC) of Sopora Radix extracts showed markedly lower on gram positive bacterias($0.0125{\sim}0.025$) such as aureus, B. subtilis, S. epidermids than that on gram negative bacterias such as E. coli, P. aeruginosa(>10). The MIC of moisture water included the Sophora Radiix which extracted by ethanol solvent was very low which meaned good antibiosis($0.0063{\sim}0.5$). The antibiosis of moisture water included the Sophora Radiix was improved by adding the ethanol on some baterias. In conclusion, Sophora Radix had excellent antibacterial effect. Therefore, it is expected that Sophora Radix can be practically used as the natural material of functional cosmetics for preventing the Bacterial inhibitory in skin.
Chronic ultraviolet (UV) radiation causes photoaging, which represents skin damage, disrupts skin barrier function, and promotes wrinkle formation. We investigated that the polysaccharide extract of an edible basidiomycetous white jelly mushroom, Tremella fuciformis, (TF-Glucan®) exhibited statistically photoprotective activity by inhibiting matrix metalloproteases (MMPs) and increasing collagen synthesis, and an anti-inflammatory activity by inhibiting nitric oxide and pro-inflammatory cytokines at the concentrations of less than 1000 ㎍/ml, which is not cytotoxic (p < 0.05). Additionally, TF-Glucan® increased the expression of involucrin and filaggrin to prevent the disruption of UVB-induced barrier function (p < 0.05). TF-Glucan® was assessed as a safe material by the human primary skin irritation (1, 3, 5%), human repeated insult patch test (no sensitization at 5%), 3T3 NRU phototoxicity assay (no phototoxicity, PIF < 2, MPE < 0.1), eye irritation test test by BCOP (no category, IVIS ≤ 3) and local lymph node assay (negative at 10, 25, 50%) for identifying potential skin sensitizing. These results suggest that TF-Glucan® may be useful as an anti-photoaging ingredient for developing cosmeceuticals.
As part of research to develop a permanent wave-reducing agent for hair, in the current study, 0.1%-1.0% of Polyquaternium-10 was added to a permanent reagent containing Nicotinoyl Dipeptide-23 to prepare the agent, which was tested on damaged hair. The pH change was relatively stable even after a date, but due to the hair's natural composition, 0.1%-0.6% of the Polyquaternium-10 concentration according to pH was suitable. The temperature safety experiment demonstrated it to be stable at a high temperature and at room temperature, but when a Polyquaternium-10 concentration of 0.9% or higher was added at 0℃, coagulation occurred. In terms of the force efficiency of permanent wave formation, wave efficiency lowered as the concentration increased: the longer the wave lasts, the lower the Polyquaternium-10 concentration. The permanent moisture content was found to be higher as the concentration of Polyquaternium-10 increased. Therefore, when Polyquaternium-10 is applied to the permanent wave-reducing agent, considering stability, permanent formation, durability, and moisture rate, the most suitable concentration was found to be that of Polyquaternium-10 in the cysteine-reducing agent. A novel finding from this study is that as the concentration of Polyquaternium-10 increases, the consistency of the permanent wave-reducing agent changes, shifting from a liquid to a viscous liquid formulation.

Research on whitening materials using natural alternatives is actively being conducted. The aim of this study was to investigate the in vitro inhibitory effects of Ranunculus chinensis Bunge (RCB) on melanogenesis and associated enzymes, such as tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2 in B16F10 murine melanoma cells. We found that RCB extract significantly attenuated melanin synthesis and reduced the activity of intracellular tyrosinase, a rate-limiting melanogenic enzyme. Western blot analysis showed that RCB extract decreased the protein expression of tyrosinase and TRP-1. In addition, it significantly decreased the expression of microphthalmia-associated transcription factor (MITF), a key regulator of melanogenesis. Extracellular signal-regulated kinase (ERK) activation has been reported to be involved in the inhibition of melanogenesis. Thus, we investigated whether the hypopigmentary effects of RCB extract were related to the activation of ERK. RCB extract induced ERK phosphorylation in a dose-dependent manner. Furthermore, it markedly inhibited body pigmentation in a zebrafish model. Our results suggest that RCB extract inhibits melanogenesis by activating ERK pathway-mediated suppression of MITF and its downstream target genes, including tyrosinase. Therefore, RCB extract can be used as a whitening agent in the development of functional cosmetics.

The anti-melanogenic activity of 259 actinomycete strains was tested, and based on the results for the inhibition of mushroom tyrosinase activity and the reduction in melanin content, Micromonospora sp. JCS1 and JCS7 were selected as the strains with the highest anti-melanogenic potential. The activity-guided fractionation of extracts from JCS1 and JCS7 led to the isolation of the dipeptides cyclo(ʟ-Phenyl alanine (Phe)-ʟ-Proline (Pro)) (1) and cyclo(ʟ-Tryptophan (Trp)-ʟ-Proline (Pro)) (2). These two compounds were tested for their inhibition of mushroom tyrosinase by monitoring ʟ-DOPA levels and melanin production. Cyclo(ʟ-Phe-ʟ-Pro) (1) and cyclo(ʟ-Trp-ʟ-Pro) (2) were thus confirmed to have the potential for use in functional whitening cosmetics containing actinomycete-derived secondary metabolites.
Nanoparticles are commonly used to avoid the opaque white color of TiO2 based sunscreen. However, a dispersing agent is typically required because of the tendency of the nanoparticles (NPs) to agglomerate. Stearic acid is one kind of dispersing agent often used for sunscreen products. However, according to the MSDS data sheet on stearic acid, stearic acid is highly hazardous to aquatic life and causes irritation on human skin. To avoid this problem, in this study a safer organic dispersing agent extracted from Korean seaweed has been studied to disperse TiO2 nanoparticles, and further use as an active agent in sunscreen products. The presence of phytochemicals in seaweed extract, especially alginate, can disperse TiO2 nanoparticles and improve TiO2 dispersion properties. Results show that seaweed extract can improve the dispersion properties of TiO2 nanoparticles and sunscreen products. Reducing the agglomeration of TiO2 nanoparticles improves sunscreen properties, by making it less opaque white in color, and increasing UV protection value. It was also confirmed that adding seaweed extract into sunscreen products had no irritating effects on the human skin, making it more desirable for cosmetics application.
The antioxidant capacity and protective effect of peptides from protein hydrolysate of Cordyceps militaris cultivated with tussah pupa (ECPs) on H2O2-injured HepG2 cells were studied. Results indicated ECP1 (<3 kDa) presented the strongest antioxidant activity compared with other molecular weight peptides. Pretreated with ECPs observably enhanced survival rates and reduced apoptosis rates of HepG2 cells. ECPs treatment decreased the ROS level, MDA content and increased CAT and GSH-Px activities of HepG2 cells. Besides, the morphologies of natural peptides from C. militaris cultivated with tussah pupa (NCP1) and ECP1 were observed by scanning electron microscopy (SEM). Characterization results suggested the structure of NCP1 was changed by enzymatic hydrolysis treatment. Most of hydrophobic and acidic amino acids contents (ACC) in ECP1 were also observably improved by enzymatic hydrolysis. In conclusion, low molecular weight peptides had potential value in the development of cosmetics and health food.

Atopic dermatitis, also called congenital fever, is a allergic eczema of chronic itching disease. It is a recurrent and familial disease and appears on a wide age group from infant to adult. It is very common, and the ratio of occurrence is about $9{\~}l2\%$ of a child. However. it is showing trend of continuous increase by social and natural environment, food culture, and life style, recently. The human skin plays a barrier role against a physical and chemical stimulus from external environment. According to the latest study, the decreased amount of ceramide in horny layer impairs the bier function and moisture-maintaining function of skin in atopic dematitis patient. Ceramide is a kind of the sphingolipid in which a fatty acid is connected to sphingosin. Ceramide constitutes about $40\%$ of total lipid between keratinocytes and has the function of defense wall and building regular structure to suppress moisture vaporization in horny layer. In horny layer of skin a comified cell is composed of multi-layer structure of a brick shape, and, as for this cornified cell, it is strongly connected by ceramide, cholesterol, and free fatty acid. Here, we described the effects of a cream containing ceramide on the recovery of skin harrier function of atopic dermatitis patient. The safety and efficacy of latex and liquid formula were evaluated as cosmetics for atopic dermatitis. The latex products was composed of intercellular lipid components-ceramide, cholesterol, and free fatty acid-to restore skin barrier function in atopic dermatitis patients. The liquid one contained beta-glucan, magnolia extracts, and licolice extracts, which have skin immunomodulatory and anti-inflammatory effects. It is also confirmed that their possibility on new cosmetic market of atopic dermatitis.
This study was carried out to evaluate cytotoxic effects of Poncirus trifoliata Raf. extract on lymphocytic leukemia tumor (L1210) cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay The comparison of Ic50 Values of Poncirus trifoliata Raf. extract in L1210 cell lines showed that their susceptibility to these fractons decreased in the following order: adriamycin > Fr.4> Fr. 6> Fr. 5> Fr. 3> Fr. 1> Fr. 2 by the MTT assay. In order to develop an antumicrobial agent, Poncirus trifoliata Raf. was extracted wit ethanol, and then it was fractionated with several mobile phase. The antitumor activities of fractions of the ethanol soluble extract was investigated. The minimal inhibitory concentrations (MIC) of fractions of the ethanol soluble extract of Poncirus trifoliata Raf. against microorganisms were also examined. Antimicrobial activities of ampicillin and ketoconazole as references were compared to those of fractions of the ethanol soluble extract of Poncirus trifoliata Raf. The antimicrobial activities of all fractions from the extract had growth inhibition activities against gram-positive bacteria, gram-negative bacteria and fungi $(MIC\;>\;200{\mu}g/ml)$. These results suggest that fraction 4 of the ethanol soluble extract of Poncirus trifoliata Raf. possessed the most antitumorous agent.
