다가알코올로부터 추출된 황금, 아카시아, 한라봉 추출물의 천연 방부시스템 연구 (Study of Natural Preservative System Using the Mixture of Scutellariae radix, Acacia nilotica and Citrus reticulata Extracted from Polyhydric Alcohols)

  • The aim of this study was to develop a new natural preservative system to improve the weak points of natural polyhydric alcohols together with the efficiency of natural plants as a preservative. Polyhydric alcohols (glyceryl caprylate and ethylhexylglycerin) and antimicrobial plants (S. radix, A. nilotica and C. reticulata) were tested using the disc diffusion method and minimum inhibitory concentration (MIC) method for their antimicrobial activity against the common poultry pathogens, respectively. A study of the preservative efficacy of the cosmetic formulations containing the optimized preservative system demonstrated sufficient preservative efficacy against bacteria and eukaryotic test microbes. These results suggest that the natural preservative system including polyhydric alcohol extracts containing natural plants could be incorporated in cosmetic formulations.

섬유문화재 보존에 활용하기 위한 천연보존제로 처리한 직물의 특성연구 (A Study on the Characteristics of Natural Preservative Agent-treated Fabrics for Textile Cultural Properties Preservation)

  • 천연보존제는 주로 식물에서 추출된 항균물질로 만들어지며 부패되기 쉬운 식품의 보존제나 천연화장품, 약품등에 주로 사용되고 있다. 본 연구는 현재 천연화장품의 보존제로 개발되어진 천연보존제를 직물에 처리하여 이것이 섬유문화재 보존에 적용이 가능한지를 탐색하고 평가하는데 그 목적이 있다. 따라서 천연보존제로 처리한 직물의 특성을 알아보고자 천연보존제 1%로 처리한 면직물과 견직물에 대해 실험한 결과 색 및 인장강도의 변화가 극히 미약하고 중성에 가까운 pH를 나타내었으며 우수한 항균성과 항곰팡이성을 가지는 것으로 나타났고 견직물에 피해를 주는 Bacillus cereus에 대해서도 우수한 항균활성을 나타내었다. 또한 섬유문화재에 적용시키기 위한 예비실험으로 출토직물 편을 천연보존제 1%로 처리한 직물로 싸서 72시간 보관한 후 출토직물편의 미생물 실험결과 미생물의 증식이 확연히 준것을 알 수 있었다.

찔레꽃 에탄올추출물의 생리활성과 화장품 방부효과 (Biological Activity and Cosmetic Preservative Effects of Rosa multiflora Ethanol Extracts)

  • Background: The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity and cosmetic preservative efficacy have not yet been studied. Thus, this species is yet to be defined as a functional cosmetic material. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora. Methods and Results: The antioxidant activity was assessed through free radical scavenging assays with 2,2-diphenyl-1-picrylhydrazyl (DPPH). Additionally, the contents of total phenols and flavonoids were analyzed. The phenolic compounds were detected using HPLC. The antimicrobial activity against Staphylococcus aureus, Escherichia coli, and Candida albicans was assessed using the disc diffusion assay. The preservative effect (challenge test) on a formulation of soothing gel was performed for 28days. The DPPH radical scavenging ability, denoted by the $SC_{50}$ (half maximal inhibitory concentration for DPPH radical scavenging) value was found to be $131.63{\mu}g/m{\ell}$. The content of total polyphenol and flavonoid content were 202 mg/g and 86.77 mg/g, respectively. In additon, astragalin and gallic acid were identified in the extract. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 5 - 0.5%, and no significant activity was noted against C. albicans. The ethanol extracts (5% and 10%) met the preservation standards of the Cosmetics, Toiletry, and Fragrance Association (CTFA). Conclusions: Thus the ethanol extract of R. multiflora can be used in cosmetics as a natural preservative and antioxidant.

A Study of Antioxidant and Antibacterial Activates of the Extraction of Perscaria hydropiper L.

  • This purpose of this study is to examine the antioxidant and antimicrobial activities of Persicaria hydropiper L. extract in 70% ethanol and in water, a medicinal herb, as an effort to examine the potential of medicinal herbs for development of antioxidants and natural preservative substitutes. The total poly-phenol content in the 70% ethanol extract and in the water extract were 19.88 mg/g and 7.46 mg/g, respectively. The DPPH radical scavenging activity was 90.99% and 64.98% in the 70% ethanol extract and water extract, in which 70% ethanol extract showed a higher activity. The antioxidant effect of Persicaria hydropiper L. extract appears to be very good and due to its excellent growth inhibitory effect on food-poisoning-causing microorganisms in the food, it is thought to be utilized as a potential natural preservative substitute in many areas.

인간 항균펩타이드인 LL-37 유래의 FK-13의 화장품보존제로 활용에 대한 연구 (A Study on the Use of Human Antibacterial Peptide LL-37-derived FK-13 as a Cosmetic Preservative)

  • 본 연구에서는 인체에 보다 안전한 천연 화장품보존제 발굴을 목표로 하여 인간유래 항균펩타이드 LL-37의 짧은 유사체인 FK-13을 화장품보존제로 사용하고자 연구를 진행하였다. 이를 위해 13개의 아미노산으로 구성된 FK-13 펩타이드를 고상 펩타이드 합성법로 합성하였고 reversed phase-high performance liquid chromatography (RP-HPLC)로 정제 후 liquid chromatography-mass spectrometry (LC-MS)를 이용하여 순도와 분자량을 확인하였다. 합성된 FK-13을 이용하여 미생물에 대한 방부력을 검정하였고 화장품에 적용 가능성을 살펴보았다. FK-13은 3개의 그람-양성균 (Staphylococcus aureus, Bacillus subtilis, Staphylococcus epidermidis)과 3개의 그람-음성균 (Escherichia coli, Salmonella typhimurium, Pseudomonas aeruginosa), 그리고 진균인 Candida glabrata 에서도 높은 항균 활성을 보였으며 넓은 항균활성 스펙트럼을 나타내었다. 이를 바탕으로 화장품보존제로서의 적합성을 확인하였다. 또한 FK-13은 고온에서 분자의 열안정성을 보였으며 기존의 천연 한방화장품 방부제 및 화학보존제들과의 항균활성 비교 실험에서도 보다 월등한 항균활성 효능을 나타내었다. 따라서 FK-13 펩타이드는 인체 친화적이고 효과적인 항균활성을 나타내므로, 기존 화학보존제를 대체할 새로운 천연 화장품보존제로 적용이 가능할 것으로 판단된다.

Antimicrobial Activity of Propolis Extract and Their Application as a Natural Preservative in Livestock Products: A Meta-Analysis

  • This study aimed to evaluate the effectiveness of propolis extract as a natural preservative for livestock products in term of chemical and microbiological characteristics by meta-analysis. The stages carried out in this study were identification, selection, checking suitability, and the resulting selected articles were used in the meta-analysis. The selection results obtained a total of 22 selected journal articles consisting of 9 articles for analysis of the antimicrobial activity of propolis extract and 13 articles for analysis of the chemical and mirobiological characteristics of livestock products. The articles were obtained from electronic databases, namely Science Direct and Google Scholar. The model used in this study is the random-effect model involving two groups, control and experimental. Heterogeneity and effect size values were carried out in this study using Hedge's obtained through openMEE software. Forest plot tests and data validation on publication bias was obtained using Kendall's test throught JASP 0.14.1 software. The results showed that there is a significant relationship between propolis extract with the results of the antimicrobial activity (p<0.05). In addition, the results of the application of propolis extract on the livestock products for the test microbes and the value of thiobarbituric acid reactive substances (TBARs) showed significant results (p<0.05). Conclusion based on the random-effect model on the effectiveness of antimicrobial activity of propolis extract and their apllication as a natural preservative of the chemical and microbiological characteristics of livestock products is valid by Kendall's test (p>0.05). Propolis in this case effectively used as natural preservatives in livestock products.

합성보존제(benzalkonium chloride)와 천연보존제(chitosan)의 세포독성 및 항균 활성에 관한 연구 (A Study on the Cytotoxicity and Antimicrobial activity of Synthetic preservative(benzalkonium chloride) and Natural preservative(chitosan))

  • 점안약에 광범위하게 사용되는 보존제(preservatives)의 세포독성 및 항균 활성 및 항진균 활성을 검정하였다. 세포독성은 L929세포를 사용하였으며, 항균 항진균 활성 검정에 사용된 균주는 눈의 정상 세균총의 일종이거나 각막염을 일으키는 세균인 Pseudomonas aeruginosa ATCC27853, Staphylococcus aureus ATCC25923, Eschenchia coli ATCC25922 등 3가지이고, 진균은 Trichoderma reesei ATCC6967을 사용하였다. 사용된 시약은 합성 보존제인 benzalkonium chloride(BAC)와 천연 보존제인 키토산이며 키토산의 분자량은 60,000, 탈아세틸화도는 95%, 점도는 2 cps였다. L929세포에 대해 24 hr, 48 hr, 72 hr 동안 각각의 농도로 세포독성 검정 결과 세포의 30% 이상 성장저해를 일으키는 농도는 BAC의 경우 0.005~0.001%였고, 키토산의 경우는 0.6~0.55%였으며, 이 결과는 시간에 따라 거의 변화가 없었다. 각각의 균에 대한 BAC의 항균 항진균 활성을 나타내는 최소저해농도는 P. aeruginosa에는 0.1~0.01%였고, S. aureus에는 0.001~0.0001%였으며 E. coli에는 0.001~0.0001%, T. reesei에는 0.1~0.01%였다. 키토산은 P. aeruginosa의 경우 2%, S. aureus가 1%에서 2일간 균 성장 정시작용을 나타내었으며, E. coli에서는 성장저해를 일으키지 않았다. 진균에 대해서는 농도 의존적으로 생육을 촉진시켰다. 천연보존제는 합성보존제에 비해 항균성은 다소 떨어지지만 세포독성을 일으키지 않으며 여러 약리 작용이 있으므로 이에 대한 연구가 더 필요하다.

수용성(水溶性) 방부처리재(防腐處理材)에서 금속류(金屬類)의 부식(腐蝕) (Corrosion of Metals in Waterborne Preservative-Treated Wood)

  • This study was performed to investigate the corrosion of metal fasteners in waterborne preservative-treated wood. Of all the metal tested, steel exhibited the greatest amount of corrosion across all preservatives and exposure conditions whereas stainless steel was totally inert. Galvanized steel corroded at a much lower rate compared to steel and the corrosion of brass was negligible. Among the preservatives, CCA-Type B was the most corrosive system tested. The sequence for the average corrosivity across all metals was: CCA-type B>CCA-Type C${\geq}$ACC${\geq}$CCA-Type A. Across all metals and retentions the salt formulations were more corrosive than the oxide and the corrosion was increased with the increse in the amount of preservative loadings. The amount of corrosion was also increased with the increase in exposure relative humidity (RH) across all metals and presevatives However, at the 60% RH exposure condition, the corrosion of metals was very negligible. Consequently, it can be concluded that across all metals oxide-type preservatives should be used to prevent the corrosion problem of metal fasteners in contact with treated wood and the use of steel nail preservative-treated wood should be avoided without distinction of end-use location, i.e., exterior or interior, and the use of stainless steel or at least brass nail in damp exterior condition was strongly recommended.

비수분산 특수 제형 화장품에 대한 방부력 시험 비교 연구 (Comparison of Preservative Efficacy Tests for Water Non-dispersible Cosmetic Formulations)

  • 본 연구에서는 수분산 제형에 사용되는 방부력 시험법과 PCPC (personal care products council) 가이드라인 대체시험법, direct contact membrane method 및 surface mold test 사이의 차이점을 연구하여 비수분산 특수 제형 방부력 시험 진행 시 고려되어야 하는 사항에 대해 알아보고자 한다. 우리는 스틱 및 프레스트 파우더에에 대한 시험법 비교 시 대체 시험법을 사용했을 때 개선된 수준으로 방부력 시험 결과를 확인할 수 있었다. 하지만 W/S 에멀젼과 루즈 파우더의 경우, 기존 시험법이나 대체 시험법간 결과 개선 차이가 크지 않았음을 확인하였다. 각각의 제품에 대해 방부시스템 존재 시와 부재 시 결과들을 확인했을 때, 시험법별로 차이가 발생함을 알 수 있었다. 결과적으로, 비수분산 특수 제형에 대한 방부력을 측정할 때 기존 및 대체 시험법을 병행 사용하여 개선된 수준의 결과를 얻을 수 있었다. 또한 본 연구를 통해 스틱 및 프레스 파우더의 경우, 소비자 사용방식을 적용하는 방식의 방부력 시험법을 적용하였을 때, 보다 더 유효성 있는 결과를 도출해 낼 수 있었다.

Status, Antimicrobial Mechanism, and Regulation of Natural Preservatives in Livestock Food Systems

  • This review discusses the status, antimicrobial mechanisms, application, and regulation of natural preservatives in livestock food systems. Conventional preservatives are synthetic chemical substances including nitrates/nitrites, sulfites, sodium benzoate, propyl gallate, and potassium sorbate. The use of artificial preservatives is being reconsidered because of concerns relating to headache, allergies, and cancer. As the demand for biopreservation in food systems has increased, new natural antimicrobial compounds of various origins are being developed, including plant-derived products (polyphenolics, essential oils, plant antimicrobial peptides (pAMPs)), animal-derived products (lysozymes, lactoperoxidase, lactoferrin, ovotransferrin, antimicrobial peptide (AMP), chitosan and others), and microbial metabolites (nisin, natamycin, pullulan, ε-polylysine, organic acid, and others). These natural preservatives act by inhibiting microbial cell walls/membranes, DNA/RNA replication and transcription, protein synthesis, and metabolism. Natural preservatives have been recognized for their safety; however, these substances can influence color, smell, and toxicity in large amounts while being effective as a food preservative. Therefore, to evaluate the safety and toxicity of natural preservatives, various trials including combinations of other substances or different food preservation systems, and capsulation have been performed. Natamycin and nisin are currently the only natural preservatives being regulated, and other natural preservatives will have to be legally regulated before their widespread use.
