STUDIES ON THE CYSTINE COMPONENT IN THE SERICULTURAL PROTEINS OF BOMBYX MORI L. (가잠사단백질의 각과정에서의 Cystine 성분에 대한 연구)

  • The purpose of this treatise is to prove the presence of cystine in silk fiber through wide sampling throughout all the sericultural processes of Bombyx mori.; also to show that disulfide cross linkages exist in the silk fiber. The conclusions reached were as follows: 1. Crystalline cystine was obtained from silk fibroin using Folin's Method. 2. Analytical data showing the cystine content of silk fiber and its related materials were obtained using Sullvan's Method as follows: Material Percent Cystine A. Mulberry leaf protein 0.175 B. Silkworm egg 0.33 C. Silkworm Body, matured, fat extracted, without silk gland 0.41 D. Silk gland, matured 1.23 E. Silkworm feces none F. Silkworm pupa, fat extracted 0.30 G. Silkworm moth, fat extracted 0.60 H. Raw Silk 0.22 I. Fibroin 0.175 J. Sericin 0.30 3. The presence of cystine in the silkworm was substantiated the existence of 0.175 % methionine in mulberry leaves and 0.12% methionine in the silk gland. 4. Part of the sulfhydryl compounds in the silk gland is believed to transfer to serine and methionine, with the former being secreted into the liquid silk finally as silk fiber and the latter used for nutritive purposes in the growing of silk gland tissue. 5. The cystine content is variable by mulberry species, silkworm species, sex, breeding process, and other culturing environments. 6. Hybrid silkworms require more nutritive amino acids for effective growth than the original parents, and secrete less of them as silk fiber. 7. From such an observation, the amino acid composition of silk fiber is believed to be fairly flexible. Cystine if included in the amorphous part of the fiber, especially in sericin. 8. The result from enriching the silkworm diet with pure cystine or wool cystine did not result in any advantage, therefore it is believed that the natural cystine and methionine contents in the mulberry leafaregoodenoughforsilkwormnutrition. 9. The disulfide cross linkage in silk fiber was verified by using the Harris Method. Contraction took place following the treatment of the fiber with various salts and acids. Comparisons were made with wool fiber. 10. During these experiments, the fibrious structure of silk fiber and the net-globular liquid form were photographed microscopically. It is believed that the globules of liquid silk are net-formed by the inter attraction of the OH ion of the globular peptide and the H ion of water as shown by the hair cracking behavior of the film. The net-globular protein precipitation from the mulberry protein solution showed that mulberry is a proper diet for the formation of fibrous protein in the silk fiber. 11. The significance of the presence of cystine in silk fiber as emphasized in this paper should result in modification of the general conception that cystine is absent from this fiber. ANTIMICROBIAL ACTIVITY OF ARTEMISIA SPECIES AGAINST CLINICALLY ISOLATED STREPTOCOCCUS MUTANS (임상 분리된 Streptococcus mutans에 대한 Artemisia species의 항균 활성도)

  • Streptococcus mutans plays a major role in the formation of dental plaque and it is considered one of the important pathogens in the development of dental caries. Established dental plaque can be more resistant to antimicrobial agents and offers nutrient rich and relatively stable cariogenic conditions for bacteria. Thus bacteria growing in dental plaque have strong resistance to antimicrobial agents and they are not removed easily by the flow of saliva. Many researchers have been performed using natural materials, especially herbal extracts to prevent dental plaque. However, the strains of mutans streptococci used in the researches were not from Koreans. Therefore, it would be necessary to evaluate the antimicrobial activity of herbal extracts against clinical isolates of Streptococcus mutans isolated from saliva of Koreans living in Jeollabuk-do. For this study four clinical isolates were isolated from saliva samples of seventeen Korean people to investigate whether essential oils of Artemisia spp. have antimicrobial activity against them including four reference strains of Streptococcus mutans. Minimum inhibitory concentrations, Minimum bactericidal concentrations and time kill studies were performed and the results showed that Artemisia lavandulaefolia DC, Artemisia scoparia, and Artemisia capillaries have antimicrobial activity against the Streptococcus mutans clinical isolates and reference strains.

An Approach of Ecological Niche to Analysis of Recognition of 5th Grade Elementary students for Conception of Photosynthesis (생태 지위적 접근을 통한 5학년의 광합성 개념 분석)

  • There have been studies about conceptual ecology making a profound study of conceptual changes in learners' cognitive structure. Because learners' cognitive structure have been compared to ecology, it is natural to think that conception in learner's cognitive structure have a niche as species in ecology have niches. Therefore, it is necessary to study niche approach about conception that learners recognize in their cognitive structure. The purposes of this study were to identify relationships among conceptions that 5th grade elementary school students recognize about photosynthesis and to identify how these relationships among conceptions about photosynthesis change before and after a class of photosynthesis in curriculum in terms of an approach of ecological niche which are composed of 3 domains - diversity of conceptions, relevance and frequency rate of conceptions, and competition among conceptions. Open ended questionnaire was developed by 4 fields: photosynthetic place, photosynthetic products, photosynthetic materials needed and environment factors of photosynthesis. The subjects sampled in this study were 310 5th grade elementary students in 5 cites. Before and after classes in photosynthesis in science curriculum, students were asked to write down conceptions that they knew about the 4 fields of photosynthesis of questionnaire and to write down scales of relevance from 1 to 30 about how they think the conceptions are related to the field of photosynthesis. The results of this study showed the following: First, most students have had a variety of conceptions and commonly recognized 'light' and 'water' as concepts in photosynthesis. Second, students still recognized their preconceptions like 'soil' and 'root,' etc. that were far from scientific conceptions of photosynthesis although they took classes in photosynthesis. Third, students needed to take the various strategies of teachers because they did not recognized scientific conceptions appropriately about photosynthetic fields. Fourth, it appeared that photosynthetic conceptions recognized by students had status in terms of relevance and frequency rate of conceptions, and competition among conceptions, and that they looked like the niche of conceptions in their conceptual ecologies.

Development of Functional Food Materials from Acanthopanax senticosus-Fermented Mushroom Mycelia (가시오가피 버섯균사체 발효물의 기능성 식품 소재 개발)

  • Three mushroom mycelia, Ganoderma lucidum, Hericium erinaceum, and Phellinus linteus, were separately diluted with the natural culture media Acanthopanax senticosus. Solid-state fermentation was used to produce three different A. senticosus-fermented mushroom mycelium groups: G. lucidum mycelia, H. erinaceum mycelia, and P. linteus mycelia. The resulting mycelia were analyzed to assess their efficacies as health functional foods. Optimized fermentation conditions were determined by considering the density and growth speed of mycelia in each A. senticosus-fermented mushroom mycelium group. The cultured mushroom mycelia under the optimized conditions were extracted using water and 70% ethanol. Extraction was followed by filtration, concentration and freeze-drying to produce extract powder of A. senticosus-fermented mushroom mycelia: Water extracts (FM-5111, FM-5121, and FM-5131) and 70% ethanol extracts (FM-5112, FM-5122, and FM-5132). Analysis of extract powder of A. senticosus-fermented mushroom mycelia was performed using the maker compounds eleutheroside B and eleutheroside E. Analysis of ${\beta}$-glucan contents was performed by enzymatic procedures.

Geochemistry and Water Quality in the Tidal Flat of Saemangum Area, West Sea of Korea in Summer (하계 새만금 갯벌의 수질 분포 및 지화학적 특성에 관한 연구)

  • Environmental quality(water and sediment) was analyzed in the tidal flat of Saemangum of Jeonbuk Province, the west coast of Korea, using the 101 sediment samples and 69 water samples collected in September 4~13, 2001. Major water quality parameters with the means of 69 surface water samples are as follows; $25.51{\pm}0.68^{\circ}C$ for water temperature, $29.88{\pm}5.01$ for salinity, $1.40{\pm}0.78mg/L$ for COD, $0.352{\pm}0.417mg/L$ for DIN, and $0.027{\pm}0.023mg/L$ for phosphate, respectively. Higher values were found at the subestuary of Dongjin and Mangyung River, and lower values at the Saemangum embayment and Gomso Bay. There was a significant negative correlation between salinity and the other water quality parameters(p<0.0001) such as COD, nutrients, SS and N/P. This correlation suggested that the major pollution sources be from terrestrial inputs through tributaries in this area. Principal component analysis clearly revealed a spatial variation of water quality; stations with higher values of nutrients and COD located subestuary of tributaries. 14 sediment quality parameters including 8 trace metals were measured using the 101 surface sediment samples. Average values for the parameters are as follows; Al $2.28{\pm}0.92%$, Cd $0.61{\pm}0.27ppm$, Cu $8.95{\pm}4.06ppm$, Fe $1.19{\pm}0.37%$, Mn $182.31{\pm}77.45ppm$, Ni $10.83{\pm}4.97ppm$, Pb $15.20{\pm}4.35ppm$, Zn $41.34{\pm}34.62ppm$, COD $2.68{\pm}1.85mg/g\;dry$, AVS $0.04{\pm}0.08mg/g\;dry$, IL $1.29{\pm}1.08%$, water content $24.11{\pm}4.49%$, TN $0.02{\pm}0.02%$, TC $0.22{\pm}0.30%$. Spatial variations of sediment quality were not clear as water quality. Some higher values were found at the subestuary of Gum River and lower values at the other area. There was a significant positive correlation between the heavy metal concentrations and organic materials within the sediment(p<0.05). Enrichment factors showed the ranges of 1~2 for most of the metals in the sediment except zinc(1~6), indicating no serious exogenous input of heavy metals in the study area. Also, the heavy metal concentrations in the sediment were within the ranges found at the natural marine environments.

Composting of the Pig Manure Used with the Ceramic Biocarrier (복합세라믹 담체를 이용한 돈분 퇴비화 연구)

  • This study was executed to evaluate the utilization and efficiency of the ceramic biocarrier as the promoter of decomposing on the organic matters for the composting using with pig manure by analyzing of the physico-chemical properties during composting. The treatments of this experiment were consisted of the control(C),microorganism(M), M with natural zeolite(M+Z), M with synthesized zeolite(M+SZ), and M with ceramic biocarrier(M+CZ). The process term of composting was conducted for 30days in the rapidly fermented machine(as pilot system). The results of the physico-chemical properties of the composts were as follows. The changes of temperature during composting was not relative with the microorganism and zeolite materials used in the composts. At all of the treatments were similar to changing of temp. from the initial stage to the final stage. But the added microorganism treatments were higher than control. And the entire pH value of treatments were appeared the same that above temperature result, also the M+CZ and M+SZ treatment among the treatment were higher. At the results of T-C, T-N and C/N ratio, in case of T-C value, the M+CZ treatment was highly more decreased than others. However at the T-N value, there were not the differences from the each treatment. And the C/N ratio was changed according to the changes of T-C and T-N value. Especially, at the M+CZ aud M+SZ treatments were remarkably reduced by about 21.4-23.3 value. In the result of G.I for evaluating of the compost humidity, the M+CZ and M+SZ treatments were close up approximately 110 value compared with the control(G.I value 100). Therefore, the examined ceramic biocarrier amended with compost-promoting-bacteria could be applied to the production of many high quality fertilizers. It is also expected that the results of this researches could be applied to the recycle of the organic wastes based on the experimental results of ceramic biocarrier and compost-promoting-bacteria application.

Hydrogeochemical Characteristics, Occurrence, and Distribution of Natural Radioactive Materials (Uranium and Radon) in Groundwater of Gyeongnam and Gyeongbuk Provinces (경상남북도 지하수 중 자연방사성물질 우라늄과 라돈의 산출특징과 함량분포에 대한 수리지화학적 연구)

  • The occurrence, distribution, and hydrogeochemical characteristics of uranium and radon in groundwater within different lithologies in Gyeongnam and Gyeongbuk provinces were investigated. A total of 201 groundwater samples from sedimentary rocks taking a large portion of the geology and from igneous rocks taking a small portion of the geology were analyzed and examined using factor analysis. Their radionuclide levels were used to construct detailed concentration maps. The groundwater types, defined using a Piper diagram, are mainly Ca-$HCO_3$ with less Na-$HCO_3$. Among the samples, one site exceeds $30{\mu}g/L$ of uranium (i.e., the maximum contaminant level of the USEPA) and three sites exceed 4,000 pCi/L of radon (i.e., the alternative maximum contaminant level). No samples were found to exceed the 15 pCi/L level of gross alpha or the 5 pCi/L level of radium. The concentration of uranium ranges from 0.02 to $53.7{\mu}g/L$, with a mean of $1.56{\mu}g/L$, a median of $0.47{\mu}g/L$, and a standard deviation of $4.3{\mu}g/L$. The mean concentrations of uranium for the different geological units increase in the following order: Shindong Group, Granites, Hayang Group, Yucheon Group, and Tertiary sedimentary rocks. The concentration of radon ranges from 2 to 8,740 pCi/L, with an mean of 754 pCi/L, a median of 510 pCi/L, and a standard deviation of 907 pCi/L. The mean radon concentrations for the investigated geological units increase in the following order: Granites, Yucheon Group, Tertiary sedimentary rocks, Hayang Group and Shindong Group. According to the factor analysis for each geological unit, uranium and radon behave independently of each other with no specific correlation. However, radionuclides show close relationships with some components. Regional investigations of radionuclides throughout the country require an integrated approach that considers the main lithological units as well as administrative districts.

The Comparison of DVH between Multiple arc FSRT and Conformal FSRT (Multiple arc FSRT와 Conformal FSRT의 DVH 비교)

  • Purpose : In FSRT (Fractionated stereotactic radiotherapy) planning, we studied the usefulness between multiple arc FSRT and conformal FSRT by comparing tumor shape and DVH (dose volume histogram). Materials and Methods In Chungnam Univ. hospital, we had treated the sixteen patients with FSRT from Aug. 1997 to Dec. 1998. In choosing multiple arc FSRT or conformal FSRT, we had considered If (irregular factor) after calculating tumor volume and surface area. We had considered multiple arc FSRT if tumor shape was similar to sphere or the value of If was less than 1.25, conformal FSRT if tumor shape was very irregular or If was more than 1.3. For evaluation of treatment planning, we had considered the appropriate DVH for tumor volume and for critical organs. Results : The errors between reference point and the coordinates point on AP, Lat radiography were less than 1 mm before treatment. We had planned $3\~$5 arcs for multiple arc FSRT, $5\~6$ports for conformal FSRT. The mean dose distribution of tumor volume of cumulative DVH between multiple arc FSRT and conformal FSRT was 90.6, 85%, respectively. The dose of critical organs irradiated was less than $5\%$ maximum dose of cumulative DVH. Conclusion : We had obtained the similar value between multiple arc FSRT and conformal FSRT, so that we had appropriate treatment planning of FSRT for multiple arc FSRT and conformal FSRT according to tumor shape and size.

Effect of Combining Wood Particles and Plastic(Polypropylene) Screen on the Physical and Mechanical Properties of Board (목재(木材)파이티클과 플라스틱(폴리프로필렌) 망(網)의 결체(結締) 보오드의 물리(物理) 및 기술적(機械的) 성질(性質)에 미치는 영향(影響))

  • As a way for the effective utilization of pallman chips and sawdusts, these furnish materials were combined with non-woody material of plastic (polypropylene) screen in board manufacturing to improve their weak physical and mechanical properties. The conventional boards were made with conditions of specific gravity 0.40, 0.55, 0.70, and 0.85, resin content 8, 10, 12 and 14%, and number of polypropylene screen 1, 2, 3 and 4, and press-lam boards were also manufactured. The physical and mechanical properties were measured and discussed on thickness swelling, bending modulus of rupture and elasticity, tensile strength, internal bond strength, and screw holding strength. The results obtained at this study were summarized as follows: 1. In thinckness swelling both of pallman chip board and sawdust board were improved by the increase of resin content, and press-lam boards showed lower thickness swelling than conventional boards. 2. Both the modulus of rupture and elasticity were increased with the increase of specific gravity, and press-lam boards showed higher modulus of rupture and elasticity than conventional boards. On the other hand, modulus of rupture was increased with the increase of number of polypropylene screen and resin content whereas these effects in modulus of elasticity was not recognized. 3. Tensile strength was increased with the increase of specific gravity, and the boards combined with polypropylene screen showed higher tensile strength than control boards. Also tensile strength was increased with the increase of number of polypropylene screen, and press-lam boards revealed higher tensile strength than conventional boards. 4. Internal bond strength was increased with the increase of specific gravity, and the boards combined with polypropylene screen were lower in internal bond strength than control boards. Also, the boards combined with odd number of polypropylene screen showed lower internal bond strength than those combined with even number of polypropylene screen. 5. Screw holding strength was increased with the increase of resin content and specific gravity but significant difference was not approved between boards combined with polypropylene screen and control boards. In press-lam boards, pallman chip boards of higher specific gravity but sawdust boards of lower specific gravity showed better screw holding strength than control boards.

Mineralogical Characteristics of Carbonate Rock-Hosted Naturally Occurring Asbestos from Asan, Muju, Jangsu Areas (국내 탄산염암 지역(아산, 무주, 장수)에서 산출되는 자연발생석면의 광물학적 특성)

  • Naturally occurring asbestos (NOA) occurs in rocks and soils as a result of natural weathering and human activities. It is proved that inhalation of asbestos fibers can lead to increase risk of developing several diseases such as lung cancer and malignant mesothelioma. The parent rocks of asbestos have been mainly associated with (ultra)mafic and carbonate rock. The previous studies on NOA were mainly limited to (ultra)mafic rock-hosted asbestos, but studies on carbonate rock-hosted asbestos are relatively rare in S. Korea. Therefore, this study was aimed to examine mineralogical characteristics of carbonate rock-hosted NOA at three sites including Muju and Jangsu, Jeonbuk province and Asan, Chungnam province. Types of rocks at the three sites mainly consisted of Precambrian metasedimentary rocks, carbonate rock, and Cretaceous and Jurassic granites. Asbestos-containing carbonate rock samples were obtained for mineralogical characterization. XRD, PLM, EPMA, SEM and EDS analyses were used to characterize mineralogical characteristics of the carbonate rock-hosted NOA. From the carbonate rock, fibrous minerals were occurred acicular and columnar forms in the three sites. Fibrous minerals were composed of mainly tremolite and associated minerals included possibly asbestos containing materials (ACM) such as talc, vermiculite, and sepiolite. The length and aspect ratios of tremolite were similar to the standard asbestiform (length >$5{\mu}m$, length:width = 3:1). These results indicate that both non-asbestiform and asbestiform tremolite with acicular forms occurred in carbonate rocks at three sites. Geological and geochemical characteristics and mineral assemblages indicate tremolite and associated minerals might be formed by hydrothermal alternation and/or hydrothermal veins of carbonate rocks due to intrusion of acidic igneous rocks.
