도심 엔터테인먼트형 복합상업시설 지하공간에 자연요소를 이용한 실내 환경디자인 방안에 관한 연구 (A Study on Application of the Interior Environmental Design Using Natural Elements in Underground Space of Urban Entertainment Center)

  • The underground space of urban entertainment center can do various roles for urban development. Especially, the design of underground space using natural elements is necessary to sustainable development of facilities. The purpose of this study is to propose the elements, characteristics and application around interior environmental design using natural elements in underground space of urban entertainment center. The research methods of this study are research references, survey and field-study. This study can be concluded as follows; First, the factors of interior environmental design using natural elements in underground space of urban entertainment center are 'landmarks expression of natural elements', 'introduction and planning of daylighting', 'planning of materials using natural images', 'planning of ecological footpath' and 'planning of energy recycling'. Second, pedestrian space and openspace considering regional context are necessary. Third, introduction of daylight and design of artificial lighting in underground space are needed to making eco-friendly environment. Fourth, landmark design using various natural elements in underground space is needed to provide eco-friendly amenity. Fifth, the interior environmental design using natural elements is needed to community space in underground space. Sixth, eco-friendly pedestrian space in underground space is necessary. Seventh, interior environmental design using natural elements is needed to the entrance of underground space of urban entertainment center.

  • This research aimed for revealing social interaction-based environmental learning elements at the natural history museum. First, we carried out a survey of the perceptual change about 'the environment' concept. It was for confirming whether environmental learning occurred in visiting. Second, we found environmental learning elements through analyzing dialogues among small group members during looking around the museum. Social interaction-based environmental learning elements were classified into five; knowledge, affection, value, skill, behaviour. Each environmental learning element included several sub-elements, so we revealed totally 9 environmental learning sub-elements. By results, we said characteristics and possibility of the natural history museum as an informal environmental educational institution. It is meaningful for extending areas of environmental education.

  • Recently, as people have more interest in health, and some change takes place in their lifestyles, a demand for environment-friendly residential space is also increasing. Research on the introduction of natural elements is mostly conducted for humans to pursue a more pleasant and comfortable life in residential space since it has one of the closest relation with humans. Therefore, it is such an important assignment at present when environmental problems are considered significant. Based on such a necessity, this study analyzed the introduction types and characteristics of natural elements in residential space by considering essential and psychological aspects more than previous researches on residential space with practical and functional aspects. As natural elements, this study put limits on plants and water that are used a lot in the interior space, and by dividing introduction types into recomposition of courtyard space, spatial partition and decorative elements, this study carried out a case analysis of residential space structures recently completed in Japan. As a basic research on space planning that is desperately required by change in the residential life paradigm and on the basis of understanding about nature, it is expected that the results of this study will be used as design basic data useful when establishing plans of introducing natural elements for residential space in the future.

유한요소법에 의한 2차원 하천 흐름 모형의 개발 (Two-Dimensional River Flow Analysis Modeling By Finite Element Method)

  • The understanding and prediction of the behavior of flow in open channels are important to the solution of a wide variety of practical flow problems in water resources engineering. Recently, frequent drought has increased the necessity of an effective water resources control and management of river flows for reserving instream flow. The objective of this study is to develop an efficient and accurate finite element model based on Streamline Upwind/Petrov-Galerkin(SU/PG) scheme for analyzing and predicting two dimensional flow features in complex natural rivers. Several tests were performed in developed all elements(4-Node, 6-Node, 8-Node elements) for the purpose of validation and verification of the developed model. The U-shaped channel of flow and natural river of flow were performed for tests. The results were compared with these of laboratory experiments and RMA-2 model. Such results showed that solutions of high order elements were better accurate and improved than those of linear elements. Also, the suggested model displayed reasonable velocity distribution compare to RMA-2 model in meandering domain for application of natural river flow. Accordingly, the developed finite element model is feasible and produces reliable results for simulation of two dimensional natural river flow. Also, One contribution of this study is to present that results can lead to significant gain in analyzing the accurate flow behavior associated with hydraulic structure such as weir and water intake station and flow of chute and pool.

Identification and Molecular Characterization of PERV Gamma1 Long Terminal Repeats

  • Porcine endogenous retroviruses (PERVs) gamma1 in the pig genome have the potential to act as harmful factors in xenotransplantation (pig-to-human). Long terminal repeats (LTRs) are known to be strong promoter elements that could control the transcription activity of PERV elements and the adjacent functional genes. To investigate the transcribed PERV gamma1 LTR elements in pig tissues, bioinformatic and experimental approaches were conducted. Using RT-PCR amplification and sequencing approaches, 69 different transcribed LTR elements were identified. And 69 LTR elements could be divided into six groups (15 subgroups) by internal variation including tandem repeated sequences, insertion and deletion (INDEL). Remarkably, all internal variations were indentified in U3 region of LTR elements. Taken together, the identification and characterization of various PERV LTR transcripts allow us to extend our knowledge of PERV and its transcriptional study.

Identification and Phylogenetic Analysis of the Human Endogenous Retrovirus HERV-W LTR Family in Placenta cDNA Library

  • Human endoqenous retroviral long terminal repeats (LTRs) have been found to be coexpressed with sequences of genes closely located nearby. It has been suggested that the LTR elements have contributed to structural changes or genetic variations of human genome connected to various diseases and evolution. Using cDNA library derived from placenta tissue, we performed PCR amplification and identified five new HERV-W LTR elements. Those LTR elements showed a high degree of sequence similarity (98-99%) with HERV-W LTR (AF072500). A phylogenetic tree obtained by the neighbor-joining method revealed that HERV-W LTR elements could be mainly divided into two groups through evolutionary divergence. Five new HERV-W LTR elements (pla-1, 4, 5, 6, 7) belonged to the group I with AX000960, AF072504, and AF072506 from GenBank database. The data suggest that several copy numbers of the HERV-W LTR elements are transcribed in placenta and may contribute to the understanding of biological function such as human placental morphogenesis.

  • Long terminal repeats(LTRs) of the human endogenous retrovirus(HERV) heve been found to be coexpresed with genes located nearby. It has been suggested that the LTR elements have contributed to the genetic variation of human genome connected to various diseases. Recently, HERV-W family was identified in the cerebrospinal fluids and brains of individuals with schizophrenia. Using cHNA library derived from human fetal brain, we performed PCR amplification and identified seven new HERV-W LTR elements. Those LTR elements showed a high degree of sequence similarity(98∼99%) with HERV-W (AF072500). A phylogentic tree obtained by the neighbor-joining method revealed that seven new HERV-W LTR elements(FB-1, 2, 4, 8, 9, 10, 12) were closely related to the AX000960, AF072504, and AF072506 from Gen Bank database. Our data suggest that several copy numbers of the HERV-W LTR elements are expressed in human feta brain and may contribute to an understanding of biological function connected to neuropsychiatric diseases.

노인요양시설 공용공간의 자연요소 도입에 관한 연구 - 매개, 교류, 휴식공간의 쾌적성 확보를 중심으로 - (A Study on the Introduction of Natural Elements in Public places of Nursing Homes - Focusing on ensuring the amenity of mediation, interaction, and rest places -)

  • As an aging society accelerates its pace for the present, providing a pleasant environment to nursing homes is such an environmental requirement with which elderly people could improve their quality of life and even acquire the effects of healing. As an optimal element of ensuring amenity, nature is a general hospital that heals human beings physically and mentally. In this vein, the investigator drew out the frame of analysis, namely emotional/healing/relating amenity, by means of theoretical examination of such natural elements as water, light, soil/stone, and plant. And on the basis of the drawn amenity, the investigator examined the application of natural elements to study subjects in order to analyze the ensured amenity. Study findings show that the existing nursing homes have ensured healing amenity sufficiently, but they are unsatisfactory in ensuring emotional and relating amenity. Based on interview survey, the investigator suggests that the introduction of natural elements into nursing homes such public space as mediation space, interaction space, and rest places to ensure emotional/healing/relating amenity must be ideally equipped with the formation of outdoor forest in which water, light, soil/stone, and plant get joined together, rather than Introducing a single element respectively. In addition, the plan of construction of nursing homes must take residing elderly people's emotional/healing/relating amenity into account on the basis of physical amenity.

Structure and Expression Analyses of SVA Elements in Relation to Functional Genes

  • SINE-VNTR-Alu (SVA) elements are present in hominoid primates and are divided into 6 subfamilies (SVA-A to SVA-F) and active in the human population. Using a bioinformatic tool, 22 SVA element-associated genes are identified in the human genome. In an analysis of genomic structure, SVA elements are detected in the 5′ untranslated region (UTR) of HGSNAT (SVA-B), MRGPRX3 (SVA-D), HYAL1 (SVA-F), TCHH (SVA-F), and ATXN2L (SVA-F) genes, while some elements are observed in the 3′UTR of SPICE1 (SVA-B), TDRKH (SVA-C), GOSR1 (SVA-D), BBS5 (SVA-D), NEK5 (SVA-D), ABHD2 (SVA-F), C1QTNF7 (SVA-F), ORC6L (SVA-F), TMEM69 (SVA-F), and CCDC137 (SVA-F) genes. They could contribute to exon extension or supplying poly A signals. LEPR (SVA-C), ALOX5 (SVA-D), PDS5B (SVA-D), and ABCA10 (SVA-F) genes also showed alternative transcripts by SVA exonization events. Dominant expression of HYAL1_SVA appeared in lung tissues, while HYAL1_noSVA showed ubiquitous expression in various human tissues. Expression of both transcripts (TDRKH_SVA and TDRKH_noSVA) of the TDRKH gene appeared to be ubiquitous. Taken together, these data suggest that SVA elements cause transcript isoforms that contribute to modulation of gene regulation in various human tissues.

Phylogenetic Analysis of HERV-K LTR Family in Human Chromosome Xq26 and New World Monkeys

  • Solitary long terminal repeats(LTRs) of human endogenous retrovirus K family(HERV-K) have been found to be coexpressed with sequences of closely located genes. It has been suggested that HERV-K LTR-like elements entered the primate genome approximately 33-40 million years ago. WE investigated the presence of HERV-K LTR elements in New World monkeys using PCR amplification. Six LTR elements of HERV-K family were identified from New World monkeys, represented by the squirrel and night monkeys. They showed a high degree of sequence homology(96-99%) with the human-specific HERV-K LTR elements. Phylogenetic analysis reveals that an LTR element (SM-1) from the squirrel monkey and another LTR element (NM-1) from the night monkey are very closely related to the human-specific HERV-K LTR elements with low degree of divergence. This finding suggests that some of LTR elements of HERV-K family have recently been proliferated in New World monkeys. A sequence in chromosome Xq26(AL034407) \ulcorner contains an HERV-K LTR element was shown to be present in the human genome, but is absent in the bonobo, chimpanzee, gorilla, orangutan, and gibbon. It has more than 99% homology to other human-specific HERV-K LTR elements. This sequence thus represents and isolated insertion of an evolving class of elements that may have made a particular contribution to human genomic plasticity.
