The degradation behavior of berberine is examined using GC-MS to select the fingerprint products that can be used to identify berberine dye in badly faded archaeological textiles. A total of $100^{\circ}C$ thermal and $H_2O_2/UV/O_2$ degradation systems were used to degrade berberine chloride 0.1% solution up to 408 hours. The samples were analyzed using the GC-MS. Dihydroberberine, 2-pteridinamine, 6,7-dimethyl-N-[(trimethylsilyl) oxy]-, and 8-methoxy-11-[3-methylbutyl]-11H-indolo[3,2-c]-quinoline, 5-oxide were detected as the major products of thermal degradation and identified as the fingerprint products for berberine dye at the early stage of degradation. Isobenzofuran-1,3-dione,4,5-dimethoxy-, 9H-fluorene,3,6-bis(2-hydroxyethyl)-,1,3-dioxolo[4,5-g]isoquinolin-5(6H)-one,7,8-dihydro-, and 3-tert-butyl-4-hydroxyanisole were detected as the major products generated by the $H_2O_2/UV/O_2$ degradation and identified as the fingerprint products for berberine dye under severe degradation conditions.
Tissue engineering is one exciting approach to treat patients who need a new organ or tissue. A critical element in this approach is the polymer scaffold, as it provides a space for new tissue formation and mimics many roles of natural extra-cellular matrices. In this review, we describe several design parameters of polymer matrices that can significantly affect cellular behavior, as well as various polymers which are frequently used to date or potentially useful in many tissue engineering applications. Interactions between cells and polymer scaffolds, including specific receptor-ligand interactions, physical and degradation feature of the scaffolds, and delivery of soluble factors, should be considered in the design and tailoring of appropriate polymer matrices to be used in tissue engineering applications, as these interactions control the function and structure of engineered tissues.
The transformation capacity (T$\_$c/) of Methylosinus trichosporium OB3b in the degradation of ethylene chlorides was determined by measuring the decrease of soluble methane monooxygenase (sMMO) activity of resting cells in batch experiments. All measurements of sMMO activity were taken in the presence of 20 mM formate to avoid the deficiency of reducing power, and within 2 hrs to avoid the effect of natural inactivation from instability of the resting cells. The constant T$\_$c/ values of 0.58 ${\pm}$ 0.132 and 0.80 ${\pm}$ 0.17 ${\mu}$mol/mg cell were obtained for trichloroethylene (TCE) and 1,2-dichloroethylene (cis and trans-1,2-DCE), respectively, regardless of their concentrations. The transformation capacity measured by this method can be used to predict the amount of cells that should be stimulated in in-situ bioremediation.
To develop an artificial bone substitute that is gradually degraded and replaced by the regenerated natural bone, the authors designed and produced a composite that is consisted of calcium phosphate and collagen. Human umbilical cord origin pepsin treated type I atelocollagen was used as the structural matrix, by which sintered or non-sintered carbonate apatite was encapsulated to form an inorganic-organic composite. With cross linking atelocollagen by UV ray irradiation, the resistance to both compressive and tensile strength was increased. Collagen degradation by the collagenase induced collagenolysis was also decreased.

Nepal is rich country in terms of its vast array of biological diversity. The relationship between local communities and the biological diversity in Nepal is a very entangle one, whereby two-thirds of our population are heavily dependent upon the biodiversity for their survival. The destruction of biodiversity signifies the destruction of people's livelihoods and survival. Biodiversity and genetic resources conservation has been a most significant part of the Nepalese society due to its socio-economic and cultural importance and provides the human race trees for timber, fuel, food, industrial raw materials, pharmaceuticals, beverages and environmental stabilization. Threats to biodiversity at global level is of greater concern, specially, human activities are the greatest factor currently affecting biodiversity, through the use of natural resources by industry, agriculture, hunting, and energy generation.
Natural lacquer was cured at room temperature at a relative humidity of 60% at $27^{\circ}C$ to obtain a lacquer film. Photolysis of lacquer was investigated by irradiating 1~75 kGy of gamma ray to this lacquer film. No significant changes were observed in the range of 1~10 kGy radiation. As the radiation doses were increased, the white spots were increased in the scanning electron microscope photographs. The increase of the white dots of the lacquer surface shows that the lacquer is degradation by radiation. As a result of IR spectral analysis of the lacquer surface with increasing irradiation dose, there was no significant change in $3,445cm^{-1}$ hydroxide group, $2,900cm^{-1}$ hydrocarbon group, and $993cm^{-1}$ triene group. However, the $1,745cm^{-1}$ carbonyl group was found to increase, and the $1,715cm^{-1}$ unsaturated hydrocarbon group and the $1,463cm^{-1}$ methylene group showed a tendency to decrease.
The purpose of this study is to explore the ecosystem service and benefit indicators of natural seaweed beds. Ecosystems of natural seaweed beds provide a wide range of services and benefits to human society including provisioning services, regulating services, supporting services, and cultural services. Indicators for each of the ecosystem services are chosen by marine plants ecologists and as follows. Ecosystem indicators of natural seaweed beds for provisioning services are well-being food(amount of seaweed harvested/amount of fish landed, fish biomass, area of natural seaweed beds, the number of species, contribution to the second production), raw materials(amount of biomass by breed, amount of aquaculture feed), genetic resources(amount of genetic material extracted, amount of genetic material contained by age and habitat), and medicinal resources(amount of medicinal material extracted). Ecosystem indicators of natural seaweed beds for regulating services are air purification(amount of fine dust/NOx or $SO_2$ captured), climate regulation(amount of $CO_2$ sequestered), waste treatment(amount of N, P stored, biochemical degradation capacity COD), and costal erosion prevention(length and change of natural coast line, amount of sediment prevented). Ecosystem indicators of natural seaweed beds for supporting services are lifecycle and maintenance(primary production, contribution to the second production) and gene pool protection(amount of compositional factors in ecosystem, introduced species). Ecosystem indicators of natural seaweed beds for cultural services are recreation and tourism(the number of visits of an area) and information for cognitive development(amount of time spent in education, research and individual learning about ecosystem of natural seaweed beds).
Focusing on quality problems of delayed chilling porcine muscle, the effects of delayed chilling and aging on the contents of ATP-related compounds and taste of pork were investigated. Twelve Landrace pigs were employed and bisected: left sides were delay-chilled(DC) at room temperature($20^{\circ}C$) for 3 hrs, whereas right sides were conventionally chilled(CC). ATP-related compounds tested were adenosine triphosphate(ATP) and its derivatives in pork muscle, inosine monophosphate(IMP), guanosine monophosphate(GMP) and L-glutamate in cooked broth. DC sides showed more rapid pH decline and degradation of nucleotides than did CC sides. The levels of ATP and adenosine monophosphate(AMP) were not changed significantly. However, adenosine diphosphate(ADP) and IMP showed the highest levels at the 1st and 5th day, respectively. Hypoxanthine(Hx) was gradually increased(p<0.05) during aging. During aging, the IMP contents cooked broth tended to decrease, while the GMP and L-glutamate contents increase. As a result of these, the taste score got better and finally the results of sensory evaluation became increased(p<0.05). However, compared to CC sides, DC sides did not seem to lower taste of pork.
Human influence on soil formation has dramatically increased with human civilization and industry development. Increase of anthropogenic soils induced researches on the anthropogenic soils; classification, chemical and physical characteristics of anthropogenic soils and plant growth from anthropogenic soils. However there have been no comprehensive analyses on soil pore or physical properties of anthropogenic soils from 3 dimensional images in Korea. The objectives of this study were to characterize physical properties of anthropogenic paddy field soils by depth and to find differences between natural and anthropogenic paddy field soils. Soil samples were taken from two anthropogenic and natural paddy field soils; anthropogenic (A_c) and natural (N_c) paddy soils with topsoil of coarse texture and anthropogenic (A_f) and natural (N_f) paddy soils with topsoil of fine texture. The anthropogenic paddy fields were reestablished during the Arable Land Remodeling Project from 2011 to 2012 and continued rice farming after the project. Natural paddy fields had no artificial changes or disturbance in soil layers up to 1m depth. Samples were taken at three different depths and analyzed for routine physical properties (texture, bulk density, etc.) and pore properties with computer tomography (CT) scans. The CT scan provided 3 dimensional images at resolution of 0.01 mm to calculate pore radius size, length, and tortuosity of soil pores. Fractal and configuration entropy analyses were applied to quantify pore structure and analyze spatial distribution of pores within soil images. The results of measured physical properties showed no clear trend or significant differences across depths or sites from all samples, except the properties from topsoils. The results of pore morphology and spatial distribution analyses provided detailed information of pores affected by human influences. Pore length and size showed significant decrease in anthropogenic soils. Especially, pores of A_c had great decrease in length compared to N_c. Fractal and entropy analyses showed clear changes of pore distributions across sites. The topsoil layer of A_c showed more degradation of pore structure than that of N_c, while pores of A_f topsoil did not show significant degradation compared with those of N_f. These results concluded that anthropogenic soils with coarse texture may have more effects on pore properties than ones with fine texture. The reestablished paddy fields may need more fundamental remediation to improve physical conditions.
This study was conducted to isolate and characterize a novel feather-degrading bacterium producing keratinase activity. A strain K9 was isolated from soil at poultry farm and identified as Xanthomonas sp. K9 by phenotypic characters and 16S rRNA gene analysis. The cultural conditions for the keratinase production were 0.3% fructose, 0.1% gelatin, 0.04% $K_2HPO_4$, 0.06% $KH_2PO_4$, 0.05% NaCl and 0.01% $FeSO_4$ with an initial pH 8.0 at $30^{\circ}C$ and 200 rpm. In an optimized medium containing 0.1% chicken feather, production yield of keratinase was approximately 8-fold higher than the yield in basal medium. The strain K9 effectively degraded chicken feather meal (67%) and duck feather (54%), whereas human nail and human hair showed relatively low degradation rates (13-22%). Total free amino acid concentration in the cell-free supernatant was about 25.799 mg/l. Feather hydrolysate produced by the strain K9 stimulated growth of red pepper, indicating Xanthomonas sp. K9 could be not only used to increase the nutritional value of chicken feather but also a potential candidate for the development of natural fertilizer applicable to crop plant soil.
