Characteristics Analysis of Site Condition and Disturbance Resistance of Tree Species for Damaged Forested Land in South Korea (산림재해지 복구를 위한 주요 수종의 입지 및 재해 저항 특성 분석)

  • We recently witnessed increasingly more natural disturbances on forested land. On the other hand, only limited number of nursery grown tree species are available for restoration projects. At the same time, so little on the disturbance resistance of species has been studied that a selection of right species for right site condition becomes very difficult. This study is a compilation of the site specificities and disturbance resistance of each species native to South Korea. Each species was surveyed for 11 items related to site specificities, and 12 items related to disturbance resistance including the resistance to snow damage. This study was conducted for 161 tree species from 44 families : 21 evergreen conifer species, 2 deciduous conifer species, 18 evergreen broad-leaved species, 118 deciduous broad-leaved species, and 2 monocotyledon species. This study suggests that native species in South Korea show resistance to all types of natural disturbances except drought.

  • To obtain the genetic information of Korean native livestock, the karyotyping of Korean native chick were performed by high-resolution banding technique. The chromosomes were prepared from lymphocyte culture and early embryos with 200 Korean native chick which have been raised at National Livestock Research Institute. There were no significant difference between Korean native chick and Leghorn in the number of chromosomes and chromosomal morphological pattern. Using high resolution banding technique, the yield of G-bands of prophase is much greater than that can be obtained by International System for Standardzed Avian Karyotypes(ISSAK, 1999). The G-band landmarks of Korean native chick were similar to those of ISSAK and Leghorn except some macrochromosomes. chromosome Z and 3 had C-band variants with heteromorphic patterns on distal and centromeric site. The proportion of constitutive heterochromatin, the heterochromatin ratio of Korean native chick was significantly more than that of Leghorn in all chromosomes.

Polymorphic Diversity of UBX Domain D from cDNA Isolated from Pectoral Muscle of Korean Native Chicken

  • The objectives of this study are to identify specific functional genes which are related with growth and protein structure of the pectoral muscle of Korean native chicken. Pectoral muscle was isolated from three Korean native chickens (KNC, red brown, 12 months old, 2.41 ${\pm}$ 0.24 kg) and three Cornish chickens (16 month old, 2.76 ${\pm}$ 3.0 kg). The subtraction cDNA library was prepared in PCR4 Blunt-TOPO vector. The DNA sequence homology was compared with other breeds and species in GenBank. A clone NDS-81 was found to be unique for the DNA sequence homology with UBX family. Their partial sequence has high homology (98%) with chicken UBX domain D. Chicken UBX domain has chicken (93%), cattle (68%), dog (67%), mouse (64%) and, human (63%) nucleotide sequence homology. Several regions were mutated from T in chicken to C or G in the NDS-81 clone. The first site is LAD in chicken, but it was expressed as (L)RM in clone NDS-81. In this site, amino acids were changed from Ala to Arg, and from Asp to Met. The second site was changed from ER (Arg) in chicken to ED (Asp) in clone NDS-81. They are both containing functional side chains and play an important role in binding other proteins. Therefore, the clone NDS-81 could be a different candidate gene for the UBX family gene and could related with pectoral muscle structure of Korean native chicken.

Correlations between the Distribution of Major Native Herb Species and Site Factors at Natural Broadleaved Forest in Mt. Gariwang and Mt. Jungwang, Pyeungchang-gun (천연 활엽수림에서 주요 자생 초본식물의 분포와 입지인자와의 상관 - 평창군 가리왕산, 중왕산 지역 -)

  • To obtain the basic data for the conservation and cultivation of major native herb species under the trees, distribution of native herb species, and corrrlations between native herb species and site factors were studied on natural broadleaved forest in Mt. Gariwang and Mt. Jungwang, Pyeunchang-gun. 136 native herb species were distributed in studied area, and the importance percentage of Pseudostellaria heterophylla was proved the highest, 10.8%. The other major herb species were the following oders, Meehania urticifolia, Corydalis remota, Pimpinella brachycarpa, Carex siderosticta, Pseudostellaria palibiniana, Erythronium japonicum, Ligularia fischeri, Asarum heterotropoides var. mandshuricum, Hylomecon vernalis and Astilbe rubra var. rubra. Highly positive correlations were proved Hepatica asiatica and Ainsliaea acerifolia, Pimpinella brachycarpa and Aconitum jaluense subsp. jaluense, Aconitum jaluense subsp. jaluense and Smilacina japonica var. japonica, Meehania urticifolia and Pseudostellaria palibiniana. Seven groups of native herb species were divided by cluster analysis. Preferring site factors for each native herb species were determined. Ligularia fischeri, Ainsliaea acerifolia, Parasenecio auriculata var. matsumurana, Erythronium japonicum and Pimpinella brachycarpa were distributed mainly on the higher sites, Ligularia fischeri were ditributed more frequently in deep soil sites. These results might be useful measures in the conservation and cultivation of native edible and medicinal herb species on the forest floor in the natual broadleaved forest, Pyeungchang-gun.

Substitutions for Cys-472 and His-509 at the Active Site of $\beta$-Galactosidase from Lactococcus lactis ssp. lactis 7962 Cause Large Decreases in Enzyme Activity

  • Structural modeling of $\beta$-galactosidase from L. lactis ssp. lactis 7962 has shown that the residues Cys-472 and His-509 are located in the wall of the active-site cavity. To examine the functions of Cys-472 and His-509, we generated five site-specific mutants: Cys-472-Ser, Cys-472-Thr, Cys-472-Met, His-509-Asn, and His-509-Phe. $\beta$-Galactosidase substituted at Cys-472 with Met or His-509 with Phe had <3% of the activity of the native enzyme when assayed using ONPG as substrate. The other mutants Cys-472-Ser, Cys-472-Thr, and His-509-Asn had ca. 10-15% of the native enzyme activity. The V$_max$ values of the five mutated enzymes were lower (60-7,000-fold) than that of native enzyme. These results show that the catalytic ability of $\beta$-galactosidase is significantly affected by mutations at Cys-472 or His-509.

Molecular Genetic Analysis of Ancient Cattle Bones Excavated from Archaeological Sites in Jeju, Korea

  • Ancient cattle bones were excavated from archaeological sites in Jeju, Korea. We used molecular genetic techniques to identify the species and establish its relationship to extant cattle breeds. Ancient DNA was extracted from four sources: a humerus (Gonae site, A.D. 700-800), two fragments of radius, and a tooth (Kwakji site, A.D. 0-900). The mitochondrial DNA (mtDNA) D-loop regions were cloned, sequenced, and compared with previously reported sequences of various cattle breeds (9 Asian, 8 European, and 3 African). The results revealed that these bones were of the breed, Bos taurus, and a phylogenetic tree indicated that the four cattle bones formed a monophyletic group with Jeju native black cattle. However, the patterns of sequence variation and reports from archaeological sites suggest that a few wild cattle, with a different maternal lineage, may have existed on Jeju Island. Our results will contribute to further studies of the origin of Jeju native cattle and the possible existence of local wild cattle.

Effect on Conception for Insemination Method of Frozen Semen in Cheju Native Cattle. II. Effect of time and site of insemination (제주한우에 있어서 동결정액 수정방법이 수태에 미치는 영향 제II보 동결정액의 수정시기와 주입부위가 수태에 미치는 영향)

  • This study was conducted to determine the optimum time and the insemination site of frozen in Cheju native cows, and 660 cows first service inseminated with rectovaginal methods by 3 skillful technicians as 3 districts near Cheju City and Sogwipo City areas obtained the following results. Optimum time for insemination was 16-20 hours after the onset of estrus and 80.6% of 67 conceived to first insemination. In the rectovaginal technique used widely at present conception rate for the palpation of ovaries showed 6.6% (index) higher than that of non-palpation. Conception rate of the insemination site; the intra-uterine horn, intra-uterus, just inside the cervix and depositing in mid-cervix were 62.5, 78.4, 74.6, and 80.9% respectively, however, there was no difference in conception rate between cervical and uterine deposition of forzon semen except for the intra-uterine horn.

Radiographic Diagnosis for Determination of Operation Site in Calves with Congenital Atresia Ani (송아지 항문폐쇄증의 수술 부위 결정을 위한 방사선 진단)

  • One day to one week-old 3 female Korean native calves were referred to the Veterinary Teaching Hospital, Chonbuk National University, with atresia ani. The authors performed radiographic and ultrasonographic diagnosis with physical examination, general hematology and blood chemical examination as investigation. The atresia ani were type I, II, III in each 3 calves. In abdominal radiography and ultrasonography, there were ventral displacement of descending colon(gas and feces filled) and typical enlargement of the blind end of colon in all cases. Operation site was determined by radiographic and ultrasonographic findings in these cases. Surgical treatment, two cases were translocation of the colon to the body wall exiting as a colostomy and one case was corrected by making a circumcision through the skin covering the site of the anus.

  • To obtain the basic data for the conservation and cultivation of native herb species under the forest, the relations between distribution of native herb species and site factors, distribution of native herb species, and site factors were surveyed on natural forest at subalpine zone in Mt. Odae. Ainsliaea acerifolia, Pseudostellaria palibiniana and Pimpinedla brachycarpa were distributed mainly on broadleaved mixed forest, Clematis fusca var. coreana, Carex siderosticta. Isodon excisus and Vicia unijuga did mainly on gab sites. The distribution of Ligularia fisheri, Aconitum pseudo-laeve var. erectum, Cacalia hastata and Artemisia stolonifera were significantly differed between slope directions. Artemisia stolonifera was distributed more frequently in south directed slope, and Ligularia fisheri, Aconitum pseudo-laeve var. erectum and Cacalia hastata were distributed more frequently in north and west directed slope than the other. The distribution of Aconitum pseudo-laeve var. erectum and Artemisia stolonifera were significantly differed between soil moisture conditions. Artemisia stolonifera was distributed mainly on the dry site, but Aconitum pseudo-laeve var. erectum did mainly on the dry site. Negative correlations between density of Clematis fusca var. coreana, Carex siderosticta. Isodon excises and Vicia unijuga and crown coverage was proved. Aconitum pseudo-laeve var. erectum, Pimpinella brachycarpa, Meehania urticifolia, Smilacina japonica and Pseudostellaria palibiniana were mainly distributed on wet and shady sites under forest canopy, but Pedicularis resupinata, Clematis fusca var. coreana and Artemisia stolonifera did on sunny sites at forest edge or gab.
