Electron microscopical study on the cecal development in fetuses and neonates in Korean native goats (한국재래산양 태아 및 신생아의 맹장 발달에 관한 전자현미경적 연구)

  • The morphological studies on the cecal development in the 60-, 90-, and 120-day-old fetuses and the newborns of Korean native goats were investigated by scanning and transmission electron microscopy. The results were summarized as follows ; Scanning electron microscopic studies : 1. In the 60-day-old fetuses, fold-like shapes protrusion on the cecal mucosa surface appeared. In the 90-day-old fetuses, the cecal villi appeared to be columnar shapes. In the 120-day-old fetuses, the cecal villi showed various tongue-like or columnar shapes. In the newborns, only the rudimental trace of the villi and the intestinal glands were observed. Transmission electron microscopic studies : 2. In the 60-day-old fetuses, the cecal epithelia were simple columnar in some areas and stratified columnar in others, and the epithelial cells contained nuclei, nucleoli, ER, mitochondria, Golgi complexes, zonula occludens, desmosomes, digitiform intercellular junctions, and large masses of the glycogen granules. 3. In the 90-day-old fetuses, the cecal epithelia were simple columnar in some area and stratified columnar in other. The microvilli of the cecal epithelia became much larger and longer than those in the 60-day-old fetuses, and intercellular junctions were developed, and increased numbers of ER, mitochondria, Golgi complexes were observed and the goblet cells contained a lot of the secretory granules. 4. In the 120-day-old fetuses, the cecal epithelia were only simple columnar in all areas. Microvilli and cytoplasmic organelles were well developed and the irregular annular nuclei were observed. 5. In the newborns, the cecal epithelia were covered with extensive microvilli, and the goblet cells with secretory granules were protruded into the lumen. And some goblet cells secreted the secretory granules into the lumen.

A Comparative Study on the Herbage Utilization from Tall type and Tall+short type Mixtures by Korean Native Goats (상번초 및 상ㆍ하번초형 혼파초지의 산양에 의한 초류이용성 비교 연구)

  • This study was conducted to investigate the effect of tall type mixtures(TM) and tall + short type mixtures(TSM) on nutritive utilization. DM intake, digestibility and utilization of nitrogen and energy of herbages from pastures by Korean native goats were determined. This experiment was conducted by total collection method in laboratory, 2000. The experimental herbage included two treatments: Tall type mixtures(orchardgrass, Potomac 40+tall fescue, Fawn 20+Festulolium braunii, Paulita 10+perennial ryegrass. Reveille 10+timothy, Climax 10+red clover, Kenblue 5+alfalfa, Vernal 5%), Tall+short type mixtures {orchardgrass, Potomac 40+tall fescue, Fawn 20+Kentucky bluegrass, New port(turf type) 10+redtop, Barricuda(turf type) 10+perennial ryegrass, Palmer III(turf type) 10+red fescue, Flyer II(turf type) 5+white clover, Regal 5%}. The voluntary dry matter(DM) intake of Korean native goats fed with herbages from tall+short type mixtures(TSM) was slightly higher than that of tall type mixtures(TM), but no difference was observed between TSM and TM(p > 0.05). The digestibility of DM, neutral detergent fiber, cellular contents and acid detergent fiber of herbage from TSM was slightly higher than that of TM, but no difference was detected between TSM and TM(p > 0.05). The utilization of nitrogen and energy by Korean native goats did not showed any difference in herbages from TSM and TM(p > 0.05). In conclusion, the herbage utilization by Korean native goats did not showed any significant difference. Thus, there is a possibility that turf grasses in mixtures can be used as a forage sources.

  • 본 연구의 목적은 초음파검사에 의하여 한국재래산양의 간장 및 담낭의 정상적인 구조와 크기를 알아보는데 있다. 30두의 한국재래산양에서 간장, 문맥, 후대정맥 및 담낭의 크기, 위치 및 구조를 측정하였다. 30마리의 산양을 체중에 따라서 5-15Kg, 15-25Kg 및 25-35Kg군으로 분류하였다. 우측복벽의 제9늑간극으로부터 12늑간극까지에서 측정을 실시하였다. 각 늑간극에서의 간장의 크기, 후대정맥 및 문맥의 위치 및 직경을 관찰하였다. 본 연구에 있어서의 간장 크기 및 위치의 초음파 측정자료는 한국재래산양의 간장의 변화에 대한 진단에 있어서 기초자료로서 사용될 수 있다는 결론을 얻었다.

Electrophoretic analysis of the major proteins of ruminant erythrocyte membrane: Their relation to slow erythrocyte sedimentation rate (반추동물 적혈구막 단백의 전기영동법에 의한 분석 -낮은 적혈구침강속도와의 관계-)

  • The proteins of the ruminant erythrocyte membranes were analysed by polyacrylamide gel electrophoresis in sodium dodecyl sulfate, and their relations to the slow erythrocyte sedimentation rate(ESR) of the ruminants were investigated by treating the erythrocytes with proteinases such as trypsin, chymotrypsin and pronase, and glycosidases such as neuraminidase and galactosidase. Protein content in the erythrocyte membrane was $2.85{\pm}0.28$ in human, $3.60{\pm}0.41$ in Korean cattle, $3.71{\pm}0.36$ in Holstein, $4.13{\pm}0.83$ in Korean native goat and $3.94{\pm}0.56mg/ml$ in sheep, showing higher in ruminant animals than in human(p<0.01). Although the general protein profiles of the ruminant erythrocyte membranes were almost similar to that of human, all the ruminant erythrocyte membranes showed one additional protein band, called band-Q in the previous report on proteins of bovine erythrocyte membrane, which migrated electrophoretically to the mid position between band-2 and band-3 in human erythrocyte membranes. The glycoprotein profiles of ruminant erythrocyte membranes revealed by periodic acid Schiff(PAS) stain showed a marked difference from that of human. The PAS-1(glycophorin) and PAS-2(sialoglycogrotein) present in human erythrocyte membranes were almost absent from the ruminant animals. Instead, a strong PAS-positive band near the origin of the electrophorograms, which was named as PAS-B in the previous report on proteins of bovine erythrocyte membranes, was shown in the ruminant animals except sheep. In addition, the erythrocyte membranes of Korean native goat and sheep showed a moderate PAS-negative band near the tracking dye of the electrophorograms, which was named as PAS-G in this study. In the erythrocyte treated with the enzymes, the migration of each protein fracture of erythrocyte membranes in response to each enzyme was diverse according to different species or breed of ruminant animals. Among others, band-Q present in ruminants was slightly or moderately decreased by trypsin-, chymotrypsin-, and pronase- treatments of the erythrocytes, but not only in sheep. It was particularly noticeable that PAS-B, a fraction of glycoprotein, present in ruminants except sheep, was better digested by proteinases than by glycosidases, showing remarkable increase(p<0.01) of the ESR in accord with complete digestion(disappearance) of the PAS-B band by pronase, trypsin or chymotrypsin treatment of erythrocytes. In sheep, there was almost no any response to the various enzymes in general protein and glycoprotein profiles of the erythrocyte membranes except PAS-G, which was markedly decreased by pronase treatment of the erythrocytes. Nevertheless, the ESRs were accelerated in erythrocytes treated with pronase, trypsin, chymotrypsin and neuraminidase. Erythrocyte osmotic fragility was increased in erythrocytes treated with only pronase among five enzymes in all the human and ruminant animals used in this study.

Comparison of sheep erythrocytes and Korean native goat erythrocytes-rosette forming rate of pig peripheral blood mononuclear cells (돼지 말초혈액 단핵세포의 면양 및 재래산양 적혈구 rosette 형성능 비교)

  • To develope the methods for isolation and enumeration of lymphocyte subpopulations in pigs, we carried out the rosette-assay using sheep erythrocytes(SRBC) and Korean native goat erythrocytes(GRBC) as a target cells. To enumerate T lymphocytes, E-rosette methods were applied with RBC treated with various concentration of polymers such as Aet and Dex, singly or in combination. And to enumerate B lymphocytes, EAand EAC-rosette assay was adopted. The results were as follows; 1. E-RFR with polymer-untreated SRBC and GRBC was $32.9{\pm}7.9%$ and $31.3{\pm}9.4%$, respectively. On the other hand, RFR with 0.1M Aet plus 8% Dex treated SRBC and GRBC was increased about two-fold($67.8{\pm}7.4%$ and $69.8{\pm}8.5%$), respectively. 2. EA-RFR with SRBC and GRBC were $ 39.1{\pm}10.2%$ and $32.6{\pm}6.1%$, respectively. 3. EAC-RFR with SRBC and GRBC were $27.6{\pm}7.0%$ arld $21.0{\pm}3.2%$, respectively. These results showed that both SRBC and GRBC could be recommanded as a binding cells for rosetteassay to isolate of lymphocyte-subpopulations in pigs.

Studies on the Improvement and Utilization of Pasture in the Foerst II. The effect of shde on the intake palptability and grazing behavior (임간초지의 개량 및 이용에 관한 연구 II. 채식기호성 및 방목습성에 미치는 비음의 영향)

  • This study was conducted to determine the effects of shade (full day light; 100, 80, 60, 40 and 20%) and fertilization levels (high; $N\;30kg-P_2O_5\;35kg-K_2O\;30kg$, moderate; $N\;20kg-P_2O_5\;25kg-K_2O\;20kg$, and low; $N\;10kg-P_2O_5\;15kg-K_2O\;10kg$) of forest pasture on intake palatability of Korean native goat, and the effects of shade under the conditions of 50% shaded forest pasture and full day light pastures on grazing behavior. This experiment was done at the experimental field of Chungnam National University, in Taejon, from 1983 to 1984. The results obtained are summarized as follows: 1. The dry matter intake, R.I.I. and Touch number of Korean native goat were affected by the shade degrees and fertilization levels, generally the increase of the fertilization level and high shading degree increased the NO_3-N$ contents in the plants, but decreased the soluble carbohydrate contents. It means that the decrease of the utilization of the shade culture grasses is due to the decreased intake palatability of forest pastures, particularly the conditions high fertilization level and above 60% of shading level seemed to have a nitrate stress. 2. Grazing time of forest pastures by Korean native goat tended to be spend more time than full day light pastures, also tendency of the major grazing activity tended to be graze continuously without difference of variation of grazing time within a day. Considering of the characteristies os shade, the intensive utilization of forest pastures seemed to be most effective during the high temperature season in August.

Effect of Whole or Steam-flaked Corn Based Diet on Ruminal fermentation Characteristics In Vitro and Ruminal Metabolism in Korean Native Goat In Vivo (통옥수수 및 Steam-flaked 옥수수 기초사료가 반추위미생물 발효성상과 한국재래산양 반추위대사 특성에 미치는 영향)

  • These study were conducted to determine the effects of a whole or steam-flaked corn based diet on rumen microbial fermentation in vitro and ruminal metabolism in the Korean Native Goat(KNG) in vivo. The experiments consisted of two dietary treatments: control, steam-flaked corn(SFC) based diet(80%) + rice straw mixed(20%)(SFCR); 100% whole corn based diet(WC). The first experiment was conducted to investigate the effect of whole corn on ruminal metabolism in vitro for 0 to 48 h. pH values were optimally maintained during incubation time, and were not significantly different between treatments. Gas production of SFCR was significantly higher than WC(p<0.01). $NH_3$-N concentration tended to increase for WC, but not significantly different between treatments. The mean value of total volatile fatty acid concentration of WC was significantly lower than SFCR(p<0.01), but SFCR and WC linearly increased as the time of incubation approached 48 h. Mean value of acetate concentration of SFCR was significantly higher than WC(p<0.01). Propionate concentration of WC for the total incubation time was significantly higher than SFCR(p<0.01). The digestibility of dry matter was not significantly different between treatments, but SFCR was somewhat higher than WC. The second experiment was conducted to effect of whole shelled corn based diet on rumen metabolism in KNG. pH values tended to decrease through all treatments. There was not a significantly difference between treatments. Microbial protein yield of SFCR was significantly higher than WC(p<0.01). $NH_3$-N concentration of WC was significantly (p<0.01) higher than SFCR. Total VFA and propionate concentration of WC was significantly higher than SFCR(p<0.01), but acetate concentrate of WC was not significantly higher than SFCR. The mean value of total lactate concentration was significantly(p<0.01) different but the value of SFCR and WC were lower than the average concentration of acidosis. In sacco DM disappearance rate of SFC was significantly(p<0.01) higher than WC.

A Survey on Some Factors Related to the Rearing of the Korean Native Goat (한국재래산양(韓國在來山羊)의 사육(飼育)에 관련(關聯)된 몇 가지 요인(要因)들에 관(關)한 조사(調査))

  • This survey was carried out for two years from June of 1982 to May of 1984 to investigate factors influencing the Korean native goat rearing. The results were summarized as follows. 1. The daily gain of female goats from weaning age to yearling was 41.9g in average. The maximum and minimum gains were 55.1 and 30.1g, respectively. 2. The mean body weights when purchased from the market were 8.07kg for survived goats and 5.89kg for dead goats. 3. The kidding months of does were distributed throughout all the seasons, and the average litter sizes were 1.2 kids for first kidding and 2.0 kids for second kidding. 4. The amount of DM intake was 2.78% of the body weight. The DM digestibility was 62.91 % for the first pregnant goats under good feeding condition. 5. The amount of DM intake was 3.92% of the body weight. The DM digestibility was 47.01% for the growing female goats under fair feeding condition. 6. The goats seemed to prefer shrub plants to grass. About 65% of the total dry forage consumed was tumbergiana (kudzu).

  • To investigate the condition of infectious rate of fascioliasis in the farm animals. This survey was carried out, by the interdermal reaction from different animals, ages and pestures in Daejeon Suburbs and Cattle market and Abattoir. The average positive rate of infectious reaction were 31.0% in Korean Cattle, 38,7% in dairy Cattle, 33.8% in Korea native Goats and 26.9% in dairy Goat. The positive rate was higher at the age of 2~6 that compares with at age of yearling. From survey on the river side area, valley and wet area, the rate of positive was occured higher, than from plain, high land and in case of animals have not been grazing. And the detective rate of liver fluke among the positive reaction was occured 92.5% in Korean Cattle and 94.7% in Korea native Goat.

Molecular Cloning of Hemoglobin Alpha-chain Gene from Pantholops hodgsonii, a Hypoxic Tolerance Species

  • To investigate the possible mechanisms of high-altitude native animals in adapting to high altitude, we cloned hemoglobin alpha-chain (alpha-chain Hb) gene from Pantholops hodgsonii, an animal species that indigenously lives at elevations of 3700-5500 m on the Qinghai-Tibetan plateau. Using reverse transcription polymerase chain reaction (RT-PCR) technique, the alpha-chain Hb gene was amplified from total RNA in the liver of the Pantholops hodgsonii. TA cloning technique was used and the PCR product was cloned into pGEM-T vector. The DNA sequence of the gene was highly homologous with sheep (99.1%), goat (98.6%), cattle (95.6%) and human (86.5%). The alpha-chain Hb gene encoded a 142-amino acid protein that could be identified with the homology of alpha-chain Hb protein in sheep (98%), goat (96%), cattle (91%) and human (87%). However, 18 alternations were detected when compared with the alpha-chain Hb gene in human, and 2 in sheep. Moreover, the alterations of a117 GluAsp and $\alpha$132 AsnSer in important regions were noted in human and sheep, respectively. Phylogenetic analysis suggested that the structure of alpha-chain Hb was highly similar to that in sheep. This study provided essential information for elucidating the possible roles of hemoglobin in adapting to extremely high altitude in Pantholops hodgsonii.
