A Histochemical Study of the Ruminant Ovary (반추수난소(反芻獸卵巢)의 조직화학적연구(組織化學的硏究))

  • The present study was underve the histochemical nature of various follicles and interstitial tissues in the ovaries of Korean native goats and cattle as well as the histochemical changes of those in the ovaries of Korean native goats treated with dexamethasone. Much more lipid granules appeared in the granulosa and theca cells of atretic follicles compared with normal follicles in these ruminant ovaries. In the ovaries of Korean native goats the interstitial tissue derived from the theca interna of atretic follicles appeared the form of patches of cells and the interstitial tissue derived from stromal cells appeared the form of diffuse, obscure bounds. In the ovaries of Korean native cattle the interstitial tissue derived from theca interna of atretic follicles showed sparsely scattered form of pathes of cells. The histochemical components were described on the basis of lipids in the granulosa and theca cells of normal follicles, atretic follicles and interstitial tissue. In the ovaries of Korean natve goats treated with dexamethasone, the granulosa and theca cells of atretic foillicle contained plenty lipid granules that were increased in size and number, however, lipid granules were markedly decreased in the interstitial tissue.

Effects of Different Feeding Systems on Nutrient Availability, Nitrogen Retention and Blood Characteristics in Native or Crossbred Korean Black Goats (사료급여체계가 재래종 및 교잡종 흑염소의 영양소 이용율, 질소축적 및 혈액성상에 미치는 영향)

  • This trial was conducted to investigate effects of different feeding types of conventional or total mixed rations (TMR) provision on feed intake, nutrient digestibility, nitrogen retention and blood metabolite characteristics in different breeds of native and cross bred Korean black goats. Ten Korean native male goats and 10 Korean crossbred black male goats (Korean native $\times$ Australian feral crossbred, 50:50), average 18 kg of BW, were used in this trial, and each breed of Korean black goats was separated into conventional and TMR diets treatments in a randomized complete block design. Conventional diet was based on forages and commercial diet. All animals were housed in individual metabolism crates and were fed ad libitum. Water was available at all times. Experimental period totally lasted for 38 days, consisting of 14 days of adaptation, 10 days of preliminary period and 14 days of sample collection period. Daily dry matter intake was significantly (p<0.05) higher in TMR treatments of the crossbred goats. Within breeds, crossbred rather than native-bred consumed more highly, and within the feeding type, TMR diets were more fed in comparison with conventional diet. Average daily gain ranked the highest in crossbred : TMR, followed by crossbred: conventional diet, native-bred: TMR, and native-bred: conventional diet (p<0.05). Dry matter digestibility was significantly (p<0.05) higher for TMR than for conventional diets, and it was higher in crossbred than in native-bred Korea black goats. Nitrogen retention was highest in TMR treatments of crossbreed, whereas it was lowest in conventional diet of native breed (p<0.05). Plasma glucose and urea nitrogen concentrations were significantly (p<0.05) higher in crossbred than in native-bred Korean black goats. Overall, the results indicated that feeding total mixed ration to crossbred rather than native-bred Korean black goats had more favourable influences on their performances, intakes, digestibility, nitrogen retention and blood metabolite characteristics. In conclusion, it is conceived that the results may contribute to economical benefit to farms producing Korean black goats together with establishment of an efficient feeding management system.

Mitochondrial DNA variation and phylogeography of native Mongolian goats

  • Objective: Mongolia is one of a few countries that supports over 25 million goats, but genetic diversity, demographic history, and the origin of goat populations in Mongolia have not been well studied. This study was conducted to assess the genetic diversity, phylogenetic status and population structure of Mongolian native goats, as well as to discuss their origin together with other foreign breeds from different countries using hypervariable region 1 (HV1) in mtDNA. Methods: In this study, we examined the genetic diversity and phylogenetic status of Mongolian native goat populations using a 452 base-pair long fragment of HVI of mitochondrial DNA from 174 individuals representing 12 populations. In addition, 329 previously published reference sequences from different regions were included in our phylogenetic analyses. Results: Investigated native Mongolian goats displayed relatively high genetic diversities. After sequencing, we found a total of 109 polymorphic sites that defined 137 haplotypes among investigated populations. Of these, haplotype and nucleotide diversities of Mongolian goats were calculated as 0.997±0.001 and 0.0283±0.002, respectively. These haplotypes clearly clustered into four haplogroups (A, B, C, and D), with the predominance of haplogroup A (90.8%). Estimates of pairwise differences (Fst) and the analysis of molecular variance values among goat populations in Mongolia showed low genetic differentiation and weak geographical structure. In addition, Kazakh, Chinese (from Huanghuai and Leizhou), and Arabian (Turkish and Baladi breeds) goats had smaller genetic differentiation compared to Mongolian goats. Conclusion: In summary, we report novel information regarding genetic diversity, population structure, and origin of Mongolian goats. The findings obtained from this study reveal that abundant haplogroups (A to D) occur in goat populations in Mongolia, with high levels of haplotype and nucleotide diversity.

Morphological Characteristics and Growth Performance of Korean Native Black Goats (재래흑염소의 형태적 특징 및 생육특성)

  • This study aims to provide basic data for investigating the phenotypic traits and growth characteristics of Korean native black goats. The Jangsu and Tongyoung strains were shown to have black hair, while the Dangjin strain was shown to have black or dark brown long hair. Only the Tongyeong strain was found to have no wattle and one goat each from the females and males of the Dangjin strain was found to have no horns. The lengths of the ears and tails were similar for the female and male goats, while the lengths of horns and beards were longer in the male goats. The birth weight was 2 kg on average, with that of male goats tending to be slightly heavier than that of the females. Weights at the weaning period were found to be slightly heavier for the males. Also, at 12 months, the weights of the male goats by strain were shown to be heavier than that of the females. The body length and depth by strain were found to be higher for the male goats, while no differences in the chest width and girth between the strains or gender were found. These findings are expected to be used as a basis for differentiating between native and crossbred Korean black goat strains and for improving Korean native black goats.

Studies on the Superovulation and Collection of microinjectable Embryos in Korean Native Goats (Capra hircus aegagrus) (한국 재래산양에서의 과배란유기와 외래유전자 주입에 적합한 수정란의 회수에 관한 연구)

  • This study was carried out to determine the hormone treatment scheme for an efficient superovulation and optimal recovery time for obtaining pronuclear embryos suitable for DNA injection in Korean native goats. For a superovulation, FSH(5.6mg) was given over four days in twice daily injections with (FSH/hCG group) or without(FSH group) hCG(100 IU) co-injection at the time of 7th FSH injection. Estrus cycle was synchronized by norgestomet ear-implantation for 11 days and its removal at the time of 6th FSH injection. Among the treated goats, the percentage of ovulated goats, which were examined at 70 to 76 h following implant removal, was greater in FSH/hCG group than in FSH group (100% vs 36.4%) but there was no significant difference in the mean numbers of ovulation points and fertilization rates between the two groups. To optimize hCG treatment scheme and recovery time, we injected hCG at the time of 7th (FSH/hCGa) or 8th(FSH/hCGb) FSH injection and then examined the developmental stage of the embryos recovered at different times after implant removal. In FSH/hCGa group, significant portions(31 to 44%) were beyond 1-cell stage, which was non-injectable, irrespective of their recovery time. However, in FSH/hCGb group recovered at 70 to 76 h after implant removal, great portions(69%) were fertilized and most of them(96.6%) were injectable 1-cell stage. Considering together the fertilization rate and developmental stage of recovered embryos, it is recommendable to administrate hCG at the time of final 8th FSH injection and collect the embryos at 70 to 76 h after implant removal to obtain injectable embryos as many as possible in Korean native goats.

Changes of plasma progesterone concentrations during the estrous cycle and its application to early pregnancy diagnosis in Korean native goats (한국재래산양(韓國在來山羊)의 발정주기중(發情週期中) 혈장(血漿) progesterone 농도변화(濃度變化)와 조기임신진단(早期姙娠診斷)에의 응용(應用))

  • A study was conducted to improve the reproductive performance of Korean native goats. The length of estrous cycle and plasma progesterone concentrations during each cycles were determined by both radioimmunoassay and estrus behaviours, and the results were used in the early pregnancy diagnosis. The estrous cycles were classified into the short(l8 days or shorter, average 16.7 days), normal(19 to 22 days, average 20.9 days) and long(23 days and longer, average 23.8 days)cycle. The average length of the 19 estrous cycles was 20.8 days. Plasma progesterone concentrations in 12 normal cycles were the lowest(0.10 ng/ml) at estrus, remained high from 6 to 16 days(range : 4.43~7.93 ng/ml) and drastically decreased thereafter to reach minimal concentrations at the next estrus. Plasma progesterone concentrations were measured for early pregnancy diagnosis at 0, 10 and 20 days after mating in the 12 Korean native goats. Plasma progesterone concentrations in the pregnant goats at 20 days after mating were significantly higher than in the non-pregnant goats(p<0.001). Of the 12 goats, 10 were confirmed pregnancy by both progesterone concentrations and kidding. The accuracy of the pregnancy diagnosis based on plasma progesterone concentrations was 100% for positive as well as for negative.

Serum cortisol concentrations on the normal condition in Korean native goats by ELISA (ELISA에 의한 국내 재래종 산양의 정상상태에서의 혈중 cortisol 농도)

  • This study was performed to examine the charges of serum cortisol concentrations on the normal conditions in Korean native goats by enzyme-linked immunosorbent assay(ELISA). The Korean native goats were 4 to 30 months old and weighted 6.0 to 28kg. The goats were allowed ad libitum access to water and food. To minimize the stress with handing and blood sampling, animals were adapted for 2 weeks before the experiment. After adaptation, intravenouse catheter was inserted left intact a jugular vein of 6 goats and blood samples were done 2~3 days later. Experimental animals were divided into 2 groups which was non-catheterized and catheterized group in order to examine the effect for changes of serum concentration and circadian rhythm of cortisol. The results were obtained as follows ; The sensitivity of serum cortisol concentration was $20pg/m{\ell}$. The intra-assay and inter-assay coefficient of variation were below 2.5% and 5%, respectively. Serum concentration of cortisol was more higher in noo-catheterized group$(32.84{\pm}16.78ng/m{\ell})$ than catheterized group$(23.20{\pm}10.29ng/m{\ell})$. The difference according to months old in serum concentration of cortisol was the more higber on 4~6 than 7~12 and lowest over 12 months old. A circadian rhythm in the serum concentration of cortisol in Korean native goats was not found significantly with 2 hours sampling intervals.

  • The purpose of present study was to investigate the progesterone concentrations throughout gestation and peripartum period, and the return to the first estrus postpartum for improvement of reproductive efficiency in Korean native goats. The average length of gestation was 148 days(range : 144~154 days) and the average number of live births was 2 kids(range : 1~5 kids) in 12 Korean native goats. Progesterone concentrations were measured in blood samples taken from 12 goats every 5 days during gestation period. Plasma progesterone concentrations were 0.10 ng/ml at Day 0 of pregnancy and increased gradually until Day 20(6.58 ng/ml). Then they decreased slightly from Day 30 to 40(range : 4.32~4.82 ng/ml), increased again after Day 40 and remained thereafter until Day 140(range : 4.32~10.36 ng/ml). The progesterone levels declined sharply to basal levels at parturitum. Plasma progesterone concentrations during the pestpartum were 6.98 ng/ml at 10 days, 4.86 ng/ml at 6 days 3.18 ng/ml at 2 days before parturition, and 0.10 ng/ml at parturition, respectively. The basal levels were maintained until the first estrus postpartum. The mean intervals from parturition to the first estrus postpartum on the basis of progesterone determination and estrus detection were $100{\pm}64(mean{\pm}S.D.)$ days in 7 Korean native goats.

Study on the Food Habits of Korean Native Goats (Capra hircus) Fed with Various Roughage Sources (여러 가지 조사료를 급여한 재래산양의 채식습성에 관한 연구)

  • The object of this experiment was to investigate the food habits of Korean native goats fed with various roughage sources. The feeding trials were conducted at Chungnam National University and Unbong Animal Genetic Resources Station in 2007. The experimental roughages sources were five including 25 species in all; grass sources: 5 species (Lolium perenne, Festuca arundinacea, Poa pratensis, Agrostis alba and mixed grass, legume sources: 5 species (Trifolium pratense, Medicago sativa, Trifolium repens, Melilotus officinalis and Vicia villosa), browse sources: 5 species(Quercus serrata Thunb., Prunus jamasakura Sieb., Quercus aliena Blume, Robinia pseudoaccacia and Pinus densiflora), weed sources: 5 species(Artemisia princeps Pampanini, Erigeron canadensis, Alopecurus aegualis Sobolewski, Echinochloa crusgalli var. frumentacea (Roxb.) Wight and Rumex crispus), and native grass sources: 5 species(Zoysia japonica Steudel, Agropyron tsukushiense var. transiens(Hack) Ohwi., Arundinella hirta(Thunb.) Tanaka, Miscanthus sinensis Anderss and Phragmites comunis Trin). Ten Korean native goats were selected which had nearly the same body weight(average 30.6kg, 남) as experimental animals. Experimental roughages were prepared by 0.5kg per 1 species fed to Korean native goats, and the experiment was carried out from 08:00 to 20:00. The chemical composition and dry matter digestibility of each roughage source and species were significantly different at the sampling area, plant species, growth stages and cutting period (p<0.05). The Korean native goats ate more roughages which had low fibrous contents, but high dry matter digestibility. Among all the 25 species of roughages, the favorite intake species order by Korean native goats was observed like this: Quercus serrata Thunb., Quercus aliena Blume, Trifolium repens, and Pinus densiflora and Artemisia princeps Pampanini which was lower intake compared to other domestic herbivores. On the other hand, compared to each roughage source, total intake amount by Korean native goats was showed as browse(29.9%), legumes(23.0%), weeds(21.6%), grasses(21.5%) and native grasses(4.0%), respectively. Based on the result, the food habit of Korean native goats seems to be closer to browser type.

The Effect of Roughage Source on Voluntary Feed Intake and Digestibility in Korean Native goats (조사료원이 한국 재래산양의 섭취량과 소화율에 미치는 영향)

  • A study was carried out to evaluate the values of roughages available in Korea on feed intake and digestibility of Korean native goats and consequently to apply its results to the feeding system of Korean goats as a basic information. The results are as follows. 1. The protein contents in Alfalfa and Acacia leaf were 17.6 and 16.3% respectively 11.7 and 6.8% higher than orchardgrass and pine needle. Ether extract tom pine needle was about two times higher than other roughage sources which are 9.2%. 2. Dry matter intake per day in goats fed alfalfa, acacia leaf and orchardgrass was higher (P < 0.05) at 590.3, 543.8 and 496.58 respectively and 217.lg in pine needle. 3. Dry matter intake per basal weight (DM glkg of B$W^{0.75}$ and DM g/kg of BW%) was higher in goats fed acacia leaf at 68.5 and 3.5% respectively than any other treatments goats fed pine needle showed the lowest (P< 0.05) dry matter intake at 28.6 and 1.5% respectively. 4. Dry matter digestibility was highest (PcO.05) in alfalfa fed goats at 61.4%. Dry matter digestibilities in orchardgrass, pine needle and acacia leaf were 58.0, 46.8 and 46.6% respectively. 5. Total digestible nutrients were highest (P< 0.05) in Alfalfa fed goats at 59.5%. Total digestible nutrients in orchardgrass, pine needle and acacia leaf were 54.2, 50.7 and 47.7% respectively. 6. In conclusion, the value of orchardgrass as feed for goats was as excellent as alfalfa and although acacia leaf had a less value than alfalfa in goats, it is considered to be used as a protein supplement in diets for goats due to its high content of protein and excellent palatability. Intake of pine needle in Korean goats in this study was satisfactory and it indicates the possibility of its use as a supplement in diets for goats. However, fkther investigation will be necessary particularly on the deleterious effects of ether extracts of pine needle
