Quercetin의 급여가 산양의 사료이용성, 혈액상 및 육질에 미치는 영향 (Effects of Dietary Quercetin on the Feed Utilization, Blood Parameters, and Meat Quality in Korean Native Goats)

  • 본 실험은 quercetin의 급여가 우리나라 재래 산양의 사료이용성, 혈액 성상 및 육질에 미치는 영향을 조사하고자 실시하였으며, 공시동물로 체중 15 kg 정도의 우리나라 재래산양 16두(2처리 8 반복)를 공시하여 TMR 사료에 quercetin을 0 및 200 mg/kg씩 첨가하여 사양실험을 실시하였다. 실험결과로서, 재래산양의 사료섭취량, 음수량, 배뇨량 및 배분량은 유의한 차이가 없었으며, 조지방, NDF 및 ADF의 소화율도 처리간에 유의한 차이가 없었으나, 건물 및 조단백질의 소화율은 첨가구가 무첨가구에 비하여 유의하게 높은 결과였다(P<0.05). 그리고 반추위액의 pH는 처리 간에 유의한 차이가 없었고, total VFA, 프로피온산 및 낙산의 농도는 첨가구가 유의하게 높았다(P<0.05). 그러나 초산, 이소 낙산, 이소 발레린산 및 발레린산은 처리구간에 유의한 차이가 없었다. A/P비는 첨가구가 유의하게 낮게 나타났다. BUN BUN/ creatinine은 quercetin 첨가구가 무첨가구에 비하여 유의하게 높았으나, ALT AST Creatinine Albumin ${\gamma}$-GT Ca Pi의 농도는 유의한 영향을 관찰할 수 없었다. 재래산양육의 pH, 보수력 및 총 페놀 함량은 유의적인 차이를 보이지 않았으며, $ABTS^+$ radical소거 능력은 첨가구가 무첨구에 비하여 도축 후 0시 간에서 유의하게 높게 나타났으나 일수가 경과함에 따라 유의한 차이는 없어졌다. 관능검사에서는 quercetin을 섭취한 산양의 육에서 색과, 조직감과 전반적인 부분의 선호도에서 유의적으로 높은 결과를 보였다. 결론적으로 건물과 조단백질 소화율, 반추위 발효성상 및 관능검사에서 긍정적인 결과를 얻었지만 육질 평가에 있어 pH, 보수력, 전자공여능 및 지방산패도에 유의적인 차이를 보이지 않았다. 그러나 재래산양육의 ABTS 환원력이나 관능적 특성에서 긍정적인 결과를 나타내어 quercetin 급여가 육질 향상에 효과적일 수 있는 가능성은 제시되었다. 따라서 추후 quercetin 급여가 육질에 미치는 영향에 대한 지속적인 연구가 필요하다고 하겠다.

한국 재래산양의 태아 및 신생아의 폐 발달에 관하여 (Development on the lungs of fetuses and neonates in Korean native goats)

  • The morphological development of lungs in fetuses of 60, 90 and 120 days of gestation and neonates of Korean native goats was investigated by light, scanning and transmission electron mictroscope. The results were summarized as follows ; 1. Gross findings ; In the 60-days-old fetus, the lung was developed and differentiated into six lobes. 2. Light microscopic findings : The gland-like bronchioles were formed in loose mesenchyme at 60 days of gestation and the bronchial wall contained smooth muscles. The loose mesenchyme had been replaced by compact parenchymal tissue at 90 days of gestation and the cartilage plates appeared in bronchial wall which contained blood vessels, submucosal glands and smooth muscles. The lung parenchyma consisted of a fine network of alveoli at 120 days of gestation and the bronchial wall contained well-developed blood vessels, submucosal glands, cartilage plates and smooth muscles. In neonates, the lung tissue was similar to the mature lung tissue and the bronchial wall contained well developed cartilage plates. 3. Scanning electron microscopic findings : The epithelial cells lining the tubules were composed of cuboidal or columnar at 60 days of gestation and the epithelial cells lining the large airways were often ciliated : some were covered with stubby microvilli. The epithelial cells lining the canals were cuboidal at 90 days of gestation and the epithelial cells lining the bronchioles were ciliated cells or nonciliated(clara) cells, The clara cells contained row microvilli. The alvealor development of this stage was rapidly progressed ; the subdivision of canals by alveolar crests and assosiated wall attenuation resulted alveoli at 120 days of gestation and the respiratory bronchioles were lined by ciliated or nonciliated epithelial cells. In neonates, the epithelial cells lining the alveolar walls were mainly covered with pneumocyte type I ; Some were covered with pneu-mocyte type II. 4. Transmission electron microscopic findings : The epithelial cells lining the tubules were adhered with tight junction at apical borders of the adjacent cells at 60 days of gestation, which contained few organells and glycogen. The epithelial cells lining the canals were composed mostly of cuboidal cell at 90 days of gestation and the epithelial cells lining of the bronchioles were ciliated of nonciliated cell, which contained few organelles and abundant glycogen. The epithelial cells lining the alveolar walls were composed of pneumocyte type I and a few pneumocyte type II at 120 days of gestation. The epithelial cells lining of the bronchioles were ciliated or nonciliated cells. In neonates, pneumocyte type I was observed as flat and thin cytoplasmic extension in shape. Otherwise, pneumocyte type II was observed as cuboidal type with apical microvilli and contained osmiophillic lamellar inclusion bodies. Putting these various experiment results together, the lung development was slowly progressed at early stage, which was rapidly progressed in the late stage of gestation.

Hydrocortisone 투여가 비유중기 재래산양의 유단백질과 유선세포 Prolactin Receptor mRNA 발현에 미치는 영향 (Effects of Hydrocortisone Administrations on Expressions of Casein and Prolactin Receptor mRNAs in Mammary Glands of Mid-Lactation of Korean Goats)

  • Glucocorticoid는 비유기 동물의 유선세포 pro-lactin receptor(PRL-R) 발현을 증가시키며, 전반적인 유선세포의 유합성 작용을 활성시킴으로 유합성 능력을 증진시킨다. 유선세포 PRL-R 발현량 증가는 유생산량 향상과 밀접한 관계를 갖는다. 본 실험은 비유중기의 재래산양의 유합성 능력을 향상시키고자, 0.05. 0.1과 0.2 g hydrocortisone을 5ml의 생리식염수에 현탁하여, 정맥투여하고 유선세포 PRL-R와 $\alpha$-유단백 질 mRNAs 발현 량을 조사하였다. 대조구로는 5$m\ell$의 생리식염수를 정맥투여 하였다. 24시간 후 유선조직을 채취하여 $\alpha$-유단백질과 PRL-R mRNA 발현량을 competitive PCR(polymerase chain reaction)로 발현량을 조사하였다. Hydrocortisone 처리 24시간 후 재래산양 유선세포의 PRL-R mRNA 발현량은 대조구의 PRL-R mRNA 발현량과 유의한 차이가 없었다. $\alpha$-유단백질 mRNA 발현은 대조구에 비하여 0.05g hydrocortisone투여구는 37%, 0.1g hydrocortisone 투여구는 630%, 0.2g hydrocortisone 투여구는 386% 증가하였다. Hydrocortison 처리에 의한 유선세포 PRL-R mRNA 발현에 변화가 없었으나 $\alpha$-유단백질 mRNA 발현 증가는 세포 내 기능성 단백질 발현과 세포 외부로 분비되는 단백질의 시간에 따른 발현양상의 차이인 것으로 사려된다. 본 연구에서 비유중기 재래산양에 hydrocortisone 투여는 $\alpha$-유단백질 mRNA 발현을 증가시키는 것으로 나타났다.

SNP 정보를 활용한 재래흑염소와 교잡종 염소의 유전적 다양성 및 유연관계 분석 (SNP-based Genetic Diversity and Relationships Analysis of the Korean Native Black Goat and Crossbred Goat)

  • 본 연구는 국내 재래흑염소 집단 (당진 계통, 장수 계통, 통영 계통 및 경상대 계통)과 교잡종 염소 집단의 유전적 다양성 및 유전적 유연관계를 조사하기 위해 수행되었다. 각 집단에 존재하는 공통 SNP 45,658개를 이용하여 분석에 이용하였다. 유전적 다양성의 지표가 될 수 있는 기대, 관측 이형접합도는 교잡종, 경상대, 장수, 통영 계통 순으로 나타났다. 집단 사이의 유전적 다양성 정도를 나타내는 분산 성분은 당진과 경상대 계통 사이에서 19.98%로 가장 높게 나타났으며, 장수와 통영 계통 사이에서 8.87%로 가장 낮게 나타났다. 또한, 집단 사이의 유전적 거리는 장수, 통영 계통에서 하나의 분지를 형성하였으며, 당진, 경상대 계통이 하나의 분지로 나타냈다. 또한, 교잡종 집단은 당진, 경상대 계통과 하나의 분지를 이루는 것으로 나타났다. 따라서 본 연구 결과는 국내 계통 간의 불필요한 근친교배와 유전자원 흐름을 줄이기 위한 기초자료 및 국내 재래흑염소 유전자원의 고유성을 나타내는 기초자료로 활용이 가능할 것으로 사료된다.

  • The development of trachea in fetuses on 60, 90 and 120 days of gestation and neonates of Korean native goats was investigated by microscopy and scanning and transmission electron microscopy. The results were summarized as follows; Light microscopic studies: 1. In the 60-day-old fetuses, the tracheal walls were differentiated and divided into four layers of the mucosa, submucosa, muscle and cartilage, and adventitia. The tracheal epithelium is composed of stratified ciliated columnar epithelium at 60- and 90-day-old fetuses while the epithelium observed at 120-day-old fetuses was pseudostraified ciliated colummar epithelium. 2. In the 90-day-old fetuses, tracheal glands extended into the submucosa and peripheral area of the tracheal cartilage. The blood vessels were observed in the submucosa and adventitia. The elastic and collagenous fibers were observed in the tracheal walls. 3. In the neonates, the tracheal walls consisted of mucosa with well-developed folds, submucosa, tracheal glands, muscle and cartilage, collagenous and elastic fibers, and adventitia, which were more developed than those of 120-day-old fetuses. The tracheal epithelium was developed as that in adults. Scanning electron microscopic studies: 4. In the 60-day-old fetuses, most of tracheal epithelial cells were nonciliated but short microvilli were sporadically observed on the luminal surface. On rare occasions, a few cells have solitary cilium. 5. In the 90-day-old fetuses, the ciliated cells appeared increasingly and cilia elongated longer than those of 60-day-old fetuses. 6. In the 120-day-old fetuses, the nonciliated cells covered with microvilli in dome-shape were barriered by thick carpet of cilia. The nonciliated cells also have many papillary projectons on the apical surface. 7. In the neonates, the nonciliated cells in tracheal epithelium were covered compactly with numerous cilia, and many secretory droplets were found on the cilia. Transmission electron microscopic studies: 8. In the 60-day-old fetuses, nonciliated cells of the tracheal epithelium contain large amounts of glycogen granules in the supernuclear and subnuclear areas meanwhile a few cell organelles were formed. Cilia were well formed along the apical cell membranes of the ciliated cells. Also found in the ciliated cells were basal corpuscles, mitochondria and short chains in granular endoplasmic reticulum(GER). Between the epithelial cells presented were well-defined junctional complex with zonula occludens and desmosomes. The nuclei were variable in size and shape. The more developed nucleoli were observed conspicuosly. 9. In the 90-day-old fetuses, nonciliated cells contained large glycogen granules. Accumulated glycogen granules were observed in the subnuclear and supranuclear portion of the cytoplasm. A few short microvilli were covered with glycocalyx. Ciliated cells contained numerous mitochondria and short chains of GER. 10. In the 120-day-old fetuses, the ciliated cells contained numerous mitochondria, abundant short chains of GER and nucleoli. Nonciliated cells contained some Golgi complex and mitochondria. The cell borders were well-defined and distinct junctional complex with zonula occludens, desmosomes, and interdigitorum. 11. In the neonates, well-developed goblet cells were observed in the tracheal epithelium. Ultrastructures of ciliated and nonciliated cells on the tracheal epithelia were similar in pattern as those in adults.

흑염소의 뇌염형 리스테리아증 발생 예 (Encephalitic listeriosis in a Korean native goat (Capra hircus))

  • This paper describes a naturally occurring case of meningoencephalitis associated with Listeria monocytogenes in a 4 month-old Korean native goat. The goat was raised in a herd consisting of seventy goats, and fed on straw and leftover vegetables including onions, cauliflower, and bean sprouts. The clinical symptoms of the goat were depression, in appetence, anorexia, circling, and recumbency before death. No conspicuous gross lesions were found except consolidation of the lung. Histopathologically, severe multifocal microabscesses and perivascular cuffings were observed in the brain stem and mid brain. In addition, mild nonsuppurative meningitis in the cerebrum were seen in this case. The organism isolated from the brain stem was confirmed as L. monocytogenes using the biochemical tests and PCR assay. Based on clinical signs, histopathology, and bacterial isolation; we confirmed that this case was a typical encephalitic listeriosis.

한국재래산양(韓國在來山羊)의 사양표준(飼養標準)에 관(關)한 연구(硏究) (Studies on the Feeding Standard of Korean Native Goat)

  • 한국재래산양(韓國在來山羊)의 사육방법(飼育方法)은 아직 산야지(山野地)에 계목(繫牧)을 시키는 등 관습적(慣習的)인 방법(方法)으로 사육(飼育)되고 있는 실정(實情)이어서 각 영양소(營養素)의 요구량(要求量)을 구명(究明)하여 좀 더 체계적(體系的)이고 과학적(科學的)인 사육방법(飼育方法)이 필요(必要)하다. 이에 유지(維持) 및 증체(增體)에 필요(必要)한 energy와 단백질(蛋白質)의 양(量)을 측정(測定)하여 사양표준작성(飼養標準作成)의 기초자료(基礎資料)로 사용(更用)하고자 하였으며, 그 시험결과(試驗結果)를 요약(要約)하면 다음과 같다. 1. 재래산양(在來山羊)의 일당(日當) 증체량(曾體量)$\bar{X}{\pm}SE$은 숫산양이 $80{\pm}13.9g$으로 암산양의 $41{\pm}8.9g$보다 현저히 높았다. 그리고 비육초기(肥肉初期)에는 조사료(粗飼料)만을 급여(給與)하고 후기(後期)에 농후사료(濃厚飼料)를 급여(給與)하는 것이 일당(日當) 증체량(曾體量)이 높았다. 2. Orchardgrass 건초(乾草)와 농후사료(濃厚飼料)를 무제한(無制限) 급여(給與)할 때 체중(體重)에 대한 섭취비율(攝取比率)은 각각 0.60~0.75%와 1.80~2.80%였고, 바랭이 생초(生草)만을 급여(給與)할 때는 10.50~14.50%, orchardgrass 건초(乾草)만을 급여(給與)할때 는 1.98~3.16%를 섭취(攝取)하였다. 3. 재래산양(在來山羊)의 유지(維持)에 요구(要求)되는 1일(日) 대사(代謝) energy는 체중(體重) 30kg, 21kg, 12kg일 때 각각 794.3kcal, 636.3kcal, $462.6kcal(Wkg{^{\circ}75}$당(當) 61.57kcal, 64.95kcal, 72.31kcal)로서 유령기(幼齡期)보다 성장(成長)함에 따라서 감소(減少)하는 경향(傾向)이었다. 그리고 절식시(絶食時)의 기초대사(基礎代謝)energy의 측정결과(測定結果)는 각각 492.3kcal, 453.9kcal, $389.9kcal(Wkg{^{\circ}75}$ 당(當) 44.4kcal, 50.4kcal, 61.1kcal)였다. 4. 유지(維持)에 요구(要求)되는 1일(日) 질소량(窒素量)은 체중(體重) 30kg, 21kg, 12kg일 때 각각 7.07g, 4.67g, 3.04g(가소화조단백질(可消化粗蛋白質) $Wkg{^{\circ}75}$당(當)2.71g, 2.35g, 2.09g)이었다. 5. 건초(乾草)와 농후사료(濃厚飼料)를 무제한(無制限) 급여(給與)할 때 energy의 가소화율(可消化率)은 65.8~74.6%였고 대사(代謝) energy의 비율(比率)은 50.7~62.9%였으며, $Wkg{^{\circ}75}$당(當) energy는 $DE=180.1{\pm}24.36kcal$, $ME=141.6{\pm}22.77kcal$ 였다. 6. Energy의 1일(日) 축적량(蓄積量) $(\bar{X}{\pm}SE)$$138.1{\pm}18.9kcal$였으며, 질소(窒素)의 축정율(蓄積率)은 13.3~22.5%였다.

Extrusion 가공처리 계분사료의 첨가수준이 재래산양의 육성능력, 영양소 소화율 및 체조성에 미치는 영향 (Effects of Feeding Level of Extruded Poultry Manure on Growth Performance, Nutrient Digestibility and Body Composition in Korean Native Goats)

  • 본 시험은 계분의 가축 사료화의 일환으로 계분, 옥수수 및 타피오카를 50:30:20의 비율로 혼합하여 extrusion 가공한 계분사료(EPM)의 급여수준이 반추가축의 증체량, 사료요구율, 영양소 소화율 및 도체성적에 미치는 영향을 구명하기 위하여 평균체중이 10kg 내외의 한국재래산양 수컷 15두를 이용하여 EPM을 시험사료중에 각각 0, 10, 20, 30 및 40%씩 첨가 급여하여 6주간 사양시험을 실시하였다. Extrusion 가공처리 계분사료의 수분, 조지방 및 조섬유 함량는 extrusion 가공처리 전보다 각각 10.8, 0.2 및 0.5% 감소하였고, NFE는 1.7% 증가하였다. 사료섭취량은 대조구의 596.1g/day에 비해 EPM 20 및 30% 첨가구에서 각각 620.6 및 698.5 g/day로 증가하는 경향을 보였으나, 40% 첨가구에서는 590.7g/day로 감소하였다. 일당 증체량은 대조구에서 119.8g으로 EPM 10, 20 및 30% 첨가구의 96.8, 98.3 및 $10^8$.2g/day와 거의 유사한 수준을 보였으나, EPM 40% 첨가구에서는 78.3g/day로 대조구에 비해 유의적으로 낮았다(p<0.05). 사료요구율은 대조구(5.01)에 비하여 EPM 첨가수준이 증가할수록 높아지는 경향을 보였으며, 특히 EPM 40% 첨가구에서는 대조구에 비해 현저하게 높았다(p<0.05). 건물 및 조단백질 소화율은 각 처리간에 큰 차이는 인정되지 않았으나, 조섬유 소화율은 대조구의 63.7%에 비해 EPM 10, 20, 30 및 40% 첨가구에서 각각 65.3, 67,5, 70.4 및 71.8%로 증가하는 경향을 보였다. 조지방 소화율은 EPM 40% 첨가구에서 68.5%로 대조구에 비해 유의적으로 낮았고(p<0.05). 다른 EMP 첨가구에서는 거의 유사한 수준을 나타내었다. 도체율은 대조구의 49.7%에 비해 EPM 10, 20, 30 및 40% 첨가구에서 각각 49.7, 48.3, 47.8 및 45.2로 감소하는 경향을 나타내었다. 산양고기중의 지방 함량은 대조구의 16.3%에 비해 EPM 첨가구 모두에서 유의적으로 감소하였다(p<0.05).

Novel Nucleotide Variations, Haplotypes Structure and Associations with Growth Related Traits of Goat AT Motif-Binding Factor (ATBF1) Gene

  • The AT motif-binding factor (ATBF1) not only interacts with protein inhibitor of activated signal transducer and activator of transcription 3 (STAT3) (PIAS3) to suppress STAT3 signaling regulating embryo early development and cell differentiation, but is required for early activation of the pituitary specific transcription factor 1 (Pit1) gene (also known as POU1F1) critically affecting mammalian growth and development. The goal of this study was to detect novel nucleotide variations and haplotypes structure of the ATBF1 gene, as well as to test their associations with growth-related traits in goats. Herein, a total of seven novel single nucleotide polymorphisms (SNPs) (SNP 1-7) within this gene were found in two well-known Chinese native goat breeds. Haplotypes structure analysis demonstrated that there were four haplotypes in Hainan black goat while seventeen haplotypes in Xinong Saanen dairy goat, and both breeds only shared one haplotype (hap1). Association testing revealed that the SNP2, SNP5, SNP6, and SNP7 loci were also found to significantly associate with growth-related traits in goats, respectively. Moreover, one diplotype in Xinong Saanen dairy goats significantly linked to growth related traits. These preliminary findings not only would extend the spectrum of genetic variations of the goat ATBF1 gene, but also would contribute to implementing marker-assisted selection in genetics and breeding in goats.

Digestibility, ruminal fermentation, and nitrogen balance with various feeding levels of oil palm fronds treated with Lentinus sajor-caju in goats

  • Objective: This study was an attempt to investigate the effect of levels of fungal (Lentinus sajor-caju) treated oil palm fronds (FTOPF) on digestibility, rumen fermentation, and nitrogen balance in goats. Methods: Four 16 month old male crossbred (Thai Native${\times}$Anglo Nubian) goats with initial body weights of $33.5{\pm}1.7kg$ were randomly assigned according to a $4{\times}4$ Latin square design. Four levels of FTOPF were assigned for feed intake. The experimental treatments consisted of 0%, 33%, 67%, and 100% of oil palm fronds (OPF) being replaced by FTOPF. Results: The results revealed that total dry matter intake and nutrient intake were not influenced (p>0.05) by the inclusion of FTOPF. However, the efficiency values of the digestibility of the dry matter, organic matter, crude protein, neutral detergent fiber, acid detergent fiber, and acid detergent lignin on FTOPF were higher (p<0.05) in treatments with 33%, 67%, and 100% of FTOPF compared with 0% of FTOPF. FTOPF feeding did not change the rumen pH, temperature, and $NH_3-N$. However, the FTOPF levels did affect the total volatile fatty acid (VFA), molar proportion of acetate, propionate, butyrate, ratio of acetic (propionic acid and acetic) plus butyric (propionic acid), and production of $CH_4$. The totals of VFA and propionate was lower in goat fed with 0% of FTOPF than in those of the other groups (p<0.05). The amount of nitrogen retention based on g/d/animal or the percentage of nitrogen retained was the lowest the goat fed with 0% of FTOPF (p<0.05), whereas nitrogen intake, excretion, and absorption were not changed among treatments. Conclusion: Based on this study, FTOPF could be effectively used as an alternative roughage source in total mixed ration diets, constituting at least up to 100% of OPF.
