The Effect of Korean Native Chicken Breed Information on Consumer Sensory Evaluation and Purchase Behavior

  • This study examined how chicken breed affects consumer behavior. The breeds evaluated were a Korean native chicken (KNC) and a commercial broiler (CB). Consumer behavior was measured by evaluating sensory attributes (taste, odor, and texture) and purchase behaviors [satisfaction, purchase intention, and willingness to pay (WTP)]. The sensory evaluation was conducted using healthy Korean consumers (n=100). The chicken was cooked in baeksuk (Korean traditional chicken soup), which is a seasonal summer health food in Korea. The participants evaluated sensory attributes and purchase behaviors between blinded samples of baeksuk (CB) and unblinded samples of baeksuk (KNC). The sensory evaluation involved chicken breasts and legs. The participants considered KNC as having a more umami taste, a chewier and juicer texture, and a less metallic odor than CB. Moreover, when participants were given KNC breed information, they expressed higher satisfaction, purchase intention, and WTP. The results of this study contribute to consumer behavior literature by revealing the effects of breed information on consumer sensory perceptions and purchase behaviors. Furthermore, the findings provide evidence that branding KNC is one way to increase its demand and low market share while contributing to sustainable consumption.

Variation in Reproductive Efficiency of Indonesian Native Cows

  • This study was conducted in North Sulawesi, Indonesia to identify the factors affecting reproductive efficiency and to determine the contribution of each factor in reproductive efficiency of Indonesian native cows. Records of reproductive efficiency, body size (hip height, body weight, body length, heart girth) as well as age on one hundred and sixty seven Indonesian native cows (seventy six cows of the Bali breed and ninety one cows of the synthetic breed from unknown proportion of th Madura, the Sumba and the Ongole breeds) of 58 farmers were available in this research. The data were analyzed by covariance analysis using SAS package including reproductive efficiency as a dependent variable and breed, body size (hip height, body weight, body length, heart girth) and age of cows as independent variables. The contribution of each independent factor to dependent variable was estimated as the percentage of the sum of square in the corrected total sum of squares. The results showed that body weight and body size of cows would be considered as th most important factors affecting reproductive efficiency of Indonesian native cows. Therefore, the reproductive efficiency could be increased by 1) using cows that reached a target weight of at least 250 kg, 2) using cows with relatively well developed height at hip of at least 117 cm, heart girth of at least 150 cm, and body length of at least 125 cm, 3) keeping breeding cows that reach a target age of 24 months old with the maximum interval between postpartum and next breeding season of 90 days for each year, and 4) using good nutritional feeding to reach the optimum weight and body condition.

Mitochondrial DNA Diversity of Korean Native Goats

  • Korean native goats have lived on the Korean peninsula for more than 2,000 years and are regarded as a valuable genetic resource for the world. As an initial step to investigate the genetic structures of this breed, phylogenetic analysis and calculation of genetic diversities have been performed using mitochondrial DNA (mtDNA) sequence variations. A total of 19 Korean native goats were grouped into six haplotypes and the large majority of haplotypes were present in 13 animals. All mtDNA of these Korean goats belonged to the mitochondrial (mt) lineage A and revealed remarkably small genetic distances within the population when compared with other Asian goat populations, indicating less genetic variation in the Korean native goats. These results indicate high-inbred status of the Korean native goats and will influence breeding and conservation strategies adopted for this breed.

Genetic Variation and Relationships of Korean Native Chickens and Foreign Breeds Using 15 Microsatellite Markers

  • The purpose of this study was to assess the genetic variation and establish the relationship amongst breeds and strains using 15 chicken specific microsatellite markers. A total of 285 unrelated DNA samples from four Korean native chicken strains (Black strain of Korean native chicken; KL, Red Brown strain of Korean native chicken; KR, Ogol strain of Korean native chicken; KS and Yellow Brown strain of Korean native chicken; KY) and three introduced chicken breeds (F strain of White Leghorn; LF, K strain of White Leghorn; LK, Rhode Island Red; RC and Cornish; CN) were genotyped to estimate within and between breed genetic diversity indices. All the loci analyzed in 15 microsatellite markers showed a polymorphic pattern and the number of alleles ranged from 5 to 14. The polymorphism information content (PIC) of UMA1019 was the highest (0.872) and that of ADL0234 was the lowest (0.562). The expected total heterozygosity (He) within breed and mean number of observed alleles ranged from 0.540 (LF) to 0.689 (KY), and from 3.47 (LK) to 6.07 (KR), respectively. The genetic variation of KR and KY were the highest and the lowest within Korean native strains, respectively. The genetic distance results showed that Korean native chicken strains were separated with the three introduced chicken breeds clustered into another group. The lowest distance (0.149) was observed between the KR and KL breeds and the highest distance (0.855) between the KR and LK breeds. The microsatellite polymorphism data were shown to be useful for assessing the genetic relationship between Korean native strains and other foreign breeds.

Length polymorphism in OGT between Korean native pig, Chinese Meishan, and the Western pig breeds

  • Background: The Korean native pig (KNP) is generally thought to have come from northern China to the Korean peninsula approximately 2000 years ago. KNP pigs were at the brink of extinction in the 1980s, since then efforts have been made to restore the breed by bringing together the remaining stocks in South Korea. As a result, KNP was registered as a breed in 2006. To find additional breed-specific markers that are distinct among pig breeds, variations in O-linked N-acetylglucosamine transferase (OGT) were investigated. OGT is located on chromosome X and catalyzes the post-translational addition of a single O-linked-${\beta}$-N-acetylglucosamine to target proteins. Findings: Length polymorphism in the intron 20 of OGT was identified. The intron 20 of OGT from Duroc, Landrace, and Yorkshire breeds was 281-bp longer than that from either KNP or Chinese Meishan pigs. The difference between the Western pig breeds (BB genotype) and KNP or Meishan pigs (AA genotype) was due to an inserted 276-bp element and the 5-bp ACTTG. Conclusions: The polymorphism in OGT identified in this study may be used as an additional marker for determining the breed of origin among Meishan and the Western pig breeds. The length polymorphism suggests that the locus near OGT is not fixed in KNP. This marker would be relevant in determining the breed of origin in crossbred pigs between KNP pigs with known genotypes and the Western pig breeds with BB genotypes, thus confirming the contribution of the X chromosome from each breed.

닭 백혈구 특이 단트론항체를 이용한 한국 재래닭의 유전 면역학적 특성에 관한 연구 (Immunogenetic characterization of Korean native chickens by monoclonal anibodies to chicken leukocyte differentiation antigens)

  • The immunogenetic analysis was performed to characterize the Korean native chickens (KNC) determined by monoclonal antibodies specific to chicken leukocyte differentiation antigens and flow cytometry. A total of 174 chickens including 58 KNC (black, brown and darkbrown colored), 77 foreign breed (Nagoya, White Reghorn, Rhode Island and Cornish) and 39 mixed breed (19 KNC with Nagoya and 20 KNC with Rhode Island) separately growing at Animal Science and Technology Institute were examined. The proportion of cells expressing MHC class II molecule (B-L in chicken) was significantly high in KNC. Proportion of CD4+ T helper cells was also higher in KNC and two mixed breed than that in foreign breed. However, proportion of CD8+ cells and TCR1 + (${\gamma}^{\delta}$ T cell receptor) cells was the lowest among the breed examined. Otherwise, those proportions were significantly high in White leghorn and two mixed breeds with two exclusive subpopulations. The two subpopulations were also typically shown in MHC class $II^+$ cells in KNC and one mixed breed, black-colored KNC with Nagoya. Although genotypic analysis was not pursued to characterize the immunogenetic properties of KNC, difference of phenotypic expression based on leukocyte differentiation molecules could be elucidated in KNC in this study.

Genetic Variability and Relationships of Native Japanese Chickens Assessed by Microsatellite DNA Profiling - Focusing on the Breeds Established in Kochi Prefecture, Japan -

  • Blood samples were collected from eight native Japanese breeds of chickens (Miyadi-dori, Ohiki, Onaga-dori, Shoukoku, Tosa-Jidori, Tosa-Kukin, Toutenkou and Uzurao) and two foreign breeds of chickens (White Leghorn and Rhode Island Red) to examine the genetic variability and relationships among the breeds by using a microsatellite DNA technique. Except for the Shoukoku breed, the other Japanese chicken breeds all originate from Kochi Prefecture. Ohiki, Onaga-dori, Tosa-Jidori, Toutenkou and Uzurao are fancy fowl, and Miyadi-dori and Tosa-Kukin are utility fowl. Among the fancy fowl, Ohiki, Onaga-dori, and Toutenkou males have thick and long feathers in the saddle and tail. Genetic variabilities of the 20 microsatellites examined, varied depending on the breed: the mean number of alleles per locus ranged from 2.05 (Miyadi-dori) to 3.90 (Rhode Island Red); proportion of polymorphic loci ranged from 0.75 (Miyadi-dori) to 1.00 (Rhode Island Red, Shoukoku and Uzurao); and mean expected heterozygosity ranged from 0.330 (Miyadi-dori) to 0.607 (Rhode Island Red). Unique microsatellite alleles were detected in each breed. Using the neighbour-joining method, phylogenetic trees were constructed based on the genetic distances of D$_{A}$ and D$_{ST}$. Among the breeds originating from Kochi Prefecture, fancy and utility breeds belonged to different clusters. Among the fancy breeds, those having thick and long feathers in the tail and saddle showed a close genetic relationship to the Shoukoku breed, which also has thick and long feathers in the tail and saddle.

  • 본 연구는 한국재래오골계의 외모형질에 대한 특징과 생장을 조사하여 품종학적인 정립과 유전적 개량 및 유전자보존을 위한 기초자료를 얻기 위하여 천연기념물 265호로 지정된 연산오골계 총 1,156수를 공시하여 실시하였다. 조사항목으로써는 우모색과 지수, 정강이와 관의 색과 정강이, 관, 우역에 따른 깃털 생장 및 생존율 등이었으며, 이들에 관한 수치들이 표의 형태로 본문에 제시되었다. 얻어진 결과들을 요약하면, 우모는 정상우에 흑색과 백색으로 구분되었으나 백색우모계의 빈도는 극히 낮고, 지색은 흑색모계에서 흑색지와 백색모계에서 백색지로 구분되고, 관은 암ㆍ수 모두 흑색계통의 단관이고, 각우가 없는 회흑색 정강이에 사지이며 흑색안을 갖는 것으로 요약된다.

Effects of Sex and Breed on Meat Quality and Sensory Properties in Three-way Crossbred Pigs Sired by Duroc or by a Synthetic Breed Based on a Korean Native Breed

  • This study was conducted to determine the effects of breed and sex on meat quality and sensory properties of the loin in three-way crossbred pigs: $Landrace{\times}Yorkshire{\times}Duroc$ (LYD) and $Landrace{\times}Yorkshire{\times}Woori$ (LYW) black pig synthesized by Korean native breed. Carcass traits did not differ by breed. Carcass weight and backfat thickness were higher in castrates than in gilts (p<0.01). LYW showed significant high values in fat content, cooking loss, and water-holding capacity (WHC) than LYD (p<0.05). Redness and yellowness of the meat were higher in LYW than in LYD (p<0.01). Further, LYW had lower pH and shear force than LYD (p<0.001). Significant high scores in color and flavor were obtained in LYW or gilts compared to LYD or castrates by sensory panel, respectively (p<0.05). However, other sensory traits did not differ by breed or sex. Capric acid (C10:0) was higher in LYD than LYW (p<0.001). However, stearic acid (C18:0) and saturated fatty acid (SFA) contents were higher in LYW than LYD (p<0.05). Eicosenoic acid (C20:2) and the n6/n3 ratio were higher in gilts than in castrates, whereas SFA content was higher in castrates than in gilts. These results suggest that certain physicochemical qualities of meat and sensory properties are improved in LYW compared to LYD. This study could provide basic data on meat quality of crossbred pigs with Woori black pig as a terminal sire.

한국재래닭 COI 유전자의 단일염기다형 분석 (Single Nucleotide Polymorphism Analysis of the COI Gene in Korean Native Chicken)

  • 조류에서 미토콘드리아내 유전자인 cytochrome c oxidase I(COI)는 종 구분을 위한 바이오마커로 많이 이용이 되고 있다. 본 연구는 이 유전자의 변이를 이용하여 닭의 품종 구분이 가능한지를 실험하기 위하여 실시하였다. 그 결과 한국 재래닭은 공시축으로 사용한 산란계인 White Leghorn과 육계인 Cornish가 가지고 있는 단일염기다형을 모두 가지고 있는 것으로 판명되어 품종구분을 위한 마커로 사용하기에는 적합하지 않은 것으로 확인되었다. 그러나 본 연구 결과를 통하여 한국재래계가 육계와 산란계의 특성을 모두 가지고 있다는 기존의 학설을 뒷받침하였으며, 품종 구분을 위하여 미토콘드리아와 genomic DNA 유래 단일염기다형 선발의 중요성을 확인할 수 있었다.
