The Effect of the Hydrogen Bond Network in the $S_1$-pocket on Catalytic Activity of Serine Protease, Achromobacter Protease I (API)

  • Crystal structural analyses of the API-TLCK complex revealed that the ${\epsilon}$-amino group (NZ) of the lysyl part of TLCK forms hydrogen bonds with OD1 of $Asp^225$ which is a substrate specificity determinant of API, OG of $Ser^214$, O of $Ser^214$, OG1 of $Thr^189$, and O of $Thr^189$ l89/. The ${\beta}$-carboxyl oxygen of $Asp^225$ forms hydrogen bonds with the NE1 of $Trp^182$. From these observations, it is thought that besides $Asp^225$, $Thr^189$, $Ser^214$, and $Trp^182$ may also contribute to the steric specificity for lysine and high proteolytic activity of API. The side-chain hydroxyl groups of $Thr^189$ and $Ser^214$ were removed to elucidate the role of these hydrogen bonds in the $S_1$-pocket. The $k_{cat}$/$K_m$ of T189V, S214A, and T189V.S214A were decreased to 1/4, 1/3, and 1/46, respectively, of the value for native API. The decreased activities were mainly due to the increase of $K_m$. The CD and fluoroscence spectra of the three mutants were similar to those of wild-type API. With regards to the kinetic parameters ($K_i\;and\;k_2$) of mutants for the reaction involving TLCK and DFP, $k_2$decreased by increase of $K_1$ only. These results suggest that the decreased catalytic activity of these mutants is caused by the partial loss of the hydrogen bond network in the $S_1$-pocket. On the other hand, the similarity of enzymatic properties between W182F and the native enzyme suggests that the hydrogen bond between OD2 of $Asp^225$ and NE1 of $Trp^182$ is not directly related to the reaction of $Asp^225$ with the substrate.

  • It is difficult to access and share design data among heterogeneous CAD systems. Usually, different CAD systems exchange the design data using a neutral format such as IGES or STEP. A prototype CAD system which uses a geometric kernel and a commercial database management system has been implemented. The prototype system used the Open Cascade geometric kernel and the commercial object-oriented database ObjectStore. STEP provides the database schema. The database can be accessed from commercial CAD systems such as SolidWorks or Unigraphics. The data access module from a commercial CAD system is developed with the CAD system's native API, ObjectStore API functions, and ActiveX.

  • Recently the market of smart phone applications grows very fast. And users want more various and rich experiments using enhanced smart phone functions. But the development of smart phone applications is not an easy job. Specially to control smart phone sensors can be realized by using each native programming languages in experts level. Moreover the process needs separate development based on each smart phone OS. Development of the Web-based smart phone application using Web Common APIs, known as 'WebApp', is one of solutions to overcome these problems. The method includes interface constructions with HTML, and Web Common API calls and accesses to smart phone device APIs with Java scripts. In this paper, the experience in development of the smart phone e-book application is introduced, which is implemented with comport and OS independent WebApp development method.

  • According to the 2021 Mobile Augmented Reality Market Report by Korea Innovation Foundation, the augmented reality market has been growing recently due to the growth of the mobile augmented reality market along with the development of smartphones. In order to provide augmented reality services to mobile users, it is necessary to create native apps for each device. However, there are problems such as maintenance costs for multi-platform and low accessibility due to app download. Recently, the construction of augmented reality system in a web-based environment by using the WebXR Device API is in progress, but it is judged that the system using the WebXR Device API is still in the research stage. In this paper, a responsive multi-platform environment was built using the WebXR Device API, Three JS, and React, and a function to provide augmented reality service to mobile and web users was implemented. As a result of the experiment, it was confirmed that the interlocking of augmented reality was successfully implemented in a responsive web environment, and look forward to the augmented reality service operating on the web in the future.

  • The teleoperation system applies all of the industrial fields due to the development of the network infrastructure. It is one of the indispensable elements for controlling the robot at a remote sight and monitoring the limit or unknown environment. The common teleoperation robot system is what has the visual module to supply the network system and realistic UI to the existed robot system. Therefore, remarked that the fusion between modules and transmission of visual data the remarked the important element to improve the robot application in the various environments. Delay of development time by robot platform and noneffective communication among developers are also problem to approach. In this paper we propose the independent teleoperation system. The main application language is JAVA in this system, which is applied JAVA API like JNI and JMF to construct the effective teleoperation system. The system has the both side communication system between sever and client as a basic structure. The visual data that is attached the robot at a remote sight is captured by JMF API and then is transmitted to the web browser called client by RTR protocol. JNI is used to connect between JAVA and the lower part application (sensor fusion, motion control.) of the robot programmed by various Native languages. The proposed system is the application that can perform the elements, for instance transmission of visual data, the fusion of various native application modules and the effective network communication, with any platform.

  • This paper is about environment-based low-code and no-code execution platform and execution method that combines hybrid and native apps. In detail, this paper describes the Low-Code/No-Code execution structure that combines the advantages of hybrid and native apps. It supports the iPhone and Android phones simultaneously, supports various templates, and avoids developer-oriented development methods based on the production process of coding-free apps and the produced apps play the role of Java virtual machine (VM). The Low-Code /No-Code (LCNC) development platform is a visual integrated development environment that allows non-technical developers to drag and drop application components to develop mobile or web applications. It provides the functions to manage dependencies that are packaged into small modules such as widgets and dynamically loads when needed, to apply model-view-controller (MVC) pattern, and to handle document object model (DOM). In the Low-Code/No-Code system, the widget calls the AppOS API provided by the UCMS platform to deliver the necessary requests to AppOS. The AppOS API provides authentication/authorization, online to offline (O2O), commerce, messaging, social publishing, and vision. It includes providing the functionality of vision.

  • In the multimedia tele-medical system the medical informations are stored and managed in multimedia database. Also, multimedia DBMS is essential in order to manage large scale medical informations, and the remote access function is necessary for the distributed processing at all around the hospital. For the multimedia tele-medical information that is composed of image/picture, data, video, and audio, a high-speed telecommunication network is necessary that can provide separated connections for each medical information type with different QoS. The commercial DBMSs are based on the TCP/IP socket API(Application Programming Interface) that does not provide multiple QoS. Also, each commercial DBMS has its own API that is incompatible with other DBMS. In this paper, we propose a multimedia DBMS agent for the remote access of the multimedia database in the tele-medical system. The proposed multimedia DBMS agent is based on the ATM API that can provide high-speed data transfer capability and multiple QoS connections. Also, the proposed multimedia DBMS agent is independent of the commercial DBMS. We explain the functional architecture of the multimedia DBMS agent, implementation technology on the ATM network environment, and the result of performance analysis.

  • Since the recent COVID-19 Pandemic, the ransomware fandom has intensified along with the expansion of remote work. Currently, anti-virus vaccine companies are trying to respond to ransomware, but traditional file signature-based static analysis can be neutralized in the face of diversification, obfuscation, variants, or the emergence of new ransomware. Various studies are being conducted for such ransomware detection, and detection studies using signature-based static analysis and behavior-based dynamic analysis can be seen as the main research type at present. In this paper, the frequency of ".text Section" Opcode and the Native API used in practice was extracted, and the association between feature information selected using K-means Clustering algorithm, Cosine Similarity, and Pearson correlation coefficient was analyzed. In addition, Through experiments to classify and detect worms among other malware types and Cerber-type ransomware, it was verified that the selected feature information was specialized in detecting specific ransomware (Cerber). As a result of combining the finally selected feature information through the above verification and applying it to machine learning and performing hyper parameter optimization, the detection rate was up to 93.3%.

  • We have come a long way in the information age. Thanks to the advancement of such technologies as the internet, we have discovered new ways to convey information on a broader scope. However, negative aspects exist as is with anything else. These may include invasion of privacy over the web, or identity theft over the internet. What is more alarming is that malwares so called 'maliciouscodes' are rapidly spreading. Its intent is very destructive which can result in hacking, phishing and as aforementioned, one of the most disturbing problems on the net, invasion of privacy. This thesis describes the technology of how you can effectively analyze and detect these kind of malicious codes. We propose sequencial hybrid analysis for API calls that are hooked inside user-mode and kernel-level of Windows. This research explains how we can cope with malicious code more efficiently by abstracting malicious function signature and hiding attribute.
