염류 스트레스에 대한 수도품종의 생리적 반응에 대한 연구 (Studies of Physiological Response to the Salt Tolerance of Rice Cultivars)

  • This study was to investigate the dry weight, the amount of Na+ and K+ water potential and leaf photosynthesis rate in plants for determining the salt tolerance mechanism in rice cultivars on soil and solution culture with NaCl. The results obtained in this study are summarized as follows ; In general, rice cultivars, cv. Tetep and Jinbu, having high salt tolerance in ID(identified on dry matter production level) showed the higher salt tolerance in RGR (relative growth rate), compared with rice cultivars(cv. Nonglim 41ho, Dunraebyeo and Sobackbyeo) having low salt tolerance. The contents of Na in rice differed depending on culivars and plant parts. Tetep contained 2.9times higher amounts of Na+ than leaf blade and root part. High salt tolerance cultivar Obongbyeo showed a larger decrease in osmotic potential than low salt tolerance cultivar Dunraebyeo suggesting that osmotic adjustment was developed under salt stress conditions in a salt tolerant cultivar . In order to know the IY(identified on grain yeild level using rice cultivars having different salt tolerance the capacity of photosyntheiss was investigated. The capapcity of photosynthesis in cv. Tetep and Obongbyeo having high salt tolerance was much higher that in cv.Dunraebyeo and Nonglim 41 having low salt tolerance.

흰쥐 적출대동맥의 수축력에 미치는 열과 Nacl의 영향

  • In this study, in order to examine whether salt and heat shock stress would alter or not contraction and relaxation of isolated rat aorta. Under anesthesia with sodium pentobarbital(50 mg Kg$^{-1}$ i.p.), male Sprague Dawley rats weighing 300-330 g were subjected to 0, heat shock combined salt stress, where as the sham group was left at modified Krebs-bicarbonate solution. To measure contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And the strip was checked for expression of heat shock protein(Hsps) by means of western blotting. The combination group of heat and 50 mM NaCl group increased vascular contractility, and the heat and 150 mM NaCl group decreased vascular contractility for 5 hours, and then recovered for 8 hours compared to that of control. Expressin of Hsp 70 of vascular muscle of rat aorta more increased by combination of heat and NaCl treatment than those of single treatment of heat or NaCl treatment, and vascular Hsp 70 showed a little decrease at 8 hours compared at 5 hours. These result indicate that mixed environmental stress either increased or decreased in vascular contractility by combination of heat and NaCl concentration.

중성염이 Poly(ethylene terephthalate) 직물의 알칼리 가수분해에 미치는 영향(II) (Effects of Neutral Salts on Alkaline Hydrolysis of Poly(ethylene terephthalate) (II) - Anionic Effect -)

  • Neutral salts have negative or positive effects on the rates of many chemical reactions and also on the rates of acidic and alkaline hydrolysis of carboxylic esters. The direction of neutral salt effects on the hydrolysis of ester depends on the charge of esters. Neutral salts accelerate alkaline hydrolysis of esters with negative charge, but decelerate alkaline hydrolysis of esters with positive charge, and have little effect on the alkaline hydrolysis of neutral esters. It is expected that the rate of the alkaline hydrolysis of Poly(ethylene terephthalte) (PET), polymeric solid carboxylic polyester with carboxyl end group at the polymer end, is also influenced positively by neutral salts. In the present work, to clarify the mechanism of the neutral salt effect on the alkaline hydrolysis of PET, many salts with different anions like NaF, NACl, NaBr, NaI were added to the aqueous alkaline solutions. Then PET was hydrolyzed with aqueous solutions of many salts in alkali metal hydroxides under various conditions. Some conclusions obtained from the experimental results were summarized as follows. The reaction rate of the alkaline hydrolysis of PET was increased by the addition of neutral salts and In k was increased nearly linearly with the square root of ionic strength of reaction medium. This fact suggested that the ionic strength effect by Debye-Huckel and Bronsted theory was exerted on the reaction. The specific salt effect was also observed. The reaction rate was increased with the decrease in the nucleophilicity of anions of neutral salts, i.e., in the order of $F^-$ <$Cl^-$<$Br^-$<$I^-$. It was thought that the reaction rate was increased in the order of $F^-$ <$Cl^-$<$Br^-$<$I^-$. because the completion of anions with $OH^-$ for carbonyl carbon became weaker with the decrease in the nucleophilicity and with the increase in the size of anions.

  • 음식물류폐기물의 무항생제 액상사료화를 통한 고급자원화를 이루기 위하여 항생제 대체용 항세균 활성 유산균주 13 종을 돼지 장관 및 분변에서 분리하였다. 분리 유산균은 모두 내염성(Nacl 4%), 내산성 (pH4.0) 및 내담즙성 (oxgall 0.8%)을 보였으며, $37^{\circ}C$에서 생장이 가장 높게 나타났다. 분리 유산균주중 Lactobacillus plantarum CJY-22, Pediococcus sp. CJY-41, Lactobacillus brevis CJY-42 및 Lactobacillus arizonensis CJY-3 균주가 가장 높은 항세균 활성 및 광역의 항세균 활성 스펙트럼을 나타냈으며, Lactobacillus plantarum CJY-22 균주의 경우 $25^{\circ}C$에서도 생장이 원활하여 음식물류폐기물에 적용시 가온없이 발효공정을 수행할 수 있는 유용 균주로 적용할 수 있을 것으로 사료된다.

  • Current drying and encapsulation methods for probiotics manufacturing are complicate and cost-burdened processes. The aim of this study was to develop a simple ingredient mixture to make probiotic granules via one-step process, providing not only a cryoprotective effect during freezing and drying but also high survival ratio in gastrointestinal tract. As cryoprotectans, commercially available ingredients including skim milk, monosaccharide (trehalose or glycerin), maltodextrins (with low or high degree of equivalents) were used. Their cryoprotective effect during lyophilization and survival ratios in artificial gastric juice and bile salt were measured against 3 strains of lactic acid bacteria (LAB) (Lactobacillus plantarum, Lb. brevis, and Lactococcus lactis). As results, 3 mixtures with different compositions showed a cryprotective effect on LAB tested and the best compostion was dependant upon LAB; skim milk 10%, trehalose 15%, glycerin 0.5%, and NaCl 1% was for Lb. plantarum and Lc. lactis, and maltodextrin 10% instead of skim milk was for Lb. brevis. In addition, those mixtures showed similar survival effect on LAB tested. These results demonstrate that skim milk or maltodextrins with trehalose, glycerin, and NACl can be effectively used for onestep lyophilization of LAB as an alternative method of encapsulation.

균류키토산의 균체생산에서 기질농도 최적화에 관한 연구 (Optimization of Substract Concentration in Cell Production of Fungal Chitosan)

  • In the process of producing chitosan from crustacean shell, the use of excessive acid and alkli is causing the problems of environmental pollution and of production cost. In this study, one way to solve these problems is to cultivate fungi, then, to extract chitosan from the cell wall. By means of flask incubation and batch cultivation, the optimum cultivation conditions for mass production of continuous cultivation was found. Four strains used for the production of fungal chitosan were Gongronella butleri IF08080, Absidia coerulea IF05301, Rhizopus delemar IF04775, Mucor tuberculisporus IF09256. In flask incubation to select strain of producing much chitosan by means of experiment of the effect of initial pH, Absidia coerulea IFO 5301 had highest yield in FCs, 258.1 $\pm$ 47.3 mg/200 $m\ell$l at pH 6.5. In flask incubation under the optimum cultivation condition, temperature 27$^{\circ}C$, culture time 6days, glucose 2%, peptone 1%, (NH$_4$)$_2$ SO$_4$ 0.5%, $K_2$HPO$_4$ 0.1 %, Nacl 0.1 %, MgSO$_4$ㆍ7$H_2O$ 0.05%, CaCl$_2$ㆍ2$H_2O$ 0.01 %, the yield of DCW brought the highest yields. In batch bioreactor, the optimum cultivation condition was that cell suspended solution was 70 $m\ell$, aeration rate 0.5 l/min, agitation rate 800 rpm, culture time 36 hr. In continuous bioreactor, the optimum substrate flow rate was 4 ι/day.

사람 NC-37 세포에서 Benzo(a)pyrene과 Doxorubicin에 의한 Nuclei내전과 c-myc 유전자의 발현에 대한 연구 (Studies on the Nuclei Adduction and Expression of c-myc Gene by Benzo(a)pyrene and Doxorubicin in Human NC-37 Cells)

  • Formation of adduct was studied in benzo(a)pyrene(BP)- and doxorubicin(Dx)-treated human NC-37 cells and isolated nuclei. Major adducts formed were determined by fluorescence absorption spectrophotometery and DNA-lin-ked protein assay. When isolated nuclei were exposed to carcinogens BP and DMBA, and anticancer drugs m-AMSA, ellipticine and Dx, varying degrees of adduct formation occured between DNA-protein complex and these drugs. When the mixture was centrifuged 1.7 M sucrose solution, binding BP and DMBA appeared to be similar between the sediment and the supernatant. When the sediment was centrifuged again with 0.35% polymin-P, the amount of BP bound was 2-fold greater in the protein(1077$\pm$55cpm) than in DNA fraction (470$\pm$20cpm), whereas that of DMBA was 1.6-fold greater in the DNA than in protein fraction. In the case of m-AMSA, ellipticine and Dx, the amount of binding was slightly greater in supernatant than in sediment in centrifugation with 1.7 M sucrose, and more than 3 times greater in the DNA- than in protein- fraction in centrifugation with 0.35% polymin P. DNA fractions which associated with a subset of nonhistone chromosomal protein were isolated from NC-37 cells exposed to $^{3}$H-BP and $^{14}$C-Dx. They were separated into two distince components DNA-S and DNA-P by centrifugation with 2M Nacl chromatin extraction. The results indicated that the amount of $^{3}$H-BP bound was 6.0-fold greater in DNA-P as compared with DNA-S, while that of $^{14}$C-Dx binding appreaed to be 6.2-fold greater in DNA-S than in DNA-P fraction. When $^{3}$H-BP binding wasdetermined in the presence of cold Dx, the amount of binding was reduced only in the DNA-P fraction, indicating that the interaction between DNA and protein is decreased. Gene expression by these drugs, BP treated cells were increased to compare with nomal cells but reduced by treatment with BP-Dx. These results suggest that the protein moiety which tightly bound to DNA-P fraction may play an important role in the regulation of gene expression.

Studies on the Sorption and Fixation of Cesium by Vermiculite

  • 천연점토 광물의 이온 교환 흡착능은 비교적 낮지만, 여러가지 활성화법이 고찰되고 있음으로서 성능의 개선이 기대되고 있다. 그중에서도 교환흡착능이 크며, 많은 양이 산출되고 있는 국산 vermiculite의 Nacl 및 KCI 처리에 의한 Cs+ ion의 교환흡착 및 고착상태에 관하여 방사성 폐액 처리의 응용면에서 연구되었으며, 그 결과는 다음과 같다. (1) pH의 증가와 더불어 Cs+ ion의 제렴율도 증가하였다. (2) 약 $10^{-3}$M CsCl 용액중 에서 Cs+ ion의 제렴효율이 가장 높았다. (3) Na-vermiculite에 의한 Cs+ ion의 교환 흡착능이 K-vermiculite에 비해 양호하였다. (4) $H_2O$, CsCl, NaCl 및 KCl 용액에 의한 추출 실험에서 80% 정도의 Cesium이 vermiculite상에 고착되었다. 이상의 결과로 부티 국산 vermiculite도 Ca+ ion의 교환흡착에 대한 친화력이 크며, NaCl 처리에 의하여 그 성능을 크게 향상시킬 수 있고, 고 준위 방사성 폐액의 지하저장에서 오는 누출천상을 저지시키기 위한 외각 충진제로서도 그 효율성을 보여 주고 있다. 또한 X-ray 회절상에서 보여 준 바와 같이 인위적으로 NaCl 처리에 의하여 biotite를 vermiculite로 전환시킬 수 있다는 것을 암시하고 있다.

중이온 빔 조사에 의한 담배의 돌연변이 유도와 내염성 식물의 선발 (Mutation Induction and Selection of Salt-tolerant Plants by Heavy-ion Beam Irradiation in Tobacco Proembryo)

  • 중이온 빔을 이용한 방사선 돌연변이의 기초자료를 얻고자 담배의 수분ㆍ수정 직후의 원배에 100 Gy의 중이온빔을 조사하여 $\textrm{M}_{1}$세대의 종자를 얻었다. NaCl 20과 25%를 포함된 MS배지에 $\textrm{M}_{1}$세대의 종자를 파종하여 17개통의 NaCl저항성 식물체를 선발하였다. 저항성 식물체는 BY-4품종에 $^{14}\textrm{N}$빔을 조사한 처리구에서만 관찰되었다. $\textrm{M}_{1}$세대의 저항성 식물체의 경우 영양생장기간 중에는 외형적 변이가 관찰되지 않았으나 생식생장기의 화기에 있어서는 다양한 변이가 관찰되었다. 특히 암술 수술의 길이의 변화, 꽃잎의 변화, 불완전 화분립, 줄기당 꽃의 착생수 감소 등의 변이가 발생하였다. $\textrm{M}_{1}$세대에서 17개의 NaCl저항성 계통 중에서 8계통은 $\textrm{M}_{2}$ 세대에도 저항성형질이 발현되었고, 또한 $\textrm{M}_{2}$세대의 3계통은 Mannitol이 포함된 배지에서 저항성을 보여주었다.

미질향상을 위한 간척지 토양 염농도별 적정 질소시비량 (Nitrogen Fertilizer Management for Improving Rice Quality under Different Salinity Conditions in Tidal Reclaimed Area)

  • 본 연구는 남서해안 간척지에서 토양 염농도(저염; 0.1%, 중염; 0.3∼0.4%)별로 쌀 품질 향상을 위한 적정 질소 시비량을 구명하기 위하여 시험한 결과를 요약하면 다음과 같다. 1.유추분화기의 초장은 질소 시비량이 많을수록 컸고, 토양염농도간에는 저염 토양에서 켰다. 2. 출수기는 저염 토양에서는 표준비인 N20kg/10a에 비해 N8-N16kg/10a까지는 같았으나 N24kg/10a에서는 1일 늦었고, 중염 토양에서는 N8-N16kg/10a 까지는 1일이 삘랐고 N24kg/10a에서는 같았으며, 토양 염농도간에는 저염 토양에서보다 중염 토양에서 4일 정도 늦었다. 3. 질소시비량이 많을수록 간장이 크고, 포장도복이 심했다. 4. 저염 토양에서 질소시비량이 많을수록 $\textrm{m}^2$ 당 립수는 많았으나 등숙비율이 낮아져 발 수량은 N12kg/10a이상에서는 유의차가 없었으며, N12kg/10a이하에서 현미의 완전미율이 높고 단백질 함량이 낮았다. 5.중염 토양에서는 질소시비량이 많을수록 $\textrm{m}^2$ 당 립수가 많고 등숙비율이 비슷하여 발 수량은 질소시비량이 많을수록 높았으나 N20kg/10a 이상에서는 유의차가 없었고, 현미의 완전미율과 단백질함량은 질소시비량간에 비슷하였다. 따라서 남서해안 간척지에서 쌀 수량 및 미질 등을 고려해 볼 때, 저염 토양에서는 N12kg/10a, 중염 토양에서는 N20kg/10a이 적당할 것으로 생각된다
