This study was conducted to determine the morphological responses of barley seedlings to NaCl stress and to investigate histological changes of cells with transmission electron microscope(TEM) after NaCl stress. Plant height and root length of 10-day old barley seedlings with NaCl stress were reduced and inhibition level was found to be more severe in the plant height than in the root length. The leaf length, leaf width and leaf area were shorter as well with NaCl stress than without NaCl stress. However, there was no difference in the number of roots between NaCl treatments. The weight of dry matter decreased at higher NaCl concentrations, especially at 100mM NaCl. The water content of shoots tend to decrease at higher NaCl concentrations, but there was no difference in the water content of roots, The reduced sugar content was greatly increase than starch. Cellulose content was higher in NaCl stressed-plant than control, and tended to decreased at higher NaCl concentrations. Lignin content also decreased NaCl stressed-plant but there was no tendency at NaCl stress concentrations. Electric conductivity of cell sap with seedlings was high with NaCl stressed-plant. Amount of cell sap gradually increased with time in the roots than in the shoots, The grana of chloroplasts was changed by 150mM NaCl concentration. The christe of mitochondria in root meristematic sells ruined in structure and cell wall of leaf and root was also ruined by NaCl stress.
The physiological and photochemical responses of rice seedling to NaCl stress were investigated through measuring leaf relative water content (RWC), root water uptake and chlorophyll fluorescence. When plants were exposed to increased salinity stress, the visual symptoms of injury were significant at ${\geq}$500 mM NaCl concentration for 4 and 5 day stress periods. The differences in Fv/Fm between control treatment and plants treated with 500 mM and 1,000 mM NaCl were evident after 5 day and 4 day, respectively, whereas in root water uptake its effect was observed at 500 mM and 1,000 mM NaCl at 2 day of salt-stressed periods. Leaf RWC in salt-stressed plants decreased gradually with increasing salinity in exogenous solution and duration of salt stress, and these decrease showed leaf RWC of 58-68% atduration over 2 day stress of 1,000 mM NaCl treatment and 88% at 1 day stress. NaCl stress led to a significant inhibition of the light-induced greening in etiolated rice plants, especially in 4 and 5 day salt-stressed plants, which linearly decreased with NaCl concentration ($R^2$=0.812 and 0.918, respectively). The effects of NaCl stress in rice seedlings indicate that water uptake in root is more sensitive to increasing NaCl concentration and stress duration than Fv /Fm in leaves compared with the same NaCl concentration.
This study was to investigate the effect of salt stress on physiological characteristics such as plant growth, photosynthesis, solutes related to osmoregulation of Beta vulgaris. A significant increase of dry weight was observed in 50 mM and 100 mM NaCl. The contents of Chl a, b and carotenoid were lower in NaCl treatments than the control. On 14 day after NaCl treatment, photosynthetic rate (PN), the transpiration rate (E) and stomatal conductance of CO2 (gs) were reduced by NaCl treatment. On 28 day after NaCl treatment, the significant reduction in gs and E was shown in NaCl 200 mM. However, PN and water use efficiency (WUE) in all NaCl treatments showed higher value than that of control. Total ion contents (TIC) and osmolality were higher than the control. On 14 day after treatment, the contents of proline (Pro) increased significantly in 200 mM and 300 mM NaCl concentration compared with control, whereas on 28 day in all treatments it was lower than that of the control. The contents of glycine betaine (GB) increased with the increase of NaCl concentration. The contents of Na+, Cl-, GB, osmolality and TIC increased with the increase of NaCl concentrations. These results suggested that under severe NaCl stress conditions, NaCl treatment did not induce photochemical inhibition on fluorescence in the leaves of B. vulgaris, but the reduction of chlorophyll contents was related in a decrease in leaf production. Furthermore, increased GB as well as Na+ and Cl- contents resulted in a increase of osmolality, which can help to overcome NaCl stress.
This atudy was conducted to determine the growth characteristics and photosynthesis of soybean (Glycine max L. cv. Keumjongkongl) 30 day old seedling to 100mM NaCl concentration containing 1/2 Hoagland`s nutrient solution in sand culture. The nodule formation of root is not found perfectly with NaCl stress. The leaf dry matter weight (g/plant) of stressed plant is more reduction in 77% to control than any other characters. The water content (%) is tend to increase but water potential (MPa) is tend to decrease at NaCl stress. The chlorophyll content (SPAD) is tend to increase at growing leaf age of control but decrease at NaCl stress. The photosynthesis, stomatal conductance and transpiration are tend to decrease sharply at NaCl stress.
The salt stress at seedling stage of winter barley was examined in different concentrations of NaCl containing 1/2 Hoagland solution. Fresh weight of seedling at 30 days after seeding was highest at 25mM of NaCl concentration containing 1/2 Hoagland solution but if the NaCl concentration was more than 50mM it began to decrease seriously. Water content in plant was decreased according to increase of NaCl concentration in 1/2 Hoagland solution, so physiological mechanism of NaCl in barley was different from saline plant. Stoma number per cm$^2$ of first leaf was higher than that of control in case of stressed by NaCl but in that case the leaf length was decreased so the number of stoma per first leaf was slightly decreased. Chloroplast shape was not changed by 75mM of high NaCl contained 1/2 Hoagland solution but cell division at root growing point was inhibited by 75mM of NaCl. As the result of salt stress mitochondria was ruined in structure and irregular solid was found to be transfered from the cytoplasm to the cell wall in root growing point.
This study conducted to elucidate the change of the cation content (Na$^{+}$, $K^{+}$, and $Ca^{2+}$), the L-proline content, and the sugar and starch content in the stems, roots, and leaves of three cultivars of the 30 days old seedling soybeans (Glycine max L. cv. Danwonkong, Hwang-keumkong, and Kwangankong) after 100 mM NaCl stress containing 1/2 Hoaglands nutrient solution in the sand culture. The reduction of the dry matter weight after 100 mM NaCl treatment among cultivars was higher in the order of Kwangankong, Danwonkong, and Hwang-keumkong. The highest reduction of the dry matter weight was occurred at the roots among three parts of plant. The Na$^{+}$ content increased with NaCl treatment in overall and specially greatly increased in roots and stems. The $K^{+}$ and $Ca^{2+}$ content decreased with NaCl treatment at the roots and stems. The $K^{+}$ content, however, at the leaves increased in all three cultivars with the NaCl treatment. The L-proline content with NaCl stress increased greatly. The increment of the L-proline content at the stems and roots of Hwangkeumkong was lower than that of other two cultivars, K wangankong and Danwonkong. The sugar content decreased with NaCl treatment at the stems and leaves. The starch content also decreased at the stems and leaves with NaCl treatment.aCl treatment.

The germination process is critical for plant growth and development and it is largely affected by environmental stress, especially salinity. Recently, hydrogen sulfide (H2S) is well known to act as a signaling molecule in a defense mechanism against stress conditions but poorly understood regulating seed germination. In this study, the effects of NaHS (the H2S donor) pretreatment on various biochemical (hydrogen peroxide (H2O2) content and amylase and protease activity) and physiological properties (germination rate) during seed germination of oilseed rape (Brassica napus L. cv. Mosa) were examined under salt stress. The seed germination and seedling growth of oilseed rape were inhibited by NaCl treatment but it was alleviated by NaHS pretreatment. The NaCl treatment increased H2O2 content leading to oxidative stress, but NaHS pre-treatments maintained much lower levels of H2O2 in germinating seeds under salt stress. Amylase activity, a starch degradation enzyme, significantly increased over 2-fold in control, NaHS pretreatment, and NaHS pretreatment under NaCl during seed germination compared to NaCl treatment. Protease activity was highly induced in NaHS-pretreated seeds compared to NaCl treatment, accompanied by a decrease in protein content. These results indicate that NaHS pretreatment could improve seed germination under salt stress conditions by decreasing H2O2 accumulation and activating the degradation of protein and starch to support seedling growth.
In head lettuce seedlings, NaCl stress was treated with hydroponic culture containing 0, 50, or with 100 mM NaCl in the seedling grown up to two leaf stages. Our focus was on the effect of NaCl on fresh and dry weights, antioxidant materials, and inorganic ion level. Fresh and dry weights of head lettuce seedlings increased with the increase in salinity while the optimal growth occured at 50 mM NaCl. The chlorophyll a (Chl a), total Chl and Chl a/b ratio increased 6 days after treatment with 100 mM NaCI. However, the Chl b content decreased. Total glutathione increased only in the root of head lettuce seedlings, whereas significant increase of total arcorbate content was observed in both shoot and root after the treatment with 100 mM NaCl. In addition, the NaCl treatment resulted in the decreased level of spermidine content, and a increased spermine content. Furthermore, $Na^+$ content in shoot and root increased significantly while $K^+$, $Ca^{2+}$, and $Mg^{2+}$ content decreased. The alteration of inorganic ion level after treatment with NaCl caused the reduction of $K^+/Na^+$, $Ca^{2+}/Na^+$, and $Mg^{2+}/Na^+$ ratio with the increase of NaCl concentration. Taken together, these findings indicate that the treatment of NaCl causes the induction of oxidative stress, and results in the alteration of metabolic mechanism in head lettuce seedlings.
Drought resistance and salt resistance of seedlings were compared with the polyethylene glycol(P.E.G) and NaCl solutions of the same osmotic potential($\Psi_\pi$=-10 bar). Plant height, seedling dry weight, chlorophyll content and leaf water potential decreased while the free proline content increased more in the P.E.G. than in the NaCl solution. Free amino acids increased 2.6 times in the P.E.G. solution and 3.6 times in the NaCl solution more than in the untreated(Hoagland's solution). Free proline occupied 66% and 61% of the content of total amino acids under water stress and salt stress, respectively. Besides free proline, phenylalanine in the P.E.G. solution and phenylalanine, alanine and asparagine in the NaCl solution increased distinctly. In short, it was shown that water and salt stress responses in seedling stage were relatively similar.
To examine whether salt stress would alter or not contractility of isolated rat aorta, under anesthesia with sodium pentobarbital(50 mg kg-1 i.p.), male Sprague Dawley rats(300-330 g) were subjected to 0, 50, and 150 mM of sodium chloride at 37$^{\circ}C$ for 60 min. where as the sham group was left at modified Krebs-bicarbonate solution. To measure contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And the strip was checked for expression of heat shock protein(Hsp) by Western blotting. One, three and eight hours later, we measured vascular contractility of isolated rat aorta treated with KCI, phenylephrine from organ bath study. The dose-vascular responses of potassium chloride and phenylephrine showed a little augmentation by NaCl concentration in the strips exposed to NaCl for 8 hours. And the response of relaxation induced by nitroprusside and acetylcholine was not influenced by NaCl stress in isolated aorta ring for 8 hours, respectively. Expression pattern of Hsp 70 of vascular muscle in isolated rat aorta showed a little increase in 150 mM NaCl group at 8 hours after NaCl treatment but not at 3 hours, and Hsp 60 expression of rat aorta was markedly increased in 50 mM NaCl group at 8 hours after NaCl treatment. Taken together, NaCl induced dose-and time dependent accumulation of the Hsp but not affected contraction of rat aorta. These data suggest that short term high salt stress was not sufficient to induce hypertension of rat aorta.
