CsI:X(X=Li+,K+,Rb+단결정의 섬광특성 (Scintillation Characteristics of CsI:X(X=Li+,K+,Rb+ Single Crystals)

  • CsI에 활성제로 Li, K, Rb를 첨가하여 CsI(Li), CsI(K) 및 CsI(Rb) 단결정을 Czochralski방법으로 육성하였다. $^{137}CS$(0.662 MeV)에 대한 CsI(Li:0.2 mole%) 섬광체의 에너지 분해능은 14.5%이었고 CsI(K:0.5 mole%) 섬광체는 15.9%이었으며 CsI(Rb:1.5 mole%) 섬광체는 17.0%이였다. 이들 CsI(Li), CsI(K) 및 CsI(Rb) 섬광체의 $\gamma$선 에너지에 대한 에너지 교정곡선은 선형적 이였다. 일정비율 시간분석법(CFT :constant-fraction timing method)으로 측정한 CsI(Li:0.2 mole%), CsI(K:0.5 mole%) 및 CsI(Rb:1.5 mole%) 성광체의 시간 분해능은 각각 9.0 ns, 14.7 ns 및 9.7 ns이였다. CsI(Li:0.2 mole%), CsI(K:0.5 mole%) 및 CsI(Rb:1.5 mole%) 섬광체의 형광감쇠시간은 각각 ${\tau}_1=41.2\;ns$, ${\tau}_2=483\;ns$, ${\tau}_1=47.2\;ns$, ${\tau}_2=417\;ns$${\tau}_1=41.3\;ns$${\tau}_2=553\;ns$이였다. 그리고 CsI(Li:0.2 mole%), CsI(K:0.5 mole%) 및 CsI(Rb:1.5 mole%) 단결정의 인광감쇠시간은 각각 0.51 s, 0.57 s 및 0.56 s이였다.

EM 및 아미노산액비 시용이 '설향' 딸기 모주의 생육에 미치는 영향 (Effect of EM and Amino acid Fertilizer Application on the Growth of 'Seolhyang' Strawberry Mother Plants)

  • The dry weight of mother plants' leaves had the highest increase rate in both NS (single-use) and NS+EM (mixed-use) mixed with NS 0.8 (customary use). In seafood amino acid fertilizer (SAF) application, the increase rate was highest in SAF solution at a 300-fold dilution. Mother plants' crown diameter, plant height, leaf length, leaf width, petiole length and leaf number showed the greatest growth amount when NS 0.8 (customary use) was mixed to NS (single-use) or NS+EM (mixed-use) solution. The growth was highest in SAF solution diluted 300 folds, but lowest in SAF solution diluted 100 folds. Of all inorganic nutrients, excluding sulfur, total amount of nitrogen, available phosphorus, potassium, calcium and magnesium had the highest increase rate in both NS (single-use) and NS+EM (mixed-use) with the treatment of NS 0.8 (customary use). Total nitrogen, in particular, was increased by 3.1% in NS 0.4, 6.0% in NS 0.8, and 4.5% in NS 0.8 with the application of NS+EM at a 500-fold dilution compared to NS alone. Total nitrogen amount showed the highest increase rate in SAF solution diluted 300 folds. Total nitrogen, available phosphorus, calcium, magnesium and EC in soils applied with culture solutions (NS, NS+EM) had increasing tendencies after fertilizer application. The results were comparable to those of SAF treatment. The increase rate of each inorganic nutrient composition declined in soils applied with NS+EM solution diluted 500 folds compared to NS alone.

The Two-Component Protease NS2B-NS3 of Dengue Virus Type 2: Cloning, Expression in Escherichia coli and Purification of the NS2B, NS3(pro) and NS2B-NS3 Proteins

  • Proteolytic processing of the dengue virus serotype 2 polyprotein precursor is catalyzed by a host signal peptidase and a virus encoded two-component protease consisting of the nonstructural proteins, NS2B and NS3. We expressed in Escherichia coli the NS2B, NS3(pro) and NS2B-NS3 proteins from the dengue virus type 2 strain 16681 as N-terminal fusions with a hexahistidine affinity tag under the control of the inducible trc promoter. All fusion proteins were purified to >90% purity by detergent extraction of inclusion bodies and a single step metal chelate chromatography. Proteins were refolded on-column and recovered with yields of 0.5, 6.0 and 1.0 mg/l of E. coli culture that was grown to $OD_{600}=1.0$ for NS2B, NS3(pro) and NS2B-NS3, respectively. Purified proteins gave strong signals in Western blots using $Ni^{2+}-nitrilotriacetic$ acid as a probe for the presence of the polyHis tag. During the purification process, $(His)_{6}NS2B-NS3$ was apparently not autoproteolytically cleaved at the NS2B/NS3 site.

  • 3-Aminopropyltrimethoxysilane(1NS) 또는 N-[3-(trimethoxysilyl)propyl]ethylenediamine(2NS)으로 표면 기능화된 실리카에 1-butyl-4-methylpyridinium chloride(Cl)와 tributylmethylammonium chloride(Amm), benzyldimethyltetradecylammonium chloride(Ben), 1-butyl-1-methylpyrrolidinium chloride(Pyr)와 같은 이온성 액체를 이용하여 표면처리한 후 메탈로센 촉매를 담지하여 에틸렌 중합을 실시하였다. $SiO_2/1NS/IL/(n-BuCp)_2ZrCl_2$ 촉매와 $SiO_2/2NS/IL/(n-BuCp)_2ZrCl_2$ 촉매의 Zr 함량은 아미노실란 화합물로만 표면 처리된 촉매보다 감소하였다. $SiO_2/1NS/IL/(n-BuCp)_2ZrCl_2$ 촉매의 중합 활성은 $SiO_2/1NS/(n-BuCp)_2ZrCl_2$ 촉매보다 증가하였다. 반면, $SiO_2/2NS/IL/(n-BuCp)_2ZrCl_2$은 Zr 함량의 감소로 중합 활성은 $SiO_2/2NS/(n-BuCp)_2ZrCl_2$ 촉매에 비해 감소하였다.

Molecular and Structural Characterization of the Domain 2 of Hepatitis C Virus Non-structural Protein 5A

  • Hepatitis C virus (HCV) non-structural protein 5A protein (NS5A), which consists of three functional domains, is involved in regulating viral replication, interferon resistance, and apoptosis. Recently, the three-dimensional structure of the domain 1 was determined. However, currently the molecular basis for the domains 2 and 3 of HCV NS5A is yet to be defined. Toward this end, we expressed, purified the domain 2 of the NS5A (NS5A-D2), and then performed biochemical and structural studies. The purified domain 2 was active and was able to bind NS5B and PKR, biological partners of NS5A. The results from gel filtration, CD analysis, 1D $^1H$ NMR and 2D $^1H-^{15}N$ heteronuclear single quantum correlation (HSQC) spectroscopy indicate that the domain 2 of NS5A appears to be flexible and disordered.

Systematic Identification of Hepatocellular Proteins Interacting with NS5A of the Hepatitis C Virus

  • The hepatitis C virus is associated with the development of liver cirrhosis and hepatocellular carcinomas. Among the 10 polyproteins produced by the virus, no function has been clearly assigned to the non-structural 5A (NS5A) protein. This study was designed to identify the hepatocellular proteins that interact with NS5A of the HCV. Yeast two-hybrid experiments were performed with a human liver cDNA prey-library, using five different NS5A derivatives as baits, the full-length NS5A (NS5A-F, amino acid (aa) 1~447) and its four different derivatives, denoted as NS5A-A (aa 1~150), -B (aa 1~300), -C (aa 300~447) and D (aa 150~447). NS5A-F, NS5A-B and NS5A-C gave two, two and 10 candidate clones, respectively, including an AHNAK-related protein, the secreted frizzled-related protein 4 (SFRP4), the N-myc downstream regulated gene 1 (NDRG1), the cellular retinoic acid binding protein 1 (CRABP-1), ferritin heavy chain (FTH1), translokin, tumor-associated calcium signal transducer 2 (TACSTD2), phosphatidylinositol 4-kinase (PI4K) and $centaurin{\delta}$ 2 ($CENT{\delta}2$). However, NS5A-A produced no candidates and NS5A-D was not suitable as bait due to transcriptional activity. Based on an in vitro binding assay, CRABP-1, PI4K, $CENT{\delta}2$ and two unknown fusion proteins with maltose binding protein (MBP), were confirmed to interact with the glutathione S-transferase (GST)/NS5A fusion protein. Furthermore, the interactions of CRABP-1, PI4K and $CENT{\delta}2$ were not related to the PXXP motif (class II), as judged by a domain analysis. While their biological relevance is under investigation, the results contribute to a better understanding of the possible role of NS5A in hepatocellular signaling pathways.

대전광역시와 충청남도 산림토양에서 분리한 국내 미기록 효모들의 미생물학적 특성과 생리기능성 (Microbiological Characteristics and Physiological Functionality of Unrecorded Yeasts from Mountains Soils in Daejeon Metropolitan City and Chungcheongnam-do, Korea)

  • 대전광역시 연자산과 충청남도 주요 산림 토양들로부터 분리, 동정한 야생효모들 중 국내 미기록종으로 Pseudozyma prolifica HL9-1, Trichosporon coremiiforme NS19-2, Candida cretensis SA4-1, Cryptococcus diffluens TJ4-3, Cryptococcus pinus YB17-2 등의 유포자효모와 Candida vartiovaarae DD2-5, Pichia galeiformis DM3-5, Candida pseudolambica JW2-3, Trichosporon xylopini NS5-1, Trichosporon moniliiforme NS5-7, Tetrapisispora iriomotensis NS14-2, Tetrapisispora nanseiensis SA17-1 등의 무포자효모들을 선별하여 이들의 미생물학적 특성을 조사하였다. 12균주 모두 구형~타원형이었고 출아법으로 영양증식하였으며 yeast extract peptone dextrose (YPD) 배지에서 잘 생육하였다. 특히 Candida cretensis SA4-1과 Tetrapisispara iriomotensis NS14-2은 10% NaCl을 함유한 YPD 배지에서 잘 생육하는 호염성 효모들이었다. Candida vartiovaarae DD2-5외 3균주들은 유당을 자화시켰으며 Tetrapisispora nanseiensis SA17-1은 xylose를 자화시키고 동시에 발효시켰다. Candida cretensis SA4-1과 Tetrapisispara iriomotensis NS14-2 두 호염성 효모들의 생리기능성을 조사한 결과 이들의 무세포추출물들의 항고혈압성 안지오텐신 전환효소 저해활성이 각각 71.3%와 68.4%로 높았다.

The Influence of Alpha-fetoprotein on Natural Suppressor Cell Activity and Ehrlich Carcinoma Growth

  • The influence of alpha-fetoprotein (AFP) on the bone marrow (BM) natural suppressor (NS) cells of intact Ehrlich carcinoma -bearing CBA mice was studied. Bone marrow NS cells were fractionated into three fractions by isopycnic centrifugation on percoll gradients: NS1 (${\rho}$=1.080 g/ml), NS2 (${\rho}$=1.090 g/ml) and NS3 (1.100> ${\rho}$ > 1.090 g/ml). These fractions were highly different in their sensitivity to known NS cell inductors (interleukin (IL)-2, IL-3 or histamine). None of the NS fractions isolated from the intact mice spontaneously produced antiproliferative activity, however, they showed a high level of NS (antiproliferative and natural killer cell inhibitory) activity under the influence of AFP. A single injection of AFP to intact mice led to an increase of spontaneous NS activity and the inhibition of natural killer cell activity. NS activity, especially NS2, was increased in when tumor cells were subcutaneously inoculated three days after AFP injection. In the AFP-treated mice, the tumor mass at 14 days was 60% larger than that in the untreated mice. Our data confirmed that AFP is a tumor marker that can inhibit cancer immunity and plays a role in cancer pathogenesis.

한국형 C형 간염 바이러스의 NS5 지역 cDNA 클로닝과 발현 (Cloning and Expression of NS5 Region of Korean Type Hepatitis C Virus)

  • Three cDNA fragments located within NS5 region of HCV were synthesized by RT using viral RNA extracted from blood sample of Korean patient as a template. The cDNAs were amplified by PCR, cloned into the T-vector, and the nucleotide sequences were determined. Comparative analysis of the nucleotide and amino acid sequence of NS5 cDNAs showed that it is closely related with HCV type 1b. The cloned NS5 cDNA showed 91-94% homology at the nucleotide sequence level and 96-98% homology at the amino acid sequence level with several strains of the HCV type 1b. The NS5 cDNAs were subcloned into E. coli expression vectors to construct pRSETA5-1, pTHAN5-1, pRSETC5-2, pRSETBB1, pRESTCB1 and pRSETB-H3. Expression of the NS5 proteins was achieved by inducing the promoter with isopropyl-thio-${\beta}$-D-galactoside (IPTG) and confirmed by SDS-polyacrylamide gel electrophoresis. The NS5 proteins were immunoreactive against sera from Korean hepatitis C patients in Western blot analysis. Among the recombinant NS5 proteins, pRSETAS-1 plasmid derived protein, coded from aa2022 to aa2521 of HCV polyprotein, showed the strongest immunoreactivity against sera from Korean hepatitis C patients in immunoblot analysis. These results suggest that NS5 proteins would be useful as an antigen for detection of antibody against HCV in the blood samples.

In Vitro Determination of Dengue Virus Type 2 NS2B-NS3 Protease Activity with Fluorescent Peptide Substrates

  • The NS2B-NS3(pro) polyprotein segment from the dengue virus serotype 2 strain 16681 was purified from overexpressing E. coli by metal chelate affinity chromatography and gel filtration. Enzymatic activity of the refolded NS2B-NS3(pro) protease complex was determined in vitro with dansyl-labeled peptide substrates, based upon native dengue virus type 2 cleavage sites. The 12mer substrate peptides and the cleavage products could be separated by reversed-phase HPLC, and were identified by UV and fluorescence detection. All of the peptide substrates (representing the DEN polyprotein junction sequences at the NS2A/NS2B, NS2B/NS3, NS3/NS4A and NS4B/NS5 sites) were cleaved by the recombinant protease NS2B-NS3(pro). No cleavage was observed with an enzymatically inactive S135A mutant of the NS3 protein, or with a modified substrate peptide of the NS3/NS4A polyprotein site that contained a K2093A substitution. Enzymatic activity was dependent on the salt concentration. A 50% decrease of activity was observed in the presence of 0.1M sodium chloride. Our results show that the NS3 protease activity of the refolded NS2B-NS3(pro) protein can be assayed in vitro with high specificity by using cleavage-junction derived peptide substrates.
