Method of Nitrate Nitrogen Determination for Plant, Soil and Water Analysis by E. coil Cells (E. coli 세포(細胞)를 이용한 식물(植物), 토양(土壤) 및 수질(水質)의 질산태(窒酸態) 질소(窒素) 분석방법(分析方法))

  • A microbiological nitrate determination method by E. coli is modified in Korea, using K12 wildtype, KCTC 1116, for the quantitative reduction of $NO_3{^-}$ to $NO_2{^-}$. The nitrate in plant, soil or water sample is determined spectrophotometrically after being diazotized with sulfaniamide and N-(1-naphthl)-ethlenediamine. The modified E. coli cell method and principle for nitrate determination using Korean wildtype E. coli strain is described, and cell culture and preparation of stock suspension for E. coli as well. This modified E. coli cell method can be managed simply and fast, it is suitable for the investigation of the large serials, it can be also automated and has a high degree of sensitivity up to 0.01ppm $NO_3{^-}-N$ in the sample solution. The applicability of the modified E. coli cell method has been tested for plant, soil and water analysis on a wide range of different samples. Recovery rates of added nitrate have been determined and comparisons with other standard nitrate analytical procedures have been carried out. The results with the modified E. coli cell method show high correlation ($r^2=0.98$) with those gained by the standard analytical procedures. The advantages and disadvantages of the method are also discussed to other nitrate determination methods.

Study on Flora Distributed of Nearby Island and Yubu-do in Seocheon, Chungnam (충남 서천 유부도와 인근 섬에 분포하는 식물상 연구)

  • This study is a survey of the flora distributed in Seocheon tidal flat wetland protected area of nearby island (Daejuk-do, Tokki-deung, Muk-do) and Yubo-do Seocheon-gun Chuncheongnam-do, Korea. The numbers of vascular plants were summarized as 231 taxa including 65 families, 167 genera, 204 species, 2 subspecies, 23 varieties and 2 forms. The halophytes were 33 taxa including 15 families, 29 genera, 29 species, 3 varieties and 1 form (Chenopodium album var. stenophyllum, Cnidium japonicum, Juncus haenkei and so on). The rare plants were 4 taxa including Pseudoraphis ukishiba (VU), Belamcanda chinensis (VU), Phacelurus latifolius (LC) and Polygonum bellardii (DD). The Korean endemic plants were 2 taxa including Salix pseudolasiogyne and Forsythia koreana. In the specific plants by floristic region were 24 taxa, a degree I were 19 taxa (Rhodotypos scandens, Zanthoxylum planispinum, Cynodon dactylon and so on), 2 taxa of a degree III (Elymus mollis and Asparagus oligoclonos), 3 taxa of a degree IV (Polygonum bellardii, Wisteria floribunda and Pseudoraphis ukishiba) and degree II, V were not found. The naturalized plants were 40 taxa including Phytolacca americana, Sicyos angulatus, Lactuca scariola and so on. Naturalization rate (NR) was 17.3% of all 231 taxa of vascular plants and urbanization index (UI) was 12.5% of all 321 taxa of naturalized plants. This study was performed to used as a basic data to identify the protected from plant ecosystems of type classify into analyze the vegetation characteristic of based on flora distributed in Yubu-do and nearby islands.

In vitro Conidial Germination and Mycelial Growth of Fusarium oxysporum f. sp. fragariae Coordinated by Hydrogen Peroxideand Nitric Oxide-signalling

  • Chemicals related to hydrogen peroxide ($H_2O_2$) and nitric oxide (NO) generations were exogenously applied to Fusarium oxysporum f. sp. fragariae (Fof) causing Fusarium wilt disease in strawberry plants, and regulations of in vitro conidial germination and mycelial growth of the fungus by the chemical treatments were evaluated. $H_2O_2$ drastically reduced the conidial germination of Fof in a dose-dependent manner, and treatment with 3-amino-1,2,4-triazole (3-AT) catalase inhibitor also led to dose-dependent inhibition of conidial germination but relatively moderately. Gradual decreases in mycelial growth of Fof were found by high concentrations of $H_2O_2$, whilst exogenous 3-AT slightly increased the mycelial growth. Increasing sodium nitroprusside (SNP) NO donor, $N^G$-nitro-l-arginine methyl ester (L-NAME) NO synthase (NOS)-inhibitor and tungstate nitrate reductase (NR) inhibitor led to dose-dependent reductions in conidial germination of Fof in quite different levels. SNP conversely increased the mycelial growth but increasing L-NAME moderately decreased the mycelial growth. Tungstate strongly enhanced mycelial growth. Differentially regulated in vitro mycelial growths of Fof were demonstrated by SNP, L-NAME and tungstate with or without $H_2O_2$ supplement. Superoxide anion production was also regulated during the mycelial growth of Fof by nitric oxide. These results show that $H_2O_2$ and NO-associated enzymes can be suggested as fungal growth regulators of Fof as well as eco-friendly disease-managing agents in strawberry production fields.

Conservation Methods and Vascular plants of Oriental Thuja Community in Dodong, Daegu (도동 측백나무군락지의 식물상 및 보전방안)

  • A plant diversity, which consists of indigenous plant community with Orientla thuja community (Natural monument no. 1) in Dodong, Daegu, is identified and analyzed as ecological characteristic to consider worth plants and vegetation resource of the region. The vascular plants of Thuja orientalis community were listed as 219 taxa (3.7% of all 4,881 taxa of Korean vascular plants); 67 families, 147 genera, 199 species, 16 varieties, and 4 forms. Vulnerable species (VU) and least concerned species (LC) were recorded based on IUCN standard; Koelretueria paniculata (VU), Thuja orientalis (LC), and Exochorda serratifolia (LC). Although the study site is a non-limestone area, a total of 15 taxa of calciphilous plants were identified; Cheilanthes argentea, Hypodematium glandulosopilosum, Asplenium retamuraria, Thuja orientalis, Spiraea blumei, Smilax sieboldii, etc. A total of 4 taxa endemic plants were identified; Prunus mandshurica for. barbinervis, Lespedeza maximowiczii var. tomentella, Forythia koreana (artificial origin), and Veronica pyrethrina. Among the list, 8 taxa of naturalized plants were identified; Fallopia dentatoalalta, Rumex crispus, Nasturtium officinale, Bidens frondosa, Erigeron annuus etc. Naturalization rate (NR) was 3.6%, of all 219 taxa of vascular plants and urbanization index (UI) was 2.2% of all 321 taxa of naturalized plants. Thuja orientalis occupies a lot of indigenous landscape in this study area which is the southern-limited part of a natural distribution where it can survive. The T. orientalis community, where indigenous plants have formed a characterful species composition based on habitat, has been confirmed as a worth national vegetation resource in an indigeous flora. It has been considered of plans for persistent conservation.

Combined Radiotherapy and Hyperthermia for Nonresectable Hepatocellular Carcinoma (절제 불가능한 원발성 간암의 온열 및 방사선 병용 요법)

  • Thirty patients with nonresectable hepatocellular carcinoma (HCC) due to either locally advanced lesion or association with liver cirrhosis, treated with combined radiotherapy and hyperthermia between April 1988 and July 1988, at Dept. of Radiation Oncology, Yonsei university College of medicine, were analysed. External radiotherapy of a total dose of 3060cGy/3.5 wks was given. Hyperthermia was given twice a week with a total of 6 treatment sessions using 8MHz radiofrequency capacitive type heating device, i.e., Thermotron RF-8 and Cancermia. In all cases hyperthermia was given within 30 minutes after radiotherapy for 30~60min. Temperature was measured by inserting thermocouple into the tumor mass under the ultrasonographic guidance only for those who had not bleeding tendency. As a result, partial response (PR) was achieved in 12 patients (40%), and symptomatic improvement was observed in 22 patients (78.6%) among 28 patients who had suffered from abdominal pain. The most significant factor affecting the tumor response rate was the type of tumor (single massive: 10/14, 71.4%; diffuse infiltrative: 2/10, 20%; multinodular:0/6, 0%; p<0.005). There were not any significant side effects relating to combined treatment. The overall 1 year survival rate was 34%, with 50% in the PR group and 22% in the no response group (NR), respectively. Median survival was 6.5 months and longer for those of PR than of NR (11 mos. vs 5, p<0.05). In conclusion, combined radiotherapy and hyperthermia appeared to be effective in local control and symptomatic palliation of HCC. Further study including a larger number of the patients to confirm its effect in survival and detrimental side effect should be urged.

Neoadjuvant Chemotherapy and Radiotherapy in Locally Advanced Hypopharyngeal Cancer (국소 진행된 하인두암의 선행 항암화학요법 후 방사선치료)

  • Purpose : To see the relationship between the response to chemotherapy and the final outcome of neoadiuvant chemotherapy and radiotherapy in patients with mocanry advanced hypopharyngeal cancer. Methods and Materials :A retrospective analysis was done for thirty-two patients with locally advanced hypopharyngeal cancer treated in the Seoul National University Hospital with neoadiuvant chemotherapy and radiotherapy from August 1979 to July 1997. The patients were treated with Co-60 teletherapy unit or 4MV or 6MV photon beam produced by linear accelerator. Daily fractionation was 1.75 to 2 Gy, delivered five times a week. Total dose ranged from 60.8 Gy to 73.8 Gy. Twenty-nine patients received continuous infusion of cisplatin and 5-FU. Other patients were treated with cisplatin combined with bleomycin or vinblastin. Twenty-four (75$\%$) patients received all three prescribed cycles of chemotherapy delivered three weeks apart. Six patients received two cycles, and two patients received only one cycle. Results :The overall 2-year and 5-year survival rates are 65.6$\%$ and 43.0$\%$, respectively. 5-year local control rate is 34$\%$. Organ preservation for more than five years is achieved in 12 patients (38$\%$). After neoadjuvant chemotherapy, 24 patients achieved more than partial remission (PR): the response rate was 75$\%$ (24/32). Five patients had complete remission (CR), 19 patients PR, and 8 patients no response (NR). Among the 19 patients who had PR to chemotherapy, 8 patients achieved CR after radiotherapy. Among the 8 non-responders to chemotherapy, 2 patients achieved CR, and 6 patients achieved PR after radiotherapy. There was no non-responder after radiotherapy. The overall survival rates were 60$\%$ for CR to chemotherapy group, 35.1$\%$ for PR to chemotherapy group, and 50$\%$ for NR to chemotherapy group, respectively (p=0.93). There were significant difference in five-year overall survival rates between the patients with CR and PR after neoadjuvant chemotherapy and radiotherapy (73.3$\%$ vs. 14.7$\%$, p<0.01). The prognostic factor affecting overall survival was the response to overall treatment (CR vs. PR, p<0.01). Conclusion :In this study, there were only five patients who achieved CR after neoadiuvant chemotherapy. Therefore the difference of overall survival rates between CR and PR to chemotherapy group was not statistically significant. Only the response to chemo-radiotherapy was the most important prognostic factor. There needs to be more effort to improve CR rate of neoadjuvant chemotherapy and consideration for future use of concurrent chemoradiotherapy.

Transcriptome Analysis of Antrodia cinnamomea Mycelia from Different Wood Substrates

  • Antrodia cinnamomea, an edible and medicinal fungus with significant economic value and application prospects, is rich in terpenoids, benzenoids, lignans, polysaccharides, and benzoquinone, succinic and maleic derivatives. In this study, the transcriptome of A. cinnamomea cultured on the wood substrates of Cinnamomum glanduliferum (YZM), C. camphora (XZM), and C. kanehirae (NZM) was sequenced using the high-throughput sequencing technology Illumina HiSeq 2000, and the data were assembled by de novo strategy to obtain 78,729 Unigenes with an N50 of 4,463 bp. Compared with public databases, about 11,435, 6,947, and 5,994 Unigenes were annotated to the Non-Redundant (NR), Gene Ontology (GO), and Kyoto Encyclopedia of Genes and Genome (KEGG), respectively. The comprehensive analysis of the mycelium terpene biosynthesis-related genes in A. cinnamomea revealed that the expression of acetyl-CoA acetyltransferase (AACT), acyl-CoA dehydrogenase (MCAD), 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA), mevalonate pyrophosphate decarboxylase (MVD), and isopentenyl diphosphate isomerase (IDI) was significantly higher on NZM compared to the other two wood substrates. Similarly, the expression of geranylgeranyltransferase (GGT) was significantly higher on YZM compared to NZM and XZM, and the expression of farnesyl transferase (FTase) was significantly higher on XZM. Furthermore, the expressions of 2,3-oxidized squalene cyclase (OCS), squalene synthase (SQS), and squalene epoxidase (SE) were significantly higher on NZM. Overall, this study provides a potential approach to explore the molecular regulation mechanism of terpenoid biosynthesis in A. cinnamomea.

Analysis of Pain Records for Cancer Patients Complaining of Moderate or Severe Pain (중등도 이상의 통증을 호소하는 암환자의 통증간호기록 분석)

  • Purpose: This is a retrospective study that investigated cancer patients' complaints of moderate or severe pain to analyze pain characteristics, pain relief interventions and their effects. Methods: The participants of this study were 363 patients who were hospitalized in the cancer ward for three to 30 days and scored 4 points or higher on the pain severity assessment. Results: The most frequent region of pain was the abdomen. The most frequent factor that exacerbated pain was movement. The most frequent pain alleviating factor was administration of analgesics. The most frequent pain type was breakthrough pain, and the most frequent non-pharmaceutical intervention for pain control was heat therapy. Among all, analgesics were routinely prescribed for 52.2% of the participants. Morphine sulfate was the most frequently used analgesic while Gabapentin was the most frequently used non-narcotic analgesic. At the time of discharge, 82.5% of the participants marked their pain intensity as 3 points or lower. Conclusion: For cancer patients complaining of moderate or severe pain, it is important to actively control pain from the beginning of admission. Thus, it is necessary to educate not only cancer patients using narcotic analgesic for pain control and their families but nurses about the effects and side-effects of drugs. Moreover, patients and their families need to learn how to assess and record pain at home to collect data that can be referred for future treatment.

A phylogenetic analysis of Korean Artemisia L. based on ITS sequences (ITS 염기서열에 의한 한국산 쑥속(Artemisia L.)의 계통분류학적 연구)

  • Taxa of Artemisia collected in Korea were constructed by molecular phylogenetic analysis based on the internal transcribed spacer(ITS) regions of nrDNA. The length of the ITS sequences aligned using the clustal X program was 636~643 bp, and the lengths of the ITS1 and ITS2 regions were 251~255 bp and 217~222 bp, respectively. The total number of variable sites was 95 for the entire sequence, and a parsimony- informative site represented an efficacious site in ITS1 rather than in ITS2. The maximum parsimony tree as calculated by the MEGA 4 program was clustered into five clades. The taxa(A. capillaris, A. japonica var. japonica, A. japonica var. hallaisanensis, A. japonica subsp. littoricora) degenerated ovary of clade 1 was supported as the subgenus Dracunculus by Ling's classification system. The results show that A. nakaii and A. fukudo were quite similar genetically(Boostrap 99%) and that the scientific name of Korean A. dubia should be reconsidered. A. sp. distributed in Ganghwa province was grouped with A. argyi(Boostrap 89%). These results suggest that the molecular techniques used in this study could be useful for the phylogenetic analysis of Korean Artemisia herbs having variations in their morphological characteristics.

Effects of Hyungbangjihwangtang Water Extract on Cultured Primary hippocampal Cell Damaged by Glucose Oxidase (형방지황탕(荊防地黃湯) 전탕액(煎湯液)이 산소자유기로 손상된 배양(培養) 해마신경세포(海馬神經細胞)에 미치는 영향)

  • The purpose of this study is to examine the toxic effects caused by glucose oxidase (GO) and the effects of Hyungbangjihwangtang (HJT) water extracts on the treatment of the toxic effects. For this purpose, experiments with the cultured hippocampal cells from new born mice were done. The results of these experiments were as follows. 1. GO decreased the survival rate of the cultured cells on MTT assay and NR assay. 2. HJT has the efficacy of decreasing the lipid peroxidation. 3. HJT has the efficacy of increasing the amount of neurofilaments. 4. HJT has the efficacy of decreasing the activation of protein kinase C(PKC). From the above results, it is concluded that HJT has marked efficacy as a treatment for the damages caused by the GO-mediated oxidative stress. Further clinical study of this pharmacological effects of H]T should be completed.
