Induction of NADPH oxidases and antioxidant proteins by Porphyromonas gingivalis in KB cells (Porphyromonas gingivalis 감염된 구강상피세포에서 NADPH oxidase와 항산화단백의 발현)

  • Porphyromonas gingivalis는 치주질환을 야기하는 독성세균으로서, 구강상피세포에 p. gingivalis가 감염되었을 때, 세포형태에 변화를 초래함으로 인해 방어기작이 작동하게 된다. 치주질환과 관련되어 생성된 활성 산소종의 소거에 관여하는 항산화성분은 p. gingivalis 이 감염된 구강상피세포에서 그 분포와 발현수준이 달라지리라 예상된다. 따라서 이번 연구에서는 구강상피세포(KB 세포)에 p. gingivalis가 감염되었을 때 야기되는 활성산소종과 이를 소거하는 역할을 하는 항산화단백들의 역할들을 규명하고자 하였다. 활성산소종 형성을 조절하는 NADPH oxidase 중 NOX4와 Rac1 전사체는 구강상피세포에서 p. gingivalis세균에 의해 증가하였으며 $gp91^{phox}$, Rac2, $p47^{phox}$$p67^{phox}$는 세균에 의한 변화가 관찰되지 않았다. 반면에 $p40^{phox}$ 전사체는 감소하는 경향을 보였다. NOX1 전사체는 p. gingivalis 처리 30분 후 감소하였다가 60분 후에는 다시 증가하는 양상을 보였다. 같은 시간에 NOX 활성화 단백인 NOXA1은 감소하고, NOX 구성단백질인 NOXO1은 증가하는 경향을 보였다. p. gingivalis가 감염된 구강상피세포를 방어하는 항산화단백 발현수준을 평가한 결과, SOD1, 2, 3 모두 p. gingivalis 처리시간에 따라 증가하는 양상을 보였다. GPx 발현 양상도 SOD와 유사하게 나타났다. $H_2O_2$를 소거하는 Prx는 감염된 KB 세포에서 Prx4와 Prx5가 4-6배 증가하는 것을 알 수 있었다. 반면 endocytosis 과정 중 $H_2O_2$ 생산은 변화되지 않았다. 이번 연구의 결과, p. gingivalis의 감염은 KB 세포의 NOX4와 Rac1의 NADPH oxidase 발현을 증가시켰으며, NOX1은 NOXA1과 NOXO1의 조절에 의해 영향을 받음을 알 수 있었다. 또한 항산화기작으로는 SOD, GPx, Prx가 증가하였는데, 이것은 Prx4와 Prx5가 중요한 역할을 할 것을 시사하였다.

  • A mobile emission laboratory (MEL) was designed to measure the amount of traffic pollutants with high temporal and spatial resolution under real conditions. Equipment for gas-phase measurements of quantity of CO, NOx, $CO_2$, and THC and for the measurement of the number density and size distribution of fine and ultra-fine particles by a FMPS and a CPC were placed in a mini-van. The exhaust of different type of vehicles can be sampled by MEL. This paper describes the construction and technical details of the MEL and presents data from the experiment in which a car chases city buses fuelled by diesel, CNG, and LPG. The diameters of most particles in the exhaust of the diesel city bus were less than 300 nm and most of the particles had a diameter of 30-60 nm. However, most particles in the exhaust of the CNG and LPG city buses were nanoparticles (diameter: less than 50 nm).

  • 우리나라 대기질은 자동차의 급격한 증가와 산업활동이 활달해짐에 따라 VOC, NOx 의 배출량이 급격히 늘어나고 있으며 이에따라 광화학 오존등 2차오염물질에 대한 대기오염문제가 심각해지고 있다. 오존에 의한 대기오염문제를 해결하기 위하여 환경부에서는 2002년부터 서울의 정동, 불광동 둥 수도권 8개 지점에서 광화학평가측정망을 운영하고 있으며, 본 연구에서는 이들 광화학평가측정망의 정도관리현황을 검토하고 측정지점별 오존전구물질(VOCs) 농도분포 특성을 분석하였다. (중략)

  • 표면연소버너의 모재 중 최근 많은 관심을 끌고 있는 금속섬유를 이용한 LPG 및 COG의 연소특성을 파악하였다. 연소모드, 매트 표면온도분포, 공해물질 배출량, 버너 전후단 압력 손실을 측정한 결과, COG 연소시가 더 넓은 범위의 적열영역을 얻을 수 있었고, 매트의 평균 표면온도도 높았다. NOx 발생량은 100ppm 이하였고, 연소부하 50kcal/$ extrm{cm}^2$hr에서의 매트 전후단의 압력손실은 30mmH2O로 압력손실은 거의 없었다. 적열범위나 표면온도분포 등이 가스별로 약간의 차이를 보였으나, 매트의 종류에 크게 상관없이 안정연소를 달성할 수 있어 가스 특성이 상이한 각종 가스의 연소에 표면연소기술의 적용이 가능할 것이다.

  • To measure the traffic pollutants with high temporal and spatial resolution under real conditions, a mobile emission laboratory (MEL) was designed. The equipment of the mini-van provides gas phase measurements of CO, NOx, CO2 and THC (Total hydrocarbon), and number density & size distribution measurements of fine and ultra-fine particles by a fast mobility particle sizer (FMPS) and a condensation particle counter (CPC). The inlet sampling port above the bumper enables the chasing of different type of vehicles. This paper introduces the technical details of the MEL and presents data from the experiment in which a MEL chases a city bus fuelled by diesel, DME and Bio-diesel. The dilution ratio was calculated by the ratio of ambient NOx and tail-pipe NOx. Most particles from the bus fuelled by diesel were counted under 300 nm and the peak concentration of the particles was located between 30 and 60 nm. However, most particles in the exhaust of the bus fuelled by DME were nano-particles (diameter: less than 50 nm). The bus fuelled by Bio-diesel shows less particle emissions compare to diesel bus due to the presence of the oxygen in the fuel.

  • In this work, a numerical investigation is performed for the combustion chamber of domestic gas boiler with 1-stage and 2-stage heat exchangers. The fluid flow and heat transfer performance is simulated with a structure change of heat exchanger. The numerical solution shows that the heat transfer of the 2-stage heat exchanger is about 24% higher than that of the 1-stage heat exchanger, while the pressure loss of the 2-stage heat exchanger increases. The temperature of combustion chamber with 2-stage heat exchanger is lower than that of 1-stage. This effect reduces thermal NOx with decrease of high temperature staying time of the combustion gas.

  • A urea-SCR system suffers from some issues associated with the ammonia slip phenomenon, which mainly occurs because of the shortage of evaporation and thermolysis time, and this makes it difficult to achieve an uniform distribution of injected urea. A numerical study was therefore performed by changing such various parameters as installed injector angle and application and angle of mixer to enhance evaporation and the mixing of urea water solution with exhaust gases. As a result, various parameters were found to affect the evaporation and mixing characteristics between exhaust gas and urea water solution, and their optimization is required. Finally, useful guidelines were suggested to achieve the optimum design of a urea-SCR injection system for improving the DeNOx performance and reducing ammonia slip.

The dilution ratio was calculated by the ratio of ambient NOx and tail-pipe NOx. Most particles from the diesel bus were counted under 300 nm and the peak concentration of the particles was located between 30 and 60 nm. The total PM number emission from diesel bus equipped with DPF was 10 orders of magnitude lower compared to those emitted from base diesel bus. And the total PM number emission from diesel bus equipped with SCR was comparable to the particle emission from base diesel bus.

The exhausts of different types of vehicles can be sampled by an MEL. This paper describes the technical details of the MEL and presents data from the experiment in which a car chases passenger vehicles fuelled by diesel and gasoline. The particle number concentration in the exhaust of the diesel vehicle was higher than that of the gasoline vehicle. However, the diesel vehicle with a DPF emitted fewer particles than the vehicle equipped with a gasoline direct injection engine, with particle diameters over 50 nm.
