Nitric oxide is an important factor to regulate the biochemical reactions in the body such as expansion of blood vessel, neural conduction and antimicrobial activity. There are two forms of nitric oxide synthase and iNOS has attracted most attention because it is involved in the development of diabetes and cardiac disease condition. There are several regulatory sequences in the promoter region of iNOS gene. One of them is (CCTTT)n. It has been reported that the number of tandem repeat of (CCTTT)n varies from population to population. So, we analyzed (CCTTT)n polymorphism in Korean genome for the purpose of comparison. According to our present study Koreans are different from other Asians reported previously because $(CCTTT)_{10}$ is the highest incidence as opposed to $(CCTTT)_{12}$ for other countries. This study should facilitate the understanding of the expression of iNOS gene in different population.
We have studied 73 pre-service science teachers' conceptions about the nature of science (NOS) using 120 true-false test items based on AAAS Benchmarks statements. We have found that participants have inadequate understanding of the NOS, especially in understanding of five categories of conceptions; change and continuity in science, bias in scientific investigations, hypothesis in scientific investigations, things common in science, and science ethics. The result also indicates that there is a difference between primary pre-service teachers and secondary pre-service teachers. From the analysis of consistency, we also found that this inadequate understanding comes from confusion rather than misconception. All the results support that there is a need of intense pre-service teacher education concerning thenature of science.
The aim of this study was to investigate the effect of TENS on muscle pain induced by carrageenan. Muscle pain was induced in male Sprague-Dowley rats by intra-muscular injection of gastrocnemius with $3\%$ carrageenan. nNOS was measured to assess the effect of TENS on muscle pain induced by Carrageenan. The lumbar enlargement of the spinal cord was removed in different groups of animals 24 h after induction of muscle pain. The level of nNOS mRNA was measured in the lumbar section of the spinal cord using RT-PCT. The expression of nNOS was analyzed in the dorsal horn of the lumbar spinal cord by immunohistochemistry. TENS decreased nNOS immunoreactivity in the dorsal horn of the lumbar spinal cord when compared with controls(p<.05). In RT-PCR, TENS decreased nNOS mRNA level of lumbar spinal cord when compared with controls(p<.05). These results suggested that application of TENS attributed to decrease muscle pain.
Lead has long been considered as a toxic environmental pollutant that severely damages the central nervous system. In various neurogenerative diseases, actrocytes become activated by proinflammatory cytokines. In the present study, we investigated whether lead (Pb$^{2+}$) affects inducible nitric oxide synthase (iNOS) expression in activated glial cells. Rat primary glial cells were stimulated with lipopolysaccharide (LPS, 1 ${\mu}$g/ml) plus IFN$_{\gamma}$(100 U/ml). Pre-treatment of Pb$^{2+}$ increased nitric oxide (NO) production in LPS/IFN$_{\gamma}$-stimulated glial cells. Lead itself, however, suppressed the basal production of NO in control glial cells. Addition of the iNOS inhibitors L-NAME (1 mM) and L-NNA (800 ${\mu}$M) prevented the Pb$^{2+}$-induced increase in NO production. Western blot analysis showed that pre-treatment of Pb$^{2+}$ augmented LPS/IFN$_{\gamma}$-induced increase in iNOS immunoreactivity, which was well correlated with the increased NO production. In addition, pre-treatment of Pb$^{2+}$ synergistically increased the iNOS mRNA expression induced by LPS and IFN${\gamma}$. The present results indicate that lead intoxication adversely affect brain function by potentiating iNOS expression and NO production in activated glial cells observed in various neurodegenerative diseases.
본 연구는 새로운 기능성 화장품 소재를 개발하기 위해 천연물 재료인 말오줌나무 추출물 활용 가능성 연구하였다. 이 목적을 이루기 위하여, 말오줌나무의 세포독성효과를 MTT assay를 통해 확인한 결과, $500{\mu}g/ml$ 농도에서 100% 이상의 세포 생존율을 나타내었다. 항염증 활성을 효과적으로 확인하기 위하여, LPS로 유도된 대식세포 내 NO 생산을 억제하는 효과를 griess의 방법으로 조사하였다. 그 결과 NO의 생성이 말오줌나무 추출물의 농도 의존적으로 저해되었음을 확인하였다. 말오줌나무 추출물을 LPS로 유도된 RAW 264.7 대식세포에서 전염증성 인자(iNOS, COX-2)들을 생성하여 측정하였다. 그 후, iNOS와 COX-2의 단백질 발현 억제 효과를 측정하기 위해 50, 100, $500{\mu}g/ml$ 농도에서 western blot을 수행하였고, ${\beta}$-actin를 양성대조군으로 사용하였다. iNOS와 COX-2의 mRNA 발현 억제 효과를 측정하기 위해 50, 100, $500{\mu}g/ml$ 농도에서 RT-PCR을 수행하였고, 양성 대조군으로 GAPDH를 사용하였다. 결과적으로, western blot으로 iNOS와 COX-2의 단백질 발현 억제 효과를 측정한결과 $500{\mu}g/ml$ 농도에서 각각 31.2%, 54.7%의 감소 효과를 보였으며, iNOS, COX-2의 mRNA 발현 억제 효과를 RT-PCR로 측정한 결과 $500{\mu}g/ml$ 농도에서 각각 72.2%, 89% 정도로 감소하였다. 이러한 결과들을 통해 말오줌나무 추출물은 항염증 효과를 가진 천연물 소재로 활용 가능할 것으로 생각된다.
본 연구는 염증 형성과정에 있어서, 해조류로부터 항염증 효과를 나타내는 물질을 분리하여 그 유도체인 HS-1580 series (HS-1580, HS-1581, HS-1582)를 합성하였다. Nitiric oxide (NO) 생성에 있어 Raw 264.7 cells에서 lipopolysaccharide(LPS) 단독으로 처리하였을 때는 대조군에서보다 4배 이상 NO 생성이 증가하였지만, HS-1580 series를 처리하고 LPS를 처리한 군에서는 농도 의존적으로 NO 생성이 억제되었다. HS-1580 series가 NO 생성 자체를 억제함으로서 NO 함량이 감소되었는지, inducible NOS (iNOS) 단백질 발현을 억제에 기인한 것인지 알아보기 위해서 Western blot으로 조사하였다. iNOS protein 발현이 HS-1580 series에 의해서 억제되었고 HS-1580 series가 cyclooxygenase-2 (COX-2), tumor necrosis factor-a $TNF-{\alpha}$ 와 $interluekin-1{\beta}\;(IL-1{\beta})$ 생성을 농도 의존적으로 억제시켰다. 이상의 결과로 HS-1580 series가 iNOS 단백질 발현 억제에 기인한 NO 생성억제, COX-2 발현 억제 및 pro-inflammatory cytokines인 $TNF-{\alpha}$ 와 $IL-1{\beta}$ 생성을 억제하는 항염증 효과를 가짐을 알 수 있다.
Objectives : In recent years, many studies have been widely researching anti-inflammation effect of various medicinal plants. $Cinnamomi$$Cortex$ was not enough in researching of the anti-inflammation. Moreover, there is no comparative study about extraction methods. Therefore, we investigated the inhibitory effects of $Cinnamomi$$Cortex$ pharmacopuncture by EtOH and Hot water extraction on Nitric oxide(NO), Prostaglandin E2(PGE2) production, Cyclooxygenase(COX)-2, inducible NOS(iNOS) expression and extracellular signal regulate kinase(ERK)1/2 phosphorylation in lipopolysaccharide(LPS) induced RAW 264.7 macrophage cell. Methods : $Cinnamomi$$Cortex$ was extracted by EtOH and Hot water. RAW 264.7 macrophage cell viability was measured by MTT assay. Effect of $Cinnamomi$$Cortex$ pharmacopuncture on NO and PGE2 production in LPS induced macrophages was accessed by Griess assay and enzyme-linked immunospecific assay(ELISA), respectively. Inhibition effect on COX-2, iNOS expression and ERK1/2 phosphorylation was examined by Immunoblotting assay. Results : 1. Cytotoxic effect of $Cinnamomi$$Cortex$ pharmacopuncture by Hot water extraction in RAW 264.7 macrophages was not appeared, except $3125{\mu}g/m{\ell}$. And cytotoxic effect was not appeared in EtOH extraction method. 2. $Cinnamomi$$Cortex$ pharmacopuncture by EtOH and Hot water extraction inhibited NO production in LPS induced macrophages significantly. 3. $Cinnamomi$$Cortex$ pharmacopuncture by EtOH and Hot water extraction inhibited PGE2 production in LPS induced macrophages significantly. 4. $Cinnamomi$$Cortex$ pharmacopuncture by EtOH and Hot water extraction inhibited COX-2, iNOS expression in LPS induced macrophages. Especially, it has been confirmed that COX-2, iNOS expression were effectively inhibited in Hot water extraction. 5. $Cinnamomi$$Cortex$ pharmacopuncture by EtOH and Hot water extraction inhibited ERK1/2 phosphorylation in LPS induced macrophages. Especially, it has been confirmed that ERK1/2 phosphorylation was effectively inhibited in Hot water extraction. Conclusions : According to the results, $Cinnamomi$$Cortex$ pharmacopuncture suppresses NO, PGE2 production, COX-2, iNOS expression and ERK1/2 phosphorylation in LPS induced macrophages. It has a potential for treating various inflammatory diseases, and Hot water extraction method could be used more extensively than EtOH extraction method.
본 연구는 고마리 추출물이 가지는 항염증 활성을 알아보기 위하여 쥐의 대식세포(RAW264.7 cell)에 Lipopolysaccharide (LPS)를 처리하여 염증반응을 유도하고 이때 발생되는 Nitric oxide (NO)의 생성 억제를 확인하였다. 또한 염증에서 중요하게 알려져 있는 Inducible nitric oxide synthase (iNOS), Cyclooxygenase-2 (COX-2), Nuclear factor-kappa B (NF-${\kappa}B$) 단백질들의 발현을 비교하였고, 추가적으로 NF-${\kappa}B$ 단백질의 핵 내부로의 이전 및 활성을 확인하였다. 메탄올 추출물은 NO 생성 및 iNOS, COX-2, NF-${\kappa}B$ 단백질의 발현을 억제하고, 세포를 보호하는 효과를 가지는 Heme oxygenase-1 (HO-1) 단백질의 발현을 증가시켰다. 위 결과를 바탕으로 하여 n-butanol, hexane, ethyl acetate 용매를 이용한 추가적인 분획을 실시하였다. 이들 분획 중 고마리의 ethyl acetate 추출물은 Prostaglandin $E_2$ ($PGE_2$), NO 생성을 억제 하였으며, iNOS, COX-2 단백질들의 발현을 감소, NF-${\kappa}B$의 핵 내부로의 이동을 억제하는 효과가 높다는 것을 확인하였다. 이러한 연구결과는 고마리 식물이 좋은 항염증 활성을 가지고 있음을 나타내며, 지속적인 분획으로 고마리 식물이 가지는 항염증 활성 물질을 선별하여 그 작용기작을 규명하는 연구가 필요하다.
Objectives : Daeganghwal-Tang(DGHT) is one of the prescriptions used for the treatment of rheumatoid arthritis(RA) in oriental medicine. The present study aimed to examine the analgesic effect of DGHT on a rat model of CFA-induced arthritis, and the relations between DGHT-induced analgesia and endogenous nitric oxide(NO) and inducible NO synthase(iNOS)/neuronal NOS. Methods : CFA-induced arthritis model used to test the effect of DGHT was chronic pain model. After the induction of arthritis, rats subsequently showed a reduced stepping force of the affected limb for at least the next 18 days. the reduced stepping force of the limb was presumably due to a painful knee. DGHT dissolved in water was orally administrated. After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 4 hours. Results : DGHT produced significant improvement of stepping force of the hindlimb affected by the arthritis lasting at least 2 hours. DGHT produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner. Both NO production and nNOS/iNOS protein expression which is increased by arthritis were suppressed by DGHT administration. Conclusions : The data suggest 1) that DGHT produces a potent analgesic effect on the chronic knee arthritis pain model in the rat and 2) that DGHT-induced analgesia modulate endogenous NO through the suppression of nNOS/iNOS protein expression.
Objectives : Inflammation is one of the self-protective abilities against tissue injury, and it has clinical symptoms like redness, heat, swelling, pain, and loss of function. The purpose of this study is to examine inhibitory effects of Naetakchunkeum-san (NTCKS) on nitric oxide (NO), Prostaglandin E2 (PGE2), inducible NOS (iNOS), cyclooxygenase-2 (COX-2), and phosphorylation of extracellular signal-regulated kinase 1/2 (ERK1/2), which play a major role in inflammatory response. Methods : The experiment was performed using Raw 264.7 cells pretreated with NTCKS extracts. Cell viability was determined by MTT assay. To evaluate anti-inflammatory effects of NTCKS, we examined NO and $PGE_2$ production in LPS-induced macrophages. We also investigated effects of NTCKS on iNOS, Cox-2, and ERK1/2 expression using western blot. Results : In MTT assay, no cytotoxicity of NTCKS (50, 100, 150, $200{\mu}g/ml$) on RAW 264.7 cell was found. LPS-induced NO production was decreased after treatment with NTCKS (150, $200{\mu}g/ml$)(p<0.05). $PGE_2$ was decreased after treatment with NTCKS (150, $200{\mu}g/ml$)(p<0.05). NTCKS inhibited LPS-induced expressions of iNOS and COX-2 in a dose-dependent manner. Increased phosphorylation of ERK1/2 by LPS was decreased by NTCKS in a dose-dependent manner. Conclusions : According to above experiments, NTCKS may be applied to inflammatory diseases such as atopic dermatitis, rheumatoid arthritis, and inflammatory bowel disease.
