Comparative Karyological Analysis of the Korean Treefrogs, Hyla japonica and Hyla suweonensis (Anura, Hylidae) (한국산 청개구리 두 종(Hyla japonica와 H.suweonensis)의 핵형 비교분석)

  • The comparative karyological analysis of the Korean treefrog, Hyla japonica and Hyla suweonensis performed by conventional giemsa-staining and NOR-staining method. The karyotypes of both species were the same (2n=24). However, according to the Karyological analysis, the 6th chromosomes of the two species distinctly different. Eventhough the 6th chromosomes of the two species shown the same % length, construction of the 6th chromosome of H.japonica was subtelocentric chromosome while H. suweonensis was submetacentric chromosome. This phenomena could explain that the morphological differences in the 6th chromosomes might be caused by pericentric inversion. The two species have 1 pair of NOR site in the 6th chromosome.

  • The random telegraph signal (RTS) for the NOR flash cell scaling is investigated. An innovative method to suppress the RTS, based on the device engineering, is proposed. By optimizing the channel doping profile and using the high-k tunnel dielectric, it is confirmed from three-dimensional (3-D) simulation, that the $V_{th}$ amplitude, dueto RTS, is significantly suppressed, from approximately 0.5 to 0.07 V in the middle of the channel at 45 nm NOR Flash technology. From this result, it is expected that the proposed method to suppress the RTS amplitude is essential for further cell size scaling in Flash memory.

  • Microbial transformation of a potent progestin, norethisterone (17��-hydroxy-19-nor-17α-pregn-4-en-20-yn-3-one) (1) was carried out by using two filamentous fungi Aspergillus niger and Penicillium citrinum. Biotransformation of 1 with A. niger yielded a hydroxylated transformed product 10��,17��-diydroxy-19-nor-17α-pregn-4-en-20-yn-3-one (2) whereas 11��,17��-diydroxy-19-nor-17α-pregn-4-en-20-yn-3-one (3) was obtained through microbial transformation of 1 by P. citrinum. It is the first report of their production from 1 by using A. niger and P. citrinum with complete 1H- and 13C-NMR assignment. The structures of both metabolites were characterized by various spectroscopic techniques and reported data.

  • A phytochemical investigation of the fruits extract of Melia toosendan afforded the isolation of two new nor-neolignans, meliasendanins E (1) and F (2), as well as twelve known compounds (3 - 14) using various separation technique such as Diaion HP20, silica, RP-18 gel column chromatography and semi-preparative HPLC. Their chemical structures were elucidated by extensive NMR spectroscopic data including 2D-NMR, and HR-ESI-MS as well as ECD data. Among the twelve known compounds, the absolute structures of 3 - 6 were determined first, and given the trivial names as meliasendanins G-J (3 - 6). Based on the evaluation of anti-inflammatory activity, compounds 7 - 8 exhibited inhibitory effects on LPS-induced nitric oxide production in RAW 264.7 macrophages with IC50 values of 34.6 and 39.5 µM, respectively.

  • For metabolic study of pinusolide, a naturally occurring platelet activating factor antagonist, a synthetic method for preparation of radiolabeled pinusolide was studied. Pinusolide was first oxidized with $OsO_4/NaIO_4 $to 17-nor-8-oxo compound (2), which was subsequently converted to pinusolide by treatment with the Lombardo reagent $(Zn/CH_2Br_2/TiCI_4)$.   • Electrochemical behavior of fluoroquinolone antibacterial agents on carbon paste electrode (CPE) were investigated by cyclic voltammetry and square wave adsorptive stripping voltammetry. The fluoroquinolone antibacterial agents tested in this study were Enrofloxacin (ENR), Norfloxacin (NOR), Ciprofloxacin (CIP), Ofloxacin (OFL) and Levofloxacin (LEV). In acetate buffer at pH 4.5, the oxidation peak potentials of the fluoroquinolone antibacterial agents of ENR, NOR, CIP, OFL, and LEV were 0.952 V, 1.052 V, 1.055 V, 0.983 V, and 0.990 V (vs. Ag/AgCl), respectively. And their oxidation peak currents from square wave adsorptive stripping voltammograms are proportional to the concentration of each antibacterial agent over the range from $0.2\;{\mu}M$ to $1\;{\mu}M$.

Effects of GamiChungYi-tang on the Caerulein-induced Acute Pancreatitis in Rats (Caerulein으로 유발된 흰쥐의 급성 췌장염에 대한 가미청이탕(加味淸胰湯)의 효과)

  • In this study, we aimed to investigate the effect of GamiChungYi-tang(GCY-t) on caerulein-induced acute pancreatitis (AP). It is performed by detecting oxidative stress markers and observing histopathological examination. Thirty adult male rats(Sprague-Dawley) were divided into six groups as follows: normal (NOR,n=5), caerulein-induced (CON,n=5), caerulein+Cefotaxime Sodium(CT,n=5), caerulein+GCY-t (130 mg/kg, CHA,n=5), caerulein+GCY-t (260 mg/kg, CHB,n=5) and caerulein+GCY-t (520 mg/kg, CHC,n=5) groups. Pancreatic tissues of rats from all groups were removed for apoptosis and light, and electron microscopic examination. Blood of rats from all groups were collected for oxidative stress markers inspection and pathological examination. Pancreatic oxidative stress markers were evaluated by the measurements of leukocyte, serum amylase and platelet activating factor (PAF), Interleukin-6 (IL-6) levels were determined spectrophotometrically. CON group has a significant increase (p<0.05) in amylase compared with NOR, but CT and CHA, CHB, CHC groups reduced the levels of these enzyme. The levels of Platelet activating factor (PAF) were increased in CON compared with NOR, but decreased in CT and CHA, CHB, CHC groups compared with CON. Interleukin-6 (IL-6) levels were increased significantly in CON compared with NOR, but reduced in CT and CHA, CHB, CHC groups. In the observations of Optical microscopy and electron microscopy, The experimental groups showed the significant decreases in pancreatic tissue inflammation, edema, vacuolization, necrosis compared to the control group. After all, GCY-t is potentially capable of limiting pancreatic damage produced during AP by restoring the fine structure of acinar cells and tissue.

  • For the evaluation of our laboratory's 11-nor-${\Delta}^9$-tetrahydrocannabinol-9-carboxylic acid (THCCOOH) urinalysis test, THCCOOH urinalysis test was carried out with Certified Reference Material (CRM). The used CRM was THCCOOH in freeze-dried urine produced by NIST as Standard Reference Material 1507b. Comparing the estimated value of our laboratory with CRM, the results was coincided in the confidence level of approximately 95%.
