Comparative toxicity of some pesticides on reproduction of Korean native freshwater Cladocerans, Moina macrocopa and Daphnia sp. (한국산 물벼룩 Moina macrocopa와 Daphnia sp.에 대한 수종 농약의 번식독성 비교)

  • The reproduction toxicity of several pesticides on two Korean water flea was investigated to develop a new standard species used for ecological risk assessment of pesticide. Moina macrocopa and Daphnia sp. were exposed to 4 different types of pesticides over 10 and 21 days, respectively. No-ohserved effect concentration (NOEC) for synthetic pyrethroid, fenpropathrin on Moina macrocopa and Daphnia sp. were 0.17 and $0.06\;{\mu}g\;L^{-1}$, respectively. Diazinon, carbofuran and myclobutanil were in the order of their reproduction toxicity to cladocerans tested. There were large differences between Moina macrocopa and Daphnia sp. in their susceptibility to fenpropathrin, diazinon and carbofuran except myclobutanil. Daphnia sp. is more sensitive than M. macrocopa to pesticides tested. Therefore Daphnia sp. may be a good surrogate species to assess the reproduction effect of pesticides on aquatic invertebrates. M. macrocopa also be a good surrogate species because it is one of the most abundant cladocera in agricultural environment, especially rice paddy in Korea. In addition to it's ecological importance of wide spread distribution, it has also economical importance to make possible to shorten period for reproduction test using M. macrocopa due to it's short life cycle.

Toxicity Test of butachlor to Medaka (뷰타클로르의 송사리에 대한 독성시험)

  • In order to evaluate the toxic effects of butachlor, a herbicide widely used for control of weeds in paddy field, on medaka (Oryzias latipes), acute toxicity tests for five developmental stages and early life stage toxicity test of were conducted. As the results of acute toxicity test, $96h-LC_{50}s$ for 1 day, 1 week, 2 weeks, 2 months and 5 months after hatching of O. latipes were 0.68, 0.52, 0.38, 1.09 and $0.45\;mg\;L^{-1}$, respectively. This indicated that the most sensitive stage was 2 weeks after hatching. The early life stage toxicity test showed that no statistically significant hatching period and hatching success of embryo was observed at all concentrations of butaclor. However, 0.05 and $0.1\;mg\;L^{-1}$ of butachlor showed statistically significant post hatching survival with p<0.1. Abnormalities of larva were 2.1, 2.3 and 10% at 0.025, 0.05 and $0.1\;mg\;L^{-1}$ of concentration, respectively. They showed abnormal vertebral axis, craniofacial alteration and retarded yolk-sac resorption. The total length and weight were decreased depending on butachlor concentration the end of test. Weight of larva was showed more sensitive toxic indicator than total length. The toxicological responses of O. latipes to butachlor expressed as LOEC(lowest observed effect concentration), NOEC(no observed effect concentration) and MATC(maximum acceptable toxicant concentration) values were 0.025, 0.013 and $0.018\;mg\;L^{-1}$, respectively.

Toxic effect of chlorothalonil, an antifouling agent, on survival and population growth rate of a marine rotifer, Brachionus plicatilis (해산 로티퍼(Brachionus plicatilis)의 생존율 및 개체군 성장률을 이용한 신방오물질(Chlorothalonil)의 독성영향)

  • In this study, the toxicity of chlorothalonil was investigated using survival rate and population growth rate of a marine rotifer, Brachionus plicatilis, typically used in live food in marine aquaculture systems. The survival rate of B. plicatilis was determined after 24 h of exposure to chlorothalonil (0.010-0.156 mg L-1). Population growth rate of B. plicatilis was calculated after 72 h of exposure to chlorothalonil (0.078-1.250 mg L-1). The survival rate and population growth rate of B. plicatilis exposed to chlorothalonil in single-dose toxicity assessment showed concentration-dependent reductions. Survival rates of B. plicatilis exposed to chlorothalonil had the following values: NOEC, 0.020mg L-1; LOEC, 0.039 mg L-1; and EC50, 0.057 mg L-1. Population growth rate of B. plicatilis exposed to chlorothalonil had the following values: NOEC, 0.156 mg L-1; LOEC, 0.313 mg L-1; and EC50, 0.506 mg L-1. When the residual concentration of chlorothalonil in the marine coastal area was more than 0.039 mg L-1, it had a toxic effect on B. plicatilis, a zooplankton. This paper provides toxicity values that can be used as baseline data for organizing environmental standards of chlorothalonil. It also provides insight into toxic effects of chlorothalonil on other non-target organisms.

Toxic Evaluation of Antifouling Paint (Irgarol and Diuron) using the Population Growth Rate of Marine Diatom, Skeletonema costatum (해산 규조류(Skeletonema costatum)의 개체군 성장률 분석을 이용한 신방오도료(Irgarol, Diuron)의 독성평가)

  • We evaluated the toxic effects of antifouling paint (irgarol and diuron) on the population growth rate (r) of the marine diatom, Skeletonema costatum. The r of S. costatum was determined after 96 hrs of exposure to irgarol (0, 0.31, 0.63, 1.25, 2.5 and 5 ㎍ l-1) and diuron (0, 7.81, 15.63, 31.25, 62.5 and 125 ㎍ l-1). It was observed that r in the control (absence of irgarol and diuron) were greater than 0.04, while r in the treatment groups decreased with increasing irgarol and diuron concentrations. Irgarol and diuron reduced r in a dose-dependent manner with significant decreases occurring at concentrations above 0.63 and 15.63 ㎍ l-1, respectively. The EC50 values of r in irgarol and diuron exposure were 1.09 and 45.45 ㎍ l-1. No observed effect concentration (NOEC) were 0.31 and 7.81 ㎍ l-1, the lowest observed effect concentration (LOEC) were 0.63 and 15.63 ㎍ l-1. This result indicate that a concentration of greater than 0.63 ㎍ l-1 of irgarol and 15.63 ㎍ l-1 of diuron in marine ecosystems induced to decreasing r of S. costatum. Also, these toxic values can be useful as a baseline data for the toxic evaluation of irgarol and diuron in marine ecosystems.

  • Toxicity assessment of antifouling agent, zinc undecylenate (ZU) has been investigated using the rate of survival and population growth in marine rotifer, Brachionus plicatilis. The survival rate of Brachionus plicatilis was determined after 24 h of exposure to ZU and was not affected up to the maximum level of $100.0mg\;L^{-1}$ of ZU. The population growth rate (r) was determined after 72 h of exposure to ZU. It was observed that r in the controls (absence ZU) was greater than 0.5 but exhibited a sudden decrease with an increase in the concentration of ZU. ZU reduced r in a dose-dependent manner and a significant reduction occurred at a concentration greater than $12.5mg\;L^{-1}$. The 50% effective concentration ($EC_{50}$) value of r during ZU exposure was $26.4mg\;L^{-1}$, No-observed-effect-concentration (NOEC) was $6.3mg\;L^{-1}$ and Lowest-observed-effect-concentration (LOEC) was $12.5mg\;L^{-1}$, respectively. Based on the results, it is apparent that ZU concentration greater than $12.5mg\;L^{-1}$ exhibited a toxic effect on the r of zooplankton, B. plicatilis in natural ecosystems.

Development of A Material Flow Model for Predicting Nano-TiO2 Particles Removal Efficiency in a WWTP (하수처리장 내 나노 TiO2 입자 제거효율 예측을 위한 물질흐름모델 개발)

  • A wastewater treatment plant (WWTP) is a major gateway for the engineered nano-particles (ENPs) entering the water bodies. However existing studies have reported that many WWTPs exceed the No Observed Effective Concentration (NOEC) for ENPs in the effluent and thus they need to be designed or operated to more effectively control ENPs. Understanding and predicting ENPs behaviors in the unit and \the whole process of a WWTP should be the key first step to develop strategies for controlling ENPs using a WWTP. This study aims to provide a modeling tool for predicting behaviors and removal efficiencies of ENPs in a WWTP associated with process characteristics and major operating conditions. In the developed model, four unit processes for water treatment (primary clarifier, bioreactor, secondary clarifier, and tertiary treatment unit) were considered. Additionally the model simulates the sludge treatment system as a single process that integrates multiple unit processes including thickeners, digesters, and dewatering units. The simulated ENP was nano-sized TiO2, (nano-TiO2) assuming that its behavior in a WWTP is dominated by the attachment with suspendid solids (SS), while dissolution and transformation are insignificant. The attachment mechanism of nano-TiO2 to SS was incorporated into the model equations using the apparent solid-liquid partition coefficient (Kd) under the equilibrium assumption between solid and liquid phase, and a steady state condition of nano-TiO2 was assumed. Furthermore, an MS Excel-based user interface was developed to provide user-friendly environment for the nano-TiO2 removal efficiency calculations. Using the developed model, a preliminary simulation was conducted to examine how the solid retention time (SRT), a major operating variable affects the removal efficiency of nano-TiO2 particles in a WWTP.

The Risk Assessment of Butachlor for the Freshwater Aquatic Organisms (Butachlor의 수서생물에 대한 위해성 평가)

  • To assess the effect of butachlor on freshwater aquatic organisms, acute toxicity studies for algae, invertebrate and fishes were conducted. The algae grow inhibition studies were carried out to determine the growth inhibition effects of butachlor (Tech. 93.4%) in Pseudokirchneriella subcapitata (formerly knows as Selenastrum capriconutum), Desmodesmus subspicatus (formerly known as Scendusmus subspicatus), and Chlorella vulgaris during the exposure period of 72 hours. The toxicological responses of P. subcapitata, D. subspicatus, and C. vulgaris to butachlor, expressed in individual $ErC_{50}$ values were 0.002, 0.019, and $10.4mgL^{-1}$, respectively and NOEC values were 0.0008, 0.0016, and $5.34mg\;L^{-1}$, respectively. P. subcapitata was more sensitive than any other algae species. Butachlor has very high toxicity to the algae, such as P. subcapitata and D. subspicatu. In the acute immobilisation test for Daphnia magna, the 24 and $48h-EC_{50}$ values were 2.55 and $1.50mg\;L^{-1}$, respectively. As the results of the acute toxicity test on Cyprinus carpio, Oryzias latipes and Misgurnus anguillicaudatus, the $96h-LC_{50}s$ were 0.62, 0.41 and $0.24mg\;L^{-1}$, respectively. The following ecological risk assessment of butachlor was performed on the basis of the toxicological data of algae, invertebrate and fish and exposure concentrations in rice paddy, drain and river. When a butachlor formulation is applied in rice paddy field according to label recommendation, the measured concentration of butachlor in paddy water was $0.41mg\;L^{-1}$ and the predicted environmental concentration (PEC) of butachlor in drain water was $0.03 mg\;L^{-1}$. Residues of butachlor detected in major rivers between 1997 and 1998 were ranged from $0.0004mg\;L^{-1}$ to $0.0029mg\;L^{-1}$. Toxicity exposure ratios (TERs) of algae in rice paddy, drain and river were 0.004, 0.05 and 0.36, respectively and indicated that butachlor has a risk to algae in rice paddy, drain and river. On the other hand, TERs of invertebrate in rice paddy, drain and river were 3.6, 50 and 357, respectively, well above 2, indicating no risk to invertebrate. TERs of fish in rice paddy, drain and river were 0.58, 8 and 57, respectively. The TERs for fish indicated that butachlor poses a risk to fish in rice paddy but has no risk to fish in agricultural drain and river. In conclusion, butachlor has a minimal risk to algae in agricultural drain and river exposed from rice drainage but has no risk to invertebrate and fish.

Initial Risk Assessment of Disodium Disulphite in OECD High Production Volume Chemical Program

  • Disodium disulphite, the HPV chemical, was assigned to Korea in order to implement OECD SIDS program in 1999. It was produced about 3,200 ton/year in 1998. This report evaluates the toxic potency of disodium disulphite based on the environmental and mammalian effects as well as human exposure. Oral $LD_{50}$ in rats is 1,540 mg/kg b.w. and effects was observed to the stomach, liver and the GI track that was filled with blood. For repeated dose toxicity, the predominant effect was the induction of stomach lesion due to local irritation. The no observed adverse effect lever for local (stomach irritation) was about 217 mg/kg bw/day. There is no evidence that disodium disulphite is genotoxic in vivo. No reproductive or developmental toxicty of disodium disulphite was observed for the period up to 2 yr and over three generation. In humans, urticaria and asthma with itching, edema, rhinitis, and nasal congestion were reported. Disodium disulphite is unlikely to induce respiratory sensitization but may enhance symptom of asthma in sensitive individuals. This chemical would be mainly transported to water compartment when released to environmental compartments since it is highly water soluble (470 g/l at 20). Low K oc (2.447) indicates disodium disulphite is so mobile in soil that it may not stay in the terrestrial compartment. The chemical has been tested in a limited number of aquatic species. hem acute toxicity test to fish, 96 hr-$LC_{50}$ was > 100 mg/1. For algae, 72 hr-$XC_{50}$ was 48.1 mg/1. For daphnid, the acute toxicity value of 48 hr-$EC_{50}$ was 88.76 mg/1, and chronic value of 21day-NOEC was > 10 mg/1. Therefore, PNEC of 0.1 mg/l for the aquatic organism was obtained from the chronic value of daphnid using the assessment factor of 100. Based on these data the disodium disulphite was recommended as low priority for further post-SIDS work in OECD.

Responses of Misgurnus anguillicaudatus and Cyprinus carpio Fed on Disease Resistant(OsCK1) Rice Variety (병저항성 GM(OsCK1)벼가 미꾸리(Misgurnus anguillicaudatus)및 잉어(Cyprinus carpio)에 미치는 영향)

  • BACKGROUND: The disease resistant (OsCK1) rice was generated by inserting choline kinase (CK1) and phosphinothricin acetyltransferase (PAT) genes isolated from Oriza sativa and Streptomyces hygroscopicus into the genome of rice (Nakdongbyeo). With the potential problems of safety, the non-target organism evaluation is required as an essential element for the environmental risk assessment of genetically modified (GM) crops. In present study, we studied the effects on survival of Misgurnus anguillicaudatus and Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. METHODS AND RESULTS: The M. anguillicaudatus and C. carpio were fed on disease resistant (OsCK1) rice and non-genetically modified (non-GM) rice (Nakdongbyeo) to 0, 10, 100, 1,000 and 5,000 mg/L, as treatment concentration respectively. The OsCK1 rice used for the test was confirmed to have the OsCK1/PAT gene expression by the PCR and ELISA analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of M. anguillicaudatus and C. carpio fed on between OsCK1 rice and non-GM rice. The 96hr-$LC_{50}$ values showed no difference between OsCK1 rice (>5,000 mg/L) and non-GM rice (>5,000 mg/L). CONCLUSION(S): The results of this study suggested that there was no significant difference in toxicity for M. anguillicaudatus and C. carpio between OsCK1 rice and non-GM counterparts.

Body Residue-based Approach as an Alternative of the External Concentration-based Approach for the Ecological Risk Assessment (외부환경농도에 기반한 생태위해성 평가방법의 대안으로서 생체잔류량 접근법)

  • 환경오염물질로부터 수생태계 보호를 위한 표준적인 평가 및 관리 수단인 수질환경기준은 오염물질의 독성작용이 일어나는 표적기관에서의 오염물질의 농도에 대한 대체측정치로서 환경 내 오염물질의 농도를 이용해 왔다. 이러한 '외부환경농도에 기반한 접근방법'은 표적기관에서의 독성물질의 농도가 생물체내 농도에 비례하고, 결국 외부환경농도에도 비례할 것이라고 가정한다. 따라서 환경오염물질의 생물이용도나 생물축적 양상의 차이 때문에 고유 독성치를 비교 평가하는데 한계가 있다. 이와 달리 '생물체내 농도에 기반한 접근방법(이하 생체잔류량 접근법)'은 환경오염물질의 생물이용도나 종 특이적 생물축적 양상과 관련된 불확실성을 제거하고, 환경오염물질 고유의 독성을 비교 평가할 수 있게 해준다. 특히 생체잔류량 접근법을 독성동태학 및 독성역학 모델과 함께 사용하는 경우는 실제 현장에서 일어나는 복잡한 노출조건에서의 독성영향을 예측하는데 활용할 수 있다. '생체잔류량 접근법'은 독성기작별 임계잔류량(Critical Body Residue)을 결정함으로써 생물모니터링의 결과를 해석하는데 적용되고 있다. 또한 생태위해성평가를 위해서 필요한 '무영향예측농도(Predicted No-effect Concentration, PNEC)를 예측하기 위한 방법으로 생체 내 잔류량에 기반해서 농도-시간-반응관계를 기술하고, 예측할 수 있는 새로운 유형의 독성역학 및 독성동태학 모델을 제시하고, 생체내 '무영향농도(No Effect Concentration, NEC)'를 추정하게 해 준다. 특히 생체내 NEC는 '무영향관찰농도(No Observed Effect Concentration, NOEC)'와 '영향농도(Effect Concentration, EC)'처럼 분산분석이나 회귀분석모델과 같은 통계적 모델에 기반해서, 농도-반응관계만을 기술할 뿐인 기존 독성모델을 대체할 대안으로 최근에 OECD와 ISO에 의해서 추천되었다.분석을 시행한 결과 인지기능 장애정도 및 MMSEK 점수 증가에 따른 사망위험도는 어느 모형에서도 인지기능 장애정도가 사망에 미치는 위험도는 통계적으로 유의하지 않았다(표 6, 표 7). 이상 본 연구는 농촌지역 노인들에서 인지기능 장애정도가 사망에 미치는 영향을 알아보고자 하였지만, 인지기능 장애정도가 사망에 미치는 영향을 통계적으로 유의하게 고찰하지 못하였다.의한 차이를 보였다. (P<0.05, P<0.001) 5. Excelco로 부식처리된 도재가 5% HF 용액으로 부식처리된 도재보다 부식정도가 더 현저하였다.은 제언을 하고자 한다. 먼저, 학교급식에 대한 식단 작성 시 학생들이 학교에서 제공되기 원하는 식단에 대한 의견을 받고 그 의견에 대한 결과를 게시하여 학생들이 제공되기 원하는 식단을 급식 시 제공하여 학생들이 식단선택에 동참할 수 있는 기회를 주는 것이 바람직하겠다. 또한 영양사는 학급의 반대표와의 정기적인 모임을 가짐으로서 학생들의 불만사항 및 개선 요구사항에대해 서로 의견을 교환하여 설문지조사가 아닌 직접적인 대화를 하여 문제점을 파악하고자 하는 적극적인 자세가 필요하겠다. 특히 아침식사의 결식 빈도가 높았고 이는 급식성과에 부정적인 영향을 줄 뿐 아니라 학교에서 제공하는 음식의 섭취정도에도 영향을 주고 있으므로 학생들에게 학부모와 전담교사 및 학교영양사는 학생들에게 이상적인 아침식사에 대한 교육은 물론이고 아침식사를 실천할 수 있도록 다양한 방안에 대해 함께 연구해야 하겠다. 정부차원에서 학교급식에 아침식사 프로그램을 도입할 수 있는 방안을 연구하고, 아침을 결식하는 학생이 학교에서 수업시작 하기 전에 간단한 식사를 할 수 있는 정책 도입이 필요하다acid의 생성량(生成量)을 측정(測定)하였는데 periodate의 소비량(消費量)은 1.23 mole, formic acid의 생성량(生成量)은 0.78 mole이다.한 경우도 비교적 많이 먹고 있었다(24.3%). 남 여
