siRNA Mediated Silencing of NIN1/RPN12 Binding Protein 1 Homolog Inhibits Proliferation and Growth of Breast Cancer Cells

  • The gene encoding the Nin one binding (NOB1) protein which plays an essential role in protein degradation has been investigated for possible tumor promoting functions. The present study was focused on NOB1 as a possible therapeutic target for breast cancer treatment. Lentivirus mediated NOB1 siRNA transfection was used to silence the NOB1 gene in two established breast cancer cell lines, MCF-7 and MDA-MB-231, successful transfection being confirmed by fluorescence imaging. NOB1 deletion caused significant decline in cell proliferation was observed in both cell lines as investigated by MTT assay. Furthermore the number and size of the colonies formed were also significantly reduced in the absence of NOB1. Moreover NOB1 gene knockdown arrested the cell cycle and inhibited cell cycle related protein expression. Collectively these results indicate that NOB1 plays an essential role in breast cancer cell proliferation and its gene expression could be a therapeutic target.

Efficacy of nobiletin in improving hypercholesterolemia and nonalcoholic fatty liver disease in high-cholesterol diet-fed mice

  • BACKGROUND/OBJECTIVES: Nobiletin (NOB), a citrus flavonoid, is reported to have beneficial effects on cardiovascular and metabolic health. However, there is limited research investigating the effect of long-term supplementation with low-dose NOB on high-cholesterol diet (HCD)-induced hypercholesterolemia and non-obese nonalcoholic fatty liver disease (NAFLD). Therefore, we investigated the influence of NOB on hypercholesterolemia and NAFLD in HCD-fed mice. SUBJECTS/METHODS: C57BL/6J mice were fed a normal diet (ND) or HCD (35 kcal% fat, 1.25% cholesterol, 0.5% cholic acid) with or without NOB (0.02%) for 20 weeks. RESULTS: HCD feeding markedly reduced the final body weight compared to ND feeding, with no apparent energy intake differences. NOB supplementation suppressed HCD-induced weight loss without altering energy intake. Moreover, NOB significantly decreased the total cholesterol (TC) levels and the low-density lipoprotein (LDL)/very-LDL-cholesterol to TC ratio, and increased the high-density lipoprotein-cholesterol/TC ratio in plasma, compared to those for HCD feeding alone. The plasma levels of inflammatory and atherosclerosis markers (C-reactive protein, oxidized LDL, interleukin [IL]-1β, IL-6, and plasminogen activator inhibitor-1) were significantly lower, whereas those of anti-atherogenic adiponectin and paraoxonase were higher in the NOB-supplemented group than in the HCD control group. Furthermore, NOB significantly decreased liver weight, hepatic cholesterol and triglyceride contents, and lipid droplet accumulation by inhibiting messenger RNA expression of hepatic genes and activity levels of cholesterol synthesis-, esterification-, and fatty acid synthesis-associated enzymes, concomitantly enhancing fatty acid oxidation-related gene expression and enzyme activities. Dietary NOB supplementation may protect against hypercholesterolemia and NAFLD via regulation of hepatic lipid metabolism in HCD-fed mice; these effects are associated with the amelioration of inflammation and reductions in the levels of atherosclerosis-associated cardiovascular markers. CONCLUSIONS: The present study suggests that NOB may serve as a potential therapeutic agent for the treatment of HCD-induced hypercholesterolemia and NAFLD.

Growth Characteristics of Nitrite Oxidizing Bacteria Isolated from Anaerobic Digestion Liquor (혐기소화액에서 분리한 아질산 산화세균의 생장특성)

  • Two nitrite oxidizing bacteria, NOB1 and NOB2, were isolated from anaerobic digester liquer of food wastewater and analyzed for their growth characteristics and the ability to oxidize nitrite under different temperature, pH, and DO( dissolved oxygen) concentrations. Both of the isolated strains have shown the best growth at pH 7.0 and at $35^{\circ}C$, and also shown higher growth rate with the increasing dissolved oxygen concentrations. As the factors to restrict the growth of these strains, parameters such as pH and DO were found to be effective ones, by increasing (up to 9.0) or decreasing pH (up to 5.0), or lowing DO below 1.0 ppm. Especially, the ability to oxidize nitrite in both strains was about 50% lower in below 1.0 ppm of DO than above of 1.0 ppm. NOB2 was found to be two times greater in both the growth rate and the nitrite-oxidizing ability than NOB1.

A Study on the Relation between Working Time and Tree Formal Characteristics (임업(林業)에서의 순수작업시간(純粹作業時間)과 임목형상조건(林木形狀條件)과의 관계연구(關係硏究))

  • The main purpose of this research is to provide scientific informations about standard wage and performance tariffs in forest management with special reference to working time for thinning. To identify relationships between net working time and tree characteristics, three geographically different sample plots were established at Yangsan, Bongpyung and Jinan and 460 oaks, 372 Japanese larches, 232 red pine and 240 pitch pine mere selected at each sample plots. The results of statistical analysis using multiple regression are as follows ; 1. Five independent variables of breast height diameter(DBH), mid-diameter(MD) large end diameter(LD), log-length(L), No. of branches(NOB) were stable independent of worker and tree species. 2. Comparing correlation coefficient of five independent variables, the best predictive variables, breast height diameter and No. of branches, were selected. Breast height diameter and No. of branches were identified as the most important independent variables in terms of effect on the dependent variable of the working time. 3. Comparing coefficient of determination (Rp) and residual mean square (MSEp), the best Linear regression equation for each tree species was selected as follower : $WT=a+b1{\times}NOB+b2{\times}DBF$ 4. Proportion of hang-up time to total working time in thinning were 66% in oak stand, 74%, in Japanese larch stand, 55%, in red pine stand and 52% in pitch pine stand, respectively. 5. Based on the best regression equation, a table of working time was made by strata of number of branches and breast height diameter. 6. Total working time using the best regression equation in Table 5 can be predicted in terms of felling time, limbing time, hang-up time, i.e., total working time increases by 11 to 13 seconds with every 1 centimeter increase in breast height diameter from 7 to 16 centimeter.

Analysis of Free Ammonia Inhibition of Nitrite Oxidizing Bacteria Using a Dissolved Oxygen Respirometer

  • Free ammonia ($NH_3$-N) inhibition of nitrite-oxidizing bacteria (NOB) has been widely studied for partial nitrification (or nitrite accumulation) and denitrification via nitrite ($NO_2^-$-N) as a low-cost treatment of ammonium containing wastewater. The literature on $NH_3$-N inhibition of NOB, however, shows disagreement about the threshold $NH_3$-N concentration and its degree of inhibition. In order to clarify the confusion, a simple and cheap respirometric method was devised to investigate the effect of free ammonia inhibition of NOB. Sludge samples from an autotrophic nitrifying reactor were exposed to various $NH_3$-N concentrations to measure the maximum specific nitrite oxidation rate ($\hat{K}_{NO}$) using a respirometer. NOB biomass was estimated from the yield values in the literature. Free ammonia inhibition of nitrite oxidizing bacteria was reversible and the specific nitrite oxidation rate ($K_{NO}$) decreased from 0.141 to 0.116, 0.100, 0.097 and 0.081 mg $NO_2^-$-N/mg NOB h, respectively, as the $NH_3$-N concentration increased from 0.0 to 1.0, 4.1, 9.7 and 22.9 mg/L. A nonlinear regression based on the noncompetitive inhibition mode gave an estimate of the Inhibition concentration ($K_I$) of free ammonia to be 21.3 mg $NH_3$-N/L. Previous studies gave $\hat{K}_{NO}$ of Nitrobacter and Nitrospira as 0.120 and 0.032 mg/mg VSS h. The free ammonia concentration which inhibits Nitrobacter was $30{\sim}50\;mg$ $NH_3$-N/L and Nitrospira was inhibited at $0.04{\sim}0.08\;mg$ $NH_3$-N/L. The results support the fact that Nitrobacter is the dominant NOB in the reactor. The variations in the reported values of free ammonia inhibition may be due to the different species of nitrite oxidizers present in the reactors. The respirometric method provides rapid and reliable analysis of the behavior and community of the nitrite oxidizing bacteria.

Changes of Nitrifying Bacteria Depending on the Presence and Absence of Organic Pollutant in Nak-Dong River (낙동강에서의 유기성 오염 유무에 따른 질화세균의 변화)

  • This study was performed at 2 sites of Nak-Dong River to investigate the changes of nitrifiers depending on the presence and absence of organic pollutants (due to the effluents of domestic wastewater treatment plant, WWTP). Conventional chemical parameters such as T-N, $NH_4$-N, $NO_2$-N, $NO_3$-N were measured and the quantitative nitrifiers at the 2 sites were analyzed comparatively by fluorescent in situ hybridization (FISH) with NSO190 and NIT3, after checking the presence of gene amoA of ammonia oxidizing bacteria (AOB) and 16S rDNA signature sequence for Nitrobacter sp. that belongs to nitrite oxidizing bacteria (NOB). Also ${\alpha}{\cdot}{\beta}{\cdot}{\gamma}$-Proteobacteria were detected using FISH to get a glimpse of the general bacterial community structure of the sites. Based on the distribution structure of the ${\alpha}{\cdot}{\beta}{\cdot}{\gamma}$-Proteobacteria and the measurement of nitrogen in different phases, it could be said that the site 2 was more polluted with organics than site 1. Corresponding to the above conclusion, the average numbers of AOB and NOB detected by NSO160 and NIT3, respectively, at site 2 [AOB, $9.3{\times}10^5$; NOB, $1.6{\times}10^6$ (cells/ml)] was more than those at site 1 [AOB, $7.8{\times}10^5$; NOB, $0.8{\times}10^6$ (cells/ml)] and also their ratios to total counts were higher at site 2 (AOB, 27%; NOB, 34%) than those at site 1 (AOB, 18%; NOB, 23%). Thus, it could be concluded that the nitrification at site 2 was more active due to continuous loading of organics from the effluents of domestic WWTP, compared to site 1 located closed to raw drinking water supply and subsequently less polluted with organics.

Partial Nitritation in an SBR Reactor by Alkalinity Control (알칼리도 제어에 의한 SBR 반응조에서의 부분아질산화)

  • In this study, major parameter of partial nitritation was investigated for the stable operation. In order to establish partial nitritation system, prevailing parameters such as temperature, BA (bicarbonate alkalinity) and pH were evaluated. As a result, it is inferred that appropriate bicarbonate alkalinity ratio (mg $NaHCO_3{\cdot}L^{-1}/mg$ Inf. $NH_4{^+}-N{\cdot}L^{-1}$) drives stable 50% partial nitritation at $32^{\circ}C$ and ambient temperature, respectively. Alkalinity ratio was proposed as new strategy for 50% partial nitritation without pH control in both temperature regimes. Because of the results, it was added amound of BA required only for 50% nitritation to inhibit nitratation. The effluent $NO_2{^-}-N/NH_4{^+}-N$ ratio reached almost 100% when initial bicarbonate alkalinity ratios (mg $NaHCO_3{\cdot}L^{-1}/mg$ Inf. $NH_4{^+}-N{\cdot}L^{-1}$) were 6.8 (R1) and 6.7 (R2), respectively. Polymerase chain reaction and denaturing gradient gel electrophoresis (PCR-DGGE) results demonstrated that AOB was the dominant nitrifying bacteria and NOB was negligible after adopting process control.

Nitrospira Community Composition in Nitrifying Reactors Operated with Two Different Dissolved Oxygen Levels

  • Nitrospira is a dominant member of nitrite-oxidizing bacteria (NOB) in nitrifying bioreactors as well as in natural habitats. In this study, Nitrospira NOB were investigated in the two nitrifying reactors operated with high and low dissolved oxygen (DO) concentrations for a period of 300 days. Phylogenetic and terminal restriction fragment length polymorphism analyses based on 168 rRNA gene sequences revealed that the Nitrospira community compositions of the two reactors during the early period related to group 1 and half of the Nitrospira community composition shifted to group 2 in the high-DO reactor after day 179, although there was no significant change in the low-DO reactor. These results suggested that DO was an important factor affecting Nitrospira community compositions in the nitrifying reactors.

Simulated Nitrogen Removal for Double-Layered PVA/Alginate Structure for Autotrophic Single-Stage Nitrogen Removal (2중 구조의 PVA/alginate 겔 비드에서의 독립영양 단일공정 질소제거효율 시뮬레이션)

  • Recently, an autotrophic single-stage nitrogen removal (ASSNR) process based on the anaerobic ammonium oxidation (ANAMMOX) reaction has been proven as an economical ammonia treatment. It is highly evident that double-layered gel beads are a promising alternative to the natural biofilm for ASSNR because of the high mechanical strength of poly(vinyl alcohol) (PVA)/alginate structure and efficient protection of ANAMMOX bacteria from dissolved oxygen (DO) due to the thick outer layer. However, the thick outer layer results in severe mass transport limitation and consequent lowered bacterial activity. Therefore, the effects of the thickness of the outer layer on the overall reaction rate were tested in the biofilm model using AQUASIM for ammonia-oxidizing bacteria (AOB), nitrite-oxidizing bacteria (NOB) and ANAMMOX bacteria. A thickness of 0.5~1.0 mm is preferred for the maximum total nitrogen (TN) removal. In addition, a DO of 0.5 mg/L resulted in the best total nitrogen removal. A higher DO induces NOB activity and consequent lower TN removal efficiency. The optimal density of AO B and NO B density was 1~10% for a 10% ANAMMOX bacterial in the double-layered PVA/alginate gel beads. The real effects of operating parameters of the thickness of the outer layer, DO and concentrations of biomass balance should be intensively investigated in the controlled experiments in batch and continuous modes.

Effects of Nitrifying Bacterial Communities with Different HRTs and Backwashing Periods in Modified BAF Process (Modified BAF 공정에서 HRT 및 역세주기가 질산화 미생물의 군집에 미치는 영향)

  • The upflow Biobead$^{(R)}$ process, one of biological aerated filters (BAF), which was used commercially, invented for removal of organic materials and nitrification. This process was modified to enhance the ability of denitrification through the induction of pre-anoxic tank. In this research, we investigated the effects of hydraulic retention time (HRT) and backwashing period in aerobic tank. The characteristics of nitrifying bacteria, which are composed of ammonia-oxidizing bacteria (AOB) and nitrite-oxidizing bacteria (NOB), also investigated using fluorescence in situ hybridization (FISH). Even though the HRT was shortened, the efficiency of nitrification was not decreased when the organic loading rate and ammonium-nitrogen loading rate were $2.10kg/m^3/day$ and $0.25kg/m^3/day$, respectively. And then the distribution ratios of AOB and NOB showed the similar patterns. However, when the backwashing period was lengthened from 12 hours to 24 hours in aerobic 1 tank, the nitrification efficiency was decreased to 63.9% from 89.2%. The results of FISH explained that this decrease of nitrification efficiency was caused by the decrease of distribution ratio of AOB in aerobic 1 tank. The nitrification efficiencies of aerobic 1 and aerobic 2 tank were increased when the backwashing period was lengthened because of relative high distribution ratios of nitrifying bacteria.
