자생 바위솔속(Orostachys) 엽의 형태적 다양성 (Morphological diversities of leaf in Korean Native Orostachys Species)

  • 우리나라의 22개 지역에서 수집한 자생 바위솔속식물의 지역적으로 나타나는 변이를 구명 하기 위하여 형태적 특징을 조사하였다. 바위 솔(No. 3∼9)은 형태적으로 유사하며 , 공통적으로 엽형은 피침형, 엽두는 예철두이며, 1∼2mm의 가시를 갖는 형태적 특성을 가지고 있다. 그러므로 바위솔은 지역종간에 변이가 적은 비교적 유전적으로 안정된 종으로 생각된다. 둥근바위솔(O. malacophyllus)의 형태 특성은 엽형이 도란형이며, 엽두가 예두인 잎을 가지고 있다. 또한 가시가 없고 잎에 붉은점으로 치밀하게 구성된 테두리선을 갖는 것이 이 종에 속한 식물의 특징이다. 연화바위솔(No. 18∼22)에 속한 식물들의 잎은 예두, 둔두 혹은 원두의 엽두 형태를 가진 도란형이나 타원형으로 수집지별로 형태가 다양하다. 그러나 잎의 끝에 가시가 없고 회분빛녹색의 잎을 갖는 등 다른 군과 구별되는 특성을 지니고 있다. 포천 수집종(ll번)과 매물도 수집종인 호랑이발톱(10번)은 이 2종은 현재 명명되지 않은 새로운 종으로 추정된다.

붕어(Carassius auratus Linnaeus)와 떡붕어(C. cuvieri Temminck and Schlegel)의 유전적 비교 (Genetic Comparison Between Crucian Carp (Carassius auratus Linnaeus) and Crucian Carp (C. cuvieri Temminck and Schlegel))

  • 한국의 예산과 당진에서 각각 채취된 붕어 (Carassius auratus)와 떡붕어 (Carassius cuvieri)로부터 genomic DNA를 분리 추출하여 반복해서 PCR로 증폭시켰다. 선택된 7개의 RAPD primer를 이용하여 primer 당 total loci, shared loci by each species, polymorphic 및 specific loci를 얻어냈다. 2종의 붕어로부터 primer와 2지역간에 banding patterns의 복잡성이 두드러지게 나타났다. DNA fragment의 분자적 크기는 150bp에서부터 1,600bp까지 커다란 차이를 나타내었다. 본 연구에서 CCY 붕어 종에서는 458개의 loci가 나타났고, CCD 떡붕어 종에서는 358개의 loci가 확인되었다. 또한 CCY 붕어 종에서는 84개의 polymorphic loci (18.3%)가 확인되었고, CCD떡붕어 종에서는 48개의 polymorphic loci (13.4%)가 확인되었다. CCY 붕어 종에서는 154개의 shared loci가 나타났으며, 이는 primer당 평균적으로 22개의 loci로 확인되었다. 또한 CCD떡붕어 종에서는 187개의 shared loci가 확인되었고, 평균해서 primer 당 26.7개의 loci가 나타났다. CCY붕어 종과 CCD 떡붕어 종의 polymorphic loci는 각각 84개와 48개로 확인되었다. 모든 붕어와 떡붕어 시료의 평균적인 BS value를 기초로 해서 CCY 붕어 종의 similarity matrix를 조사해 본 결과 0.434로부터 0.868까지 나타났고, CCD 떡붕어 종의 값은 0.449로부터 0.924까지 확인되었다. CCY 붕어 종내의 평균적인 BS value는 0.641±0.013이고, CCD 떡붕어 종내의 BS value의 평균값은 0.684±0.013을 나타내었다. 결과적으로 CCD 떡붕어 종내의 개체의 BS value 평균값이 CCY 붕어 종내의 평균값보다 높게 나타났다. 2 붕어와 떡붕어간의 평균적인 BS value은 0.484±0.007 (0.307~0.682)를 나타내었다. 7개의 primer를 사용하여 얻어진 dendrogram은 cluster 1 (AURATUS no. 01~AURATUS no. 11), cluster 2 (CUVIERI no. 12~CUVIERI no. 21) 및cluster 3 (CUVIERI no. 22)와 같이 3개의 유전적 클러스터로 나뉘어졌다. CCY 붕어 종내의 8번째 개체 (AURATUS no. 08)와 9번째 개체 (AURATUS no. 09) 사이가 가장 가까운 유전적 관계 (0.064)를 나타내었다. 또한 CCY붕어 종의 11번째(AURATUS no. 11)와 CCD떡붕어 종의 17번째 (CUVIERI no. 17) 사이가 가장 먼 유전적 거리 (0.477)를 나타내었다. 결과적으로 볼 때 한국 및 대서양산 lobster (0.612), 갈치 (0.708), 동자개(0.714)에 비해서 상대적으로 낮은 유전적 거리를 나타내었다.

국내 자생 민들레 5종의 항산화 활성 및 Nitric Oxide 생성억제 활성 (Effect of Five Korean Native Taraxacum on Antioxidant Activity and Nitric Oxide Production Inhibitory Activity)

  • The effect on the antioxidant activity and Nitric Oxide activity production inhibitory activity of Taraxacum has not been known. Therefore, phenolics and flavonoid contents were investigated from the ethanol extracts of five different Taraxacum species. The results showed that, among the five Taraxacum, T. hallaisanensis contains the highest total phenolic and flavonoid contents. When the antioxidant activity was measured by DPPH, $ABTS^+$ and reducing power activity, the free radical scavenging activity of T. hallaisanensis was also the highest among five Taraxacum species. However, measurement by CCK-8 assay in Raw264.7 cells indicated that the extracts of Taraxacum species have no effect on cell viability. Moreover, we also investigated the effect of Taraxacum species on NO scavenging activity in lipopolysaccharide (LPS)-stimulated Raw264.7 cells. The results clearly showed that Taraxacum species inhibited NO production, and the inhibitory effect of T. hallaisanensis was the strongest. The above results suggested that Taraxacum species affected the antioxidant and NO scavenging activity, and among the five species, antioxidant and NO scavenging activity assay of T. hallaisanensis was significantly higher than those of other four Taraxacum species. Therefore, T. hallaisanensis could be used as a potential drug with anti-oxidant and anti-inflammatory effect.

RAPD를 이용한 자생 바위솔속(Orostachys) 식물의 유연관계 분석 (Analysis of Genetic Relationship among Korean Native Orostachys Species Using RAPD)

  • 한국에 자생하는 22지역의 바위솔속 식물들의 유전적 근연관계를 구명하기 위하여 RAPD분석을 실시하였다. 18종의 바위솔속 식물은 3개의 군(A, B, C 군)으로 집괴되었으며, 나머지 4종은 어떤 분류군에도 속하지 않았다. A군인 연화바위솔(No.18-No.22)은 유사도가 66.3-73.9%로 낮게 나타났으며, B군인 둥근바위솔(No.12-No.17)도 유사도 지수가 69.7-83.7%로 비교적 낮게 나타났다. C군인 바위솔(No.3-No.9)은 안면도 수집종(No.9)을 제외한 유사도 지수가 84.2-92.3%로 높아, 지역종 간에 변이가 적은 비교적 유전적으로 안정된 종으로 생각되었다.

Effect of island geography on plant species on uninhabited islands in southeastern South Korea

  • We investigated the pattern of floral diversity including naturalized plant species and three ecological factors (area, elevation and distance from mainland) of plant species on 53 uninhabited islands in Gyungsangnam-do, southeastern South Korea. A total of 206 taxa in 67 families were observed, and the species of Compositae was most common. Thirteen taxa in eight families of the naturalized plants were observed on 33 islands. The numbers of total plant species, area and elevation were significantly correlated, but no relationship with distance from the mainland was observed. In addition, no relationship was found among the numbers of naturalized plants, area and elevation. However, the average rate of naturalization on islands with different elevations differed significantly, indicating the smallest proportion of naturalized plant species was on high islands. Multiple regression of total species richness identified elevation as a significant factor, while no significant variables were correlated with naturalized plant species. Nonmetric multidimensional scaling (NMS) ordination identified three major variables, distance from mainland, number of naturalized plant species and elevation. These findings indicate that the geography of islands such as area and elevation affected the species richness of plants on uninhabited islands, while human disturbance had a greater effect than geography on the species richness of naturalized plants on islands in southeastern South Korea.

회문산 일대의 관속식물 분포 특성 (Distribution Characteristics of Vascular Plants in the Mt. Hoemun)

  • The vascular plants in the Mt. Hoemun were listed 541 taxa composed of 113 families, 354 genera, 473 species, 1 subspecies, 64 varieties and 3 forms. Divided into 541 taxa; woody plants were 151 taxa (27.9%) and herbaceous plants were 390 taxa (72.1%). Based on the list of rare plants by the Korea Forest Service, 10 taxa were recorded in the studied areas; Lilium distichum (Preservation priority order: No. 159), Lilium callosum (No. 191), Tricyrtis dilatata (No. 97), Iris ensata var. sponianea (No. 197), Gastrodia elata (No.9), Aristolochia contorta (No. 151), Paeonia obovata (No. 30: Legal protection species), Prunus yedoensis (No. 110 : Planted species), Viola albida (No. 202), Schpolia japonica (No. 208). Based on the list of Korean endemic plants, 15 taxa were recorded; Cephalotaxus koreana, Lilium amabile, Salix purpurea var. japonica, Pseudostellaria multiflora, Aconitum pseudolaeve var. erectum, Filipendula glaberrima, Weigela subsessilis, etc. Based on the list of approved for delivering overseas of plants, 10 taxa were recorded; Carex okamotoi, Lilium distichum, Clematis trichotoma, Vaccinium oldhami, Paulownia coreana, Asperula lasiantha; etc. Specific plant species by floral region were total 4S taxa (8.3% of all 541 taxa of vascular plants); Gastrodia elata, Paeonia obovata in class V, Carex arenicola, Corydalis grandicalyx in class IV, 5 taxa (Spiraea salicifolia, Schpolia japonica, Asperula lasiantha, etc.) in class III, 7 taxa (Lilium distichum, Iris ensata var. spontanea, Caryopteris incana, etc.) in class II and 28 taxa (Camptosorus sibiricus, Orixa japonica, Lonicera praeflorens, etc.) in class I. The naturalized plants in the surveyed sites were 13 families, 38 genera, 43 species, 2 varieties, 45 taxa (Phleum pratense, Medicago sativa, Sonchus asper, etc.) and naturalization rate was 8.3% of all 541 taxa of vascular plants. Ambrosia artemisiifolia var. elatior an ecosystem disturbing wild plants have been increasing. Therefore, the counter plants for continuous control and conservation are needed on the ecosystem of Mt. Hoemun.

영산강의 식물플랑크톤 군집 동태 (Dynamics of Phytoplankton Community in Youngsan River)

  • Dynamics of phytoplankton communities were investigated at five stations in Youngsan River from December 1995 to November 1996. Phytoplankton were identified to the total of 466 taxa, which were composed of 136 genera, 438 species, 27 varieties and 1 forma. The standing crops ranged 1,163-50,765 cells ${\cdot}ml^{-1}$ during the investigation periods. The variation of classes was in order to Chrysophyceae - Bascillariophyceae - Chlorophyceae at St. 1 and was only Bacillariophyceae at the other stations. The dominant species were 12 taxa including Aulacoseira ambigua, A. garnulata, Cyclotella meneghiniana, C. pseudostelligera, Chlorella vulgaris, Dinobryon sertularia, Flagilaria elliptica, Monoraphidium contortum, Micractinium pusillum, Nitzschia palea, Scenedesmus grahneisii and Stephanodiscus hantzschii f. tenuis. The standing crops of dominant species ranged from 10.6% to 94.7%. The genus Dinobryon in Chrysophyceae was dominant species at St. 1 in December 1995, but not recorded in the other months. The dominant species were composed with the planktonic diatoms from winter to spring and were the tychoplanktonic and the benthic species at St. 2-5 from summer to fall. The relationships between total standing crops and water temperature, pH, $NH_4$, $NO_2$, $NO_3$ and $PO_4$ showed low positive or negative coefficients. Stephanodiscus hantzschii f. tenuis had low positive or negative coefficients with water temperature, $NH_4$, $NO_2$, $NO_3$ and $PO_4$. The fluctuations of standing crops in the Youngsan river phytoplankton community were not caused by a single factor but controlled by the complex factors with interaction between phytoplankton community and envirowmental factors.

Genetic Distances and Variations of Three Clupeid Species Determined by PCR Technique

  • In this study, seven oligonucleotides primers were shown to generate the shared loci, specific loci, unique shared loci to each species and shared loci by the three species which could be obviously calculated. Euclidean genetic distances within- and between-species were also calculated by complete linkage method with the sustenance of the hierarchical dendrogram program Systat version 13. The genomic DNA isolated from herring (Clupea pallasii), Korean anchovy (Coilia nasus) and large-eyed herring (Harengula zunashi), respectively, in the Yellow Sea, were amplified several times by PCR reaction. The hierarchical dendrogram shows three chief branches: cluster 1 (PALLASII 01, 02, 03, 04, 06 and 07), cluster 2 (NASUS 08, 09, 10, 11, 12, 13 and 14), and cluster 3 (ZUNASHI 15, 16, 17, 18, 19, 20, 21 and PALLASII 05). In three clupeid species, the shortest genetic distance displaying significant molecular difference was between individual PALLASII no. 03 and PALLASII no. 02 (0.018). Individual no. 06 of PALLASII was most distantly related to NASUS no. 11 (genetic distance = 0.318). Individuals from herring (C. pallasii) species (0.920) exhibited higher bandsharing values than did individuals from Korean anchovy (C. nasus) species (0.872) (P<0.05). As a result, this PCR analysis generated on the genetic data displayed that the herring (C. pallasii) species was widely separated from Korean anchovy (C. nasus) species. Reversely, individuals of Korean anchovy (C. nasus) species were a little closely related to those of large-eyed herring (H. zunashi) species.

낙동강 중,하류의 식물플랑크톤 군집의 월 변화 (Monthly Variations of Phytoplankton Communities in the Mid and Lower Parts of the Nakdong River)

  • Monthly variations of phytoplankton communities were conducted to the investigation at five stations in mid and lower parts of the Nakdong River from December 1995 to November 1996. The phytoplankton communities were identified a total 456taxa which were composed of 136 genera, 427 species, 27 varieties and 2 forma. The standing crops of phytoplankton communities ranged from 2.7 to 52.8 (${\times}$10³)cells·ml$^{-1}$ during the investigation periods. Bacillariophyceae was the dominant classes at all stations. The dominant species were 7taxa and the standing crops of these ranged from 18.3 to 95.1% of the standing crops of phytoplankton communities. The dominant species in the mid parts (st. 1, 2) of this river system were benthic species such as Navicula gregaria, N. viridula var. rostellata and Nitzschia palea, however it was planktonic species (Stephan discus hantzschii, Golenkinia radiata) and meroplanktonic species (Aulacoseira ambigua, A. granulata) in the lower parts (st. 3, 4, 5). The seasonal variations of the dominant species in the lower parts were appeared to the planktonic species (S. hantzschii) from winter to spring, and were the meroplanktonic species (A. ambigua and A. granulata) in from summer to fall. The lower parts of the Nakdong river were entropic states because the dominant species were composed A. ambigua, A. granulate and S. hantzschii which were indicators of entropic water quality. The interrelationships between total standing crops and environmental factors(water temperature, pH, NH$_4$, NO$_2$, NO$_3$ and PO$_4$) were low positive or negative coefficients. S. hantzschii had low positive or negative relations with water temperature, NH$_4$, NO$_2$, NO$_3$ and PO$_4$. As the result, the variations of standing crops of phytoplankton were not caused by a single factor but controlled by the complex factors.

녹화용(綠化用) 자생(自生) 목본식물(木本植物)과 초본식물종자(草本植物種子)의 혼파처리(混播處理)에 관(關)한 연구(硏究) (Studies on Mixed-Seeding of Native Woody and Herb Species)

  • 이 연구는 비탈면 녹화용(綠化用) 자생목본(自生木本)과 초본종자(草本種子) 적정혼파비(適定混播比)를 결정하기 위해, 서울대학교 농업생명과학대학 실내온실에서 목본(木本) 초본종자(草本種子)의 파종실험(播種實驗)을 실시하였으며, 파종(播種)은 목본(木本)의 경우 초기발아기대본수 60립(粒)/$m^2$을 기준으로 초본종자(草本種子)와 혼합하여 복토량(覆土量), 시비처리(施肥處理), 파종량별(播種量別)로 실험하여 얻은 결과를 분석한 것이다. 1994년 6월 파종후(播種後) 목본(木本)과 초본(草本)의 개체수(個體數), 생중량(生重量), 수고(樹高)와 초고변화(草高變化), 분얼수(分蘖數)를 조사한 결과, 목본(木本)의 개체수(個體數)는 1995년에 싸리가 파종구(播種區)($m^2$)별(別)로 평균 1.14본(本), 족제비싸리 0.496본(本)이였으며, 조팝나무와 물오리나무는 파종 발아후 당해년도(1994)에 고사하였다. 초본(草本)의 경우는 1995년에 달맞이꽃이 5.06본(本), 솔새 1,072본(本), 개솔새는 0본(本)이였으며, 억새는 412.53본(本), 쑥 88.6본(本), 비수리 8.9본(本), 새 57.46본(本)으로 조사되었다. 지상부생장(地上部生長)에 있어서, 목본(木本)의 싸리와 족제비싸리는 급격한 지상부생장(地上部生長)과 생중량(生重量)의 증가를 보이고, 초본(草本)의 경우에는 파종 당해년도에 솔새, 개솔새, 억새, 달맞이꽃, 새 등은 생장(生長)이 빠르나 파종 이듬해에는 솔새와 억새가 지상부(地上部) 및 생중량(生重量)의 증가가 높았다. 파종량(播種量)에 따른 목본(木本)의 변화를 보면, 파종 당해년도에는 수종(樹種)간에 유의성(有意性)이 없었으나, 파종 이듬해에는 파종량(播種量)에 따라 유의(有意)하였고, 초본(草本)의 경우는 파종량(播種量)과 개체수(個體數)의 변화는 파종당해년도와 이듬해 모두 유의(有意)하였다. 분얼(分蘖)이 가장 우수한 것은 억새(21개)로 나타났으며, 다음은 솔새, 새 순으로 나타났다. 녹화용(綠化用) 자생목본종자(自生木本種子)와 초본종자(草本種子)의 혼파(混播)에서, 초본(草本)의 종자량(種子量)이 많을수록 목본(木本)의 개체수(個體數), 지상부(地上部) 생장량(生長量), 생중량(生重量), 초본분얼수(草本分蘖數) 등은 감소하였다. 따라서, 비탈면의 녹화식생(綠化植生)으로, 목본(木本)은 싸리와 족제비싸리, 초본(草本)으로는 분얼(分蘖)이 우수한 새류(類)를 이용하는 것이 효과적이라 분석되며, 목본(木本)의 성립(成立)을 위해서는 파종량(播種量)에 대한 연구가 더욱 수행되어야 할 것이다.
