Microbiological Safety of Various Gouda Cheeses Produced from Raw Milk (비살균 원유로 제조된 다양한 Gouda 치즈의 미생물 안전성)

  • Since 2018, the production and sales of ram-milk cheese ripened for over 60 days has been permitted in South Korea. Hence, this study aimed to examine the microbiological changes in 7 different types of Gouda cheese. During the aging period, traditional raw-milk Gouda Cheeses 1 and 2 did not contain Salmonella spp. during the 60-day storage period and no E. coli after 20-day storage. Coliform bacteria were not detected in Cheese 1 after 40 days; however, they were detected in Cheese 2 up to 60 days. Salmonella spp. were inhibited during the 60-day storage period in Cheese 3 (Salmonella spp.-contaminated raw-milk Gouda cheese), Cheese 4 (Cheese 3 contaminated with lactic acid bacteria DH 5 isolated from Kefir) and Cheese 5 (Cheese 3 contaminated with lactic acid bacteria DN1 isolated from Kefir). In particular, inhibition of Salmonella spp. was more prominent in Cheese 4 and Cheese 5 than in Cheese 3. During 60-day storage, Cheese 6 had a significantly reduced lactic acid bacteria. Furthermore, in Cheese 7, E. coli, E. Salmonella ssp. were rarely detected, and lactic acid bacteria were slightly greater in Cheese 7 than in other cheeses during the 60-day period. Moreover, all samples from Cheese 1 to Cheese 7 were not contaminated with Listeria monocytogenes, Staphylococcus aureus, Clostridium perfringens, and E. coli O157:H7.

Exogenous DNA Transfer by Intracytoplasmic Sperm Injection in Porcine Oocytes (돼지에 있어서 난자내 정자 직접 주입에 의한 외래 유전자 도입에 관한 연구)

  • Sperm-mediated DNA transfer has a potential to markedly simplify techniques for the generation of transgenic animals. The exogenous DNA transfer by intracytoplasmic sperm injection (ICSI) procedure has been recently introduced in the production of transgenic animals. In this study, the developmental competence and tile expression rates of transgene were investigated after injection of spermatozoon or sperm head with enhanced green fluorescent protein (EGFP) gene into the mature porcine oocytes. The porcine oocytes were injected with intact sperm, membrane-disrupted sperm or sperm head. After injection. embryos were cultured in NCSU23 medium up to the blastocyst stage, and the developmental competence and expression rates were studied. The developmental rate (67.0%) of sperm injection group was higher than that (59.7%) of sperm head injection group, and the rates of EGFP expression were also significantly different between sperm injection and sperm head injection groups (42.1 vs 20.0%) (F<0.05). In the porcine oocytes injected with sperm treated with different methods of membrane disruption, the removal of sperm membrane did not alter the developmental competence of embryos. The rate of blastocysts at 7 days after injection with intact and membrane disrupted sperm were 15.0 and 14.2%, respectively. The EGFP expression rates, 38.4% in embryos injected with frozen-thawed sperm was higher than that, 22.4% of embryos injected with the Triton X-100 treated sperm. Prior to injection, sperm were cultured in different EGFP gene concentrations from 0.Ol to 1ng/u${mu}ell$. However, no significant difference in developmental rates of embryos among different concentrations of EGFP gene were observed. The highest expression rate of EGFP gene, 37.4% was obtained from the embryos injected with spermatozoa treated with 0.1 ng/${mu}ell$ EGFP gene. These results suggested that exogenous DNA could be attached to the membrane disrupted sperm, and that these sperm could be used as a vector carrying foreign DNA into embryos.

Effects of Ferrous Sulfate and Ascorbic Acid on In Vitro Fertility and Sperm Lipid Peroxidation in the Pig (돼지의 체외수정능력과 정자의 Lipid Peroxidation에 있어서 Ascorbic Acid와 Ferrous Sulfate의 영향)

  • This study investigated the effect of ferrous sulfate (Fe$^{2+}$) and/or ascorbic acid (Asc) on fertilizing ability in vitro of frozen-thawed boar spermatozoa. Using chlortetracycline (CTC) fluorescence, the spermatozoa was treated in preincubation medium with control, Fe$^{2+}$(1 mM), Asc (0.5 mM) and Fe$^{2+}$Asc to assessed for acrosome reaction, and the oocyte penetration test to determine whether the Fe$^{2+}$ and/or Asc can promote the penetration ability in vitro. When frozen-thawed spermatozoa was washed with preincubation medium, there were significantly (P < 0.05) more acrosome-reacted in medium with Fe$^{2+}$Asc (38%) than control (27%). The penetration rates were also significantly (P < 0.05) higher in medium with Fe$^{2+}$Asc (76%) than control (55%). Next, the lipid peroxidation of sperm was evaluated on the basis of malondialdehyde production following same treatments. The addition of Fe$^{2+}$Asc to sperm suspension increases the formation of malondialdehyde. However, there were not significantly different under the all conditions. The sperm suspension were also treated with control, Fe$^{2+}$, Asc and Fe$^{2+}$/Asc and assayed for sulfhydry1(-SH) group content. In the Fe$^{2+}$/Asc group, sperm-SH group were higher than another groups. In spermatozoa treated with Fe$^{2+}$ and/or Asc, however, no changes in sperm -SH-groups were detected when compared to controls. In another experiment, the activity of sperm binding to zona pellucida was evaluated through binding to salt-stored porcine oocytes. In control and Asc treatment groups, sperm binding to zona pellucida were significantly (P < 0.05) higher than in medium with Fe$^{2+}$. On the other hand, there is not a significant increase in binding to zona pellucida with spermatozoa treated by Fe$^{2+}$/Asc. In summary, the present study suggests that Fe$^{2+}$/Asc causes an enhancement in fertilizing ability that is associated with penetration rate increased without change of spermatozoa binding capacity to homologous zona pellucida.o homologous zona pellucida.

Effect of Feeding Colored Barley and Whole Crop Barley on Performance, Egg Quality and Blood Composition of Laying Hens (유색보리 및 청보리의 급여가 산란계의 생산성과 계란의 품질 및 혈액성상에 미치는 영향)

  • This study was conducted to investigate the effect of feeding colored barley (CB) and whole crop barley (WCB) on performance, egg quality and blood composition of laying hens. A total of 280, 35 weeks old Brown Nick laying hens were allocated into the individual cage for the period of four weeks. Experimental diets contained 2,750 kcal/kg ME and 16% CP, respectively. The treatments consisted of 5, 10, 15% CB and WCB as feed ingredients with control and there were five replications in each treatment. Higher egg production was noticed at 10% level of CB and 5% level of WCB in the diets, respectively. Feed intake was higher in WCB and CB treatments compared to those of the control (P<0.05), but the feed conversion was not different. Yolk color tended to increase both in WCB and CB groups. Haugh unit seemed to increase WCB treatments. When the CB and WCB were fed at 15% level, blood total proteins, albumin, total cholesterol and triglyceride contents were decreased than that of the control birds, but total cholesterol and HDL-cholesterol tended to be higher with the increasing level of WCB. As results, no significant differences were observed in performance and egg quality with different levels of dietary WCB and CB. Thus, CB and WCB can be substituted as a feed ingredient up to 15% level in the laying hens' diet. However, further studies are required by feeding more than 15% levels of CB and WCB in the diet of laying hens.

Effects of Supplementation of Ruminally Protected Amino Acids on In vitro Ruminal Parameters and Milk Yield and Milk Composition of Dairy Cows in Mid-lactation (보호아미노산의 추가 공급이 반추위 발효성상 및 비유중기 착유우의 유량 및 유성분에 미치는 영향)

  • This study was undertaken to investigate the effects of ruminally protected amino acids (Methionine and Lysine) on in vitro ruminal parameters, and in vivo milk yield and milk composition in mid-lactating cows. In the first in vitro experiment, there were no statistical significances between treatments in ruminal pH and dry matter digestibility during various incubation times. In the second in vivo experiment, milk yield decreased by 11.92% in control and 5.68% in the treatment respectively, but decrease rate of milk yield in the treatment was lower than control. Milk yields naturally decreased as time goes by since the DIMs(Days in milk) of the cows in experiment were in mid-lactation period. 4% FCM(Fat corrected milk) and milk protein yields also, respectively, decreased by 11.25% and 11.09% in control and 6.16% and 5.47% in the treatment as compared with the intial. Milk protein and milk fat production were higher in the treatment(0.90kg, 1.10kg) than those of control(0.66kg, 0.79kg). Milk fat content significantly increased with supplementing protected amino acids as compared to control(P<0.05). From the above results, protected amino acids were positively utilized in the performances of mid-lactating cows without inhibiting rumen fermentation. Further investigation is suggested for essential amino acid composition and intestinal digestion rate out of rumen bypass protein in dietary protein to be estimated.

Greenhouse Environment and Growth of Green Pepper (Capsicum annuum L.) in Greenhouse Covered with CEM BIO Film (CEM BIO Film 피복시설의 환경특성과 풋고추 생육)

  • Spectroradiometric light transmittance from 300 to 1,100nm in the greenhouse covered with the CEM BIO polyethylene film was greater than that in the greenhouse covered with polyethylene film (control). As a whole, solar radiation transmittance into greenhouse was a half level, due to shades caused by double layer covering, frame and equipment. Net radiation energy emitted throughout surface of the greenhouse covered with CEM BIO polyethylene film was 5,424.5W.m$^{-2}$ , which was lower by 2.9% as compared to that of the greenhouse covered with polyethylene film. Photosynthetically active radiation from 400 to 700nm of the greenhouse covered with CEM BIO polyethylene film was 3,861.2W.m$^{-2}$ , which was higher by 3.8% as compared to hat of the greenhouse covered with polyethylene film. Accumulated minimum air temperature from Oct. 7, 1997 to Oct. 16, 1997 of the greenhouse covered with CEM BIO polyethylene film was 100.5$^{\circ}C$, which was higher by 2.5$^{\circ}C$ as compared to that of the greenhouse covered with polyethylene film. As results, height, stem diameter, leaf count, leaf area, fresh weight and dry weight of green pepper plants and canopy production structure measured at 30 days after transplanting were enhanced. Mean fruit weight n the greenhouse covered with CEM BIO polyethylene film was 11.28 g and 1.25 g greater as compared to that in the greenhouse covered with polyethylene film, due to increased fruit diameter and flesh thickness. Percent marketable fruits produced in the greenhouse covered with CEM BIO polyethylene film were 96.1%, and was greater by 2.7% thant that of the greenhouse covered with polyethylnee film due to decreased infection, sterility, severe curve and twisted fruits. The green pepper yield of the greenhouse covered with CEM BIO polyethylene film from Nov. 19, 1997 to Feb. 3, 1998 was greater by 974 kg per hectare than that of the greenhouse covered with polyethylene film, but the total fruit had no difference.

Effects of Donor Cells and Estrus Synchronization on the Production of Cloned Korean Native Goat (공핵 세포 및 발정 동기화가 복제 재래 산양 생산에 미치는 영향)

  • The objective of this study was to examine the effect of donor cell types, the source of recipient oocytes and estrous synchronization on pregnancy and delivery rates of somatic cell nuclear transfer (SCNT) embryos in Korean native goats. Recipient oocytes were surgically collected after superovulation. Ear cells and fetal fibroblasts were collected and cultured in serum-starvation condition (TCM-199 + 0.5% FBS) for cell confluence. The zonae pellucidae of in vivo- and in vitro-matured oocytes were partially drilled using a laser system. Single somatic cell was transferred into the enucleated oocyte. The reconstructed oocytes were electrically fused with 0.3 M mannitol. After the fusion, embryos were activated by Ionomycin+6-DMAP. NT embryos were cultured in mSOF medium supplemented with 0.8% BSA at $39^{\circ}C$ in an atmosphere of 5% $CO_2$, 5% $O_2$, 90% $N_2$ for 12 to 20 hr. One hundred and two SCNT embryos were transferred into 20 recipients and pregnancy rate at days 30 was 20.0%. Of them, one developed to term and delivered 1 kid. Ear cells showed significantly higher fusion (63.8 vs. 26.5%) and pregnancy rates (20.0 vs. 0.0%) than those of fetal fibroblast (p<0.05). The recipients synchronized by CIDR showed significantly lower pregnancy rates compared to that of recipient in natural estrus ($0.0{\sim}25.0%$ vs. 100%) (p<0.05). Cloned kid was born from the recipient in natural estrus. For the synchronization of estrus between recipient and donor, there was no difference between treatments (${\pm}0$ vs. +12 hr) in pregnancy rate. The first healthy cloned kid (Jinsoonny) was produced by transfer of SCNT embryos derived from in vivo oocytes and ear cells into a recipient goat whose estrus was synchronized with the donor. These results imply that donor cells for nuclear transfer may affect the success rate, and the estrus synchronization between donor and recipient animals can also be important.

Main constituents and bioactivities of different parts of aronia (Aronia melanocarpa) (아로니아 부위별 주요 성분 정량 및 생리활성 평가)

  • This study was designed to evaluate the biological activities and main constituents of different parts (fruit, leaf, and stem) of aronia (Aronia melanocarpa). The total phenolic and flavonoidcontents, DPPH and ABTS+ radical-scavenging activity, reducing power, and ferric reducing/antioxidant power were observed to follow the order of: leaves > stems > fruits, regardless of extraction solvents. The inhibitory activity against lipopolysaccharide-induced NO production in Raw 264.7 cells was significantly higher in the aronialeaf extract-treated group than in the groups treated with stem and fruit extracts. The ultra-performance liquid chromatography (UPLC) analysis was mainly composed of routine. In addition, the highest content level was measured in the case of the catechinmemberepigallocatechin witha higher value than that found in green tea. Theresults of this studyprovide useful information for understanding the chemical constituents and biological activities of aroniafruits and byproducts.

Dry Weight and Nitrogen Contents in Different Parts of 'Fuyu' Persimmon as Affected by Application Timing and Methods of Supplemental Nitrogen (질소 추비 시기와 방법이 '부유' 단감나무의 건물중과 질소함량 및 수체 부위별 분포에 미치는 영향)

  • We studied the accumulation and partitioning of dry weight (DW) and nitrogen (N) in different parts of field-grown 'Fuyu' persimmon to elucidate that the foliar applications of supplemental N in June or September compared favorably with the traditional soil application in securing leaf area and fruit production. We also estimated the proportion of N permanently removed from the tree at the end of a growing season. Urea was applied either to leaves in June and/or September or to the soil in June and September for three consecutive years, and the trees were excavated in November for analyses. Total DW ranged from 4.2-4.8, 8.7-9.2, and 17.1-21.5 kg in a 4-, 5-, and 6-year-old tree, respectively, without statistical difference among the four treatments. Of the total DW, 3.3-10.2% was in shoots, 5.7-10.5% in leaves, 8.3-31.4% in aerial woods, 13.0-27.0% in root, and 28.0-59.3% in fruits. As the trees became more productive, DW proportion of fruits significantly affected that of the root: in 6-year-old trees, root DW accounted for only 10.6-15.8% of the tree total when fruit DW accounted for 50-60%. N contents ranged from 24.6-28.3, 48.3-53.5, and 98.3-122.6 g in a 4-, 5-, and 6-year-old trees, respectively, without statistical difference among the treatments. Of the total N, 6.2-11.5% was in shoots, 16.7-24.3% in leaves, 17.6-23.5% in aerial woods, 17.2-37.5% in roots, and 16.9-34.4% in fruits. As in DW, the increase in the proportion of N in fruits decreased in the root most significantly. Application methods for supplemental N did not affect the proportion of DW and N removed from the tree through abscising leaves and harvested fruits. Percentage of DW removal was 41 in 4- and 5-year-old trees, but it was 61 in more productive 6-year-old trees; that of N was 39, 43, and 49%, respectively. No significant changes in the contents of DW and N in field-grown trees, as well as their percentages removed from the tree at the end of the season, demonstrated that foliar application of supplemental N was as good as soil applications with much less N.

Effect of Peel and Whole Crop of Kale and Angelica Keiskei Koidz on Fatty Acid Composition and Quality of Eggs (케일과 명일엽 및 부산물의 급여가 계란 품질 및 지방산 조성에 미치는 영향)

  • The effects of various levels of dietary dried whole crop of kale (0.1%, 0.3%), dried peel of kale (0.3%), dried whole crop of Angelica keiskei (Koidz) (0.1%, 0.3%), and peel of Angelica keiskei (Koidz) (0.3%) in egg-laying performance were studied with 560 ISA brown layers for a period of 12 weeks. No significant differences were recognized between the treatments and control in egg production, feed intake and egg mass. The quality of egg and eggshell were not different among treatments but egg yolk color was improved in the both of whole crop and peel of kale and Angelica keiskei (Koidz). Vitamin and polyunsaturated fatty acid concentration of eggs increased in treatments compared to control but were not significantly different from controls. In conclusion, dried peel of kale and Angelica keiskei (Koidz) supplementation in laying hens diet improved egg yolk color, and improve vitamin concentration of chicken eggs.
