Investigations of Soil Chemical Properties in the Cultivation Fields of Rubus coreanus with Different Growth Ages (복분자 재배 기간에 따른 토양의 화학적 특성 변화)

  • This study was conducted to investigate the relations between soil chemical properties and Korean raspberry (Rubus coreanus Miq.) growth. Soils were collected from 54 sampling sites in 1 to 6 years old Korean raspberry cultivation fields (RCFs) at Gochang-gun, Jeollabuk-Do, Korea. In RCF with two to six years old plants, soil pH and exchangeable $Ca^{2+}$ and $Mg^{2+}$ contents were higher at the leaf-emergence time, but EC, available phosphorus content, CEC, and total nitrogen content were higher at the harvesting time; especially at the harvesting time, the content of available phosphorus in the RCF with 3~6 years old plants were at least three times higher than in the RCF with 2 years old plants. Water-soluble anions ($Cl^-$, ${NO_3}^-$, ${PO_4}^{2-}$, and ${SO_4}^{2-}$) in the RCF were also measured. The contents of ${NO_3}^-$ and ${SO_4}^{2-}$ were always higher in the RCF with older plants than in the RCF with younger plants, which were not affected by sampling time and sampling locations (rhizosphere and non-rhizosphere sites). However, soils collected from non-rhizosphere at the leaf-emergence time contained higher ${PO_4}^{2-}$ content, but $Cl^-$ content was higher in the soils collected from rhizosphere at the harvesting time. In general, soils in the RCFs contained excess amounts of inorganic nutrients such as available phosphorus and exchangeable $K^+$. Thus, optimal levels of soil properties for Korean raspberry cultivation should be reconsidered.

Blood Pressure Modulating Effects of Black Raspberry Extracts in vitro and in vivo (복분자 추출물의 항고혈압 활성)

  • This study aimed to investigate the effects of 50% ethanol extract of ripe black raspberry (Rubus occidentalis, RBR) on hypertension in human umbilical vein endothelial cells (HUVECs) and in spontaneously hypertensive rats (SHR). Angiotensin converting enzyme (ACE) inhibition and activation of nitric oxide production by endothelial nitric oxide synthase were significantly regulated by RBR in HUVEC cells. Moreover, the SHR showed significantly higher levels of blood pressure, ACE, renin, endothelin-1, and interleukin-6 than Wistar Kyoto rats (WKY). However, treatment with captopril and RBR decreased the levels of these hypertension-related events in the SHR. The renal arteriole showed greater media thickness/lumen diameter (%) in the SHR than in the WKY. However, media thickness/lumen diameter (%) was reduced in SHR by treatment with captopril and RBR. In addition, the number of eosinophilic cardiac muscle cells was decreased in the heart muscles after treatment with captopril and RBR. Therefore, this study suggests that 50% ethanol extract of RBR may be useful for the prevention and treatment of high blood pressure.

  • The sensitivity and specificity of crude and affinity-purified antigens of Clcnorchis sinensis obtained from the infected rabbits were studied. Stage-specific antigenic proteins from the eggs, metacercariae and adult worms were characterized by SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and enzyme-linked immunosorbent astray (ELISA). The results were as follows: 1. The antibody.binding antigen (ABA) purified from whole worm crude antigen (IVWA) by CNBr-activated Sepharose 4B affinity chromatography made :l specific bands against rabbit antisera on Ouchterlony gel diffusion plate, while WWA made 7 bands. Major WWA protein bands by SDS-PAGE were found at 16, 300~18, 500 and 28, 000~29, 000 daltons, while major ABA protein bands were at 18, 000~21, 000 and 29, 000~31, 000 daltons. The reactivity of ABA with rabbit anti-sera in ELISA was remarkably less sensitive than that of WWA. 2. Molecular weights of egg antigen (EGA), metacercarial antigen (MEA) and adult worm antigen (WWA) of C. sinensis ranged from 15, 000-200, 000 daltons, 15, 000-100, 000 daltons and 11, 000~80, 000 daltons, respectively. Major WWA proteins consisted mainly of polypeptide bands of low molecular weight, less than 31, 000 daltons, while those of EGA and MEA consisted of higher molecular T.eights than 30, 000 daltons. 3. The ELISA reactivities of WWA to rabbit anti.sera were remarkably greater than those of MEA. EGA showed negative reaction throughout the experiments. WWA showed higher optical density (O.D.) than 1.0, when reacted with rabbit anti-sera obtained at 4~6 weeks after the infection. In the rabbit anti-sera later than 12 weeks after the infection, the O.D. reacting witll WWA showed a plateau without variation. MEA shoT.ed relatively low O.D. values (<0.6), when reacted with anti-sera from lightly in(ected groups throughout the experiments, althougll there were some wealth positive cases (O.D.>0.6) ill heavily infected groups. MEA reacted with rabbit anti-sera showed negative results on Ouchterlony gel diffusion plates. Summarizing the above results, it is suggested that the whole worm antigen prepared from the adult worms of C. sinensis is most highly antigenic. However, this antigen might reveal cross reactions with other trematodes such as Paragonimus westermani, therefore, purification of antigenic proteins from the crude antigen is essential 18 increase the sensitivity and specificity for the immuncdiagnosis of clonorchiasis.

Function and regulation of nitric oxide signaling in Drosophila

  • Nitric oxide (NO) serves as an evolutionarily conserved signaling molecule that plays an important role in a wide variety of cellular processes. Extensive studies in Drosophila melanogaster have revealed that NO signaling is required for development, physiology, and stress responses in many different types of cells. In neuronal cells, multiple NO signaling pathways appear to operate in different combinations to regulate learning and memory formation, synaptic transmission, selective synaptic connections, axon degeneration, and axon regrowth. During organ development, elevated NO signaling suppresses cell cycle progression, whereas downregulated NO leads to an increase in larval body size via modulation of hormone signaling. The most striking feature of the Drosophila NO synthase is that various stressors, such as neuropeptides, aberrant proteins, hypoxia, bacterial infection, and mechanical injury, can activate Drosophila NO synthase, initially regulating cellular physiology to enable cells to survive. However, under severe stress or pathophysiological conditions, high levels of NO promote regulated cell death and the development of neurodegenerative diseases. In this review, I highlight and discuss the current understanding of molecular mechanisms by which NO signaling regulates distinct cellular functions and behaviors.

Anti-inflammatory effect of beluga lentil extract in RAW 264.7 macrophages (RAW 264.7 대식세포에서 벨루가 렌틸 추출물의 항염증 효과)

  • The anti-inflammatory effect of beluga lentil extract (BLE) and its underlying mechanisms were investigated in lipopolysaccharide (LPS)-treated RAW 264.7 cells. Treatment with BLE significantly decreased nitric oxide (NO) production and protein and mRNA expressions of inducible NO synthase (iNOS) in LPS-treated RAW 264.7 cells. Down-regulation of this inflammatory gene expression was not associated with NF-κB/MAPK signaling pathways, and further mechanistic studies demonstrated that BLE decreased LPS-induced iNOS expression through upregulation of the nuclear factor erythroid 2-related factor 2 (Nrf2)-mediated heme oxygenase-1 (HO-1) expression. These results suggest that beluga lentil represent a potential source of natural anti-inflammatory agents, and further studies will be necessary to determine its anti-inflammatory effects in vivo.

The Availability of Automobile Catalytic Convert of Copper Based on the DFT Calculations of Cu-NO Complexes (Cu-NO 복합체에 대한 DFT 계산에 따른 Cu의 자동차 촉매변환기 적합성)

  • The purpose of this study is to show the possibility of using Cu catalyst in removal of $NO_x$ from automobile exhaust which is regarded as the primary source of fine dust PM2.5. The energy and the bond lengths of the three possible structures of Cu-NO complex, which is formed by binding NO molecule to Cu, and the changes in IR and Raman spectra are calculated using MPW1PW91 method on the level of 6-311(+)G(d,p) of basis sets with Gaussian 09 program. As a result, the enthalpy of formation of the Cu-NO complexes are obtained as ${\Delta}H=104.89$, 91.98, -127.48 kJ/mol for the linear, bent, and bridging forms of them, respectively. And the bond lengths between N and O in NO complexes, which becomes longer than NO molecule, indicates that O is easily reduced from Cu-NO. In addition, the Cu-NO complexes using Cu catalyst can be easily measured by infrared or Raman spectroscopy because in the IR and Raman spectra of the NO and Cu-NO complexes the positon and the intensity of bands are definitely different in each vibration mode.

Product Quality and Shelf-life Effect of Low-fat Functional Sausages Manufactured with Sodium Lactate and Chitosans During Storage at 15℃ (젖산나트륨과 키토산을 첨가한 저지방 기능성 소시지의 15℃에서 저장 중 품질 및 저장성 효과)

  • This study was performed to measure physicochemical and textural characteristics, and shelf-life effect of low-fat functional sausages(LFFSs) manufactured with sodium lactate(SL, 3.3%), lac pigment and various molecular weights(MWs) of chitosan (Low=1.5 kDa, Med=30-50 kDa and High=200 kDa) during storage at 15$^{\circ}C$ for 18 days. LFFSs had 73.7-76.0% moisture, lower than 3% fat and 14-15% protein, respectively. pH values were 6.05-6.44 and the control(150 ppm, $NaNO_2$) was the lowest among LFFSs (p<0.05). Increasing storage time decreased pH values, but no differences in pH values were observed up to 6 days of storage (p>0.05). LFFSs containing SL and low MW of chitosan improved water holding capacity (WHC) and different from those with SL and medium-MW chitosan. WHC was decreased with increased storage time and differences of WHC were observed from 18 days of storage. The addition of chitosan reduced both lightness and redness values, as compared to 150 ppm sodium nitrite(SN), and increased storage time decreased yellowness(p<0.05), especially at 12 days of storage. LFFSs with SL and medium-MW chitosan increased most textural properties compared to the control(p<0.05). The addition of SN of 150 ppm in LFFSs retarded microbial growth for E. coli 0157:H7, while those with SL tended to have an antimicrobial effect for Listeria monocytogenes(LM). The growth rate of LM was delayed by addition of various MW of chitosans in LFFSs, especially high MW chitosan, as compared to LFFSs containing SL alone. These results indicated that the functional, textural and antimicrobial effects of LFFSs were improved by addition SL and various MW of chitosan combinations. In addition, 0.05% lac pigment improved the cure color of LFFSs similar to those of 150 ppm SN.

A molecule has a cis-typed molecular structure having the form of "the substituted butene backbone, C-C(S)=C(NO$_2$)-C, connecting to a sulfur atom with the methylbenzene ring and to a nitro group. The methylbenzene ring and the substituted butene moiety are nearly planar with the maximum deviations from their own molecular planes, 0.018 ${\AA}$ for the C(1) atom of the benzene group and 0.045 ${\AA}$ for the N atom of the NO$_2$ group, respectively. The angles to the plane of the butene backbone are 88.5$^{\circ}$from the plane of the methyl-benzene and 78.6$^{\circ}$from the plane of the nitro group. Rotation of the nitro group from the butene plane seems to reduced contribution of resonance structure involving the nitro group, and resultant repulsion between the O(2) atom of SO$_2$ and the O(3) atom of NO$_2$ appears to be 2.894 ${\AA}$ longer than an expected van der Waals distance of 2.80 ${\AA}$.

  • The crystal and molecular structure of acetone 4-benzylthiosemicarbazone, $C_{11}H_{15}N_3S$, has been determined by the single crystal X-ray diffraction methods. The crystals are monoclinic, space group $P2_1/c$ with unit cell dimensions, a = 10.249(7), b = 11.403(9), c = 10.149(7)TEX>${\AA}$, ${\beta}$ = 90.9$(1)^0$ and z = 4. The intensities were collected on an automatic four-circle diffractometer with graphite-monochromated Mo-$K_{\alpha}$ radiation. The structure was solved by direct methods and refined by full matrix least-squares methods. The final R was 0.045 for 1554 observed reflections. S-C(8)-N(2)-N(3)-C(9)-C(10) atoms make a zigzag planar chain. There are no unusual bond lengths and angles. There are two independent hydrogen bonds in the crystal structure. One is N-H${\cdots}$S intermolecular hydrogen bond with the length of 3.555${\AA}$ and makes dimer-like units. The other is N-H${\cdots}$N intramolecular hydrogen bond with the length of 2.568${\AA}$. The structure was compared with those of other thiosemicarbazone derivatives.

Effects of Buckwheat Polysaccharides on the Digestibilty of Casein in vitro (In vitro에서 메밀 다당분획이 casein 가수분해에 미치는 영향)

  • This study was conducted to determine the effect of soluble polysaccharides and indigestible polysaccharides on the digestibility of casein in vitro and the structure of polysaccharide. The digestibility of casein by trypsin in vitro was reduced to $80{\sim}89%$ and $69{\sim}99%$ by high molecular soluble polysaccharide (HMS-P) and low molecular insoluble polysaccharide (LMI-P) prepared from buckwheat, respectively. The digestibility of casein by chymotrypsin was reduced to $63{\sim}88%$ and$71{\sim}79%$ by HMS-P and LMI-P, respectively. But casein digestibiliy by trypsin and chymotrypsin was slightly reduced by LMS-P. The casein hydrolyzates inhibited by HMS-P gave a main peak in the void volume on Sepadex G-100, but the peak in the total volume was only appeared in case of LMS-P having no inhibito교 effect. It was suggested that the HMS-P was consisted of 4-linked and 4,6-dissubstituted glucose from the structural analysis by GC.
