구강세치제에 함유된 SLS(Sodium lauryl Sulfate)가 HaCaT 세포와 NIH-3T3 세포에 미치는 독성 효과 (The effect of the cytotoxicity of sodium lauryl sulfate containing toothpaste on HaCaT and NIH-3T3 cells)

  • Objectives: The purpose of this study was to determine the toxic effects of sodium lauryl sulfate(SLS) in human keratinocyte HaCaT cells and mouse fibroblast NIH-3T3 cells. Methods: The effect of sodium lauryl sulfate(SLS) cell viability and proliferation were determined by WST-1 assay and changes shape of nucleus were evaluated by Hoechst staining under fluorescence microscopy. Additionally, observation of cell morphological changes under light microscopy. Results: SLS induced cytotoxicity and a marked apoptosis in both HaCaT and NIH-3T3 cell lines. With the result of the WST-1 assay, SLS induced the cytotoxicity of 0.005% and 0.0075%, 0.01% SLS for 24 h after HaCaT and NIH-3T3 cells in time and dose-dependent manner(p<0.005). SLS inhibited cell growth and caused apoptosis as evidenced by nuclear fragmentation and condensation. Thus, determination of the morphological changes to define apoptosis was visualized using inverted phase contrast microscopy. Conclusions: SLS had toxicity of the human keratinocyte cells and mouse fibroblast cells and this study will provide the basic data for the development of proper SLS concentration in dentifrice.

Toxic Effect on Phenolic Compound by Colorimeteric Assay in Normal NIH 3T3 Fibroblasts

  • This study was carried out to evaluate the cytotoxic effect of phenolic compound on normal NIH 3T3 fibrolasts. The colorimetric assay for phenol compound, syringic acid was performed by MTT assay or XTT assay. MTT or XTT assays are known as a very sensitive method in measuring the cytotoxic effect of chemical agents in vitro. In the present study, syringic acid on normal Nlli 3T3 fibroblasts did not show any cytotoxicity for MTT assay or XTT assay compared with control after cells were treated with various concentrations of syringic acid for 48 hours. MTT/sub 50/ and XTT/sub 50/ were 3,340.9 μM and 2,462.4 μM of syringic acid, respectively. From the above the results, it is suggested that phenolic compound of syringic acid did not have any cytotoxicity on normal NIH 3T3 fibroblasts.

Up-Regulation of Glutathione Biosynthesis in NIH3T3 Cells Transformed with the ETV6-NTRK3 Gene Fusion

  • The ETV6-NTRK3 gene fusion, first identified in the chromosomal translocation in congenital fibrosarcoma, encodes a chimeric protein tyrosine kinase with potent transforming activity. ETV6-NTRK3-dependent transformation involves the joint action of NTRK3 signaling pathways, and aberrant cell cycle progression resulting from activation of Mek1 and Akt. The level of glutathione (GSH) was found to be markedly increased in ETV6-NTRK3-transformed NIH3T3 cells. The activities of the two GSH biosynthetic enzymes as well as of glutathione peroxidase, together with their mRNAs, were also higher in the transformed cells. The transformed cells were able to grow in the presence of GSH-depleting agents, whereas the control cells were not. L-Buthionine-(S,R)-sulfoximine (BSO) inhibited activation of Mek1 and Akt in the transformed NIH3T3 cells. These observations imply that up-regulation of GSH biosynthesis plays a central role in ETV6-NTRK3-induced transformation.

차가버섯과 어성초 함유 발효 조성물이 인체 위암 AGS 및 대장암 HCT-15 세포 생육에 미치는 영향 (Effect of Fermented Compositions Containing Inonotus obliquus with Houttuynia cordata on Growth of Human AGS Gastric and HCT-15 Colon Cancer Cells)

  • 어성초 분말을 포함하는 차가버섯 발효 조성물의 물 추출물이 인체 위암 세포 AGS 및 대장암 세포 HCT-15, 그리고 정상세포 NIH3T3 fibroblast의 세포 생육에 미치는 영향을 검토하였다. 세포독성 실험은 세포수 count와 MTT 방법으로 측정하였다. 어성초 분말을 포함하는 차가버섯 발효 조성물의 제조는 차가버섯과 어성초 분말을 혼합하고, 여기에 대두 발효 미생물원을 다시 혼합시켜 습도 $50{\sim}60%$,온도 $30{\sim}37^{\circ}C$에서 30일 정도 발효시킨 후 건조 시킨 분말에서 5% 물 추출물을 얻어 동결건조 시켜 실험에 제공하였다. MTT assay 방법에서 차가버섯 발효 조성물의 수용성 추출물 0.16, 0.4, 0.8, 1.6 및 4.0 mg/ml 첨가 농도에서 AGS 세포 생육은 13, 25, 40, 67 및78% 억제 되었으며, HCT-15세포 생육은 22, 40, 50, 69및 76%씩 각각 억제되었다. 그러나 동일한 실험조건에서 정상 세포수 NIH3T3은 86% 이상의 생존율을 나타내었다. 차가버섯 발효 조성물의 수용성 추출물은 인체 대장암 세포주 HCT-15와 위암 세포수 AGS에 대해 생육억제 작용이 강한 반면, 정상세포 NIH3T3에 대해서는 세포 독성을 거의 나타내지 않아 가장 바람직한 암예방 또는 항암식품 개발가능성을 제시하였다.

Carbofuran이 NIH 3T3섬유모세포와 흰쥐의 신장에 미치는 영향과 Phenobarbital sodium의 보상효과에 관한 연구 (Effects of Carbofuran and Compensation of Phenobarbital sodium in the NIH 3T3 Fibroblast and Rat Kidney)

  • This study was carried out to investigate toxicity of insecticide carbofuran and compensatory effects of phenobarbital sodium (PB) in vivo and in vitro. Sprague Dawley male rats were used as experimental animals and divided into carbofuran only administered group and simultaneous application group of carbofuran and PB. At 30 rain and 1, 3, 6, 12, 24, 48 and 96 hrs after each treatment, the animals were sacrificed by decapitation. Kidney were immediately removed, immersed in fixatives, and processed with routine method for light microscopic study. Paraffin sections were stained with H-E, PAM and PAS. $5.0\times 10^4$ cell/ml of NIH 3T3 fibroblast in each well of 24 multidish were cultured: After 24 hours, the cells were treated with solution of six groups; control group cultured in media only, carbofuran $MTT_50$ or $NR_50$ group cultured in the media containing carbofuran $MTT_50$ or $NR_50$ and four experimental groups cultured in the media containing carbofuran $NR_50$ plus various concentratins of PB. After the NIH 3T3 fibroblast of all groups were cultured in same condition for 48 hours, Tetrazolium MTT (MTT) and NR (neutral red) assay were performed to evaluate the cytotoxicity of cell organelles. Under the light microscope, atrophic change of renal corpuscles were frequently observed in 1 and 2 days after carbofuran treatment. The increase of the mesangium was apparent in 1 and 2 days after carbofuran treatment. Necrotic changes of the epithelium and loss of brush border of proximal tubules were most severe at 2 and 3 days after carbofuran treatment, respectively. In contrast, there were no evidences of the toxic effects on renal tissues at 48hrs in carbofuran-PB treated groups. Carbofuran $MTT_50$ and $NR_50$ were 78$\mu M$, 82.5$\mu M$ respectively. MTT and NR quantities were significantly increased in carbofuran-PB 100$\mu M$ treatment group and carbofuran-PB 100$\mu M$ treatment group. On the basis of these results, it is obvious that PB has compensatory effects against carbofuran toxicity.

고삼 추출물을 이용한 해독물질의 개발 (Development of Antitoxic Agents from Sophora flavescens Ait.)

  • 고삼 메탄올 추출물로부터 분리한 flavonoids를 IR, NMR등의 분광학적인 방법으로Leachianone A(LA)과 Sophoraflavanone G(SFG)로 동정하였고, 카드뮴의 세포독성효과를 고삼 및 고삼추출물이 방어할 수 있는 지를 검색하기 위하여 NIH 3T3 세포에 고삼 및 LA와 SFG, LA+SFG를 처리하고 MTT assay 및 광학현미경으로 세포의 생존률을 검색하여 다음과 같은 결론은 얻었다. MTT의 흡광도는 카드뮴의 농도에 의존하여 감소 하였으며, $IC_{50}$/인 MTT50은 12.5$\mu$M이었으며 카드뮴을 $IC_{50}$/농도로 처리하고 고삼 및 LA, SFG, LA+SFG를 각 화합물의 자체 세포독성을 갖지 않는 농도로 처리한 후 MTT assay로 세포 생존률을 측정한 결과 LA, SFG, LA+SFG 처리군들은 카드뮴 처리군(MTT$_{50}$)에 비해 세포생존률이 증가되었으며 이들은 고삼 및 고삼 추출물의 농도에 의존적으로 증가되었고 각각의 단일 화합물보다 두 화합물 LA+SFG병용처리군에서 수복 효과가 더 좋은 것으로 나타났으며 광학현미경적 소견에서도 세포재생이 뚜렷하게 보였다. 이상과 같은 고삼에서 분리한 flavonoids가 카드뮴 독성에 의하여 손상된 NIH 3T3 섬유아세포의 재생효과에 영향이 있는 것으로 판단된다.다.

Comparison of Bradykinin- and Platelet-Derived Growth Factor-Induced Phosphoinositide Turnover in NIH 3T3 Cells

  • Phosphoinositide turnover in response to platelet-derived growth factor, epidermal growth factor, and bradykinin was evaluated in NIH 3T3 cells. Platelet-derived growth factor and bradykinin induced a significant increase in incorporation of $^{32}P$ into phosphatidylinositol (PI), phosphatidylinositol 4-monophosphate (PIP), and phosphatidylinositol 4.5-bisphosphate ($PIP_2$) in serum-starved NIH 3T3 cells. However, epidermal growth factor increased incorporation of $^{32}P$ into these phosphoinositides by only a small amount. Stimulation with platelet-derived growth factor, not bradykinin, caused a rapid elevation of PI and PIP kinase activities that were maximally activated within 10 min. The maximal levels of their elevation in cells with plateletderived growth factor stimulation were 3.2-fold for PI kinase, and 2.1-fold for PIP kinase. Short term pretreatment of NIH 3T3 cells with phorbol 12-myristate 13-acetate, activator of protein kinase C. caused an approximately 60% decrease in platelet-derived growth factor-induced PI kinase activities, indicating the feedback regulation of phosphoinositide turnover by protein kinase C. These results suggest that although the enhancement of phosphoinositide turnover is a rapidly occurring response in platelet-derived growth factor- or bradykinin-stimulated NIH 3T3 cells, phosphoinositide kinases may be associated with initial signal transduction pathway relevant to platelet-derived growth factor but not to bradykinin.

유근피(楡根皮) 추출물의 유방암 세포주 MCF-7 생장 억제 효과 (Anti-proliferative Effect of Ulmi Pumilae Cortex Extracts on MCF-7 cells)

  • Purpose: This study was conducted to investigate the anti-proliferative effects of Ulmi Pumilae Cortex Extracts(UPCE) on MCF-7(human, breast, adenocaecinoma) and NIH3T3 (human, murine, fibroblast). Methods: MCF-7 cells and NIH3T3 cells were cultured and seeded in cell culture plates, respectively. UPC was extracted with hot water and then further fractionated it into five types: hexane, chloroform, ethyl acetate, butanol, and water soluable fractions. These five different fractions from UPCE were tested for their anti-proliferative effects on MCF-7 cells and NIH3T3 cells by MMT assay. Results: Among the five solvent-fractions of UPCE, n-hexane fraction and ethyl acetate fraction showed a strong anti-proliferative effects on MCF-7 cells but they displayed significant cytotoxicity on NIH3T3 cells, too. On the other hand, chloroform fraction showed a marked anti-proliferative effects on MCF-7 cells and low cytotoxicity on NIH3T3 cells. Conclusion: Chloloform fraction from UPCE showed selective anti-cancer activities on human breast cancer cell MCF-7 relatively to the other fractions.

IL-6 mRNA Expression in Mouse Peritoneal Macrophages and NIH3T3 Fibroblasts in Response to Candida albicans

  • Despite extensive investigation, the mechanisms of immune responses to Candida albicans infection remain poorly understood. Using RT-PCR and Northern blot analysis, this study demonstrates the pattern of IL-6 mRNA expression in thioglycollate-elicited mouse peritoneal macrophages and NIH3T3 fibroblasts (NIH3T3) in response to C. albicans. The expression of IL-6 mRNA was detectable in both cell types. However, IL-10 mRNA was only expressed in the macrophages, and IL-4 mRNA was not expressed in neither of the two cell types. Although the phagocytic function of the macrophages was inhibited by Cytochalasin D, these macrophages could still induce the expression of IL-6mRNA. These findings indicated that the phagocytosis of C. albicans is not pivotal in the induction of IL-6 mRNA expression. A Northern blot analysis was used to investigate the dose effects of C. albicans and time-course kinetics of IL-6 mRNA expression at various time points. IL-6 mRNA was expressed in a dose-independent manner, and was detectable as early as 30min after C. albicans stimulation. It was evenly sustained up to 4h. These results can contribute to understanding the mechanism of IL-6 mRNA expression in macrophages and NIH3T3 cells in response to C, albicans.
