Effect of Different Pretreatments on Indium-Tin Oxide Electrodes

  • The effect of pretreatment on indium-tin oxide (ITO) electrodes has been rarely studied, although that on metal and carbon electrodes has been enormously done. The electrochemical and surface properties of ITO electrodes are investigated after 6 different pretreatments. The electrochemical behaviors for oxygen reduction, $Ru(NH_3){_6}^{3+}$ reduction, $Fe(CN){_6}^{3-}$ reduction, and p-hydroquinone oxidation are compared, and the surface roughness, hydrophilicity, and surface chemical composition are also compared. Oxygen reduction, $Fe(CN){_6}^{3-}$ reduction, and p-hydroquinone oxidation are highly affected by the type of the pretreatment, whereas $Ru(NH_3){_6}^{3+}$ reduction is almost independent of it. Interestingly, oxygen reduction is significantly suppressed by the treatment in an HCl solution. The changes in surface roughness and composition are not high after each pretreatment, but the change in contact angle is substantial in some pretreatments.

  • Effects of chemical treatments on in sacco and in vitro digestibility of barley straw by rumen fluid and pure cultures of cellulolytic bacteria were studied to evaluate the pretreatment and to improve the poor quality feed. Chemicals were applied by dissolving them in water equivalent to 40% of the weight of the straw (dry matter basis). Pretreatment with 5% NaOH yielded the largest increase in sacco digestion followed by pretreatment with 2% $(NH_4)_2SO_3$, 2.6% $NH_4OH$, 1.6% $NaHSO_3$ and untreated straw (control). In sacco dry matter digestibility of straw treated with NaOH and $(NH_4)_2SO_3$ continued to increase as the concentration of chemical increased (1 to 7.5%), as it was the in vitro dry matter loss by leaching. Treatment of barley straw with 5% NaOH enhanced significantly (p < 0.01) in vitro digestibility by rumen fluid, Fibrobacter suceinogenes and Ruminococcus albus though the fermentation products by cellulolytic bacteria were low, whereas the treatment with 5% $(NH_4)_2SO_3$ inhibited in vitro digestibility by F. succinogenes and R. albus together with lower fermentation products. Dry matter loss by leaching and bacterial digestion from barley straw treated with NaOH and $(NH_4)_2SO_3$ suggested the effect of pretreatment with these chemicals were based on leaching, and the cellulolytic bacteria had little to do with digestion.

Retrospect on Refractories in Water Treatment (기존 정수처리방법으로 제거가 어려운 유기물에 대한 실험적 연구)

  • As a basic experiment to develope biological pretreatment proces~ in water treatment, the experiments on biodegradability and isothermal adsorption of activated carbon were performed on refractories such as humic acid, $NH_3-N$, phenol and ABS which caused the problems in drinking water treatment. Also, the treatabilities on humic acid were examined in the continuous flow type reactors. The removal efficiencies of humic acid, $NH_3-N$, phenol and ABS in the biodegradable experiments for 5 days were 20.1%, 73.4%, 91.7% and 97.5%, respectively. In the isothermal adsorption test of refractories on activated carbon to be used as a media in the continuous flow type reactors, ABS and phenol are adsorbed easily, but humic acid and $NH_3-N$ are difficult to be done. The removal efficiencies of humic acid in granular activated carbon(GAC) reactor were about 7-8% higher than in biological activated carbon(BAC) reactor. The removal efficiencies of humic acid in biological fluidized bed(BFB) reactor were about 30% in GAC media, but were almost zero in sea sand media.

Bioconversion of Straw into Improved Fodder: Preliminary Treatment of Rice Straw Using Mechanical, Chemical and/or Gamma Irradiation

  • Crude protein (CP) content of mechanically ground rice straw into small particles by an electric grinder and reducing value (RV) and soluble protein (SP) in the culture filtrate were lower than that of the chopped straw into $5{\sim}6\;cm$ lengths when both ground and chopped straws were fermented with Aspergillus ochraceus, A. terreus or Trichoderma koningii, at steady conditions. The reduction rate of RV, SP and CP was 22.2, 2.4, 7.3%; 9.1, 4.9, 8.5% or 0.0, 0.0, 3.6% for the three fungi, respectively. Chemical pretreatment of straw by soaking in $NH_{4}OH$ for a day caused significant increase in CP of the fermented straw than the other alkali and acidic pretreatments. Gamma irradiation pretreatment of dry and wet straw with water, specially at higher doses, 100, 200 or 500 kGy, caused significant increase in RV and SP as CP in the fermented straw by any of these fungi. Chemical-physical combination pretreatment of rice straw reduced the applied dose of gamma irradiation required for increasing fermentable ability of fungi from 500 kGy to 10 kGy with approximately the same results. Significant increases in RV and SP of fermented straw generally occurred as the dose of gamma irradiation for pretreated straw, which combined with $NH_{4}OH$, gradually rose. Whereas, the increase percentage in CP of fermented straw that was pretreated by $NH_{4}OH-10\;kGy$ was 12.4%, 15.4% or 8.6% for A. ochraceus, A. terreus or T. koningii, respectively.

  • Efficient extraction of total proteins from soil microorganisms is tedious because of small quantity. In this regard, an improved method for extraction of whole cell proteins is developed from soil microorganisms, Saccharomyces cerevisiae and Pichia pastoris. of which the cell wall are very strong. Pretreatment with NH4OH prior to the final extraction using NaOH/SDS was tried under the basis that ammonium ion was possible to enhance the permeability and/or to weaken the yeast cell walls. The pre-treatment of yeast cells with NH4OH drastically enhanced the protein extraction when it was compared with control (without NH4OH pre-treatment). At the pre-treatment of 0.04 N NH4OH at pH 9.0, about 3 fold of proteins was obtained from p. pastoris. Ammonium hydroxide appears to penetrate into the yeast cell walls more readily at basic pH. The effect of NH4OH pretreatment was pH dependent. The methods developed in this experiment might be applicable for an effective extraction of yeast proteins for the purpose of biochemical studies, especially proteomic analysis.

Dyeing of Cotton Fabrics Using Zizania latifolia Turcz. Extracts (줄풀을 활용한 면직물의 염색)

  • This study examined the pretreatment effect, appropriate dyeing conditions, color fastness and functionality of cotton fabrics dyed with Zizania latifolia Turcz. Dye uptake was substantially increased by pretreatment and chitosan pretreatment was much more effective than tannic acid pretreatment. Optimal dyeing conditions were colorant concentration of 100%, dyeing temperature of $100^{\circ}C$, dyeing time of 80 minutes and dyebath pH of 5.5. Color fastness of chitosan pretreatment and dyed cotton to washing, rubbing, perspiration and light was 4, 5, 3-4(acidic), 3(alkaline) and 3 respectively. Color fastness of tannic acid pretreatment and dyed cotton to washing, rubbing, perspiration and light was 4, 5, 3-4(acidic), 4-5(alkaline) and 4 respectively. Deodorization rates of ammonia(NH3) were 99.9% in both cases. UV protection rate were 98.3% of UV-A and 98.5% of UV-B in case of chitosan pretreatment. UV protection rate were 98.2% of UV-A and 98.5% of UV-B in case of tannic acid pretreatment. UV protection factor(UPF) was 50+ in both cases. Reduction rate of Staphyloccus aureus were 99.9% in both cases. Therefore Zizania latifolia Turcz. could be used a new functional colorant.

Effect of Feed Monochloramination on Performance of RO Membranes (NH2Cl 사용으로 인한 RO 막의 성능 향상)

  • The 15 month pilot study was performed to investigate the effect of pretreatment by monochloramine ($NH_2Cl$) on the performance of RO membranes made of cellulose acetate (CA) and polyamide (PA). Both RO membranes experienced severe biological fouling without any pretreatment during the treatment of highly organic surface water in Florida, USA. Feed monochloramination at 5 mg/L significantly minimized productivity loss by effective control of biofouling. The CA membrane did not show any structural damages by monochloramine, while the PA membrane suffered from a gradual loss of membrane integrity by chlorine oxidation, which was characterized as an increase in productivity and a decrease in selectivity. The degradation of PA membrane increased with increasing monochloramine dose.

  • It is difficult to obtain high Cu nucleation density and continuous Cu films in Cu-MOCVD without cleaning the TiN substrate prior to Cu deposition. In this study effects of plasma precleaning on the Cu nucleation density were investigated using SEM, XPS, AES, AFM analyses. Direct plasma pretreatment is much more effective than remote plasma pretreatment in enhancing Cu nucleation. Cleaning effects are enhanced with increasing the rf-power and the plasma exposure time in hydrogen plasma pretreatment. The mechanism through which Cu nucleation is enhanced by plasma pretreatment is as follows: Hydrogen ion\ulcorner in the hydrogen plasma react with TiN to form Ti and $NH_3$ Cu nucleation is easier on the Ti substrate than TiN substrate.

  • Soil N mineralization and nitrification can be measured conveniently using mixed bed (cation and anion) exchange resin bags. However, appropriate use of these resin bags requires pretreatment to avoid colorimetric interference and standardize N ion adsorption. Three pretreatments were evaluated : control (untreated), 2 M NaCl with a distilled water rinse, and 4 M NaCl. The 4 M NaCl treatment was effective at removing background levels of $NH_4{^+}$ and $NO_3{^-}$, but adsorbed low amounts (about 40%) of inorganic N from standard solutions. Untreated resin bags adsorbed a constant, higher amount of $NO_3{^-}$ (60%), but did not remove background levels of $NH_4{^+}$. The 2 M NaCl treatment followed by a distilled water rinse performed best : it removed background $NH_4{^+}$ and adsorbed a constant amount of both $NH_4{^+}$ (70%) and $NO_3{^-}$ (60%). Because the ion exchange resin is fairly expensive, we also tested if the resin bags could be reused. Resin bags were either loaded with $NH_4{^+}$ and $NO_3{^-}$ in the laboratory or incubated in soil in the field, desorbed with the 2 M NaCl treatment, and then loaded with standard $NH_4{^+}$ and $NO_3{^-}$ solutions. Lab loaded resin bags adsorbed about 60% of inorganic N then loaded with 2.5 or $5.0mgN\;1^{-1}$ and 70% when loaded at 10 or $20mgN\;1^{-1}$, whereas reused field incubated bags showed the opposite adsorption efficiency. These results demonstrate that resin bags can give reproducible results, but care must be taken to evaluate the effect of pretreatment and potential for reuse.

Effect of Ultrasonic Pretreatment on Analysis of Potassium Ion in Human Urine Using 15-Crown-5-Anthracene-based Membrane (15-Crown-5-Anthracene 막을 이용한 요 중의 칼륨이온 분석에 미치는 초음파 전처리의 효과)

  • The effect of ultrasonic decomposition was introduced to develop a pretreatment method for the analysis of potassium ion in human urine by potentiometry. N-(4’-benzo-15-crown-5)-anthracene-9-imine, which has a good selectivity coefficient for potassium against ammonium, was used as an ion-selective material for the determination of potassium in urine with relatively high concentration of $NH_4{^+}$. Protenis in urine be removed by 85.1% when the sample acidified with 1.0 M $HNO_3$ was preteated for 100 s by sonication. Potential response of the membrane electrode in the pretreated urine had a slope of 54.6(${\pm}0.2,\;n=5$) mV/decade over the linear range of log $[K^+]$=-5~-1(r=0.9997). When an oxidant, $H_2O_2$, was addwd to the urine sonicated with $HNO_3$, the deproteinization increased 10% more than that in case if only $HNO_3$ and then the maximum ratio of ca. 95% was obtained. Moreover, the Nernstian slope for $K^+$ added to the urinary sample increased to 56.7(${\pm}0.1,\;n=3$) mV/decade. When the calibration curves were measured, the slopes did not vary even after the electrode was successively used 20 times with ultrasonic cleaning. The results showed that an ultrasonic pretreatment method provides simplicity in use, reduced treatment time and improved potentiometric characteristics of the membrane as the method effectively removes ca. 95% of proteins in urine.
