Effect of Hyunggaeyunkyotangbalhyobang (HYBH) on Atopic Dermatitis in NC/Nga Mice Model (형개련교탕발효방(荊芥連翹湯醱酵方)이 NC/Nga mouse 동물병태에 미치는 영향)

  • Various related factors and tissue changes in vitro and in vivo were observed to investigate the efficacy of HYBH on atopic dermatitis. The results are described below. HYBH improved the atopic dermatitis symptoms by naked eye examination, and significantly decreased dermatitis clinical index at 14 weeks. HYBH significantly decreased CD4+/CD45+, CD4+, CD8+, CD3+/CD69+ immune cell ratios in PBMC by 28%, 16%, 30%, 26% and 22% respectively. HYBH significantly decreased CD11b+/Gr-1+, CD3 immune cell ratios in dorsal skin by 35.3% and 67.5% respectively. HYBH significantly decreased the expression of IL-4 and IFN-${\gamma}$ in spleen by 23% and 15% respectively. HYBH significantly decreased the production rate of IL-5, IL-13 and histamine in serum by 17%, 23%, and 8.8% respectively and increased IL-17 production by 17%. HYBH significantly decreased immunoglubulins IgG1 and IgE production in serum. The results above indicated that treatment of HYBH improved atopic dermatitis symptoms by anti-oxidant activity and immune modulation activity as a clinical evidence. Also, different fermentation conditions using various microbial strains should be accumulated as the clinical evidence for broad application in the future.

Inhibitory Effects of Gosambaeksunpibokhap-bang(GBBB) Extract in an Atopic Dermatitis Animal Model (아토피 동물모델에서 고삼백선피복합방(GBBB)의 아토피 억제효과)

  • In order to clarify the Gosambaeksunpibokhap-bang(GBBB)'s therapeutic possibility on atopic dermatitis (AD), influences of GBBB on the changes of various immune-related factors and histological changes in NC/Nga mice were evaluated. Experimental results are as follows. Sign of recovery from AD was observed in GBBB treated group with naked eye test. The ratio of white blood cells, neutrophil, lymphocytes, monocyte in blood were decreased to 54%, 63%, 57% and 86% respectively in the GBBB group. IL-4, IL-5, IL-13, Histamine and IgE were significantly decreased to 40%, 80%, 62%, 61% and 57% respectively in the GBBB group. H&E staining showed thickness of epidermis and dermis were decreased by GBBB and inhibited the infiltration of lymphocytes. On the basis of these results, GBBB was confirmed that the possibility as an AD treatment applied externally to the skin. In the further study, immune control mechanism of GBBB will be demonstrated through the additional molecular biological research.

Effects of Chunggi-san Administration along with Samhwangseze-gamibang on NC/Nga Atopic Mice (청기산(淸肌散)과 삼황세제가미방(三黃洗劑加味方) 병용이 NC/Nga mice의 아토피 피부염에 미치는 영향)

  • Atopic dermatitis(AD) is a chronic inflammatory skin disease. AD has increased gradually, many people are tortured with AD. Chunggi-san(CG) and Samhwangseze-gamibang(SG) has been used for many kinds of skin disease in the Oriental medicine. But reports about the effect of CG and SG are insufficient. So, author investigated the effect of CG and SG on NC/Nga atopic mice. Major findings are summarized as follows: The clinical skin severity scores of experimental group in 13 and 16 week were decreased by 42% and 50% compared to the control group. Serum IgE, IL-4, IL-5, IL-6, IgM, IgGI levels of experimental group were significantly decreased compared to the control group. Serum $IFN-\nu$ was significantly increased in the experimental group compared to the control group. mRNA expression levels of IL-4, IL-5, and CCR3 in the skin tissues of experimental group were significantly decreased, and expression level of IL-6 in the skin tissues of experimental group was significantly decreased compared to the control group. $IFN-\nu$ mRNA expression levels was increased compared to the control group. According to biopsy reports of the ear and skin tissues showed that the tissue damage, experimental group were highly reduced compared to the control group. Judging from that $IL-1{\beta}$, $TNF-{\alpha}$, IL-6 express of gene, the effects of inflammatory cytokines revelation were significantly decreased compared to the control group. Depending on the density of CG, inflammatory RAW 264.7 in the serum of CG were significantly inhibited compared to the control serum that leaded a COX-2 activity model.

Hwangnyeonhaedok-tang Extracts Ameliorates Atopic Dermatitis via Epidermal Lipid Barrier Regeneration in NC/Nga Mouse (황련해독탕의 피부지방장벽개선을 통한 Th2 분화조절이 아토피피부염 완화에 미치는 효과)

  • Objectives Hwangnyeonhaedok-tang is a Korean herbal medical treatment that removes toxic heat, fever and inflammation. The purpose of this study was to investigate the effect of Hwangnyeonhaedok-tang treatment on the relief of atopic dermatitis (AD) through regeneration of skin lipid barrier. Methods Male NC/Nga mice (20 g, 6 week age) were used. Each 10 mice were allocated to the control group (Ctrl), the AD-induced with no treatment group (AE), and the group which induced AD after administering Hwangnyeonhaedok-tang extract (HT). To induce AD-like skin lesions, sodium dodecyl sulfate (SDS) (Sigma-Aldrich, USA) was rubbed on the back of each mouse to remove the lipid lamella of the stratum corneum, and Dermatophagoides (D.) farinae crude extract was applied. HT group was orally administered Hwangnyeonhaedok-tang after induction of AD. IL-4 IL-13, $p-I{\kappa}B$, iNOS, Sudan Black B (SB), loricrin, and filaggrin were observed to confirm the effect. Results In HT group, AD skin score was decreased by 46%. The cytokine IL-4 and IL-13, which can identify Th2 differentiation, was reduced by 73% and 58% each. Anti-inflammatory effects were observed in $p-I{\kappa}B$ and iNOS by 69% and 54%, respectively. Finally, SB showed that the regeneration of the lipid layer and the increase of the regeneration power of loricrin and filaggrin were increased by 437% and 464%, respectively. Conclusions From the study result, we observed that Hwangnyeonhaedok-tang treatment alleviates AD by decreasing skin score, reducing Th2 differentiation, inducing anti-inflammatory, and increasing skin lipid barrier regeneration. Thus, Hwangnyeonhaedok-tang treatment would be considered as an effective AD relieving treatment.

The effects of HYT on various immunological factors related to pathogenesis of allergic dermatitis in NC/Nga mice induced by Biostir AD (아토피피부염 동물 병태모델에서의 형개연교탕(荊芥連翹湯)의 면역조절작용)

  • Atopic dermatitis induced NC/Nga mice were used to investigate the efficacy of HYT on the recovery of dermatitic symptoms by how HYT influenced the immune related factors. The results are as below: 1. Compared to the control, HYT treated group showed recovery of atopic dermatitis by the naked eye observation, and significant reduction of dermatits index was observed after 14 weeks. 2. HYT treated group showed significant decrease of the ratio of CCR3+, B220+/IgE+, and Gr-1+/CD11b+ immune cells in dorsal skin by 40.5%, 34.2%, and 48.1%, respectively. 3. HYT treated group showed increase in the ratio of CD19+ immune cells within PBMC by 10.8%, as well as decrease in CD3+, CD3+/CD69+, NKT+ ratios by 5.3%, 35.2%, and 44.9%, respectively. 4. HYT treated group showed increase in the expression of IFN-$\gamma$ in serum by 589.3%, whereas the expression of IL-4, IL-5, IL-6, IL-13, TNF-$\alpha$, MCP-1 and RANTES were decreased by 31.4%, 82.1%, 97.1%, 39.5%, 83.7%, 26.1%, 48.6%, respectively. A 47.2% decrease in IgE expression was also observed. The results above strongly supported the improvement of atopic dermatitis by HYT treatment through immune modulation. Further studies on the synergistic effect of each ingredients of HYT and therapeutic effects according to the dosage of each ingredient should be followed for clinical applications.   • Atopic dermatitis induced NC/Nga mice were used to investigate the efficacy of Hyunggaeyunkyotanggamibang(HYGB) on the recovery of dermatitic symptoms through its influence on the immune related factors. The results are as below: 1. HYGB treated group showed improvement of atopic dermatitis with naked eye observation, and significant decrease of dermatitis index was observed after 14 weeks. 2. HYGB treated group showed significant decrease of the ratio of CCR3+, B220+/IgE+, and CD11b+/Gr-1+ immune cells in dorsal skin by 41.7%, 21.5%, and 23.8%, respectively. 3. HYGB treated group showed an increase of CD19+ immune cells by 10.3% in PBMC, whereas CD3+, CD3+/CD69+, NKT+ immune cells were decreased by 4.3%, 42.9%, and 21.7%, respectively. 4. HYGB treated group showed an increase in the expression of IFN-$\gamma$ in the serum by 514.3%. However, the expressions of IL-4, IL-5, IL-6, IL-13, TNF-$\alpha$, MCP-1 and RANTES were decreased by 21.2%, 69.8%, 90.5%, 28.7%, 72.2%, 26.1%, and 19.9%, respectively. Also, the expression of IgE was decreased by 44.3%. 5. HYGB treated group showed a decrease of the expression of IL-4 and IL-5 by 43% and 44.3%. The results above indicated that HYGB clinically used for atopic dermatitis treatment has objective validity, and therefore can be provided as the basic data for EBM(Evidence based medicine) construction for anti-allergic and anti-inflammatory studies.

Effect of Skin Lipid Barrier Formation on Hataedock Treatment with Douchi

  • Objectives: Hataedock is indigenous treatment of Korean medicine that administers herbal extracts orally to newborn infants for remove the fetal heat. The purpose of this study was to evaluate skin lipid barrier formation effect of Hataedock treatment with Douchi. Methods: We measured the Western blot to observe the expression of protein such as involucrin and loricrin. Moreover, we observed immunohistochemical changes in NC/Nga mice. The 3-week-old NC/Nga mice were divided into 3 groups: the 3-week-old control group (3w-Ctrl), 5-week-old control group (5w-Ctrl), and the Hataedock-treated group (5w-FGT). Only the 5w-FGT group was treated with Douchi at the 3rd week. We identified the changes of the lipid skin barrier and protein differentiation through immunohistochemical changes of involucrin, loricrin, filaggrin and acid sphingomyelinase (ASM) in the stratum corneum. Results: The expression of involucrin and loricrin was increased in the Western blot that was treated with concentration of Douchi extracts. In 5w-FGT group, loricrin-positive reaction was increased by 54.0%, involucrin-positive reaction was increased by 84.0%, filaggrin-positive reaction was increased by 108.0% and ASM-positive reaction was increased by 91.0% in the stratum corneum. Conclusions: These results suggest that Hataedock treatment with Douchi promoted skin lipid barrier formation by promoting differentiation of keratinocytes.

Effects of Ato-tang on DNCB-induced atopic dermatitis in Mice (아토피피부염 동물 병태 모델에서 아토탕의 개선효과)

  • Objectives : The aim of this study is to investigate anti-atopic dermatitis effect using ato-tang.Methods : Ato-tang was external treatment to NC/Nga mice for 4 weeks, where atopic dermatitis was induced by DNCB at 1% and 0.4% for 3 weeks. Atopic dermatitis index score was measured using eye observation and picture evaluation. The histopathological change of dorsal skin was observed by hematoxylin and eosin (H&E) staining. Cytokines including IL-4, IL-5 and IL-13 were measured by Luminex or reverse transcriptase polymerase chain reaction (RT-PCR) analysis and immunoglobulin E (IgE) was measured by ELISA reader.Results : The dorsal skin of Ato-tang group showed decrease in erythema, pruritus, dry skin, edema, excoriation, erosion and lichenification level through naked eye observations. Immunoglobulin cell infiltration and the thickness of epidermis were significantly decreased in the dorsal skin compared to control. Production of Th2 cytokines (IL-4, IL-5, IL-13) and IgE level in serum were all significantly decreased, in comparison with control. In addition, mRNA expression level of Th2 cytokines (IL-4, IL-5, IL-13) in spleen was decreased, in comparison with control.Conclusion : The results indicated that external treatment of ato was improved skin barrier function in the symptoms of atopic dermatitis disease. Also, atopic dermatitis factors where cytokine as well as immunoglobulin E in serum and mRNA expression were decreased, respectively, in comparison with control. Therefore, we suggest that ato could be effectively used as a external therapeutic drug based on atopic dermatitis factors.

Effects of Natural Herb Mixture on Anti-atopic Dermatitis and Skin Regeneration (천연 소재 복합물이 항아토피 피부염 및 피부재생에 미치는 영향)

  • Objectives : This study aims to evaluate the effects of natural herb mixutre (NHM) on atopic dermatitis and skin regeneration using in vivo test. Methods : NHM was prepared with DW. 25% of NHM was applied to skin lesion, where atopic dermatitis was induced by DNCB in NC/Nga mice. The levels of cytokines (IL-4, IL-5, IL-6, IL-13, TNF-a, and $IFN-{\gamma}$), and IgE in serum were measured by Luminex. Immune cells (WBC, eosinophil, lymphocyte, and monocyte) in blood were counted by coulter counter. The gross investigation of atopic dermatitis index score test were performed during the NHM treatment period. Also, the histopathological change of dorsal skin was observed by H&E and M&T staining. Results : NHM showed the levels of IL-4, IL-5, IL-6, IL-13, IgE, WBC, eosinophil, lymphocyte, and monocyte in serum or blood were significantly decreased. On the contrary, the productions of FGF, and VEGF were increased in the serum. Also, atopic dermatitis index score in NHM-treated mice were observed in the similar levels to those of normal group. Histological examination demonstrated that NHM suppressed immune cell infiltration and thickening of epidermis, meanwhile the extraction induced collagen production in the dorsal skin. Conclusion : This study demonstrated that NHM is appeared to be effective on atopic dermatitis and skin regeneration efficacy based on the observations with hematologic, gross, and histologic examinations. Therefore, we suggest that NHM could be effectively used as an external therapeutics against atopic dermatitis and a consequence skin damage.
