A Study on Thermal Diffusivity Measurement by Improvement of Laser Flash Uniformity Using an Optical Fiber (광섬유를 이용한 레이저섬광의 균일분포 증진효과에 따른 열확산계수 측정에 관한 고찰)

  • When thermal diffusivity is measured by laser flash method, the thermal diffusivity call be calculated front the assumption of the uniformly heated whole surface of the specimen. It has been known that the approximate 5% error is made by the non-uniform energy distribution on the specimen surface of laser pulse heat source. In this study, to obtain the highly-uniformed laser beam, which has both the low non-uniform heating error from non-uniform laser beam and the energy loss, research was carried out on no transmitting loss by optical fiber and high repetitions. In addition, heating error and thermal diffusivity were measured as the measuring positions were varied and compared with the results using the uniform and the non-uniform laser beams. In addition, dole to using the uniformalized laser beam, the whole surface of the specimen was heated uniformly and as a result, it was the thought that this was very effective to reduce the variations of the errors of the thermal diffusivity as the measuring positions were varied. It can be obtained that when the thermal diffusivity of POCO-AXM-5Q1 of SRM in NBS was measured with both the uniform and the non-uniform laser beams, the dispersion error of the former was from 2 to 2.5%, which was more improved than that of the latter.

  • Acid protease produced from Aspergillus tubingensis was pruified by ethanol fractionation, dialysis, and DEAE cellulose column chromatography. As a result of purification its specific activity increased to 5.4 times, and percent recovery was 39. The kinetic constants of the enzyme were studied. Km and Vmax was $1.5{\times}10^{-7}M\;and\;0.11{\Delta}O.D/min$ , respectively, when casein was used as substrate. The order of Km value of several proteins is : casein

  • The chemical composition of the volatile constituents from the leaves, stems, and roots of Acanthopanax senticosus growing wild in Mt. Deok-Yu was determined by GC and GC/MS spectrometric analysis with the aid of NBS, Wiley Library and RI indice searches. The major constituents identified were ${\delta}-elemene\;(23.14%),\;{\beta}-elemene\;(5.59%),\;{\gamma}-cadinene\;(5.43%),\;and\;{\alpha}-bergamotene\;(28.54%)$ in the leaves, ${\alpha}-pinene\;(11.50%),\;{\delta}-elemene\;(17.62%),\;{\alpha}-bergamotene\;(27.21%)$, and bicycloheptane derivative (6.38%) in the stems, and ${\alpha}-pinene\;(21.90%)$, and (+)-aromadendrene (3.77%) in the roots.

  • Breast cancer is the most common malignancy in women around the world. About one in 12 women in the West develop breast cancer at some point in life. It is estimated that 5%-10% of all breast cancer cases in women are linked to hereditary susceptibility due to mutations in autosomal dominant genes. The two key players associated with high breast cancer risk are mutations in BRCA 1 and BRCA 2. Another highly important mutation can occur in TP53 resulting in a triple negative breast cancer. However, the great majority of breast cancer cases are not related to a mutated gene of high penetrance, but to genes of low penetrance such as CHEK2, CDH1, NBS1, RAD50, BRIP1 and PALB2, which are frequently mutated in the general population. In this review, we discuss the entire spectrum of mutations which are associated with breast cancer.

The Roles of Tryptophan and Histidine Residues in the Catalytic Activities $\beta$-Cyclodextrin Glucanotransferase from Bacillus firmus var. alkalophilus

  • In order to investigate the critical amino acid residues involved in the catalytic activities of $\beta$-cyclodextrin glucanotransferase ($\beta$-CGTase) excreted by Bacillus firmus var. alkalophilus, the amino acid residues in $\beta$-CGTase were modified by various site-specific amino acid modifying reagents. The cyclizing and amylolytic activities of $\beta$-CGTase were all seriously reduced after treatment with Woodward's reagent K (WRK) modifying aspartic/glutamic acid, N-bromosuccinimde (NBS) modifying tryptophan, and diethylpyrocarbonate (DEPC) modifying histidine residues. The roles of tryptophan and histidine residues in $\beta$-CGTase were further investigated by measuring the protection effect of various substrates during chemical modification, comparing protein mobility in native and affinity polyacrylamide gel electrophoresis containing soluble starch, and comparing the $K_m$ and $V_{max}$ values of native and modified enzymes. Tryptophan residues were identified as affecting substrate-binding ability rather than influencing catalytic activities. On the other hand, histidine residues influenced catalytic ability rather than substrate-binding ability, plus histidine modification had an effect on shifting the optimum pH and pH stability.

Metarizium anisopliae (Metschn.) Sorok이 생산하는 Lactobacillus plantarum 용균효소의 분리, 정제 및 특성

  • To improve the preservation of Kimchi, we isolated Lactobacillus plantarum lytic enzyme-producing strain from soil, and the enzyme was purified and characterized. From the observation of cultural and morpho- logical characteristics, the isolated strain was identified as Metarrisium anisopliae (Metschn.) Sorok. The enzyme was purified to 75-folds with 40% yields through affinity adsorption and CM-Sephadex C-50 column chromatog- raphy. The optimum pH and temperature for lytic activity are 4.0 and 40$\circ$C, respectively, and the enzyme acitvity is stable between pH 3.0 and 9.0, and up to 50$\circ$C. The enzyme is a monomeric protein with molecular weight of 40,000 daltons by SDS-PAGE and gel filtration. The enzyme is endopeptidase which breaks the peptide linkage of Lactobacillus plantarum peptidoglycan. The lytic action spectra confirmed that Leuconostoc mesenteroides, a useful strain for the fermentation of Kimchi, is not lysed by the enzyme. The enzyme activity is inhibited by N-bromosuccinimide (NBS), which probably indicates the involvement of tryptophan residue in active site of the enzyme, and also inhibited by Ag$^{+}$. The amino acid composition analysis showed that the enzyme contains more acidic amino acids than basic ones, and composition of alanine, glycine, proline and tyrosines was very high.

Machine's Determination of Main Color and Imbalance in a Drawing for Art Psychotherapy (그림진단을 위한 주제색 및 불균형 판단의 자동화)

  • Art psychotherapy is widely accepted as an effective tool for diagnosis and treatment of psychological disorders. Important factors for art psychotherapy diagnosis, based on the projection theory that the world of the inner mind appears in drawings, include main color and imbalance of a drawing. This paper develops a system for a machine to determine the main color and the imbalance of a drawing by color recognition and edge detection. Our proposed color recognition procedure adopts NBS(National Bureau of Standards) distance between colors in HVC(Hue, Value, Chroma) color space which is most similar to the human eye's color perception. Our edge detection procedure applies blurring, clustering and transformation to a standard color in a series. Our system considers the numbers of pixels and clusters for each color as a criterion for main color and the frequency of edge coordinates for each region for imbalance. The proposed machine procedure, verified through case studies, can help overcome the subjectivity, ambiguity and uncertainty in human decision involved in art psychotherapy.

  • Uranium radionuclides are electrodeposited on inexpensive stainless steel cathode from a mixed oxalate-chloride electrolyte. The factors affecting the optimum condition for the deposition are determined by studying the effects of deposition time, initial current, electrode spacing, pH of electrolyte and uranium concentration in the electrolyte at fixed cathode area. The experiment which was repeated 3 times at each uranium concentration with 60 minutes of deposition time, gave an error of less than 4% standard deviation at the 90% confidence level with average yield greater than 99%.

  • The changes of amount in flavor components of chewing gum during storage of various relative humidity were studied by GC/MS. The volatile components in chewing gum were extracted by sample preperation and 15 volatile components were identified by NIST/NBS library searching. According to the period of storage, quantities of major volatile components extracted by tetrahydrofuran-methanol solutions were determined by using tetradecane as internal standard. Until 4 weeks, comparing with starting peak area ratio, the decrease of chewing gum flavor components were rapid at 33, 75 than 53 R.H(%). And after 16 weeks, amount of chewing gum flavor at all storage conditions similarly decreased to $45{\sim}49%$ of the initial amount.

  • In this paper, using the bargaining game theory, we propose a bandwidth management scheme that allocates bandwidth in an efficient and proportionally fair manner between the service classes with different service requirements. Since the traffic input rates of the classes are asymmetric in most of the time, the proposed scheme allocates bandwidth in proportion to the traffic input rates to increase the bandwidth utilization while protecting the quality of service of a class against the excessive traffic input of the other classes. In addition, the proposed method considers the weights of classes so that the bandwidth is allocated differentially among the classes.
