Induction of Anticarcinogenic Enzymes by Dichloromethane-soluble Fraction of Physalis alkekengi var. francheti Hort. in Mouse Hepatoma Cells

  • Physalis alkekengi var. francheti Hort. is known as an insecticide and traditional remedy for liver related diseases. Therefore, this study investigated the chemopreventive effects of extracts and several solvent fractions (n-hexane, dichloromethane, n-butanol, water) of Physalis alkekengi var. francheti Hort. First, their cytotoxicity and NQO1 activity were measured using an MTT assay, plus a quinone reductase [NAD(P)H dehydrogenase (quinone); NAD(P)H: (quinone acceptor) oxidoreductase, EC 1.6.99.2]-inducing activity assay was performed using cultured murine hepatoma cells (Hepa1c1c7) and its mutant cells(BpRc1). The reduction of electrophilic quinones by NQO1 is an important detoxification pathway and major mechanism of chemoprevention. When compared with the other solvent soluble fractions with different polarities, the dichloromethane fraction of Physalis alkekengi var. francheti Hort. showed a higher NQO1-inducing activity that was also dose-dependent. Moreover, the dichloromethane fraction of Physalis alkekengi var. francheti Hort. induced ARE-luciferase activities in HepG2-C8 cells that were generated by transfecting the ARE-luciferase gene construct, suggesting the Nrf2-ARE-mediated induction of anti-oxidative enzymes. In conclusion, the dichloromethane-soluble fraction of Physalis alkekengi var. francheti Hort. showed a relatively strong induction of detoxifying enzymes, thereby meriting further study to identify the active components and evaluate their potential as cancer preventive agents.

  • NAD(P)H:quinone oxidoreductase (NQO1), an obligatory two-electron reductase, is a ubiquitous cytosolic enzyme that catalyzes the reduction of quinone substrates. The NQO1- mediated two-electron reduction of quinones can be either chemoprotection/detoxification or a chemotherapeutic response, depending on the target quinones. When toxic quinones are reduced by NQO1, they are conjugated with glutathione or glucuronic acid and excreted from the cells. Based on this protective effect of NQO1, the use of dietary compounds to induce the expression of NQO1 has emerged as a promising strategy for cancer prevention. On the other hand, NQO1-mediated two-electron reduction converts certain quinone compounds (such as mitomycin C, E09, RH1 and β-lapachone) to cytotoxic agents, leading to cell death. It has been known that NQO1 is expressed at high levels in numerous human cancers, including breast, colon, cervix, lung, and pancreas, as compared with normal tissues. This implies that tumors can be preferentially damaged relative to normal tissue by cytotoxic quinone drugs. Importantly, NQO1 has been shown to stabilize many proteins, including p53 and p33ING1b, by inhibiting their proteasomal degradation. This review will summarize the biological roles of NQO1 in cancer, with emphasis on recent findings and the potential of NQO1 as a therapeutic target for the cancer therapy.

  • Induction of NAD(P)H:(quinone-acceptor) oxidoreductase (QR) which promotes obligatory two electron reduction of quinones and prevents their participation in oxidative cycling and thereby the depletion of intracellular glutathione, has been used as a marker for chemopreventive agents. Induction of phase II enzyme is considered to be an important mechanism of cancer prevention. In our previous study, we assessed the quinone reductase QR-inducing activities of 216 kinds of medicinal herb extracts in cultured murine hepatoma cells, BPRc1 and hepalc1c7 cells. Among the 216 herbal extracts tested in that study, extracts from Chrysanthemum zawadskii showed significant induction of QR. In this study, we examined QR-inducing activity of solvent fractions of the herbal extract. The dichloromethane fraction of the herb showed the highest QR induction among the samples fractionated with four kinds of solvents with different polarity. The fraction also significantly induced the activity of glutathione S-transferase (GST), one of the major detoxifying enzymes, at $4{\mu}g/mL\;and\;2{\mu}g/mL$ in hepalc1c7 and BPRc1 cells, respectively. In conclusion, dichloromethane-soluble fraction of Chrysanthemum zawadskii which showed relatively strong induction of detoxifying enzymes merits further study to identify active components and evaluate their potential as cancer preventive agents.

파프리카 추출물이 quinone reductase 유도활성에 미치는 영향 (Induction of Quinone Reductase Activity in Hepatoma Cells by Paprika (Capsicum annuum L.))

  • Phase II 효소계는 자연계에 존재하는 다양한 화학물질과 천연 소재들에 의해 유도되며, 이들의 유도는 화학적 발암물질과 그 밖의 여러 가지 독성물질들로부터 생체를 보호하는데 중요한 역할을 한다. 본 연구에서는 파프리카의 메탄올 추출물을 제조하고, 이를 각 용매별로 분획하여 Hepa1c1c7 세포주를 이용하여 대표적인 암예방 효소인 quinone reductase의 유도 활성을 조사하였다. 그 중 QR 유도활성이 가장 높았던 EtOAc 분획물은 $10-500\;{\mu}g/mL$ 농도로 처리했을 때, $200\;{\mu}g/mL$의 농도에서 QR 활성이 약 3.3배로 가장 높게 나타났으며, 파프리카의 추출물의 총 폴리페놀 및 플라보노이드 함량 역시 EtOAc 분획물에서 가장 높게 나타났다. 또한 $BP^rcl$ 세포주에서도 농도 의존적으로 높은 QR 유도활성을 보여 파프리카의 EtOAc 분획물의 항암성분은 monofunctional inducer인 것으로 생각된다.

Cirsium japonicum var. maackii inhibits hydrogen peroxide-induced oxidative stress in SH-SY5Y cells

  • Over-produced reactive oxygen species (ROS) exert oxidative damage on lipids, proteins, and DNA in the human body, which leads to the onset of neurodegenerative diseases such as Alzheimer's disease (AD). In this study, we explored the cellular antioxidant effect of Cirsium japonicum var. maackii (CJM) against hydrogen peroxide (H2O2)-induced oxidative stress in neuronal cells. The antioxidant activity was assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, 2',7'-dichlorofluorescin diacetate and nitric oxide (NO) assays, and the molecular mechanisms were examined by Western blot analysis. H2O2 treatment of SH-SY5Y cells decreased cell viability and increased ROS and NO production compared to H2O2-untreated cells. However, CJM increased cell viability and decreased ROS and NO accumulation in the H2O2-treated SH-SY5Y cells compared to H2O2-treated control cells. Especially, the EtOAc fraction from CJM showed the strongest antioxidant effect compared with the other extracts and fractions. Therefore, we further examined the CJM mechanism against oxidative stress using the EtOAc fraction from CJM. The EtOAc fraction up-regulated the expressions of heme oxygenase-1, NAD(P)H quinone oxidoreductase 1, and thioredoxin reductase 1. These results indicate that CJM promotes the activation of antioxidative enzymes, which eliminate ROS and NO, and further leads to an increase in the cell viability. Taken together, our results show that CJM exhibited an antioxidant activity in H2O2-treated SH-SY5Y cells, and it could be a novel antioxidant agent for the prevention or treatment of neurodegenerative disease such as AD.

Antioxidant mechanism of black garlic extract involving nuclear factor erythroid 2-like factor 2 pathway

  • BACKGROUN/OBJECTIVES: Although studies have revealed that black garlic is a potent antioxidant, its antioxidant mechanism remains unclear. The objective of this study was to determine black garlic's antioxidant activities and possible antioxidant mechanisms related to nuclear factor erythroid 2-like factor 2 (Nrf2)-Keap1 complex. METHODS/MATERIALS: After four weeks of feeding rats with a normal fat diet (NF), a high-fat diet (HF), a high-fat diet with 0.5% black garlic extract (HF+BGE 0.5), a high-fat diet with 1.0% black garlic extract (HF+BGE 1.0), or a high-fat diet with 1.5% black garlic extract (HF+BGE 1.5), plasma concentrations of glucose, insulin,homeostatic model assessment of insulin resistance (HOMA-IR) were determined. As oxidative stress indices, plasma concentrations of thiobarbituric acid reactive substances (TBARS) and 8-isoprostaglandin $F2{\alpha}$ (8-iso-PGF) were determined. To measure antioxidant capacities, plasma total antioxidant capacity (TAC) and activities of antioxidant enzymes in plasma and liver were determined. The mRNA expression levels of antioxidant related proteins such as Nrf2, NAD(P)H: quinone-oxidoreductase-1 (NQO1), heme oxygenase-1 (HO-1), glutathione reductase (GR), and glutathione S-transferase alpha 2 (GSTA2) were examined. RESULTS: Plasma glucose level, plasma insulin level, and HOMA-IR in black garlic supplemented groups were significantly (P < 0.05) lower than those in the HF group without dose-dependent effect. Plasma TBARS concentration and TAC in the HF+BGE 1.5 group were significantly decreased compared to those of the HF group. The activities of catalase and glutathione peroxidase were significantly (P < 0.05) increased in the HF+BGE 1.0 and HF+BGE 1.5 groups compared to those of the HF group. The mRNA expression levels of hepatic Nrf2, NQO1, HO-1, and GSTA2 were significantly (P < 0.05) increased in the HF with BGE groups compared to those in the HF group. CONCLUSIONS: The improvements of blood glucose homeostasis and antioxidant systems in rats fed with black garlic extract were related to mRNA expression levels of Nrf2 related genes.

들송이(Agrocybe cylindracea)와 말똥진흙버섯(Phellinus igniarius)을 배양한 대두추출물의 암예방 효과 (Chemopreventive Effects of the Extracts from Soybean Fermented with Basidiomycetes)

  • 본 연구에서는 버들송이(Agrocybe cylindracea)와 말똥진흙버섯(Phellinus igniarius)을 배양한 대두 추출물의 암예방 효과를 검토하기 위하여 in vitro상에서 암발생 억제물질 연구에 널리 이용되고 있는 기작 즉, Quinone reductase(QR), glutathione S-transerase(GST)의 활성 유도, glutathione(GSH)의 생성 촉진 그리고, polyamine 합성대사의 억제능을 측정하였다. 그 결과, 버들송이와 말똥진흙버섯를 배양한 대두 추출물이 마우스 hepatoma Hepa1c1c7 cell 배양에서 QR과 GST 유도활성이 있음이 확인되었다. 또한 GSH 생성량은 버들송이를 배양한 대두 추출물에서 20%, 말똥진흙버섯를 배양한 대두 추출물에서 40%로 증가하였다. 한편, 버들송이와 말똥진흙버섯을 배양한 대두 추출물의 polyamine 합성대사의 억제효과를 검토하기 위해 Acanthamoeba castellanii의 증식을 측정하였다. 그 결과, 버들송이와 말똥진흙버섯을 배양한 대두 추출물 20mg/3ml와 40mg/3ml를 처리할 경우 A. castellanii의 증식을 억제하는 것으로 나타났다. 이러한 결과 담자균을 배양한 대두 추출물은 in vitro상에서 나타난 암과 관련된 각종 활성으로부터 암예방 효과가 있는 것으로 나타나 기능성 식품으로서의 가능성을 보여주었다.

Protection by Chrysanthemum zawadskii extract from liver damage of mice caused by carbon tetrachloride is maybe mediated by modulation of QR activity

  • Our previous study demonstrated that methanolic extract of Chrysanthemum zawadskii Herbich var. latilobum Kitamura (Compositae) has the potential to induce detoxifying enzymes such as NAD(P)H:(quinone acceptor) oxidoreductase 1 (EC 1.6.99.2) (NQO1, QR) and glutathione S-transferase (GST). In this study we further fractionated methanolic extract of Chrysanthemum zawadskii and investigated the detoxifying enzyme-inducing potential of each fraction. The fraction (CZ-6) shown the highest QR-inducing activity was found to contain (+)-(3S,4S,5R,8S)-(E)-8-acetoxy-4-hydroxy-3-isovaleroyloxy-2-(hexa-2,4-diynyliden)-1,6-dioxaspiro [4,5] decane and increased QR enzyme activity in a dose-dependent manner. Furthermore, CZ-6 fraction caused a dose-dependent enhancement of luciferase activity in HepG2-C8 cells generated by stably transfecting antioxidant response element-luciferase gene construct, suggesting that it induces antioxidant/detoxifying enzymes through antioxidant response element (ARE)-mediated transcriptional activation of the relevant genes. Although CZ-6 fraction failed to induce hepatic QR in mice over the control, it restored QR activity suppressed by $CCl_4$ treatment to the control level. Hepatic injury induced by $CCl_4$ was also slightly protected by pretreatment with CZ-6. In conclusion, although CZ-6 fractionated from methanolic extract of Chrysanthemum zawadskii did not cause a significant QR induction in mice organs such as liver, kidney, and stomach, it showed protective effect from liver damage caused by $CCl_4$.
