Changes of Chloroplast Number per Guard Cell pairs of Leaves by Ploidy Level in Nicotiana tabacum L. cv. BY-4 (담배 식물체[Nicotiana tabacum L. cv. BY-4]의 배수성에 따른 공변세포의 엽록체 수 변화)

  • We evaluated a possibility of the use of chloroplast number per guard cell pairs as a measure for ploidy level in the different ploidy levels of tobacco plant (Nicotiana tabacum L. cv. BY-4) . The guard-cell chloroplast numbers of leaves of haploid plant were a half of wild-type plant. Furthermore, the number of chloroplast per guard cell pairs of the leaves of doubled-haploid plant increased in two times compared with that of haploid plant. In addition, the chloroplast number was not changed in the F$_1$ progenies. The change of the chloroplast number by leaf stage was not observed. The results indicate that there is a strong relationship between ploidy level (2x and 4x) and chloroplast number per guard cell pairs. This relationship was also, observed in both in vitro and pot cultured plants. It was determined that the measurement of chloroplast number in guard cells of leaf epidermis is simple to use and less labour intensive, and hence can be considered a practical alternative to the chromosome counting methods or flow cytometry in the tobacco plant.

Pathogenicity of a Korean isolate of Pepper mild mottle virus and development of full-length cDNA clone for infectious in vitro transcripts

  • A Korean isolate of Pepper mild mottle virus (PMMoV-Kr) was isolated from a diseased pepper crop in Chunchon, Korea. The isolate was biologically purified on Nicoticaa tabacum cv. Xanthi-nc by successive single local transfer steps, and propagated on N. tabacum cv. Samsun. PMMoV-Kr could systemically infect on N. glauca, N. benthmiana, N. occidentalis and Lycopersicon esculentum, which is typical of known isolates of PMMoV. PMMoV-Kr belongs to the pathotype P1,2 based on pepper-tobamoviral indicator experiments; Capsicn chinone harboring L3 gene revealed resistant (necrotic local lesion on inoculated leaf, HR) whereas L+, L1 and L2 pepper plants expressed susceptible reactions of mosaic systemic symptoms for the isolate. To confirm the pathology and delineate symptom determinant of the isolate, full-length cDNAs of PMMoV-Kr were amplified by RT-PCR with a primer set corresponding to the 5'- and 3'-ends of PMMoV. The RT-PCR molecules amplified from genome RNA of the isolate was cloned into the pUC18 vector. Full-length cDNA clones constructed under the control of the T7 RNA promoter could be successfully transcribed to produce in vitro transcript RNA. Infectivity of the capped transcripts and its progeny virus was verified by Western blot and RT-PCR analyses.

Degree of Heterosis in Diallel Crosses of Burley and Chinese Sun-cured Tobacco (Nicotiana tabacum L.) (버어리종 연초와 중국 양건종의 이면교잡에 의한 양적형질의 잡종강세정도)

  • This study was conducted to obtain the basic information for breeding of burley(N. tabacum cv. burley) and sun-cured tobacco(N. tabacum cv. sun-cured) variety. Two burley(Burley 21, KB 108), and T.I. 1068 and three sun-cured varieties(Yeonbyeon 3, Jaheungyeon and Jaraehong) were diallel crossed and 15 combinations of F$_1$generations were made in 1998, respectively. Six parental varieties, 15 F$_1$s were grown under the complete randomized block design with 3 replications at Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute in 2000. The data of 12 quantitative characters were measured for degree of heterosis. The results obtained are as follows. Heterosis of F$_1$generation was positive in weight per leaf, plant height, stem diameter, yield, leaf length and total alkaloid content. Cross combinations between either Burley 21 or KB 108 and sun-cured varieties showed comparatively high heterosis in each characters.

  • Plantlets derived from leaf blade-segment culture of a variegated tobacco (Nicotiana tabacum L. cv. BY-4) that was induced by a heavy-ion ($^{14}N$) beam irradiation to proembryos, were characterized. When explants from both white and green sections of leaves of the variegated plant were cultured on MS medium containing 0.1 mg/L NAA and 1.0 mg/L BAP, the white sections yielded only white shoots, whereas the green sections generated approximately 47.2% green, 37.4% white and 15.4% variegated shoots. In the F1 generation of a green tobacco derived from the leaf blade-segment culture, the segregation ratio of green to white was 1,651:54. Furthermore, reciprocal crosses showed that all of the progenies was green, indicating that the variegation is not maternally inherited. When the signal intensity of photosynthesis genes was determined by DNA gel blot analysis using the variegated leaves derived from green sections of variegated leaves, there were more of the rbcL, psbA, 16S rDNA and 23S rDNA chloroplast genes in the white sections than the chloroplast genes in wild type and green sections of the variegated plants.

  • This study was conducted to investigate the utility and agronomic characteristics and use of cytoplasmic. male-sterile (CMS) haploid plants derived from interspecific cross between (male-sterile NC82$\times$burley21) F$_1$ plant (Nicotiana tabacum L.) and Nicotiana africana. Abundant seeds of high germinability were obtained when Nicotiana tabacum (cytoplasmic male-sterile F$_1$ plants) is pollinated by Nicotiana africana. Most of seedlings died at the cotyledonary stage. The remaining seedlings are viable F$_1$ hybrids or maternal haploids that can be easily distinguished. Number of interspecific Fl hybrids and matermal haploids per capsule obtained from the interspecific cross between cytoplasmic male-sterile tobacco F$_1$ plants and N.africana yielded 2.2 and 0.5 plants, respectively. Out of 149 CMS haploid plants obtained from interspecific cross, 102 plants showed green type while the others were yellow type for leaf and stem. This results agreed with the genetic ratios expected among haploid plants from the F$_1$ hybrids heterozygous for two recessive genes of yellow color of burley tobacco plant. Out of 83 CMS haploid plants inoculated with TMV, 48 plants showed resistant, while the others was susceptible. It agreed with the expected genetic ratios for a single dominant gene of TMV resistance. CMS haploid plant will be useful as a source material for breeding of CMS doubled haploid lines

Fertilizer Placement Effect on Agronomic Characteristics of Burley Tobacco(N. tabacum L.) (시비방법이 버어리종 잎담배의 농경적 특성에 미치는 영향)

  • Recently the method of basal compound fertilizer (N : P : K = 18.2 : 9.8 : 35.0) Placement has been changed from banding to broadcasting in tobacco cultivation. The effects of the compound fertilizer placement, level of fertilizer and additional urea application on the agronomic characteristics, two chemical compositions and physical Properties of burley tobacco(Nicotiana tabacum L. cv. KB 108) were investigated at Chonju Experiment Station, Korea Ginseng and Tobacco Research Institute in 1995-1996. Six treatments consisted of (1) band (method of fertilizer Placement) + In kg/10a(compound fertilizer) + 0(additional urea applied), (2) broadcast+140+0, (3) broadcast+140+25, (4) band+180+0, (5) broadcast+180+0, and (6) broadcast +180+25. The additional urea was applied at hilling. No significant differences were detected between banding and broadcasting method of compound fertilizer Placement in field 9rowth, wield, organoleptic qualify(price Per kilogram), chemical composition and Physical properties of cured leaf, The increased compound fertilizer by 30%(40k/10a) or the additional urea application by 25kg per 10a produced slightly higher yield than the recommended amount of basal compound fertilizer without additional urea application did. It also increased the total alkaloid content of cured leaf. It is recommended that no more basal fertilizer above the recommended amount and no additional urea application are needed in burley tobacco fertilization, even though the method of basal fertilizer placement being chanced from banding to broadcasting.   • The field evaluation were conducted on dihaploid progenies derived from anther culture of F1 plant of Drama x Sohyang (N. tabacum L.) The mean values of agronomic and chemical traits of dihaploids such as Nicotine, length/width, plant height, Leaf length, leaves per plant and Yield were greater than those of tar parents. The correlation coefficient of dihaploids were similar to those of conventional lines and varieties. In path analysis, in relating yield components to yield, leaves per plant and leaf width had a direct effect on Yield, but the other components such as plant height, leaf length and days to flowering influenced Yield indirect way.

  • In order to obtain the basic information for the successful breeding of aromatic tobacco (N. tabacum L.), the combining ability and the mode of inheritance in 28 $F_2$ hybrids by diallel crosses between Korea local and oriental varieties were investigated. Estimates of general combining ability were significant for all characters investigated, and the specific combining ability was significant for stalk height, leaves per plant, leaf length, leaf width and days to flower. The partial dominance was exhibited by all characters investigated as well as additive effects were larger than dominance effects for all characters.

  • Transgenic tobacco plants were selected by using the transformation of putrescine N-methyltransferase(PMT) gene, the key enzyme in diverting polyamine metabolism towards the biosynthesis of nicotine. PMT was fused in reverse orientation to the CaMV 35S promoter of the plant expression vector pBTEX(pPAB3) to produce tobacco plants of low nicotine content. To compare nicotine content, only pBTEX vector and PMT gene which was fused in forward orientation to the CaMV 35S promoter(pPAB2) were also transformed to the leaf tobacco plants(Nicotiana tabacum cv. NC82 and N. tabacum cv. Br2l). The presence of sense- and antisense-PMT gene, and pBTEX vector in the transgenic plant was confirmed by genomic PCR.
