Role of N-terminal Hydrophilic Amino Acids in Molecular Translocation of CTLA-4 to Cell Surface (CTLA-4 항원의 세포막 도달 기작에서 친수성 N말단 아미노산 잔기의 역할)

  • Background: This study was aimed to differentiate two forms of CTLA-4 (CD152) in activated peripheral blood lymphocyte and clarify the mechanism how cytoplasmic form of this molecule is targeted to cell surface. Methods: For this purpose we generated 2 different anti-human CD152 peptide antibodies and 5 different N'-terminal deletion mutant CTLA4Ig fusion proteins and carried out a series of Western blot and ELISA analyses. Antipeptide antibodies made in this study were anti-CTLA4pB and anti-CTLA4pN. The former recognized a region on extracellular single V-like domain and the latter recognized N'-terminal sequence of leader domain of human CD152. Results: In Western blot, the former antibody recognized recombinant human CTLA4Ig fusion protein as an antigen. And this recognition was completely blocked by preincubating antipeptide antibody with the peptide used for the antibody generation at the peptide concentration of 200 ug/ml. These antibodies were recognized human CD152 as a cytoplasmic sequestered- and a membrane bound- forms in phytohemagglutinin (PHA)-stimulated peripheral blood lymphocyte (PBL). These two forms of CD152 were further differentiated by using anti-CTLA4pN and anti-CTLA4pB antibodies such that former recognized cytosolic form only while latter recognized both cytoplasmic- and membraneforms of this molecule. Furthermore, in a transfection expression study of 5 different N'-terminal deletion mutant CTLA4Ig, mutated proteins were secreted out from transfected cell surface only when more than 6 amino acids from N'-terminal were deleted. Conclusion: Our results implies that cytosolic form of CTLA-4 has leader sequence while membrane form of this molecule does not. And also suggested is that at least N'-terminal 6 amino acid residues of human CTLA-4 are required for regulation of targeting this molecule from cytosolic- to membrane- area of activated human peripheral blood T lymphocyte.

IN VITRO MICRO-SHEAR BOND STRENGTH OF FIVE COMPOSITE RESINS TO DENTIN WITH FIVE DIFFERENT DENTIN ADHESIVES (미세-전단 결합 강도 시험을 이용한 상아질 접착제와 수복용 복합 레진의 호환성에 관한 연구)

  • The purpose of this study was to compare and to evaluate the combination use of 5 kinds of dentin adhesive systems and 5 kinds of composite resins using micro-shear bond test. Five adhesive systems (Prime & Bond NT (PBN). Onecoat bond (OC), Excite (EX), Syntac (SY), Clearfil SE bond (CS)) and five composite resins (Spectrum (SP), Synergy Compact (SC), Tetric Ceram (TC), Clearfil AP-X (CA), Z100 (Z1)) were used for this study ($5{\;}{\times}{\;}5{\;}={\;}25group$, n =14/group). The slices of horizontally sectioned human tooth were bonded with each bonding system and each composite resin, and tested by a micro-shear bond strength test. These results were analyzed statistically. The mean micro-shear bond strength of dentin adhesive systems were in order of CS (22.642 MPa), SY (18.368 MPa), EX (14.599 MPa). OC (13.702 MPa). PBN (12.762 MPa). The mean bond strength of self-etching primer system group (CS, SY) in dentin was higher than that of self-priming adhesive system groups (PBN, EX, OC) significantly (P<0.05). The mean bond strength of composite resins was in order of SP (19.008 MPa), CA (17.532 MPa). SC (15.787 MPa), TC (15.068 MPa). Z1 (14.678 MPa). Micro-shear bond strength of SP was stronger than those of other composite resins significantly (P < 0.05). And those of TC and Z1 were weaker than other composite resins significantly (P < 0.05). No difference was found in micro-shear bond strength of composite resin in self-etching primer adhesive system groups (CS, SY) statistically. However, there was significant difference of micro-shear bond strength of composite resin groups in self-priming adhesive systems group (PBN, EX, OC). The combination of composite resin and dentin adhesive system recommended by manufacturer did not represent positive correlation. It didn't seem to be a significant factor.

  • Shin Hae-Chul's music opened a new horizon in the Korean pop music market where only popular music is produced and the music of a similar genre or pattern is mainstream, which has dissolved the complaints of the public who longed for new music. He experimented with his musical potential every time he released his work, which received favorable reviews in that he raised the level of Korean pop music. To solve the unstable factors in today's pop music, it is important to appeal to the public by capturing quality work through continued studies and creative activities. In that context, the attitude of "Next" is led by the late Shin Hae-Chul, and his qualities as an artist greatly contributed to the development of Korean pop music. This indicates that if musicians strive to develop themselves, and strengthen their creative ability to make bold and new attempts, Korean pop music will lead to another fashionable "K-pop" and will open up a new horizon.

ANALYSIS OF HIGH-FIELD MAGNETIZATION PROCESS IN $Sm_{2}Fe_{17}N_{3.0}$

  • The observed high-field magnetization curves of $Sm_{2}Fe_{17}N_{3.0}$ at 4.2 K and 296 K are well reproduced by the calculation using the Sm-Fe exchange field $2\mu\textrm{B}H_{ex}\;=\;320\;K$ and two crystalline electric field parameters ${A_{0}}^{2}=\;-910\;K$ and ${A_{1}}^{0}=\;200\;K$. The calculation shows that during the magnetization process along the hard axis at 4.2 K, the Sm moment rotates toward the direction antiparallel to H when H < 110 kOe and then returns to the field direction with further increase of the field. At 296 K, the Sm moment rotates toward the direction antiparallel to H monotonously with increasing field and finally becomes antiparallel to H when $H{\geq}H_{A}=210\;kOe$. The particular magnetization process of the Sm moment can be explained by the field-induced noncollinear coupling between the spin and orbital moments of the Sm ion.

Thermal Resistance and daf-16 Regulation of Fermented Zizyphus jujuba Fruits in Caenorhabditis elegans

  • The mechanism of anti-aging of fermented jujube (Zizyphus jujuba fruits (ZJF)) was investigated using transgenic daf-16 and mev-1 strains of C. elegans. Jujube extracts fermented for 7 days (F7-ZJF) and 14 days (F14-ZJF) with Laetiporus sulphureus were treated to a NGM agar plate with 10-15 transgenic daf-16 and mev-1 strains of the synchronized age. There was no difference of lifespan between the drug-treated group (7-day fermented ex. (F7-zjf-200 ㎍/mL), 14-day fermented ex. (F14-zjf-200 ㎍/mL)) and the non-treatment group in both daf-16 and mev-1 strains. In the thermal stress experiment, F7-zjf-200 ㎍/mL showed a significant (t = 4.017) activity in thermal stress resistance with a 12% higher survival rate than the control group. In the survival test in H2O2, F7-zjf-200 ㎍/mL and F14-zjf-100 ㎍/mL have significant activity in oxidative stress resistance compared to the control group. This study indicates that life span expand of N2 strain of the jujube extract is related to the regulation of daf-16 and inhibition of mev-1 signal in C. elegans.

Compare Physicochemical Properties of Topsoil from Forest Ecosystems Damage patterns (산림생태계 훼손 유형별 표토의 이화학적 특성 비교)

  • This study was carried out to evaluate the physicochemical properties of different types of topsoil in forest ecosystems by damage pattern and analyse the possibility of using the topsoil as a planting ground construction material. There were 72 samples from 36 sites of 12 damaged areas and 36 sites of 12 non-damaged areas. The results showed that the physicochemical properties of topsoil from non-damaged areas of forest ecosystems were on an average clay loam~sandy loam in soil texture, showing $0.95{\sim}1.10Mg/m^3$ in soil bulk density, $35.7{\sim}44.0m^3/m^3$ in solid phase, 56.0~64.3 in soil porosity, 8.4~35.8% in aggregate stability, 5~13 mm in soil hardness, 5.3~6.1 in pH, 0.14~0.65 dS/m in EC, 0.28~0.42% in T-N, $14{\sim}22cmol^+/kg$ in CEC, $0.15{\sim}0.31cmol^+/kg$ in Ex. $K^+$, $2.07{\sim}2.84cmol^+/kg$ in Ex. $Ca^{2+}$, $0.45{\sim}1.97cmol^+/kg$ in Ex. $Mg^{2+}$, 17~96 mg/kg in Av. $P_2O_5$ and 3.2~5.6% in OM. On the other hand, damaged areas were on an average clay loam~loamy sand in soil texture, showing $1.54{\sim}1.75Mg/m^3$ in soil bulk density, $52.8{\sim}58.0m^3/m^3$ in solid phase, 42.0~47.2 in soil porosity, 4.2~22.5% in aggregate stability, 13~25 mm in soil hardness, 4.8~5.5 in pH, 0.13~0.62 dS/m in EC, 0.02~0.12% in T-N, $5{\sim}15cmol^+/kg$ in CEC, $0.11{\sim}0.18cmol^+/kg$ in Ex. $K^+$, $0.45{\sim}2.36cmol^+/kg$ in Ex. $Ca^{2+}$, $0.39{\sim}0.96cmol^+/kg$ in Ex. $Mg^{2+}$, 15~257 mg/kg in Av. $P_2O_5$ and 0.4~2.2% in OM. After conducting a comparison of physicochemical characteristics of non-damaged forest area and damaged areas, it was found that the physicochemical characteristics of damaged areas were more deteriorated compared to that of non-damaged areas. Therefore, it is judged that it is necessary to establish countermeasures for the conservation and management of the damaged areas for topsoil recycling in the future.

HIGH RESOLUTION $HC_3N$ OBSERVATIONS TOWARD THE CENTRAL REGION OF SAGITTARIUS B2

  • We have observed the emission of $HC_3N$ J=4-3, 5-4,10-9 and 12-11 transitions toward the Sgr B2 central region in an area of $150'\times150'$ with resolutions of 16'-48'. The intensities and central velocities of line profiles show significant variations with positions. In contrast to the intensities of the low J-level transitions which gradually increase from the central source toward the outside region, the $HC_3N$ emission of the high J-level transition become stronger toward 'the central radio continuum source MD5. Systematic change in the radial velocity of each line profile occurs along north-south direction. There are a few peaks in most line profiles, and these indicate that there are multiple velocity components along the line of sight. Distributions of excitation temperature and column density which were estimated from the excitation calculations show the existence of a small $(1\times2pc),\;hot\;(T_{ex}>50K)$ core which contains two temperature peaks at-15' east and north of MD5. The column density of $HC_3N\;is\;(1-3)\times10^{14}cm^{-2}$ Column density at distant position from MD5 is larger than that in the central region. We have deduced that this 'hot-core' has a mass of 105M 0, which is about an order of magnitude larger than those obtained by previous studies.

The Cut Off Values for Diagnosing Hot flashes by Using Digital Infrared Thermographic Imaging (적외선 체열 촬영을 이용한 안면홍조 진단의 절단값 산정)

  • Objectives: The purpose of this study is to find diagnostic points and define the cut off values of hot flashes by using digital infrared thermographic imaging. Methods: Thermographic images of 75 patients with hot flashes (HF, n=35) and non-hot flashes (NHF, n=40) were retrospectively reviewed. We used the temperature difference between Ex-HN3 and CV17, LU4, CV12, CV4 for diagnosing hot flashes. The temperature differences of between two groups were analysed using independent samples t-tests. The cut off values were calculated by received operating characteristic curve analysis. Analyses were undertaken using SPSS version 17.0. and p-value of <0.05 was considered significant. Results: The temperature difference Ex-HN3 and LU4 were the most significantly different between groups (p<0.001). Using receiver operating characteristic curve analysis, the sensitivity, specificity, and area under the curve were 65.7%, 72.5%, 0.729, respectively. The optimum cut off value was defined as $1.00^{\circ}C$. Conclusions: These results suggest that the digital infrared thermographic imaging is a reliable instrument for estimating hot flashes.
