Inhibitory effects of advanced glycation end products formation and free radical scavenging activity of Cirsium setidens (곤드레 추출물의 최종당화산물의 생성저해 및 라디칼소거 활성)

  • Naturally occurring antioxidants, such as polyphenols are widely found in fruits, vegetables, wines, juices, and other plant-based dietary sources and are divided into several sub classes, including phenylpropanoids, flavonoids, stilbenoids, and lignans. As part of the our ongoing search for bioactive food ingredients, the antioxidant and advanced glycation end products (AGEs) formation inhibitory activities of the methanolic extract of the aerial parts of Cirsium setidens were investigated in vitro bioassay system. The antioxidant properties were evaluated through radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radicals. In addition, the activity of C. setidens against diabetes complications was also tested via AGEs formation inhibition assay. The total phenolic contents were determined using a UV-VIS spectrophotometric method. All tested samples showed a dose-dependent radical scavenging and AGEs inhibitory activities. In particular, the n-butanol (BuOH)-soluble portion showed the most potent radical scavenging activities against DPPH and $ABTS^+$ radicals with $IC_{50}$ values of $24.3{\pm}1.7$ and $25.0{\pm}3.3{\mu}g/mL$, respectively. Futhermore, the inhibition of AGEs formation by the n-BuOH-soluble portion ($IC_{50}$ value; $46.0{\pm}1.5{\mu}g/mL$) was higher than that those of the soluble portions for the other solvent. The results showed that C. setidens could be considered as an effective source of natural antioxidants and other ingredients.

Biological Activity of Sorghum bicolor M. cv. Bulgeunjangmoksusu Extracts (붉은장목수수 추출물의 생리활성)

  • The objective of this study was to determine the biological activities of Sorglum bicolor extracts. Organic fractions, including n-Hexane, EtOAc, and n-BuOH fractions were obtained from the methanol extract of Sorglum bicolor M.. In DPPH radical scavenging activity, $SC_{50}$ values of methanol extract and EtOAc fraction were exhibited $0.66{\pm}0.26{\mu}g/mL$ and $1.03{\pm}0.02{\mu}g/mL$, respectively. Contents of total polyphenol and flavonoids in EtOAc fraction, which were much higher than those of other fractions, were 58.12 mg/g and 4.79 mg/g respectively. Also, effects of reducing power was strongly showed in EtOAc fraction. ${\alpha}$-Glucosidase and ${\alpha}$-amlyase inhibition activities were showed the higher effect in D.W. fraction ($2.83{\mu}g/mL$, $36.64{\mu}g/mL$). In MTT assay in the AGS, HT29 and HCT116 cell lines were significantly higher in the n-BuOH fraction than in the other fractions at $50{\mu}g/mL$ concentration of extracts.

  • In this study, the effects of dibenzocyclooctadiene on seed germination were investigated in pepper. Four $C_{18}$ dibenzocyclooctadiene lignans - schisandrin (1143.7 mg), schisandrin C (317.3 mg), gomisin A (261.4 mg) and gomisin N (213.4 mg) - were isolated from hexane extracts of the fruits of Schisandra chinensis. The molecular structures of the four lignans were elucidated based on spectroscopic analyses including 1D NMR experiments, and bi comparing their spectroscopic data with those of previous literatures. Seeds were immersed in $10^{-5}\;M$ schisandrin, $10^{-6}\;M$ schisandrin C and $10^{-7}\;M$ gomisin A and gomisin N for 1 hr and incubated at $25^{\circ}C$ in the dark for germination. Compared to untreated control, treatment with schisandrin C and gomisin A suppressed seed germination at 48 hrs after incubation, whereas treatment with gomisin N increased germination rate at 48 hrs after incubation. The results of the germination activity of the lignans from S. chinensis confirm their potential plant growth role, and the relative natural abundances of these metabolites suggest their potential use as natural plant growth regulators.

Detection of Hydrocarbons Induced by Electron Beam Irradiation of Almond (Prunus amygosalus L.) and Peanut (Arachis hypogaea) (전자선 조사한 아몬드(Prunus amygosalus L.)와 땅콩(Arachis hypogaea)에서 유래한 지방분해산물 분석)

  • Food irradiation has recently become one of the most successful techniques to preserve food with increased shelf life. This study aims to analyze hydrocarbons in almonds (Prunus amygosalus L.) and peanuts (Arachis hypogaea) induced by electron beam irradiation. The samples were irradiated at 0, 1, 3, 5 and 10 kGy by e-beam and using florisil column chromatography fat, and content was extracted. The induced hydrocarbons were identified using gas chromatography-mass spectrometry (GC/MS). The major hydrocarbons in both irradiated samples were 1,7-hexadecadiene ($C_{16:2}$) and 8-heptadecene ($C_{17:1}$) from oleic acid, 1,7,10-hexadecatriene ($C_{16:3}$) and 6,9-heptadecadiene ($C_{17:2}$) from linoleic acid and 1-tetradecene ($C_{14:1}$) and pentadecane ($C_{15:0}$) from palmitic acid. Concentrations of the hydrocarbons produced by e-beam were found to be depended upon the composition of fatty acid in both almonds and peanuts. The $C_{n-2}$ compound was found to be higher than $C_{n-1}$ compound in oleic acid and palmitic acid, while in case of linoleic acid, $C_{n-1}$compound was higher than $C_{n-2}$ compound. The radiation induced hydrocarbons were detected only in irradiated samples, with 1 kGy or above, and not in the non-irradiated ones. The production of 1,7-hexadecadiene ($C_{16:2}$), 8-heptadecene ($C_{17:1}$), 1,7,10-hexadecatriene ($C_{16:3}$) and 6,9-heptadecadiene ($C_{17:2}$), in high concentration gave enough information to suggest that these may be the possible marker compounds of electron beam irradiation in almonds and peanuts.

Inhibitory Effects of Aralia cordata Thunb Extracts on Nitric Oxide Synthesis in RAW 264.7 Macrophage Cells (독활(Aralia cordata Thunb) 추출물의 Nitric Oxide Synthesis 저해효과)

  • Assessment was made of the effects of Aralia cordata Thunb (DH) on the cell proliferation, inducible nitric oxide synthase (iNOS) mRNA gene expression and nitric oxide (NO) production in RAW 264.7 macrophage cells. For the screening of anti-inflammatory activities, ethanolic extracts of 55 species of traditional herbal medicines were examined for inhibitory effects, and it was confirmed that DH possessed inhibitory effects on NO production. As a result, DH significantly decreased the production of NO and iNOS gene expression at a concentration of $250{\mu}g/mL$. The chloroformsoluble fractionates have the strongest No synthesis inhibitory effect. It is presumed that the inhibition of NO production in LPS-stimulated RAW 264.7 cells by DH components occurred via the modulation of iNOS and DH, and that the active compound from DH may be useful for therapeutic management of inflammatory-associate diseases.

Insecticidal activity of the crude extract and its fractions of Custard apple (Annona reticulata L.) (커스타드애플(Annona reticulata L.)씨 추출물과 그 분획물의 살충활성)

  • In recent years, many agricultural scientists are studying on eco-friendly farming methods. Among of lots of the methods, the natural insecticides are highly motivated and interested due to their safety and biodegradable issues, and readily available source of bioinsecticides. In this study, the crude extract of custard apple (Annona reticulata L.) seed and its three fractions which were separated based on polarity indexes were examined for their insecticidal activities against Myzus persicae Sulzer and Nilaparvata lugens S.. The crude extract (Aceton/MeOH) showed strong insecticidal activities against both insects at 3.00 mg/mL ($LD_{50}$=0.45mg/mL for M. persicae S. and 1.42 mg/mL for N. lugens S.). Furthermore, simple fractionation with hexane, chloroform, and water lead to increase three-folds insecticidal activity on chloroform fraction ($LD_{50}$=0.13mg/mL for M. persicae S. and 1.14 mg/mL for N. lugens S.). The results suggest that A. reticulata extracts might be used to control for M. persicae effectively.

Antioxidant Activities of Cryptomeria japonica Leaves Extracts by Extraction Methods (추출 방법에 따른 삼나무 잎 추출물의 항산화 활성 평가)

  • This study was to investigate the antioxidant activities of Cryptomeria japonica leaves extracts such as essential oil, methanol extract and hot water extract and to evaluate its potential as a natural antioxidant. Antioxidant activities of extracts were evaluated by DPPH radical scavenging activity, FRAP activity, zanthin oxidase inhibitory activity, and iron ion chelate activity, comparing with the positive controls. According to the results, methanol and hot water extracts showed higher antioxidant activities than essential oil. This fact suggested that terpenoids have lower antioxidant effect than phenolic compounds which were found in methanol and hot water extracts. Especially, acetone and water soluble fractions of methanol extract and ethanol fraction of hot water extract contained a lot of phenolic compounds and hydroxyl group, indicating that many hydroxyl groups and glycosidic bonds showed higher antioxidant effect than the other fractions. These results can suggest that the phenolic compounds which contained hydroxyl group or glycosidic bonds have a certain role for effective antioxidant activities.

Structures and Antioxidant Activity of Diketopiperazines Isolated from the Mushroom Sarcodon aspratus (능이버섯(Sarcodon aspratus)으로부터 분리한 diketopiperazine계 화합물의 화학구조 및 항산화활성)

  • Three antioxidants, Sa-1, Sa-2 and Sa-3, were isolated from the MeOH extract of the mushroom Sarcodon aspratus through EtOAc extraction, silica gel and Sephadex LH-20 column chromatography, and $C_{18}$ HPLC. The structures of the compounds were determined mainly by NMR and mass spectroscopic data analyses. Sa-1, Sa-2 and Sa-3 were identified as diketopiperazines, and their structures were determined to be cyclo(prolyl-valyl), cyclo(prolyl-leucyl) and cyclo(prolyl-isoleucyl), respectively. This is the first time that these compounds were isolated from this mushroom. They showed antioxidant activity by scavenging DPPH radical $(EC_{50}\;0.15{\sim}0.18\;mM)$ and superoxide radical $(EC_{50}\;0.21{\sim}0.24\;mM)$.

Protective Effects of HEAC and its Primary Active Compound on HCl-ethanol Induced Gastritis in Rats (흰쥐에서 염산-에탄올로 유발된 위염에 대한 HEAC 추출물과 그 주성분의 억제효과)

  • The purpose of this study is to evaluate the gastric mucosal damage effect by Hericium erinaceus cultibated with Artemisia capillaris (HEAC) with 0.15 M HCl in ethanol in rats. Among them, 80% ethanol extract showed potent inhibitory effect (67.7%) on gastritis. Using solvent fractionation, 80% ethanol extract of HEAC was separated into five fractions with $n$-hexane, methylene chloride, ethyl acetate, $n$-butanol and water. As a result, fractions obtained with methylene chloride exhibited strong inhibitory effects on gastritis. With this result, the methylene chloride extract was analyzed by GC/MS, HPLC, IR and NMR($^1H$, $^{13}C$) to identify primary active components and the main component in the extract was identified as ethyl linoleate. The effective does ($ED_{50}$) values of 80% ethanol extract of HEAC and ethyl linoleate showed 22.6 and 6.4 mg/kg, respectively and these values were higher than those of stillen (44.2 mg/kg) and selbex (46.5 mg/kg). Therefore, the administration of 80% ethanol extract of HEAC and ethyl linoleate have a strong protective effect agasint the gastritis induced by HCl-ethanol and may be promising new drug for treating gastritis and gastric ulcer.

Studies on the Immobilization of Enzymes and Microoganism Part 1. Immobilizing Method of Glucose Oxidase by Gamma Radiation (효소 및 미생물의 고정화에 관한 연구 제1보. 방사선조사에 의한 Glucose Oxidase의 고정화법)

  • A new method for immobilization of glucose oxidate by the aerobic gamma radiation of synthetic monomers was developed. The radiocopolymerization was conducted aerobically at -70 to -8$0^{\circ}C$ with the mixture of several polyfunctional esters, acrylates and native enzyme. The retained activity of immobilized glucose oxidase was about 50 to 55% when a NK 23G ester, acrylamide-bis and water mixture (1:1:2) in cold toluene treated with 450 krad of gam-ma radiation. The radiation dose did not influence significantly to the enzyme activity. The solvents used to prepare the beads of glucose oxidase and monomers were toluene, n-hexane, petoleum ether and chloroform. 0.05M tris-gycerol (pH 7.0) was a more suitable bugger solution for immobilizing the enzyme than was 0.02M phosphate. Immobilization of glucose oxidase shifted the optimum pH for its reaction from 6.0 to 6.5. The pH profile for the immobilized enzyme showed a broad range of optimum activity while the native enzyme gave a sharp pick for its optimum pH value. The immobilized enzyme reaction temperature was at the range of 30~4$0^{\circ}C$.
