중합도 n$\leq$10을 갖는 N-아세틸-D-글루코사민(GlcNAc)을 미생물 Serratia marcesce-ns QM B14f6의 최소배지 회분식 발효를 이용하여 키틴 및 키토산을 분해하는 효소 키티나제(1.5 unit/mL) [키토바이아제(3.48 unit/mL)가 포함됨]를 생성시킨다. 효소 반응에 의한 키토올리고당의 생성 활성을 규명하기 위하여 부분적으로 탈아세틸화된 키토산을 효소적 가수분해 반응시킴으로써 생성된 N-아세틸-D-글루코사민과 D-글루코사민의 키토올리고당의 생성 패턴을 확인 조사한다. 이 혼합 올리고머로부터 CM-Sephadex의 컬럼 크로마토그래피에 의하여 당별로 분획시켜 추출한 헤테로 키토 올리고당들을 각각 N-아세틸화하고 이 최종 생성물을 전기투석 장치로서 정제하여 키토올리고 1-7당을 제조하였다. 부분적으로 탈아세틸화 키토산(환원당으로서 2697 mg/mL)을 효소반응에 의해 생성시킨 키토올리고당은 1당으로서 GlcNAc=4.25%, 2당 $(GlcNAc)_2$=4.49%, 3당 $(GlcNAc)_3$=11.1%, 4당 $(GlcNAc)_4$=2.5%, 5당 $(GlcNAc)_{5}$ =0.64%, 6당$(GlcNAc)_{6}$=2.12%, 7당 $(GlcNAc)_{7}$=1.21%가 각각 제조되었다.
Purpose: Poly-N-acetyl glucosamine(PGlcNAc) nanofiber-based materials, produced by a marine microalga, have been characterized as effective hemostatic and angiogenic agents. The similarity between PGlcNAc patch and the natural extracellular matrix allows it to support new healthy tissue growth in an injured area and to encourage fluid absorption. In this study, we hypothesized that a poly-N-acetyl glucosamine fiber patch(PGlcNAc patch) may enhance wound healing in the db/db mouse. Methods: PGlcNAc patches were applied on one square centimeter, full-thickness, skin wounds in the db/db mouse model. Wounds(n=15 per group) were dressed with a PGlcNAc nanofiber patch for 1 hour(1 h), 24 hours(24 h) or left untreated(NT). After the application time, patches were removed and wounds were allowed to heal spontaneously. The rate of wound closure was evaluated by digital analysis of unclosed wound area in course of time. At day 10, wounds(n=7 per group) were harvested and quantified with immunohistochemical markers of proliferation(Ki-67) and vascularization (platelet endothelial cell adhesion molecule, PECAM-1). Results: Wounds dressed with PGlcNAc patches for 1 hour closed faster than control wounds, reaching 90% closure in 16.6 days, nine days faster than untreated wounds. Granulation tissue showed higher levels of proliferation and vascularization following 1 h treatment than the 24 h and NT groups. In addition to its hemostatic properties, the PGlcNAc material also appears to accelerate wound closure in healing-impaired genetically diabetic mice. Conclusion: This material, with its combination of hemostatic and wound healing properties, has the potential to be effective agent for the treatment of complicated wounds.
경골어류 4종(조피볼락, 용치놀래기, 송곳니베도라치 및 졸복) 장관의 복합당질 성상의 차이를 밝히기 위해 장을 네 부위로 나누워 아홉 종류의 biotinylated lectin들(DBA, SBA, PNA, BSL- I , RCA- I , sWGA, UEA- I 및 Con A)로 반응시켰다. 조피볼락을 제외하면 부위에 따른 차이는 없었다. 용치놀래기 장 상피는 모든 부위에서 D-glucose/mannose 잔기를 함유하고 있었다. 조피볼락, 송곳니베도라치 및 졸복 장 모든 부위에서는 정도의 차이는 있지만 ${\beta}$-N-acetyl-D-gal-actosamine가 함유되어 있었다. 또한 조피볼락의 근위장, 중간장 및 원위장에는 galactose-${\beta}$-1,3-N-acetyl-D-galactosamine 및 ${\alpha}$-D-galactose도 함유되어 있었으나 직장에는 이들 당 잔기들이 함유되어 있지 않았다. 송곳니베도라치에는 ${\beta}$-N- acetyl-D-galactosamine 외에 ${\beta}$-N-acetyl-D-glucosamine 및 D-glucose/mannose도 발견되었으며, 졸복에서는 Galactose-${\beta}$-1,3-N-acetyl-D-galactosamine, D-galactose 및 D-glucose/mannose도 발견되었다.
Ulex europaeus agglutinin I(UEA I), succinylated wheat germ agglutinin(sWGA), Griffonia simplicifolia lectin-I(GSL-I)을 이용하여 생쥐 부정소 조직내 당쇄의 분포를 조사하였다. 당쇄의 측쇄 말단의 $\alpha$-L-fucose 잔기에 특이적으로 결합하는 UEA I은 체부 및 미부 부정소를 제외한 두부 부정소 선단의 세정관 상피를 강하게 표지하였으나 관강은 중간 정도의 강도로 모두 표지되어 $\alpha$-L-fucose 잔기를 갖는 부정소 항원들은 부정소 선단부에서 주로 합성된 후 분비되는 것으로 추정된다. 이와 반대로 당쇄 말단의 $\alpha$-D-galactose 잔기에 특이적으로 결합하는 GSL-I은 두부 부정소를 제외한 체부와 미부 부정소 상피의 투명세포의 세포질과 섬모 및 기저세포를 표지하였다. 다당 사슬 및 복잡한 구조를 갖는 glycan의 당쇄의 N-acetyl-giucosamine 잔기에 특이적으로 결합하는sWCA는 부정소 부위별로 커다란 차이를 보이지 않았으나, 체부와 미부 부정소의 투명세포는 주세포에 비해 더 강하게 표지되어 sWGA 표지는 투명세포 기능분화의 표식자로 추측된다. UEA I 및 sWGA에 의한 관강의 표지 강도는 체부보다는 미부에서 약하게 관찰되어 미부 부정소 관강내에 $\alpha$-L-fucose 및 N-acetyl-glucosamine 잔기의 절단과 관련된 효소활성이 존재하는 것으로 추정된다. 요약하면 $\alpha$-L-fucose, $\alpha$-D-galactose, N-acetyl-glucosamine 잔기를 갖는 당쇄의 분포가 상피세포의 종류 및 부정소의 길이를 따라 차이를 보임을 확인하였다. 이는 정자의 성숙을 조절하는 부정소 각 절편 및 특정 절편 내 소관상피 세포의 기능적 분화를 대변하는 것으로 사료된다.

BACKGROUND: Glucosamine hydrochloride (GlcN HCl) has been shown to inhibit cell growth and matrix synthesis, but not with N-acetyl-glucosamine (GlcNAc) supplementation. This effect might be related to an inhibition of critical growth factors (GF), or to a different metabolization of the two glucosamine derivatives. The aim of the present study was to evaluate the synergy between GlcN HCl, GlcNAc, and GF on proliferation and cartilage matrix synthesis. METHOD: Bovine chondrocytes were cultivated in monolayers for 48 h and in three-dimensional (3D) chitosan scaffolds for 30 days in perfusion bioreactors. Serum-free (SF) medium was supplemented with either growth factors (GF) $TGF-{\beta}$ ($5ng\;mL^{-1}$) and IGF-I ($10ng\;mL^{-1}$), GlcN HCl or GlcNAc at 1mM each or both. Six groups were compared according to medium supplementation: (a) SF control; (b) SF + GlcN HCl; (c) SF + GlcNAc; (d) SF + GF; (e) SF + GF + GlcN HCl; and (f) SF + GF + GlcNAc. Cell proliferation, proteoglycan, collagen I (COL1), and collagen II (COL2) synthesis were evaluated. RESULTS: The two glucosamines showed opposite effects in monolayer culture: GlcN HCl significantly reduced proliferation and GlcNAc significantly augmented cellular metabolism. In the 30 days 3D culture, the GlcN HCl added to GF stimulated cell proliferation more than when compared to GF only, but the proteoglycan synthesis was smaller than GF. However, GlcNAc added to GF improved the cell proliferation and proteoglycan synthesis more than when compared to GF and GF/GlcN HCl. The synthesis of COL1 and COL2 was observed in all groups containing GF. CONCLUSION: GlcN HCl and GlcNAc increased cell growth and stimulated COL2 synthesis in long-time 3D culture. However, only GlcNAc added to GF improved proteoglycan synthesis.
N-Acetyl-D-glucosamine (NAG)는 보습제로 사용되어지는 히아루론산의 구성물질인 뮤코 다당류의 일종이며, 특히 화장품으로서 응용은 보습제로서의 사용이 최초이다. 본 실험에서는 게나 새우의 껍데기에서 추출된 키틴을 탈아세틸화 하여 얻은 NAG를 화장품 원료로서 적용하고자 하였다. 현재 기능성 주름 원료로 알려진 레티놀(retinol)과 NAG를 비교하기 위하여 섬유아세포의 활성능력 및 콜라겐 생성촉진 효과를 비교 실험하였으며, 제형 내에서의 안정성을 위하여 HPLC로 역가를 측정하였다. 실험결과, 게의 껍질로부터 유도된 NAG는 피부에 자극을 전혀 주지 않으면서 섬유아세포의 세포활성 및 콜라겐의 생성을 촉진시키는 효과를 나타내었으며, 헤어리스 마우스를 대상으로 실험을 실시한 결과 피부층의 변화를 통하여 주름의 감소 효능을 볼 수 있었다.
Chitin synthases(UDP-N-acetyl-D-glucosamine: chitin 4-$\beta$-N-acetyl-D-glucosaminyl transferase, EC 2.4.1.16) catalyze the synthesis of chitin from UDP-N-acetyl-D-glucosamine. Two zymogenic type of chitin synthase gene(TmCHS1 and TmCHS2) were amplified and its nucleotide sequences were determined. By the amino acid comparison and UPGMA tree grouping, TmChs1 and TmChs2 were classified as class II and class IV chitin synthases respectively. The class II type TmChs1 was grouped with others of Agaricales ectomycorrhizal mushroom. Additionally the phylogenetic tree was well adapted to Hymenomycete previously classified by morphological and physiological characteristics.
The strain of Serratia marcescens QM B1466 produces selectively large amount of chitinolytic enzymes (about 1mg/L medium). Enzymatic hydrolysis of chitin to N-acetyl-${\beta}$-D-glucosamine (NAG) was performed with a system consisting of two hydrolases (chitinase and chitobiase) produced by optimization of a microbial host consuming chitin particles. For the development of Large-scale biological process for the production of NAG from chitinaceous waste, the selection and optimization of a microbial host, particle size of crab/shrimp chitin sources and initial induction time using chitin as a sole carbon source on chitinase/chitobiase production and NAG production were examined. Crab-shell chitin(1.5%) treated by dilute acid and , ball-milled with a normal diameter less than 250m gave the highest chitinase activity over a 7 days culture. Crude chitinase/ chitobiase solution obtained in a 10 L fed-batch fermentation showed a maximum activities of 23.6 U/mL and 5.1 U/mL, respectively with a feeding time of 3 hrs, near pH 8.5 at 30$^{\circ}C$.
Selenomonas ruminantium 균체(菌體)를 TCA로 가열(加熱) 분해(分解)한 후 chloroform : methanol (1 : 3)로 추출(抽出)한 당지질(糖脂質)을 분리(分離)하고 이 당지질(糖脂質)을 acetone가용부분(可溶部分) spot A화합물(化合物)과 acetone불용부분(不溶部分)에서 ether로 다시 가용부분(可溶部分)을 추출(抽出)한 spot B화합물(化合物)의 두 부분(部分)으로 분리(分離)하고 이 두 화합물(化合物)에 대(對)하여 각각(各各) 그 화학구조(化學構造)를 구명(究明)하며 당지질(糖脂質)의 구조(構造)를 추정(推定)한 바 다음과 같은 결과(結果)를 얻었다. 1. 두 화합물(化合物)의 적외선흡수분석결과(赤外線吸收分析結果) spot A는 amino당(糖)에 O-acyl 및 N-acyl지방산(脂肪酸)이 결합(結合)하였으며 spot B는 amino당(糖)에 O-acyl 및 N-acyl지방산(脂肪酸)이 결합(結合)하고 인(燐)을 함유(含有)하고 있음을 알았다. 2. 두 화합물(化合物)을 GLC에 의(依)하여 지방산조성(脂肪酸組成)을 조사(調査)한 바 spot A.B화합물중(化合物中)에 있는 O-acyl 및 N-acyl 지방산(脂肪酸)은 ${\beta}-OH\;C_{13:0}$지방산(脂肪酸)이 대부분(大部分)이 였는데 저급(低級)의 hydroxy지방산(脂肪酸) ${\beta}-OH\;C_{9:0}$도 특이적(特異的)으로 함유(含有)되여 있음을 알았다. 3. 두 화합물(化合物)을 hydrazine분해(分解)를 시킨 결과(結果)를 paper chromatography로 조사(調査)한 바 spot A화합물(化合物)은 glucosamine 이 2분자(分子) 결합(結合)하여 있는 chitobiose와 같은 Rf 치(値)를 나타냈음으로 2분자(分子)의 glucosamine이 결합(結合)됨을 확인(確認)하고 spot B 화합물(化合物)의 낮은 Rf치(値)는 glucosamine에 인(燐)이 결합(結合)되여 있음을 알았다. 4. spotA화합물(化合物)의 산분해물(酸分解物)을 다시 ninbydrine으로 산화분해(酸化分解) 시키면 arabinose만이 생기는 것으로 보아 glucosamine의 amino기(基)는 $C_2$의 위치(位置)에 결합(結合)하여 있음을 알았다. 5. N-acetyl화(化)한 spot A에 $NaBH_4$를 추리(處理)한 결과(結果) glucosamine의 전량(全量)이 반감(半減)하는 것으로 보아 2분자(分子)의 glucosamine이 결합(結合)되여 있는 것을 알 수 있고 Morgan-Elson반응(反應) 및 $NaIO_4$분해(分解)에 의(依)하여 2개(個)의 glucosamine은 1.6결합(結合)임을 확인(確認)하였다. 6. N-acetyl화(化)한 spot A.B화합물(化合物)에 ${\beta}-N-acetyl$ glucosarninidase를 반응(反應)시킨 결과(結果) spot A화합물(化合物)은 100% N-acetyl glucosamine으로 분해(分解)되고 spot B화합물(化合物)은 분해(分解)되지 않았으므로 spot A화합물(化合物)만이 ${\beta}$결합(結合)을 하고 있음을 알았다. 7. $^{32}P$함유(含有) spot B화합물(化合物)에 phosphodiesterase 및 phosphomonoesterase를 작용(作用)시킨 결과(結果) phosphodiesterase는 반응(反應)치 않고 phosphomonoesterase에 의(依)하여 100% $^{32}P$가 유리(遊離)되는 것으로 보아 glucosamine 2분자(分子)에 한계의 인(燐)이 monoester결합(結合)을 하고 있음을 알 수 있다. 8. spot A화합물(化合物)은 glucosaminiyl ${\beta}-1.6-glucosamine$의 결합(結合)을 하였고 O-acyl 및 N-acyl지방산(脂肪酸)이 결합(結合)되여 있으며 주지방산(主脂肪酸)은 ${\beta}-OH\;C_{13:0}$임을 알았다. 9. spot B화합물(化合物)도 glucosaminyl ${\beta}-1.6-glucosamine$의 결합(結合)을 하고 O-acyl 및 N-acyl지방산(脂肪酸)이 결합(結合)되여 있으며 주지방산(主脂肪酸)은 ${\beta}-OH\;C_{13:0}$이나 인(燐)이 monoester결합(結合)을 하고 있는 것이 spot A화합물(化合物)과 특이(特異)함을 알았다.

제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
