Appropriate nitrogen application enhances saponin synthesis and growth mediated by optimizing root nutrient uptake ability

  • Background: Cultivation of medicinal crops, which synthesize hundreds of substances for curative functions, was focused on the synthesis of secondary metabolites rather than biomass accumulation. Nutrition is an important restrict factor for plant growth and secondary metabolites, but little attention has been given to the plasticity of nutrient uptake and secondary metabolites synthesis response to soil nitrogen (N) change. Methods: Two year-field experiments of Sanqi (Panax notoginseng), which can synthesize a high level of saponin in cells, were conducted to study the effects of N application on the temporal dynamics of biomass, nutrient absorption, root architecture and the relationships between these parameters and saponin synthesis. Results: Increasing N fertilizer rates could improve the dry matter yields and nutrient absorption ability through increasing the maximum daily growth (or nutrient uptake) rate. Under suitable N level (225 kg/ha N), Sanqi restricted the root length and surface and enhanced the root diameter and N uptake rate per root length (NURI) to promote nutrient absorption, but the opposite status of Sanqi root architecture and NURI was found when soil N was deficient. Furthermore, increasing N rates could promote the accumulation of saponin in roots through improving the NURI, which showed a significant positive relationship with the content of saponin in the taproots. Conclusion: Appropriate N fertilizer rates could optimize both root architecture and nutrient uptake efficiency, then promote both the accumulation of dry matter and the synthesis of saponins.

맹장 결찰계(Cecal-ligated Chicken)를 이용한 [15N]urea의 이용성에 관한 연구 (A Study on the Utilization of Dietary [15N]urea in Cecal Ligated Chickens)

  • 본 연구는 맹장 결찰계를 이용하여서 [15N]urea의 이용성을 조사하였다. 질소의 balance 및 이용률은 맹장 결찰 수술에 의해서 높아지는 경향이 인정되었다. 이는 맹장 결찰 수술로 인한 요산 배설량의 유의한 감소로 인한 결과였다(사료 및 요소 유래 질소 이용률, P<0.01). 닭에 있어서 맹장 결찰 수술은 사료 유래 ammonia 배설량은 증가(P<0.01), 요소 유래 ammonia배설량은 감소(P<0.01)시켰다. 요소 유래, 요소질소의 배설량은 맹장 결찰 수술에 의해서 유의하게 증가하였지만(P<0.01), 맹장 결찰계도 5% 단백질 사료와 같이 급여한 요소를 51.6%정도 이용하는 것으로 나타났다. 결론적으로 닭에 있어서 단백질 수준이 결핍된 사료와 같이 급여된 요소의 이용에 있어서 맹장은 유의한 역할을 한다고 할 수 있다. 하지만 질소의 이용에 있어서는 반드시 긍정적인 역할만을 한다고는 결론내릴 수 없다.

  • The objectives of the present study were to investigate whether or not dietary lysine addition could improve N balance of female Japanese Saanen goats at 15 to 32 months of age, weighing 31 to 40 kg, fed on a wheat bran-hay cube diet supplemented with methionine, and whether or not ruminally protected lysine supplementation could give as good an N balance performance as lysine in the presence of ruminally protected methionine when given orally to the goats. It was considered from changes in N balance and N utilization that the first-and second-limiting amino acids in the diet were methionine and lysine respectively, under the present experimental conditions. The ruminally protected lysine in addition to the ruminally protected methionine gave no improvement in N balance and N utilization compared with the ruminally protected methionine alone, suggesting that the ruminally protected lysine used in the present study was not effectively utilized by the goats.

Mycorrhiza 및 Rhizobium 접종이 알팔파-페레니얼 라이그라스 혼파에 의한 질소와 인의 이용성 및 성장에 미치는 영향 (Effects of Inoculation of Mycorrhiza and Rhizobium on N, P utilization and Vegetative Growth in Alfalfa/Perennial Ryegrass Intercropping)

  • To investigate the effect of Mycorrhiza (Glomus intradics) and Rhizobium inoculation on the N, P utilization and growth response of Alfalfa (Medicago saliva L.) and Perennial ryegrass (Lolium perenne L.) in mixed sward, four treatments (non-inoculation, Control; Mycorrhiza inoculation: M; Rhizobium inoculation, R and Mycorrhiza and Rhizobium inoculation, M+R) were applied. The associated analyses were canied out on the early vegetative growth stage (DAS 56, 56 days after seeding) and on the early flowering stage (DAS 126). The decreased rate of total N and P content in soil, with advancing plant growth, was relatively higher in the M and M + R treatment than control. The content of availabie phosphorus in soil at DAS 126 increased by about 34 and 38 % in M and M+R treatment compared to control (189.2 mg PzOskg DM), while non-significant changes was observed in R treatment. Total N uptake and P uptake in the control at DAS 126 were 44.71 and 3.52 mglplant in mixed sward, respectively. About 71, 98 and 197 % of increases in total N uptake and 70, 72 and 11 1 % of increases in total P uptake were estimated in M, R and M+R treatment. Comparing to control, total dry matter yield significantly increased by 27, 33 and 53 %, and crude protein yield also by 78, 83 and 204 %, respectively, in M, R and M+R treatment. The present data indicated that mycorrhiza orland rhizobium inoculation improved N, P utilization of both alfalfa and perennial ryegrass plants, and consequently increased total yield (especially by dual inoculation, M+R). The content of available phosphorus in soil at DAS 126 increased by about 40 % in M and M+R treatment compared to control (141.0 mg PzOg'kg DM), while non-significant changes was observed in R treatment. Total N uptake and P uptake in the control at DAS 126 were 33.91 mglplant and 2.42 mdplant, respectively, about 21, 50 and 51 % of increases in total N uptake and 30, 11 and 47 % of increases in total P uptake were estimated in M, R and M+R treatment. Comparing to control, dry matter yield significantly increased by 8, 27 and 28 %, and crude protein yield also by 21, 42 and 39 %, respectively, in M, R and M+R treatment. The present data indicated that mycorrhiza orland rhizobium inoculation improved N, P utilization, and consequently increased the yield of alfalfa. Change in Nitrogen Fractions and Ruminal Nitrogen Degradability of Orchardgrass Ensiled at Various Moisture Contents and the Subsequent Effects on Nitrogen Utilization by Sheep

  • The effect of various moisture contents of fresh forage on the change in nitrogen (N) fractions, in vitro ruminal N degradability, and the subsequent N utilization of silage in sheep were evaluated. Orchardgrass (Dactylis glomerata L.) with high (HM, 76%), medium (MM, 65%) and low (LM, 40%) moisture contents were ensiled into silos of 120 L capacity for 120 days. A nitrogen balance trial was conducted using a 4${\times}$4 Latin square design consisting of four dietary treatments (i.e. fresh forage, HM, MM and LM silages) and four wethers. With respect to N fractions, fraction 1 (buffer solution soluble N), fraction 2 (buffer solution insoluble N-neutral detergent insoluble N), fraction 3 (neutral detergent insoluble N-acid detergent insoluble N), and fraction 4 (acid detergent insoluble N) were determined. The proportion of fraction 1 in silages tended to decrease, while the in vitro ruminal degradability of insoluble N increased (p<0.05) with lower moisture contents at ensiling. Consequently, nitrogen utilization in sheep tended to improve as the moisture content of ensiled grass was decreased, with a negative correlation (p<0.01) between urinary N and the in vitro ruminal degradability of insoluble N. The averaged N retentions for HM, MM, and LM silage treatments were 59, 73 and 79% of that for fresh forage, respectively.

Denitrifying PAO와 SBBR-MSBR을 이용한 생물학적영양소제거공정에서 탄소원 절약에 대한 연구 (Evaluation of COD Utilization for Biological Nutrient Removal with dPAO in SBBR-MSBR System)

  • The combined system of sequencing batch biofilm reactor (SBBR) and membrane SBR (MSBR) was operated with sewage to evaluate the COD utilization for biological nutrient removal (BNR). The SBBR was operated for nitrification reactor, while denitrifying PAO (dPAO) was cultivated in MSBR with anaerobic-anoxic operation. In the SBBR and MSBR system, the enhanced biological phosphorus removal (EBPR) was successfully achieved with higher N removal. The COD utilization in combined SBBR-MSBR system was significantly reduced compared to ordinary BNR (up to 3.1 g SCOD/g (N+P) and 1.6 g SCOD/g (N+P) with different C/N/P ratio). The results suggest that a dPAO process could effectively reduce carbon energy (=COD) requirement. The combination of oxic-SBBR and anaerobic-anoxic MSBR for dPAO utilization could be an attractive alternative to upgrade the process performance in weak sewage.

Quality Properties and Flavor-Related Components of Beef Longissimus Lumborum Muscle from Four Korean Native Cattle Breeds

  • This study was carried out to assess the quality properties, components associated with taste and aroma of beef as a function of breed. For this purpose, steers from four Korean native cattle breeds: Hanwoo (n=10), Chikso (n=10), black Hanwoo (n=12, BHW) and Jeju black cattle (n=12, JBC) were used. The steers all were raised under identical conditions and finished at a similar age of around 30-months old. Following 24 h of slaughter, all longissimus lumborum muscles were collected and used for analysis of meat quality, fatty acids, and flavor-related components (metabolic compounds, free amino acids, and aroma volatiles). The Hanwoo presented a significantly higher intramuscular fat content (IMF, 22.85%) than the BHW (11.78%), Chikso (9.25%), and JBC (9.14%; p<0.05). The meat of Hanwoo breed showed lighter and redder color, and lower shear force value (p<0.05). The JBC presented a "healthier" fatty acid profiles as it had a higher total unsaturated fatty acids content (p<0.05). With regard to flavorrelated components, Hanwoo also had higher total contents of free amino acids and metabolites associated with umami and sweet tastes, and fat-derived volatile compounds (aldehydes, alcohols, and ketones) associated with fatty aroma. It may be concluded that there was a considerable difference in the meat quality properties among breeds. The variations of IMF content and flavor-related components may be the main factors contributing to the typical flavors of beef among the four Korean native cattle breeds.
